Action spectrum for bergamot-oil phototoxicity measured by sunburn cell counting.

The present study investigated the phototoxic effect of bergamot oil and its photosensitive component, bergapten, on sunburn cell (SBC) production in guinea pig skin. The back skin was pretreated with bergamot oil or bergapten and exposed to monochromatic light under various conditions. After irradiation, skin specimens were excised, and histological sections were prepared. The number of sunburn cells in the interfollicular epidermis was counted. The SBC formation by bergamot oil or bergapten plus UVB radiation was the same as that without pretreatment with any photosensitizer. In contrast, a

Fetal platelet counts correlate with the severity of the anemia in red-cell alloimmunization.

OBJECTIVE: To determine whether fetal anemia secondary to maternal red-cell alloimmunization is associated with thrombocytopenia. METHODS: The records of 78 patients undergoing intrauterine transfusion for red-cell alloimmunization were reviewed. Pre-transfusion fetal platelet counts were compared between hydropic and nonhydropic fetuses. A regression analysis was performed between the fetal platelet counts and the fetal bilirubin levels, hematocrits, and reticulocyte counts taken at the initial transfusion. The hematocrits, reticulocyte counts, and bilirubin levels were adjusted for gestatio

Performance of a T-cell-based diagnostic test for tuberculosis infection in HIV-infected individuals is independent of CD4 cell count.

The performance characteristics of the enzyme-linked immunospot assay (ELISPOT) assay (T-SPOT TB) for the diagnosis of latent tuberculosis infection in HIV-infected individuals are unknown. Given that ELISPOT enumerates Mycobacterium tuberculosis antigen-specific IFN-gamma-secreting T cells, HIV-associated immunosuppression might adversely affect test performance. However, we found that 28 out of 29 HIV-positive individuals (97%) gave evaluable test results, and performance was independent of the CD4 T-cell count. ELISPOT test performance appears to be independent of HIV-associated immunosuppr

Differentiating large cell lymphoma from indolent small B-cell lymphoma in fine needle aspirates using p53, PCNA and transformed lymphocyte count.

OBJECTIVE: To determine the usefulness of proliferating cell nuclear antigen (PCNA), p53 protein expression and transformed lymphocyte count (TLC) as adjunctive tests to differentiate indolent small B-cell lymphoma from large cell lymphoma in fine needle aspiration biopsies. STUDY DESIGN: Aspirates of lymphoproliferative disorders from April 1993 to January 1997 were reviewed. The percentage of TLCs was determined on the Papanicolaou smear. The percentage and intensity of p53 and PCNA immunocytochemical staining was evaluated on cell block sections. These results were compared and correlated

Clinical features and outcome of T-lineage acute lymphoblastic leukemia in adults: a low initial white blood cell count, as well as a high count predict decreased survival rates.

Although biological and clinical features differ between B-lineage acute lymphoblastic leukemia (ALL) and T-lineage ALL (T-ALL), there have been few reports that focused on the prognosis for T-ALL in adults, primarily due to its rarity. Here, we studied the long-term outcomes and prognostic factors specific for adult T-ALL by combining patient data from the three prospective trials conducted by the Japan Adult Leukemia Study Group (JALSG). Among 559 patients whose immunophenotypes could be evaluated, 87 (15.6%) were identified as T-ALL. Of them, 66 patients (75.8%) achieved complete remission,

Low reticulocyte count at infusion is a risk factor for ICANS in CAR-T cell therapy.

INTRODUCTION: Chimeric antigen receptor (CAR)-T cell therapies targeting CD19 have shown efficacy against B-cell malignancies. However, they frequently cause immune effector cell-associated neurotoxicity syndrome (ICANS), which can be life-threatening and require intensive care. Even when it is not severe, ICANS can lead to prolonged hospitalization and limit treatment options, especially for elderly patients. Despite its clinical significance, a reliable, early predictive marker for ICANS has not been identified. METHODS: To identify risk factors for ICANS, we retrospectively analyzed B-cell

Nonlinear relationships between somatic cell count and technological properties of bovine milk: A longitudinal observational study.

