TNFα Blockade Inhibits Both Initial and Continued Control of Pulmonary Coccidioides.

Tumor necrosis factor alpha (TNFα) is a pluripotent cytokine that is important in many infections, though its role in Coccidioides infection remains poorly understood. The need to understand TNFα in Coccidioides infection has increased recently with the widespread use of TNFα inhibitors for a wide variety of autoimmune conditions. Here, we couple the newly developed Coccidioides infection model using strain Cp1038 and C57BL/6 × DBA/2J F1 (B6D2F1) mice. B6D2F1 mice develop long-lasting control of Cp1038. Treatment of B6D2F1 mice with anti-TNFα antibodies permits significant fungal proliferation…

Factors Influencing Distribution of Coccidioides immitis in Soil, Washington State, 2016.

Coccidioides immitis and Coccidioides posadasii are causative agents of Valley fever, a serious fungal disease endemic to regions with hot, arid climate in the United States, Mexico, and Central and South America. The environmental niche of Coccidioides spp. is not well defined, and it remains unknown whether these fungi are primarily associated with rodents or grow as saprotrophs in soil. To better understand the environmental reservoir of these pathogens, we used a systematic soil sampling approach, quantitative PCR (qPCR), culture, whole-genome sequencing, and soil chemical analysis to iden…

Dendritic cell-based immunization induces Coccidioides Ag2/PRA-specific immune response.

Valley Fever, or coccidioidomycosis, is caused by a soil-borne, highly virulent fungal pathogen, Coccidioides spp. Infection with Coccidioides can be life-threatening. Since an effective treatment is not available and the T cell-mediated immune response is protective, vaccine development is of interest. In this study, a primary dendritic cell (DC)-vaccine was evaluated for its ability to stimulate Coccidioides antigen-specific immune response in an extremely susceptible BALB/c mouse model. The DC-vaccine (Ag2-DC) was prepared by non-virally transfecting the primary bone marrow-derived DCs with…

Susceptibility of TLR4-defective C3H/HeJ mice to Coccidioides posadasii infection.

Coccidioides posadasii is one of the two fungal pathogens that cause coccidioidomycosis. The inhalation of air-borne arthroconidia leads to the formation of endospore-forming spherules in the lungs and pulmonary infection. In severe condition, the endospores are disseminated to other non-pulmonary organs in the body. The Toll-like receptors (TLR) expressed by a number of immune and non-immune cells can significantly impact the host defense and susceptibility to C. posadasii infection. In this study, we infected TLR4-defective C3H/HeJ mice with a sublethal dose of C. posadasii and studied funga…

Pulmonary Coccidioides nodule in a Swiss patient with chronic lymphatic leukaemia.

The dimorphic fungus Coccidioides immitis, causative agent of coccidioidomycosis, is endemic in semi-arid regions of the Americas. The disease is rarely encountered in Europe. We describe the case of a 61-year-old Swiss patient with chronic lymphatic leukaemia who was eventually diagnosed with an initially unnoticed pulmonary coccidioidomycosis.

Ecological niche modeling of Coccidioides spp. in western North American deserts.

Coccidioidomycosis is an endemic infectious disease in western North American deserts caused by the dimorphic ascomycete Coccidioides spp. Even though there has been an increase in the number of reported cases in the last years, few positive isolations have been obtained from soil samples in endemic areas for the disease. This low correlation between epidemiological and environmental data prompted us to better characterize the fundamental ecological niche of this important fungal pathogen. By using a combination of environmental variables and geospatially referenced points, where positive isol…

Detection of Coccidioides species in clinical specimens by real-time PCR.

Coccidioides spp. are dimorphic fungal pathogens endemic to the semiarid regions of North, Central, and South America. Currently, direct smear and culture are the most common means of identifying Coccidioides spp. While these methods offer relatively sensitive and specific means of detecting Coccidioides spp., growth in culture may take up to 3 weeks, potentially delaying the diagnosis and initiation of appropriate antifungal therapy. In addition, growth of the organism represents a significant safety risk to laboratory personnel. The need for a rapid and safe means of diagnosing coccidioidomy…

Molecular markers reveal cryptic sex in the human pathogen Coccidioides immitis.

Coccidioides immitis, cause of a recent epidemic of "Valley fever" in California, is typical of many eukaryotic microbes in that mating and meiosis have yet to be reported, but it is not clear whether sex is truly absent or just cryptic. To find out, we have undertaken a population genetic study using PCR amplification, screening for single-strand conformation polymorphisms, and direct DNA sequencing to find molecular markers with nucleotide-level resolution. Both population genetic and phylogenetic analyses indicate that C. immitis is almost completely recombining. To our knowledge, this stud…

Identification of antigens of Coccidioides immitis which stimulated immune T lymphocytes.

T-cell mediated immune response to coccidioidomycosis has been shown to be the principal mechanism of resistance to this respiratory fungal disease in experimental animals. In this study, a Coccidioides immitis antigen-specific murine T-cell line was used to identify macromolecules capable of eliciting an immune mouse T-cell proliferative response. The murine T-cell line was selected on the basis of its strong positive response to a soluble conidial wall fraction (SCWF), which had previously been shown to be reactive in humoral and cellular immunoassays. An antigen-specific T-cell line rather …

Interaction of human peripheral blood mononuclear cells with Coccidioides immitis arthroconidia.

