Effect of vagal stimulation and parenteral acetylcholine on canine trachealis muscle shortening.

Canine trachealis smooth muscle shortening (TMS) in response to vagal nerve stimulation is approximately 30%, far less than the 70% predicted from in vitro studies. We hypothesized that in vivo airway smooth muscle activation during vagal stimulation may be submaximal, and in this study we wished to determine TMS during maximal activation. TMS was studied in 12 alpha-chloralose-anesthetized dogs during vagal stimulation, systemic acetylcholine injection, and local acetylcholine injection. Bilateral vagal stimulation produced TMS of 26 +/- 5% (SE) length at functional residual capacity (LFRC). …

A rat model for predicting orthostatic hypotension during acute and chronic antihypertensive drug therapy.

An experimental model has been developed to determine postural hypotension in chloralose-urethane-pentobarbital (CUP) anesthetized rats. Using this technique, mean arterial pressure, heart rate, and blood pressure compensation to 60 degree tilt were examined in rats treated acutely and chronically with a spectrum of standard antihypertensive agents. The results closely parallel the orthostatic profiles seen clinically with these drugs. Significant orthostatic hypotension was seen with the acute intravenous administration of the alpha-adrenergic antagonists phenoxybenzamine, phentolamine, and p…

Cardiovascular effects of prostaglandin D3 and D2 in anesthetized dogs.

Prostaglandin (PG) D3 has been identified as an inhibitor of human platelet aggregation, but little is known of the hemodynamic activity of this material. In morphine pretreated, chloralose-urethan anesthetized dogs, bolus intravenous injections (1, 3.2 and 10 microgram/kg) of PGD3 and also PGD2 were associated with marked, dose-related increases in pulmonary arterial pressure. Cardiac index and rate increased, while peripheral vascular resistance decreased in response to injections of PGD3. A biphasic (depressor followed by a pressor phase) effect on systemic arterial pressure was observed af…

Changes in rat hind limb vascular resistance following intracerebroventricular drug administration.

The right hind limbs of rats (which had previously implanted intraventricular guide cannulae) were isolated from the systemic circulation, but with the nerves to the limb remaining intact, and perfused using a constant output blood pump. Using this preparation, changes in vascular resistance, blood pressure and heart rate were monitored following injection of noradrenaline, phentolamine and propranolol into the lateral cerebral ventricles (i.c.v.) of rats anaesthetised with alpha-chloralose. All three drugs lowered blood pressure. Noradrenaline administered i.c.v. induced a nervously mediated …

Spectral analysis of blood pressure and heart rate, catecholamine and neuropeptide Y plasma levels in a new model of neurogenic orthostatic hypotension in dog.

The aim of the study was to compare changes in blood pressure (BP) and heart rate (HR) variability, catecholamine and neuropeptide Y (NPY) plasma levels induced by passive head-up tilt in normal and sino-aortic denervated (SAD) chloralose-anaesthetized dogs. In controls, 80 degrees head-up tilt test failed to change BP and increased HR. Plasma noradrenaline and NPY levels (but not adrenaline) significantly rose. In SAD dogs, head-up tilt test induced a marked and reproducible decrease in BP without any change in HR or noradrenaline and NPY plasma levels. In SAD dogs, spectral analysis in supin…

Carbonic anhydrase and control of breathing: different effects of benzolamide and methazolamide in the anaesthetized cat.

1. The effect of inhibition of erythrocyte carbonic anhydrase on the ventilatory response to CO2 was studied by administering benzolamide (70 mg kg-1, i.v.), an inhibitor which does not cross the blood-brain barrier, to carotid body denervated cats which were anaesthetized with chloralose-urethane. 2. In the same animals the effect on the ventilatory response to CO2 of subsequent inhibition of central nervous system (CNS) carbonic anhydrase was studied by infusing methazolamide (20 mg kg-1), an inhibitor which rapidly penetrates into brain tissue. 3. The results show that inhibition of erythro…

Measurements of duodenal wall motility, mucosal fluid transport and alkaline secretion. Description and evaluation of a methodological approach in the anaesthetized cat.

This paper describes and evaluates a method for simultaneous recordings of duodenal motility and mucosal secretions in chloralose-anaesthetized cats. Biliary and pancreatic secretions were diverted by separate cannulation of each duct. A 2 cm segment of the proximal duodenum was isolated between two luminally situated balloons (the proximal balloon being positioned immediately distal to the pylorus). Each balloon was connected to a barostatic device maintaining a constant pressure within the balloon independent of the degree of contraction. Motor activity in the duodenal wall was recorded as c…

Effect of glutamate stimulation of bed nucleus of the stria terminalis on arterial pressure and heart rate.

Experiments were done in the chloralose-anesthetized, paralyzed, and artificially ventilated rat to determine the cardiovascular responses elicited during chemical stimulation of bed nucleus of the stria terminalis (BST) and to investigate the components of the peripheral autonomic nervous system that mediate these responses. Neurons in BST were selectively stimulated by the microinjection (10-20 nl) of the excitatory amino acid L-glutamate (1 M). Stimulation of BST elicited decreases in mean arterial pressure (n = 105) of -6 to -55 mmHg. These depressor responses were on occasion (n = 60) acc…

Control of tissue environment during vital microscopy of the microcirculation in the m. tenuissimus in cat.

