Alleviation of high-fat diet-induced lipid metabolism disorders: role of quinoa peptides in reducing high-activity BSH-producing gut microbiota abundance and modulating BA-FXR/TGR5 signaling.

INTRODUCTION: Lipid metabolism disorders contribute significantly to various metabolic diseases and are closely related to gut microbiota dysbiosis. Quinoa intake has been increasingly linked to improved metabolic regulation and body weight control. In our earlier work, oral administration of quinoa-derived peptides (QPep) modulated gut microbiota composition and mitigated hepatic lipid dysregulation in high-fat diet (HFD)-induced obese mice. Nevertheless, the specific mechanisms responsible for these effects remain incompletely understood. OBJECTIVES: This study aimed to elucidate the mechan

Possible Role of High-Dose Barbiturates and Early Administration of Parenteral Ketogenic Diet for Reducing Development of Chronic Epilepsy in Febrile Infection-Related Epilepsy Syndrome: A Case Report.

We describe the efficacy of high-dose barbiturates and early administration of a parenteral ketogenic diet (KD) as initial treatments for acute status epilepticus (SE) in an 8-year-old girl with febrile infection-related epilepsy syndrome (FIRES). The patient was admitted to our hospital with refractory focal SE. Abundant epileptic discharges over the left frontal region were observed on electroencephalogram (EEG). Treatment with continuous infusion of thiamylal for 4 hours, increased incrementally to 40 mg/kg/h, successfully ended the clinical SE, and induced a burst-suppression coma. The inf

A possible role for ghrelin, leptin, brain-derived neurotrophic factor and docosahexaenoic acid in reducing the quality of life of coeliac disease patients following a gluten-free diet.

PURPOSE: A gluten-free diet (GFD) has been reported to negatively impact the quality of life (QoL) of coeliac disease (CD) patients. The gut-brain axis hormones ghrelin and leptin, with the brain-derived neurotrophic factor (BDNF), may affect QoL of CD patients undergoing GFD. Our aims were to evaluate whether: (a) the circulating concentrations of leptin, ghrelin and BDNF in CD patients were different from those in healthy subjects; (b) GFD might induce changes in their levels; (c) BDNF Val66Met polymorphism variability might affect BDNF levels; and (d) serum BDNF levels were related to dieta

Green tea extract protects against hepatic NFκB activation along the gut-liver axis in diet-induced obese mice with nonalcoholic steatohepatitis by reducing endotoxin and TLR4/MyD88 signaling.

Green tea extract (GTE) reduces NFκB-mediated inflammation during nonalcoholic steatohepatitis (NASH). We hypothesized that its anti-inflammatory activities would be mediated in a Toll-like receptor 4 (TLR4)-dependent manner. Wild-type (WT) and loss-of-function TLR4-mutant (TLR4m) mice were fed a high-fat diet containing GTE at 0 or 2% for 8 weeks before assessing NASH, NFκB-mediated inflammation, TLR4 and its adaptor proteins MyD88 and TRIF, circulating endotoxin, and intestinal tight junction protein mRNA expression. TLR4m mice had lower (P<.05) body mass compared with WT mice but similar ad

Optimized amino acid ratios in reduced crude protein diets sustain broiler performance, gut health, and resilience to necrotic enteritis.

This study investigated the effects of essential-to-total amino acid (E:T) ratios in reduced crude protein (CP) diets on broiler performance, gene expression, and cecal microbiota under necrotic enteritis (NE) challenge. A total of 756 mixed-sex 0-d-old Cobb 500 broilers were assigned to six treatments in a 2 × 3 factorial arrangement, with two challenge conditions (yes or no) and three dietary treatments: 17% reduced CP (RCP) with E:T ratio of 0.64 (RCP0.64), 17% RCP with E:T ratio of 0.68 (RCP0.68), and 19% normal CP as control (NCP) from d 9 to 18. Performance was measured from d 8 to 18 po

Incorporating Euglena gracilis into the habitual diet modulates immune function and reduces common cold-like symptoms: a randomized controlled trial.

BACKGROUND: Dietary approaches to support immune function have emphasized the importance of dietary fiber, including non-digestible polysaccharides, although intake remains below recommended levels in many populations. In this context, microalgae have attracted attention as sustainable sources of these components. Euglena gracilis, a microalga used in foods, contains paramylon, a β-1,3-glucan with reported immunomodulatory properties; however, its effects on plasmacytoid dendritic cell (pDC) activation in humans within a dietary context remain incompletely understood. METHODS: This randomized

High-fat diet feeding reduced smooth muscle contractility and increasing tissue programmed death activity in rats bladder tissue.

PURPOSE: High-fat diet (HFD)-induced metabolic disturbances are recognized risk factors for bladder dysfunction, yet the underlying mechanisms remain incompletely understood. METHODS: Twelve male Wistar rats were allocated to normal diet (ND) or HFD groups for 12 weeks. Physiological parameters, cystometry, and detrusor muscle contractility assays were performed. Histological analyses assessed vascular remodeling and fibrosis, while immunohistochemistry and western blotting evaluated programmed cell death pathways and receptor expression. RESULTS: HFD-fed rats exhibited significant body weig

Targeting the Gut-Kidney-Heart Axis in Chronic Kidney Disease: The Mediterranean Diet as a Strategy to Reduce Uremic Toxins and Cardiovascular Risk.

