Simultaneous catalepsy and apomorphine-induced stereotypic behavior in mice.

Intraventricular administration of haloperidol or chlorpromazine produces catalepsy and blocks apomorphine-induced stereotypic behavior. Low intraventricular doses of domperidone, sulpiride and spiperone, equally cataleptogenic as haloperidol or chlorpromazine, augment rather than diminish stereotypic behavior produced by subsequent apomorphine treatment. The resultant stereotypic behavior continues even while the animal is in a rigid cataleptic posture and is marked by persistent gnawing and licking. Prior to the induction of catalepsy and after recovery from it, mice display the entire range

Drug-induced Sudden Death: A Scoping Review.

BACKGROUND: The risk of sudden cardiac death (SCD) can be increased with the use of drugs. However, it has been described heterogeneously in the literature. OBJECTIVE: This study aims to systematically review epidemiological studies dealing with druginduced sudden death, describe their methodologies, and summarize the results found. METHODS: A scoping review has been carried out using Medline electronic database. The search was limited up to 2020. Epidemiological studies were included, and case reports or case series were excluded. RESULTS: Out of 3,114 potential articles, 74 were included.

Carotid body dopaminergic mechanisms are functional after acclimatization to hypoxia in goats.

Ventilatory acclimatization to sustained hypoxia (VASH) is the time-dependent increase in ventilation that occurs during prolonged exposure to hypoxia. We tested the hypothesis that carotid body (CB) dopaminergic mechanisms are down-regulated during VASH, which would allow CB afferent discharge and ventilation to increase beyond the initial response to hypoxia. Domperidone (DOM; 1.0 mg.kg-1) was administered intravenously to block CB dopamine (DA) receptors after VASH was complete in awake goats. DOM caused a significant augmentation of the ventilatory response to hypoxia in acclimatized goats

Exploration of potential mechanisms and biomarkers related to ERS-associated RCD in steroid-induced osteonecrosis of the femoral head based on bioinformatics, with experimental validation.

BACKGROUND: The pathogenesis of osteonecrosis of the femoral head (ONFH) is associated with endoplasmic reticulum stress (ERS) and regulated cell death (RCD), but reliable linking biomarkers and their mechanisms remain unclear. This study aimed to screen SONFH biomarkers related to ERS/RCD and explore their mechanisms. METHODS: Using datasets from public databases on ONFH, ERS-related genes (ERGs), and RCD-related genes (RRGs), candidate genes were isolated by intersecting the differentially expressed genes (DEGs) with the ERG and RRG sets. We identified biomarkers by integrating a protein-pr

Class side effects: decreased pressure in the lower oesophageal and the pyloric sphincters after the administration of dopamine antagonists, neuroleptics, anti-emetics, L-NAME, pentadecapeptide BPC 157 and L-arginine.

The ulcerogenic potential of dopamine antagonists and L-NAME in rats provides unresolved issues of anti-emetic neuroleptic application in both patients and experimental studies. Therefore, in a 1-week study, we examined the pressures within the lower oesophageal and the pyloric sphincters in rats [assessed manometrically (cm H2O)] after dopamine neuroleptics/prokinetics, L-NAME, L-arginine and stable gastric pentadecapeptide BPC 157 were administered alone and/or in combination. Medication (/kg) was given once daily intraperitoneally throughout the 7 days, with the last dose at 24 h before pre

Side effects of drugs stimulating prolactin secretion on the behavior of male rats.

The behavioral effects of drugs stimulating prolactin (PRL) secretion have been studied in male rats. Acquisition of active avoidance behavior was facilitated by short- (SHPRL) and long-term hyperprolactinaemia (LHPRL) induced by pituitary homografts under the kidney capsule. Dopamine antagonists, such as haloperidol and bromperidol, though causing hyperprolactinaemia, suppressed the acquisition of active avoidance behavior. Sulpiride was not effective in this respect. SHPRL facilitated and LHPRL inhibited sexual behavior of male rats. Haloperidol, but not sulpiride, suppressed sexual capacity

Testosterone-induced hyperprolactinaemia in a patient with a disturbance of hypothalamo-pituitary regulation.