This longitudinal observational study investigated the impact of somatic cell count (lnSCC) on milk composition and technological traits in 332 clinically healthy cows. Over 2 years, 865 samples were collected at 4 lactation stages. The analysis covered milk yield, major components (fat, protein, lactose, FFA, citric acid, urea contents), and processing-related traits (casein and TS contents, acidity, pH, coagulation time, and curd/whey quality and curd yield). The mixed model included cow as a random effect, while HYS, HYM, parity, and milking time were fixed effects. DIM (quadratic) and lnSC

Duct-level evaluation of mammary gland health, somatic cell count and bacterial flora in lactating jennies.

BACKGROUND: Assessing mammary gland health in donkeys is challenging due to the lack of species-specific diagnostic criteria for subclinical mastitis. Although somatic cell count (SCC) is widely used in dairy ruminants, its diagnostic value in jennies remains unclear. Each donkey teat contains two anatomically independent duct systems, potentially influencing microbial distribution. OBJECTIVES: To evaluate mammary gland health at the individual duct level in lactating jennies by assessing SCC, bacteriological findings, and total bacterial count (TBC), and to investigate associations with host

Predicting Iron Deficiencies Using Routine Complete Blood Cell Count Parameters: A Machine Learning Approach and Evaluation.

Background/Objectives: Iron deficiency remains a prevalent condition, needing specific laboratory tests for diagnosis. This study aimed to evaluate whether routine complete blood cell count (CBC) parameters can be used within a machine learning framework to predict low ferritin and low transferrin saturation, used as biochemical markers of altered iron status, potentially supporting more targeted laboratory test utilization. Methods: In this single-center retrospective outpatient study, we analyzed 32,437 records from subjects undergoing both complete blood cell count and iron metabolism testi

Pilot assessment of point-of-care differential white blood cell count analysers in schoolchildren with schistosomiasis.

Public health monitoring of schistosomiasis needs appropriate field-based technology, so to expand this portfolio, we conducted a first pilot assessment of point-of-care differential white blood cell count analysers. In total, 120 Malawian primary school children were examined with six key haematological parameters recorded. A notable proportion had abnormal differential white blood cell profiles with eosinophilia (48.3%) positively associated with schistosomiasis (OR 3.2, 95% CI 1.3 to 7.7). Whilst costs of equipment and associated tests were non-trivial, both machine performance and haematol

Prognostic impact of lymph node count features in total laryngectomy for advanced laryngeal squamous cell carcinoma.

INTRODUCTION: The optimal surgical management in advanced laryngeal squamous cell carcinoma (ALSCC) is still under debate. The extent of neck dissection as well as the nodal involvement affect survival metrics in head and neck cancer (HNSCC) patients. Despite pN status, other parameters like nodal yield (NY) or lymph node ratio (LNR) have been investigated before. There are data showing that log odds of positive lymph nodes (LODDS) are a good survival prognosticator in HNSCC in general but specific data on ALSCC is missing. This study aims to assess the prognostic value of lymph node count fea

Markedly elevated cerebrospinal fluid white blood cell count in multiple sclerosis: How high is too high?

BACKGROUND: The diagnosis of multiple sclerosis (MS) is based on the revised McDonald's criteria, which includes clinical history, neurological examination, MRI findings and cerebrospinal fluid (CSF) analysis necessitating the exclusion of alternative explainable diagnoses. Markedly elevated CSF white blood cells (WBCs) over 50 (units) is considered a "red flag" for an alternative diagnosis apart from MS. OBJECTIVE: To present a man with typical relapsing-remitting MS (RRMS) despite markedly elevated CSF WBCs. CASE: A 33-year-old man presented with progressive bilateral numbness and weakness

Prediction of white blood cell count during exercise: a comparison between standalone and hybrid intelligent algorithms.