Chronic kidney disease (CKD) is associated with a markedly increased risk of cardiovascular (CV) morbidity and mortality that cannot be fully explained by traditional risk factors. Emerging evidence highlights the central role of the gut-kidney-heart axis, whereby gut microbiota dysbiosis promotes the generation and systemic accumulation of uremic toxins, including indoxyl sulfate (IS), p-cresyl sulfate (PCS), and trimethylamine N-oxide (TMAO). These metabolites contribute to endothelial dysfunction, oxidative stress, inflammation, and vascular remodeling, thereby accelerating CV disease progr

Adverse effects of high-fat diet consumption on contractile mechanics of isolated mouse skeletal muscle are reduced when supplemented with resveratrol.

Increasing evidence indicates resveratrol (RES) supplementation evokes anti-obesogenic responses that could mitigate obesity-induced reductions in skeletal muscle (SkM) contractility. Contractile function is a key facet of SkM health that underpins whole body health. For the first time, the present study examines the effects of a high-fat diet and RES supplementation on isolated soleus (SOL) and extensor digitorum longus (EDL) contractile function. Female CD-1 mice, ∼6 weeks old (n = 38), consumed a standard laboratory diet (SLD) or a high-fat diet (HFD), with or without RES (4 g kg-1 diet) fo

Nutritionist-Guided Video Intervention Improves Adherence to Mediterranean Diet and Reduces the Rate of Gestational Diabetes Mellitus: A Randomized Clinical Trial.

Aims: Gestational diabetes mellitus (GDM) represents an increasing global challenge. Mediterranean diet interventions have proven benefits, but their implementation is limited by the absence of nutritionists in many public health systems. This study aimed to evaluate whether a video intervention guided by a nutritionist could increase compliance to Mediterranean diet and reduce the incidence of GDM and adverse maternal-neonatal outcomes. Methods: In this randomized controlled trial, 1750 consecutive pregnant women were allocated (1:1) to standard care (verbal, printed advice) or to a video des

Reducing the dietary starch:protein ratios in low-protein diets enhanced the growth performance of goslings from 1 to 28 days of age.

The current study aimed to explore the suitable starch: protein ratios under different dietary protein levels for goslings. A total of 360 male 1-day-old Jiangnan White goslings were randomly divided into 6 groups with six replicates containing ten goslings each. The experimental design consisted of a 3 × 2 factorial array of treatments. Three protein levels (18%, 16%, 14%) and two starch: protein ratio (S: P ratio) types (standard, reduced) were formulated. The results showed that: reducing the S: P ratio at the same dietary protein level increased weight gain (WG), average daily gain (ADG),

Anti-inflammatory diet reduces risk of metabolic dysfunction-associated fatty liver disease among US adults: a nationwide survey.

BACKGROUND: While dietary intervention was an important public health strategy for the prevention and intervention of metabolic dysfunction-associated fatty liver disease (MAFLD), the effect of diet-induced inflammation on MAFLD has not been studied in detail. Therefore, we aimed to analyze the relationship between dietary inflammatory index (DII) and MAFLD. METHODS: This study included data from the National Health and Nutrition Examination Survey 2017-2018. MAFLD was diagnosed based on the presence of hepatic steatosis, as determined by transient elastography, along with evidence of either

A Combination of a Dopamine Receptor 2 Agonist and a Kappa Opioid Receptor Antagonist Synergistically Reduces Weight in Diet-Induced Obese Rodents.

As the global obesity rate increases, so does the urgency to find effective anti-obesity drugs. In the search for therapeutic targets, central nervous system (CNS) mechanisms engaged in the regulation of energy expenditure and food intake, such as the opioid and dopamine systems, are crucial. In this study, we examined the effect on body weight of two drugs: bromocriptine (BC), a D2R receptor agonist, and PF-04455242, a selective κ opioid receptor (KOR) antagonist. Using diet-induced obese (DIO) rats, we aimed to ascertain whether the administration of BC and PF-04455242, independently or in c

Obesogenic cafeteria diet induces dynamic changes in gut microbiota, reduces myenteric neuron excitability, and impairs gut contraction in mice.

The cafeteria diet (CAF) is a superior diet model in animal experiments compared with the conventional high-fat diet (HFD), effectively inducing obesity, metabolic disturbances, and multi-organ damage. Nevertheless, its impact on gut microbiota composition during the progression of obesity, along with its repercussions on the enteric nervous system (ENS) and gastrointestinal motility has not been completely elucidated. To gain more insight into the effects of CAF diet in the gut, C57BL/6 mice were fed with CAF or a standard diet for 2 or 8 wk. CAF-fed mice experienced weight gain, disturbed gl

Semaglutide reduces tumor burden in the GAN diet-induced obese and biopsy-confirmed mouse model of NASH-HCC with advanced fibrosis.

Non-alcoholic steatohepatitis (NASH) is emerging as a major cause of hepatocellular carcinoma (HCC), however, it is not resolved if compounds in late-stage clinical development for NASH may have additional therapeutic benefits in NASH-driven HCC (NASH-HCC). Here, we profiled monotherapy with semaglutide (glucagon-like-receptor-1 receptor agonist) and lanifibranor (pan-peroxisome proliferator-activated receptor agonist) in a diet-induced obese (DIO) mouse model of NASH-HCC. Disease progression was characterized in male C57BL/6 J mice fed the GAN (Gubra Amylin NASH) diet high in fat, fructose an

Acute stress and restricted diet reduce bill-mediated heat dissipation in the song sparrow (Melospiza melodia): implications for optimal thermoregulation.

We used thermal imaging to show that two environmental factors - acute stress and diet - influence thermoregulatory performance of a known thermal window, the avian bill. The bill plays important roles in thermoregulation and water balance. Given that heat loss through the bill is adjustable through vasoconstriction and vasodilation, and acute stress can cause vasoconstriction in peripheral body surfaces, we hypothesized that stress may influence the bill's role as a thermal window. We further hypothesized that diet influences heat dissipation from the bill, given that body condition influence