A case of a patient with hypopituitarism due to a disturbance of hypothalamo-pituitary regulation is presented, who developed high-grade hyperprolactinaemia after the initiation of substitutive therapy with testosterone esthers. The increase in serum Prl was strictly related to testosterone aromatization to oestradiol, since anti-oestrogen compounds were effective in reducing (clomiphene) or abolishing (tamoxifen) the enhanced Prl secretion. The oestrogen effect in raising Prl release was not attributable to a reduction in the dopamine inhibition of Prl-secreting cells, as the dopamine-antagon

Effects of N-methyl-D-aspartic acid and kainic acid on prolactin secretion in hyper- and hypoprolactinaemic conditions.

OBJECTIVE: The stimulatory and inhibitory effects of N-methyl-D-aspartic acid (NMDA) and kainic acid on prolactin (PRL) secretion have been correlated with the serum prolactin concentrations before drug administration. In the present experiments, we analysed the role of NMDA and kainic acid in PRL secretion in females with different serum concentrations of PRL. METHODS: Hypoprolactinaemic females were obtained by ovariectomy or after administration of diethyldithiocarbamate (an inhibitor of dopamine-beta-hydroxylase). Chronic hyperprolactinaemia was induced by neonatal administration of testo

Does B-HT 920 lower intraocular pressure in cats by interacting with alpha 2- and/or DA2 adrenoceptors?

Topical administration of B-HT 920 (50 micrograms) to the eyes of normal unanesthetized cats produced decreases in intraocular pressure and pupil diameter. The ocular hypotensive effect of B-HT 920 was eliminated by sympathectomy and pretreatment with sulpiride (2 mg/kg, s.c.). B-HT 920 also produced dose-related inhibition of contractions of the cat nictitans elicited by stimulating the pre- and postganglionic sympathetic trunks. B-HT 920-induced suppression of the contracting nictitans was antagonized more effectively by relatively selective DA2 antagonists, sulpiride and domperidone, than b

A case report on pheochromocytoma mimicking as fulminant myocarditis-a diagnostic challenge.

We present an exceptional case of a 53-year-old female, initially misdiagnosed with fulminant myocarditis, but later correctly diagnosed with pheochromocytoma. The presentation of the patient included a spectrum of symptoms such as headache, chest discomfort, palpitations, and dyspnea, following the intake of Domperidone. Two weeks prior to admission, the patient had experienced episodes of diarrhea and a low-grade fever. Unresolved symptoms and an unmanageable surge in blood pressure despite comprehensive fulminant myocarditis treatment prompted further investigation. The discovery of an adre

[Antiemetic properties of levo-sulpiride].

Levo-sulpiride is a substituted benzamide with antiemetic activity 3-8 times more potent than the racemic form and the d-isomer. Its mode of action is partially central (inhibition of dopaminergic receptors at the trigger zone for vomiting) and partially peripheral (normalization of motor activity of stomach and gall-bladder). The drug was found effective in the prevention of chemotherapy-induced and post-operative vomiting as well as in the treatment of nausea and vomiting during hepatic, biliary and gastroduodenal disorders, organic and functional dyspepsia, motion sickness and vertigo. Novel synaptic responses mediated by dopamine and gamma-aminobutyric acid in neuroendocrine cells of the intermediate pituitary.

Neuroendocrine cells of the pituitary pars intermedia contain pro-opiomelanocortin and secrete mainly alpha-melanocyte-stimulating hormone and, in lesser amounts, corticotropin and endorphin. We have recorded intracellularly from rat pars intermedia cells in vitro and analysed the synaptic inputs to these cells. Electrical stimulation of the pituitary stalk evoked a biphasic postsynaptic response in pars intermedia cells. An initial inhibitory postsynaptic potential was most likely mediated by gamma-aminobutyric acid (GABA) because there was a conductance increase, and it was blocked by bicucu

Effect of stage of lactation and gestation on milking-induced hormone release in lactating dairy cows.