Decades of research in exercise immunology have demonstrated the profound impact of exercise on the immune response, influencing an individual's disease susceptibility. Accurate prediction of white blood cells (WBCs) count during exercise can help to design effective training programs to maintain optimal the immune system function and prevent its suppression. In this regard, this study aimed to develop an easy-to-use and efficient modelling tool for predicting WBCs count during exercise. To achieve this goal, the predictive power of a range of machine-learning algorithms, including six standal

Tumor mutational load is prognostic for progression to therapy among high-count monoclonal B-cell lymphocytosis.

High-count monoclonal B-cell lymphocytosis (HCMBL) is a precursor condition to chronic lymphocytic leukemia (CLL). We have shown that among individuals with HCMBL, the CLL-International Prognostic Index (CLL-IPI) is prognostic for time-to-first therapy (TTFT). Little is known about the prognostic impact of somatically mutated genes among individuals with HCMBL. We sequenced DNA from 371 individuals with HCMBL using a targeted sequencing panel of 59 recurrently mutated genes in CLL to identify high-impact mutations. We compared the sequencing results with that of our treatment-naïve CLL cohort

Decreased Peripheral Blood Natural Killer Cell Count in Untreated Juvenile Dermatomyositis Is Associated with Muscle Weakness.

Juvenile Dermatomyositis (JDM) is the most common inflammatory myopathy in pediatrics. This study evaluates the role of Natural Killer (NK) cells in Juvenile Dermatomyositis (JDM) pathophysiology. The study included 133 untreated JDM children with an NK cell count evaluation before treatment. NK cell subsets (CD56low/dim vs. CD 56bright) were examined in 9 untreated children. CD56 and perforin were evaluated in situ in six untreated JDM and three orthopedic, pediatric controls. 56% of treatment-naive JDM had reduced circulating NK cell counts, designated "low NK cell". This low NK group had mo

Prognostic Relevance of Negative Lymph Node Count in Resected Stage I-IIIa Small-cell Lung Cancer.

OBJECTIVES: The prognostic significance of the negative lymph node (NLN) count has been confirmed in various cancers but not in small-cell lung cancer (SCLC). We aimed to evaluate the correlation between the NLN count and the prognosis of patients with stages I-IIIa SCLC who underwent lobectomy. METHODS: Data on the clinical characteristics of SCLC patients who underwent lobectomy between 2000 and 2019 were collected from the SEER database and organized based on the X-tile plots to identify the optimal cutoff point for the NLN count. Kaplan-Meier curves and a Cox proportional hazard model wer

A case of fatal acute bacterial meningoencephalitis with extremely high cerebrospinal fluid white blood cell count.

Acute bacterial meningoencephalitis is still prevalent despite the widespread vaccination and still fatal despite the advances in antimicrobial therapy. Identifying patients at risk, lowering the threshold of clinical diagnosis and early treatment of such a curable disease will save patients' lives.

High pre-transplant Mucosal Associated Invariant T Cell (MAIT) count predicts favorable course of myeloid aplasia.

AIMS: Mucosal Associated Invariant T (MAIT) cells are unconventional T cells with anti-infective potential. MAIT cells detect and fight against microbes on mucosal surfaces and in peripheral tissues. Previous works suggested that MAIT cells survive exposure to cytotoxic drugs in these locations. We sought to determine if they maintain their anti-infective functions after myeloablative chemotherapy. METHODS: We correlated the amount of MAIT cells (measured by flow cytometry) in the peripheral blood of 100 adult patients before the start of myeloablative conditioning plus autologous stem cell t

Reduced neural progenitor cell count and cortical neurogenesis in guinea pigs congenitally infected with Toxoplasma gondii.

Toxoplasma (T.) gondii is an obligate intracellular parasite with a worldwide distribution. Congenital infection can lead to severe pathological alterations in the brain. To examine the effects of toxoplasmosis in the fetal brain, pregnant guinea pigs are infected with T. gondii oocysts on gestation day 23 and dissected 10, 17 and 25 days afterwards. We show the neocortex to represent a target region of T. gondii and the parasite to infect neural progenitor cells (NPCs), neurons and astrocytes in the fetal brain. Importantly, we observe a significant reduction in neuron number at end-neurogene
