A mouse model of allergic conjunctivitis with mucosal immune-related gene and circRNA dysregulation.

Allergic conjunctivitis (AC) is an inflammatory ocular condition triggered by allergens like pollen. Although general allergic mechanisms are well characterized, the immune specificity of the ocular mucosa remains unclear. This study investigates immune-related mRNAs and circRNAs to uncover novel molecular pathways in AC pathogenesis. Using a ragweed pollen-induced murine AC model, conjunctival tissues were subjected to RNA sequencing. DESeq2-based differential expression analysis revealed dysregulated mRNAs and circRNAs, with significant enrichment in the IL-17 signaling pathway. Key IL-17-as

Eosinophilic esophagitis is associated with increased risk of allergic conjunctivitis: a multicenter cohort study.

BACKGROUND: Eosinophilic esophagitis (EoE) is a chronic, Th2-driven inflammatory esophageal disease frequently associated with atopic disorders. However, ocular comorbidities in EoE, particularly allergic conjunctivitis, have not been well characterized. In this study, we compare the incidence of ocular outcomes, with a focus on allergic conjunctivitis, between patients with EoE and matched controls with gastroesophageal reflux disease with esophagitis (GERD-E). METHODS: We conducted a retrospective, propensity score-matched cohort study using the TriNetX Global Collaborative Network, includi

The effectiveness of olopatadine hydrochloride eye drops for allergic conjunctivitis: Protocol for a systematic review.

BACKGROUND: Allergic conjunctivitis (AC) is a multifactorial and common type of ocular surface disease that affects many people. The quality of life for AC patients can be significantly decreased caused by symptoms of ocular itching, swelling, redness, and tearing. Topical antihistaminics, mast cell stabilizers, non-steroidal anti-inflammatory drugs (NSAIDs), and steroids have been widely used to treat AC. Many clinical trials have indicated that olopatadine hydrochloride eye drops can provide quick relief of symptoms and signs. The purpose of this review is to evaluate systematically the effe

Investigation of the Possible Role of Tie2 Pathway and TEK Gene in Asthma and Allergic Conjunctivitis.

Tie2, coded by the TEK gene, is a tyrosine kinase receptor and plays a central role in vascular stability. It was suggested that variations in the TEK gene might influence the susceptibility to asthma and allergic conjunctivitis. The aim of this study was to further investigate these suggestions, involving different populations and to study the Tie2 related pathway on a mouse model of asthma. The discovery, stage I cohort involved 306 patients with moderate and severe allergic rhinitis, the stage II study consisted of four cohorts, namely, adult and pediatric asthmatics and corresponding contr

Topical Administration of β-1,3-Glucan to Modulate Allergic Conjunctivitis in a Murine Model.

PURPOSE: Recent studies on β-1,3-glucan (BG), a cell wall component of a variety of fungi, yeasts, and bacteria, demonstrated that it affects the balance of Th1/Th2 immune responses. We therefore determined whether topical application of BG modulates ocular allergy in a murine model. METHODS: We sensitized 7- to 8-week-old BALB/c mice once with ovalbumin (OVA) and aluminum hydroxide via intraperitoneal injection. Mice were rested for 2 weeks and then challenged by instillation of OVA eye drops once daily for 13 days. We administered BG eye drops 5 minutes after OVA challenge once daily. Clini

Interaction between nitric oxide and prostaglandin synthesis in the acute phase of allergic conjunctivitis.

Both nitric oxide and prostaglandins induce vasodilatation which is an important feature of local inflammation. The purpose of the study described here was to investigate a possible interaction between these two types of mediators in an experimental model of allergic conjunctivitis. A conjunctival allergic reaction was induced with antigen in sensitized guinea pigs. Conjunctival vascular permeability changes were evaluated with the prophylactic use of an inhibitor of nitric oxide synthase (L-NAME) and a cycloxygenase inhibitor (indomethacin). To study a possible interaction between nitric oxid

Novel Polyvinyl Pyrrolidone-Loaded Olopatadine HCl-Laden Doughnut Contact Lens to Treat Allergic Conjunctivitis.

Olopatadine HCl is an antiallergic drug used for the management of allergic conjunctivitis. Currently, it is delivered via eye drop solution, which is highly inefficient due to low bioavailability. Silicone contact lenses can be used to sustain the release of ophthalmic drugs. However, the presence of drug alters the optical transmittance and physical properties of the contact lens. The objective was to design a novel polyvinyl pyrrolidone (PVP)-coated olopatadine-ethyl cellulose microparticles-laden doughnut contact lens to sustained ocular delivery with limited alteration to the optical and

The C-C chemokine receptor 6 (CCR6) is crucial for Th2-driven allergic conjunctivitis.

Allergic conjunctivitis from an allergen-driven Th2 response is characterized by conjunctival eosinophilic infiltration. Although CCL20-CCR6 axis has been reported to play a proinflammatory role in several murine models of autoimmune diseases including allergic diseases, their underlying mechanism needs to be investigated. We here examined whether CCL20-CCR6 axis could play a role in the development of allergic conjunctival inflammation using murine experimental allergic conjunctivitis (EAC) model induced by ovalbumin (OVA) allergen. Mice were challenged with consecutive 10days of OVA via conj

Multiple cedar pollen challenge diminishes involvement of histamine in allergic conjunctivitis of Guinea pigs.

It has been reported that antihistamines do not fully modify symptoms of allergic conjunctivitis in clinical settings, suggesting that histamine is not the only contributor to symptom generation in the disease. However, in the majority of experimental allergic conjunctivitis models, antihistamines are very effective in the reduction of symptoms. In the present study, we used our recently developed guinea pig model of allergic conjunctivitis and evaluated whether involvement of histamine in the induction of symptoms of allergic conjunctivitis is altered by multiple antigen challenges.