Three experiments were conducted to better understand why milking-induced prolactin (PRL) release decreases as lactation advances. Experiment 1 compared the effects of milking, 2-min manual stimulation of the mammary gland (without milking), or injection of 1 IU of oxytocin (without milking) on hormonal release in early lactation cows, late-lactation and nongestating cows, and late-lactation and gestating cows (n = 6 per physiological status). Blood samples were collected from 20 min before the start of the treatments to 60 min after. During milking, PRL release (area under the curve above the

Significance of dopamine receptor antagonists in human postural control.

The purpose of this study was to investigate the effect of different dopamine receptor antagonists on the function of proprioceptive reflex mechanisms involved in the regulation of stance and gait. Haloperidol as preferential D2 and the 'pure' dopamine D2 receptor antagonist sulpiride significantly reduced the level of the reflex response in the gastrocnemius muscle following backward perturbation. Flupentixol, however, which in addition blocks D1 receptors, had no effect on compensatory gastrocnemius EMG responses. Furthermore, neither the peripherally acting dopamine D2 receptor antagonist d

The acute effects of central- and peripheral-acting dopamine antagonists on plasma HVA in schizophrenic patients.

The short-term effects of fluphenazine on plasma HVA concentrations were compared with the effects of fluphenazine and concurrent administration of debrisoquin, a monoamine oxidase inhibitor which does not cross the blood brain barrier and is used to enhance the CNS contribution to circulating plasma HVA concentrations. Fluphenazine significantly increased plasma HVA with or without debrisoquin 24 hours following the initiation of treatment. Domperidone, a butyrophenone dopamine antagonist which acts only in the peripheral nervous system, failed to alter plasma HVA concentrations. These data s

[Receptive relaxation of the canine stomach].

Receptive relaxation of the stomach by feeding was studied in 5 conscious dogs by means of chronically implanted force transducers on the gastric body and antrum. It was found that only the gastric body but not antrum relaxed by feeding, and magnitude of the relaxation of the gastric body was linearly related to the volume (5, 10 and 20 g/kg) of test meals, given once or in multiple times. Secondly, in the study of pharmacological blockers, it was found that atropine, hexamethonium and phentolamine significantly inhibited relaxation, while naloxone and proglumide significantly suppressed relax

Dihydroergotoxine decreases blood pressure in spontaneously hypertensive rats by interacting with peripheral dopamine receptors.

Dihydroergotoxine (10 micrograms/kg s.c.) decreased mean carotid blood pressure in urethane-anaesthetized spontaneously hypertensive rats but failed to modify the same parameter in normotensive rats. The effect was statistically significant 20 min after the injection and relatively long lasting (up to 90 min). Pharmacological characterization of the phenomenon indicated that it is mediated by stimulation of dopamine receptors, since pretreatment with haloperidol, cis-flupentixol but not with trans-flupentixol, completely prevent the reduction in blood pressure induced by dihydroergotoxine. [Hypogalactia: therapeutic possibilities].

Having briefly summarized the physiology of lactogenesis and gactopoiesis, the authors stress the fundamental role played by prolactin. In this context, they suggest medical treatment intended to improve mammary function in cases of deficient lactation. Two different types of treatment were tried: direct administration of prolactin or administration of antidopaminergic drugs capable of increasing endogenous prolactin production. In addition to confirming once more the usefulness of early physiological stimulation by suckling, experience in a group of puerperae showed that treatment apt to incr

Continuous dopaminergic stimulation in cranial dystonia.

Meige's disease is a distressing complaint, the treatment of which often poses a challenge to the neurologist. The patient described here had blepharospasm-oromandibular dystonia, which responded transiently to oral lisuride. On three occasions, drug holidays successfully restored efficacy but thereafter further trials proved fruitless. Continuous subcutaneous lisuride administration in 0.35 mg doses per day, by means of a portable infusion pump, led to sustained improvement for 7 months. No major side effects were observed. Our findings suggest that this treatment deserves further trials.