The physiological preservation of the tenuissimus muscle preparation in cat during vital microscopy of the microcirculation is assessed, com … eria of tissue normalcy. The microcirculation and metabolism are evaluated during anesthesia (alpha-chloralose) at rest as well as in hemorrhagic shock. In the resting state, muscle temperature drops to 28 degrees C with the irrigation technique, whereas the Mylar technique keeps the temperature at 34--35 degrees C. Neither technique causes deviations in nor …

Interaction between centrally acting hypotensive drugs and tricyclic antidepressants.

From clinical experience it is known that the hypotensive action of clonidine and alpha-methyl-DOPA is antagonized by desipramine. This antagonism was investigated in detail in chloralose-anaesthetized cats. Both the centrally acting hypotensive agents and the tricyclic antidepressants where infused into the left vertebral artery. (1) The central hypotensive action clonidine was antagonized by desipramine, imipramine, amitriptyline, protriptyline and mianserine. For the inhibition of the hypotensive action of clonidine by protriptyline a parallel shift of the dose-response curve was obtained, …

Reflex respiratory and cardiovascular effects of stimulation of receptors in the nose of the dog.

1. In forty-one out of forty-seven dogs under chloralose-urethane or Nembutal anaesthesia, mechanical stimulation of the nasal mucous membrane caused a reduction or inhibition of respiration, bradycardia, variable changes of arterial blood pressure, and a small rise in venous pressure.2. Contractions of the caudal region of the oesophagus of the cat.

1. The motility of the most caudal part of the oesophagus has been investigated in cats which were decerebrated, had the whole of the central nervous system pithed or were anaesthetized with chloralose.2. Contractions of the most caudal region of the oesophagus were evoked by its stretch and the presence in it of HCl. These reactions were obtained in all types of preparations used.3. In decerebrate preparations oesophageal contractions occurred in response to the introduction of acid into the stomach. These were not obtained after the vagus nerves had been cut.4. Slight degrees of gastric dist…

Suppression of bladder reflex activity in chronic spinal cord injured cats by activation of serotonin 5-HT1A receptors.

The effects of 8-OH-DPAT (5-HT1A receptor agonist) and WAY100635 (5-HT1A receptor antagonist) on reflex bladder activity were investigated in alpha-chloralose anesthetized or conscious chronic spinal cord injured cats. The results were similar in both anesthetized and conscious animals. Cystometrograms revealed that 8-OH-DPAT (0.5 mg/kg, s.c.) significantly increased the bladder volume threshold for eliciting a large amplitude micturition contraction, but only slightly reduced the amplitude of the contractions and did not alter the small amplitude pre-micturition contractions. 8-OH-DPAT also r…

Involvement of vagal opioid receptors in respiratory effects of morphine in anaesthetized rats.

Respiratory effects of morphine injection to the femoral vein were investigated in urethane and chloralose anaesthetized and spontaneously breathing rats, prior to and after midcervical vagotomy. Bolus injection of morphine HCl at a dose of 2 mg/kg of body weight induced depression of ventilation in all rats, due to the significant decrease in tidal volume and to the decline in respiratory rate both pre- and post-vagotomy. Expiratory apnoea of mean duration of 10.0+/-3.4 s was present in the vagally intact rats only. Bilateral midcervical section of the vagus nerve precluded the occurrence of …

Neuromuscular mechanisms of esophageal responses at and proximal to a distending balloon.

To determine the neuromuscular mechanisms involved in esophageal responses at and proximal to a distending stimulus, a manometric catheter incorporating a latex balloon at its distal end was positioned in the smooth muscle esophagus of alpha-chloralose-anesthetized opossums and used to record intraluminal pressures over the balloon and at several sites proximal to the balloon. Air inflation of the balloon evoked simultaneous phasic contractions at several sites proximal to the balloon that were abolished by bilateral cervical vagotomy. Effects of central nervous system-acting drugs on urinary bladder contraction in unanesthetized rats.

We studied the effects of drugs on urinary bladder contraction in unanesthetized (UA) rats using the same method as that previously employed to investigate similar effects in urethane and alpha-chloralose-anesthetized (A) rats. The surgical procedure was performed under halothane anesthesia, and after the recovery, the rats were restricted in a Ballman cage during the experiment. The pattern of the cystometrogram obtained in UA rats was very similar to that in A rats, and almost the same pattern was maintained for at least three hours. Baclofen (10 mg/kg, i.p.), morphine (10 mg/kg, i.p.) and p…

The lateral vestibular nucleus is a site for the depressant action of benzodiazepines on the crossed extensor reflex.

The effects of diazepam microinjected into the subcerebellar nuclei, the lateral vestibular nucleus (LVN), the red nucleus and the substantia nigra were investigated using the crossed extensor reflex (CER) model in chloralose-anesthetized rats. Microinjections of diazepam at doses of 1 and 25 ng/animal into the left LVN, ipsilateral to the testing muscle, produced a dose-related CER depression, while injections of diazepam at a dose of up to 100 ng/animal into the right LVN and into other regions tested produced little effect. The CER depression produced by diazepam could be clearly reversed b…

Reversible blockade of cardiac efferents with procaine instilled into the pericardium of cats.

The blocking of afferent signals from the heart by the instillation of local anaesthetics into the pericardium has frequently been used for studying cardiac reflexes. Yet no attempts have been made to define what effects this has on efferent cardiac drive. Therefore, in nine chloralosed cats with open chests and catheters sewn into the pericardium the procaine concentrations were determined which would block the heart rate responses to electrostimulation of the vagi nerves or the stellate ganglia. The procaine effects on arterial pressure and left ventricular peak positive dP/dt were also eval…