We explored the in vitro interaction of human peripheral blood mononuclear cells with the arthroconidial stage of the fungus Coccidioides immitis. Fresh peripheral blood monocytes in an adherent monolayer were capable of ingesting C. immitis. Further, peripheral blood monocytes from either skin-test-positive or skin-test-negative donors significantly decreased the in vitro growth of C. immitis when coccidioidal arthroconidia were incubated with monocytes. Peripheral blood mononuclear cells also reduced fungal incorporation of the chitin precursor N-acetyl glucosamine. Cell fractions consisting…

Electron-microscopic observations of the Coccidioides immitis parasitic cycle in vivo.

BALB/c mice were inoculated by intranasal challenge with viable arthroconidia of C. immitis and in-vivo morphogenesis of the fungal pathogen was investigated by electron-microscopic examination of pulmonary lavage and cryofractured lung specimens. Samples were prepared at intervals over an 11-day period. Stages of spherule and endospore development were easily identified by scanning and thin-section electron microscopy. Details of morphogenesis of the pathogen in vivo closely resemble developmental aspects reported from in-vitro studies.

Human sex hormones stimulate the growth and maturation of Coccidioides immitis.

Because men and pregnant women show increased susceptibility to extrapulmonary dissemination of coccidioidomycosis, studies were conducted to determine the direct effect of human sex hormones and related compounds on the growth and maturation of Coccidioides immitis in vitro. 17 beta-Estradiol, progesterone, and testosterone were highly stimulatory for the parasitic phase of C. immitis growth whereas cholesterol, ergosterol, and 17 alpha-estradiol (a physiologically inactive stereoisomer of 17 beta-estradiol) lacked such effects. Rates of spherule maturation and endospore release were accelera…

Uniform susceptibility of various strains of Coccidioides immitis to amphotericin B.

The susceptibility of 12 strains of Coccidioides immitis to amphotericin B (amB) was studied in vitro and in vivo. The minimal inhibitory concentration (MIC) of the mycelial phase of these strains was 0.078 to 0.16 mug/ml after 3 days of incubation, but by 15 days all strains were inhibited by 2.5 mug/ml. Mice infected intraperitoneally with these strains were sucessfully treated with 0.5 mg of amB per kg per day. These strains included several studied by others and which reportedly varied widely in susceptibility (MIC from 0.24 to 24.01 mug/ml) to amB. Four of these strains representing this …

Studies on coccidioides immitis. II. Physiological studies on in vitro spherulation.

Brooks, Lula D. (Arizona State University, Tempe) and William T. Northey. Studies on Coccidioides immitis. II. Physiological studies on in vitro spherulation. J. Bacteriol. 85:12-15. 1963.-Studies on the amino acids assayed in Edamine revealed that the amino acids phenylalanine, tyrosine, and tryptophan, in addition to the amino acid derivatives dihydroxyphenylalanine (DOPA), epinephrine, tyramine monohydrochloride (tyramine HCl), and pyrocatechol play an important role in the Coccidioides immitis conversion process. In the presence of phenylalanine, tyrosine, tryptophan, DOPA, pyrocatechol, d…

Detection of Coccidioides immitis in Archived Soils from Madera County, California.

In the 1970s, archaeologists excavating sites in Madera County, California, which is now submerged by Hensley Reservoir, contracted coccidioidomycosis (Valley fever), a fungal disease caused by Coccidioides immitis. Soil samples from these sites were archived but not processed due to concerns about infectious arthroconidia. This study investigates whether these decades-old samples, stored at room temperature, still contain Coccidioides and pose a risk to researchers and collections managers. We analyzed 176 subsamples using molecular and culture-based methods, with DNA extractions performed in…

Spatiotemporal analysis of lung immune dynamics in lethal Coccidioides posadasii infection.

Coccidioidomycosis, or Valley fever, is a lung disease caused by inhalation of Coccidioides fungi, prevalent in the Southwestern United States, Mexico, and parts of Central and South America. Annually, the United States reports 10,000-20,000 cases, although those numbers are expected to increase as climate change expands the fungal geographic range. While 60% of infections are asymptomatic, 40% symptomatic infections are often misdiagnosed due to similarities with bronchitis or pneumonia. A small subset of infection progress to severe illness, necessitating a better understanding of immune res…

Life Cycle Dominates the Volatilome Character of Dimorphic Fungus Coccidioides spp.

Valley fever (coccidioidomycosis) is an endemic fungal pneumonia of the North and South American deserts. The causative agents of Valley fever are the dimorphic fungi Coccidioides immitis and C. posadasii, which grow as mycelia in the environment and as spherules within the lungs of vulnerable hosts. Current diagnostics for Valley fever are severely lacking due to poor sensitivity and invasiveness, contributing to a 23-day median time to diagnosis, and therefore, new diagnostic tools are needed. We are working toward the development of a breath-based diagnostic for coccidioidomycosis, and in t…

Transcriptional Analysis of Coccidioides immitis Mycelia and Spherules by RNA Sequencing.

Coccidioides immitis and C. posadasii are dimorphic fungi that transform from mycelia with internal arthroconidia in the soil to a tissue form known as a spherule in mammals. This process can be recapitulated in vitro by increasing the temperature, CO2 and changing other culture conditions. In this study, we have analyzed changes in gene expression in mycelia and young and mature spherules. Genes that were highly upregulated in young spherules include a spherule surface protein and iron and copper membrane transporters. Genes that are unique to Coccidioides spp. are also overrepresented i…
