"Lego-Brick"-Style Supramolecular Self-Assembly for Naringin-Chlorogenic Acid-Glucosylrutin Carrier-Free Nanoparticles: Self-Assembly Mechanism, Release Behavior, and Anti-Inflammatory Activity.

Naringin (NG), a citrus flavonoid with anti-inflammatory properties, suffers from low solubility, limiting the full realization of its health benefits. Inspired by the modular assembly concept of "Lego-bricks", this study constructed naringin-chlorogenic acid-glucosylrutin carrier-free nanoparticles (NCGN) with high apparent co-assembly efficiency (95.38 ± 0.77%) and loading capacity (32.54 ± 0.47%) through supramolecular self-assembly, leading to 4.6-fold increase in the aqueous solubility of NG. Investigation into the supramolecular self-assembly mechanism revealed that the process was prima

A Natural Polyphenol, Chlorogenic Acid, Attenuates Obesity-Related Metabolic Disorders in Male Rats via miR-146a-IRAK1-TRAF6 and NRF2-Mediated Antioxidant Pathways.

Chronic high-fat diet (HFD) feeding in male rats causes significant metabolic as well as inflammatory disturbances, including obesity, insulin resistance, dyslipidemia, liver and kidney dysfunction, oxidative stress, and hypothalamic dysregulation. This study assessed the therapeutic effects of chlorogenic acid (CGA), a natural polyphenol, administered at 10 mg and 100 mg/kg/day for the last 4 weeks of a 12-week HFD protocol. Both CGA doses reduced body weight gain, abdominal circumference, and visceral fat accumulation, with the higher dose showing greater efficacy. CGA improved metabolic par

Evaluating the antiviral efficacy and specificity of chlorogenic acid and related herbal extracts against SARS-CoV-2 variants via spike protein binding intervention.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and influenza A virus infect the respiratory tract through surface proteins, causing similar symptoms. Since the onset of the COVID-19 pandemic, both viruses have posed significant ongoing global health threats. Like the influenza virus, SARS-CoV-2 evolves into variants that can reduce vaccine efficacy. Thus, herbal medicines are being explored as supplementary options to enhance protection against these infections. This study aimed to investigate the therapeutic potential of chlorogenic acid (3-CQA) and related extracts from green c

Elucidation of intermolecular interactions between chlorogenic acid and glucose-6-phosphate translocase: A step towards chemically induced glycogen storage disease type 1b model.

We have developed 3D coculture models, which we term MAME (mammary architecture and microenvironment engineering), and used them for live-cell imaging in real-time of cell:cell interactions. Our overall goal was to develop models that recapitulate the architecture of preinvasive breast lesions to study their progression to an invasive phenotype. Specifically, we developed models to analyze interactions among pre-malignant breast epithelial cell variants and other cell types of the tumor microenvironment that have been implicated in enhancing or reducing the progression of preinvasive breast ep

Green coffee bean extract rich in chlorogenic acids prevents muscle capillary regression via inhibiting oxidative stress and enhancing angiogenesis.

Reactive oxygen species (ROS) production in disused skeletal muscle induces capillary regression, which disrupts the balance of pro- and anti-angiogenic factors. We tested whether green coffee bean extract containing rich chlorogenic acid, which has antioxidant properties, can prevent capillary regression caused by muscle atrophy. The study included 24 female Sprague-Dawley rats, randomly assigned to four groups: control (CON), 2-week hindlimb unloading (HU), 2-week hindlimb unloading supplemented with coffee extract rich in chlorogenic acids (HU+50%CGA), and 2-week hindlimb unloading suppleme

Effects of chlorogenic acid on the interfacial properties and physical stability of sunflower oil body emulsions: Insights from a reconstructed protein-emulsion model.

The stability of sunflower oil body emulsions is mainly governed by interfacial phospholipids and proteins, whereas the role of endogenous polyphenols remains unclear. This study aimed to evaluate the effects of chlorogenic acid (CGA) on physical stability. Reconstructed oil body proteins (ROBPs) and emulsions (ROBEs) were used as model systems. CGA decreased β-turn content from 43.06% in 0 mg-CGA-ROBPs to 26.50% in 38 mg-CGA-ROBPs. For ROBPs, as CGA addition increased from 0 to 38 mg, the emulsifying activity index increased from 27.91 to 77.32 m2/g and the emulsion stability index from 11.04

The Impact of Chlorogenic Acid Liposomes Dip-Coating on the Physicochemical Quality and Microbial Diversity of Low-Salt Cured Fish During Refrigerated Storage.

Low-salt cured fish is prone to deterioration due to lipid oxidation and microbial proliferation during refrigeration. Chlorogenic acid (CGA), with excellent antioxidant and antimicrobial activity, is a promising candidate for the preservation of cured fish. However, its instability in the presence of environmental factors significantly confines its direct application. In this research, CGA was encapsulated in liposomes and utilized as a dip-coating for cured fish. The effects of varying concentrations of CGA-loaded liposomes (L-CGA) coating on the physicochemical quality and microbial diversi

Chlorogenic acid regulates the expression of protein phosphatase 2A subunit B in the cerebral cortex of a rat stroke model and glutamate-exposed neurons.

