Polymorphisms in the methylenetetrahydrofolate reductase and methionine synthase reductase genes and homocysteine levels in Brazilian children.

Hyperhomocysteinemia is a risk factor for thrombosis, and methylenetetrahydrofolate reductase (MTHFR) and methionine synthase reductase (MTRR) polymorphisms, folate, and B12 levels could contribute to plasma homocysteine (Hcy) variation. Although well established in adults, few studies have been performed in childhood. In this study, we investigated association of polymorphisms C677T and A1298C in the MTHFR gene and A66G in the MTRR gene with Hcy levels in children. These polymorphisms, as well as Hcy, folate, and vitamin B12 levels were investigated in 220 normal children with ages ranging fr

Analysis of spermatogenesis and fertility in adult mice with a hypomorphic mutation in the Mtrr gene.

Recent research has focussed on the significance of folate metabolism in male fertility. Knocking down the mouse gene Mtrr impedes the progression of folate and methionine metabolism and results in hyperhomocysteinaemia, dysregulation of DNA methylation and developmental phenotypes (e.g. neural tube, heart and placenta defects). The Mtrrgt mouse line is a model of transgenerational epigenetic inheritance (TEI), the hypothesised cause of which is the inheritance of a yet-to-be determined epigenetic factor via the germline. We investigated Mtrrgt/gt testes and sperm function compared with contro

Ferredoxin:NADP+ oxidoreductase in junction with CdSe/ZnS quantum dots: characteristics of an enzymatically active nanohybrid.

Ferredoxin:NADP(+) oxidoreductase (FNR) is a plant and cyanobacterial photosynthetic enzyme, also found in non-photosynthetic tissues, where it is involved in redox reactions of biosynthetic pathways. In vivo it transfers electrons to nicotinamide adenine dinucleotide phosphate (NADP(+)), forming its reduced version, NADPH, while in vitro it can also use NADPH to reduce several substrates, such as ferricyanide, various quinones and nitriles. As an oxidoreductase catalyzing reaction of a broad range of substrates, FNR may be used in biotechnological processes. Quantum dots are semiconductor nan

[The role of homocysteine and methylenetetrahydrofolate reductase, methionine synthase, methionine synthase reductase polymorphisms in the development of cardiovascular diseases and hypertension].

Cardiovascular diseases (CVDs) are the leading causes of death in the developed countries. Elevated homocysteine level is as an independent risk factor of CVDs. The C677T and A1298C variants of methylenetetrahydrofolate reductase gene (MTHFR) have been shown to influence folate and homocysteine metabolisms. However, the relationship between MTHFR polymorphisms and hyperhomocysteinemia has not been well established yet. The gene variants were also reported to be associated with CVDs. In addition, the C677T polymorphisms may play a role in the development of hypertension. Recent research evidenc

Methionine synthase reductase (MTRR) A66G polymorphism is not related to plasma homocysteine concentration and the risk for vascular disease.

Epidemiological evidence has revealed that hyperhomocysteinemia increases the risk for vascular disease. Methionine Synthase Reductase (MTRR) is one of several key enzymes in the homocysteine metabolic pathway and its mutant forms have been implicated in abnormal homocysteine accumulation. In this study, we determined total plasma homocysteine levels and MTRR A66G polymorphism in 114 patients with vascular disease: 58 patients with deep-vein thrombosis, 56 patients with arterial thrombosis, and 95 healthy subjects from the Sicilian population. Our data confirmed that, as already reported, mode

Polymorphic background of methionine synthase reductase modulates the phenotype of a disease-causing mutation.

Methionine synthase reductase (MTRR) is the locus of the cblE class of inborn errors of cobalamin metabolism that is characterized by megaloblastic anemia and homocystinuria. Two highly prevalent SNPs, c.66A>G (p.Ile22Met) and c.524C>T (p.Ser175Leu), are found in the MTRR gene. On the basis of the allele frequency of these amino acids and sequence comparison with members of the same family of proteins, the p.Ile22/p.Ser175 sequence is designated as wild type. While characterizing a pathogenic methionine synthase reductase (MSR) mutation, c.166G>A (p.Val56Met), we discovered an interaction betw

A three-component dicamba O-demethylase from Pseudomonas maltophilia, strain DI-6: purification and characterization.

Dicamba O-demethylase is a multicomponent enzyme that catalyzes the conversion of the herbicide 2-methoxy-3,6-dichlorobenzoic acid (dicamba) to 3,6-dichlorosalicylic acid (DCSA). The three components of the enzyme were purified and characterized. Oxygenase(DIC) is a homotrimer (alpha)3 with a subunit molecular mass of approximately 40 kDa. FerredoxinDIC and reductaseDIC are monomers with molecular weights of approximately 14 and 45 kDa, respectively. EPR spectroscopic analysis suggested the presence of a single [2Fe-2S](2+/1+) cluster in ferredoxinDIC and a single Rieske [2Fe-2S](2+; 1+) clust

Nature of oxygen inhibition of nitrogenase from Azotobacter vinelandii.

The reduction of nitrogen, acetylene, azide, and cyanide at various oxygen concentrations by nitrogenase from Azotobacter vinelandii was measured with a well-defined system. Oxygen inhibited the reduction of each substrate uncompetitively. The inhibition constants (K(i)) were 0.014, 0.023, 0.008, and 0.003 atm of oxygen for reduction of nitrogen, acetylene, azide, and cyanide, respectively. The system used included ATP-generating components, subcellular particles from A. vinelandii with high nitrogenase specific activity, and illuminated spinach chloroplasts plus carriers to supply electrons.

Drugs Metabolism-Related Genes Variants Impact on Anthracycline-Based Chemotherapy Induced Subclinical Cardiotoxicity in Breast Cancer Patients.

