Lysosomal degradation of ganglioside GM2 by hexosaminidase A requires the presence of a small, non-enzymatic cofactor, the GM2-activator protein (GM2AP). Lack of functional protein leads to the AB variant of GM2-gangliosidosis, a fatal lysosomal storage disease. In the framework of an extensive health survey, 119 viscose rayon workers exposed to carbon disulfide (CS2) and 79 workers from other plants not exposed to any toxic agent in the working environment underwent a gastrointestinal examination including a self-administered questionnaire, abdominal palpation, percussion and a number of liver function tests. The histological examination of testes was carried out in white male rats poisoned by the intraperitoneal administration once a week for 6 months of carbon disulphide at different doses of 5, 10, 15, 25 mg/kg body weight. Krill chitin modified with carbon disulfide was used as a support of enzymes. The dithiocarbamino groups created after reaction of CS(2) with chitinous NH(2) can participate in binding the enzymes without denaturation and loss in activity. The stability of these bonds depends on pH and decreases gradually at ionic strengths higher than 0.01. Chitin modified with CS(2) can be used as a support for enzymes having high activity at mild conditions of pH and ionic strength.

When rats were exposed to 2 mg l-1 (approximately 640 ppm) of carbon disulfide (CS2) for 4 h, the concentration of free CS2 in the red blood cells (RBCs) approached a plateau within 2 h. Free CS2 in plasma reached a steady state concentration within 15 min of exposure. More than 90% of the free CS2 in blood was found in the RBCs regardless of the length of exposure. In 48 men occupationally exposed to CS2 the following glycoproteins were estimated in blood serum, using radial immunodiffusion on M-Partigen plates: alpha1-acid glycoprotein, alpha1-antitrypsin, alpha2-HS-glycoprotein, alpha2-macroglobulin, Gc glycoprotein, hemopexin, haptoglobin, ceruluplasmin and beta2-glycoprotein I. The studies indicated a highly statistically significant increase of: alpha1-acid--glycoprotein and Gc glycoprotein, and decrease of: alpha1-antitrypsins, alpha2-HS--glycoprotein and beta2-glycoprotein.