BACKGROUND: Ischemic stroke is a serious neurological disorder caused by blockages in cerebral artery. Protein phosphatase 2A (PP2A) is a phosphatase that performs a critical role in cell signaling and growth. PP2A subunit B acts as a neuroprotective agent in the nerve system. Chlorogenic acid, which is mainly found in roasted coffee, has antioxidant, anti-inflammatory, and anti-apoptotic effects. We hypothesized that chlorogenic acid modulates PP2A subunit B expression in ischemic stroke models and glutamate-mediated neurons. Middle artery occlusion (MCAO) surgery was operated and chlorogenic

DNA Mutagenicity of Hydroxyhydroquinone in Roasted Coffee Products and Its Suppression by Chlorogenic Acid, a Coffee Polyphenol, in Oxidative-Damage-Sensitive SAMP8 Mice.

Hydroxyhydroquinone (HHQ) is an oxidative component produced by roasting coffee beans and has been reported to generate relatively large amounts of reactive oxygen species (ROS). In this study, we used senescence-accelerated mouse prone 8 (SAMP8) mice to determine whether HHQ consumption increases oxidative-stress-induced injury, because in SAMP8 mice, the activity of 8-oxoguanine DNA glycosylase 1, which repairs oxidative modifications in DNA, is decreased. The results showed that two out of twelve (16.7%) HHQ-treated mice presented polyuria and glucosuria around 2 months after the start of t

Chlorogenic acid-grafted chitosan as a natural cryoprotectant preserves the quality and myofibrillar protein stability of pork patties during repeated freeze-thaw cycles.

Repeated freeze-thaw (F-T) cycles during cold-chain storage severely deteriorate the quality of minced meat products. This study evaluated the cryoprotective efficacy of chlorogenic acid-grafted chitosan (CA-g-CH) on the quality stability and structural integrity of myofibrillar proteins (MPs) in pork patties. Pork patties supplemented with 0%, 0.3%, 0.6%, or 0.9% (w/w) CA-g-CH were subjected to six F-T cycles, and their physicochemical quality, water mobility, texture, rheological properties, lipid and protein oxidation, microstructure, and MP conformation were systematically evaluated. Chlorogenic Acid Alleviates Heat Stress-Induced Fetal Growth Restriction Through Gut-Placental Crosstalk by Reshaping Gut Microbiota and Mediating Keap1-Nrf2 Antioxidant Signaling.

BACKGROUND: Climate change-driven heat stress (HS) poses a growing threat to pregnancy outcomes; however, the mechanisms linking maternal HS to fetal growth restriction (FGR) remain incompletely understood, and effective nutritional interventions are lacking. The gut-placenta axis critically governs maternal-fetal health, but its role in mediating HS-related FGR has yet to be elucidated. METHODS: A murine model of gestational HS (38.5 °C, 2.5 h daily from E0.5 to E12.5) was established to investigate the protective effects of chlorogenic acid (CGA) on HS-induced FGR. Placental efficiency, oxi

Chlorogenic acid targets SLC37A2 to inhibit macrophage activation via ER-dependent NF-κB and NLRP3 signaling pathways against sepsis-induced acute lung injury.

Sepsis-induced acute lung injury (SI-ALI) requires urgent treatment due to severe inflammation. Our study found chlorogenic acid (CGA) suppressed LPS-induced macrophage activation by lowering NO, TNF-α, and IL-6. TPP-based strategies identified SLC37A2 as the direct target of CGA, validated by CETSA/MST. Molecular docking indicated CGA-SLC37A2 hydrogen bonding. CGA alleviated endoplasmic reticulum stress via SLC37A2, inhibiting TLR4/NF-κB and NLRP3 pathways to reduce inflammation. In SI-ALI mice and zebrafish models, CGA mitigated lung injury through these mechanisms taken together. This work

Upregulation of caveolin-1 by mulberry leaf extract and its major components, chlorogenic acid derivatives, attenuates alcoholic steatohepatitis via inhibition of oxidative stress.

Excessive alcohol uptake exerts hepatocellular toxicity, ultimately leading to multiple liver diseases such as steatohepatitis and liver cirrhosis. Here, we aimed to explore the effects of mulberry leaf extract (MLE) and its major components chlorogenic acid (CGA) and neochlorogenic acid (nCGA) on alcoholic steatohepatitis. We determined the composition of MLE using liquid chromatography-mass spectrometric (LC-MS) analysis, which showed that MLE consisted of mainly chlorogenic acid derivatives and other polyphenols. Next, we utilized a high alcohol-fed mouse model and demonstrated that MLE all