Breast cancer is the most common cancer in women worldwide. Anthracyclines (doxorubicin, epirubicin, daunorubicin, idarubicin) are among the most used drugs for the treatment of breast cancer. Unfortunately, anthracyclines cause cardiotoxicity, which is a limiting factor for its use, and the lifetime cumulative dose of anthracyclines is the major risk factor for cardiotoxicity. In our study, we focused on acute and subacute heart damage. One of the main factors is a genetic predisposition, which determines individual susceptibility to anthracycline cardiotoxicity. The main idea of this study w

Functional variant in methionine synthase reductase intron-1 is associated with pleiotropic congenital malformations.

Congenital malformations, such as neural tube defects (NTDs) and congenital heart disease (CHD), cause significant fetal mortality and childhood morbidity. NTDs are a common congenital anomaly, and are typically induced by higher maternal homocysteine (Hcy) levels and abnormal folate metabolism. The gene encoding methionine synthase reductase (MTRR) is essential for adequate remethylation of Hcy. Previous studies have focused on the coding region of genes involved in one-carbon metabolism, but recent research demonstrates that an allelic change in a non-coding region of MTRR (rs326119) increas

[Microbe-inspired system enzymology of vitamin B₁₂ metabolism].

Vitamin B₁₂ is produced only by prokaryotes and utilized by animals as an essential micronutrient. Genetic complementation analysis of cell lines from patients indicated that at least eight gene products are involved in intracellular B₁₂ metabolism and utilization. We have investigated bacterial adenosylcobalamin-dependent enzymes and elucidated their structure-based fine mechanisms. They tend to undergo mechanism-based inactivation during catalysis, because they use highly reactive radicals for catalyzing chemically difficult reactions. We have discovered molecular chaperone-like reactivating

Spectroscopic characterization and co-crystal structure of P450Blt reveal the molecular basis for biarylitide formation.

P450Blt is a cytochrome P450 enzyme (CYP) that forms crosslinked biarylitides by installing an intramolecular aromatic CN bond on a ribosomally synthesized pentapeptide. Closely related CYPs, such as the nitrating enzyme RufO, exhibit distinct catalytic selectivity on the same peptide scaffold. Although both P450Blt and RufO have been characterized, direct mechanistic comparison has been limited by the lack of spectroscopic studies of P450Blt and differences in their crystallization methods. Here, we performed spectroscopic and structural characterization of P450Blt to establish a framework fo

GE11/RGD Dual-Ligand Copper Nanoassemblies Synchronize Cuproptosis and Photothermal Therapy for Targeted Cervical Cancer Ablation.

Cervical cancer remains a major global health burden. Conventional surgery, radiotherapy, and chemotherapy are limited by insufficient tumor targeting, severe systemic toxicity, and drug resistance, highlighting an urgent need for safe, precise, and combinatorial therapeutic strategies. In this work, we fabricated one-step self-assembled GE11/RGD dual-ligand-modified copper nanoassemblies (GR@Cu NPs). These nanoparticles display densely modified targeting peptides on a copper-rich surface and enable efficient tumor accumulation with minimal off-target distribution. Under 980 nm near-infrared i

Mixotrophic denitrification with Arundo donax straw and elemental sulfur in constructed wetlands: enhanced nitrogen removal, carbon-sulfur interactions, and greenhouse gas mitigation.

Surface flow constructed wetlands (SFCWs) treating low carbon-to-nitrogen ratio wastewater are often limited by insufficient electron donor availability, leading to inefficient denitrification and greenhouse gas emissions. This study evaluated mixotrophic SFCWs integrated with Arundo donax straw and elemental sulfur (S0) over 160 days. The total nitrogen (TN) removal rate of 794.45 mg N/(m2·d) was achieved, exceeding those of the straw-only and S0-only SFCWs by 24.16% and 213.15%, respectively. Sequential operation further reduced effluent TN to 3.07 ± 0.16 mg/L (78.90% removal). The global wa

Why is the chloroplast redox regulatory machinery so complex?

Dithiol-disulfide exchange of cysteine residues has deep effects on protein conformation, hence on enzyme activity, which is the basis of redox regulation. The redox state of cells, including cellular compartments, is dynamically adjusted by reducing, i.e. NAD(P)H, and oxidizing signals, such as reactive oxygen species (ROS), generated during metabolic activity. Among ROS, H2O2 triggers the oxidation of cysteine thiols and the formation of disulfide bridges, thus having a relevant contribution to redox regulation. Disulfide reduction is catalyzed by thioredoxins (Trxs), small polypeptides with

AR-targeted therapies sensitize prostate cancer to cuproptosis by transcriptionally activating FDX1.

Androgen receptor (AR) signaling is central to prostate cancer progression, yet resistance to AR-targeted therapies remains a major clinical challenge. Understanding the molecular consequences of AR pathway inhibition is therefore essential for improving therapeutic outcomes. Here, we identify a previously unrecognized link between AR antagonism and cuproptosis, a copper-dependent form of regulated cell death. Using integrated genomic profiling, we find that AR-targeted agents transcriptionally activate the key cuproptosis regulator Ferredoxin-1 (FDX1), thereby rendering prostate cancer cells

Cuproptosis and copper metabolism regulate hepatocellular carcinoma progression and therapeutic response to sorafenib.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related death, and systemic therapy with sorafenib is limited by primary and acquired resistance. Copper is now recognized both as a growth-promoting cofactor in HCC and as the trigger of cuproptosis, a copper-dependent form of regulated cell death executed through ferredoxin-1 (FDX1)-mediated reduction of Cu2+, aggregation of lipoylated tricarboxylic acid (TCA) cycle proteins such as dihydrolipoamide S-acetyltransferase (DLAT), loss of Fe-S cluster proteins and proteotoxic mitochondrial stress.
