We report herein Rhodococcus equi infection in an 11-y-old, male llama with a history of diarrhea and endoparasitism. Postmortem examination revealed granulomatous and ulcerative enteritis, pyogranulomatous mesenteric lymphadenitis, fibrinosuppurative peritonitis, and granulomatous hepatitis. Intralesional macrophages were laden with gram-positive cocci. Bacteriology identified R. equi, and cultures tested positive for R. equi choE and vapA genes by PCR. This case expands the reported spectrum of lesions associated with R. equi infections in llamas from pyogranulomatous bronchopneumonia and pe

Congenital malformations, including cephalic anomalies such as diprosopia and dicephalia, represent rare but significant developmental abnormalities in humans and animals. While these conditions are primarily attributed to incomplete embryonic splitting, their genetic origins in domestic animals remain poorly understood. This study presents a detailed molecular and anatomical investigation of three cases of cephalic malformations in ruminants: two Holstein-Friesian calves and one Mediterranean Italian River Buffalo. Anatomical analyses classified the malformations as diprosopus in two cases an

A 14-month-old female Miniature Poodle dog with an enlarged clitoris and asymmetry in the placement of the teats was subjected to clinical, histopathological, and genetic studies. Macroscopically, the uterus and fallopian tubes appeared normal, while both ovaries were diffusely altered. At histology, the ovarian parenchyma was almost completely effaced by a diffuse hyperplasia of theca cells with atretic primary follicles. Chromosome analysis showed pure (non-mosaic) X monosomy (77,X). This finding was confirmed by the highly sensitive droplet digital PCR (ddPCR) approach. Despite the observed

The ability to absorb exogenous pigments (Pgm+) has, until now, been considered an established virulence factor of Yersinia pestis, the causative agent of plague. This property correlates with the sensitivity to the bacteriocin pesticin (Psts). Both functions are chromosomally encoded. In the present study, using Hfr donors and isogenic Pgm-Psts and Pgm-Pstr mutants, these functions were shown to be determined by discrete but closely linked genes. These markers designated pgm and psn, respectively, were preliminarily located within a linkage group including 11 loci. It was also found that pigm

Kallmann de Morsier Syndrome is defined by the association of an hypogonadism with an anosmia. The hypogonadism is due to a deficiency of GnRH (gonadotropin-releasing hormone). Olfactory bulbs and tracts are underdeveloped in the patients. Embryological studies have indicated that the migration of GnRH neurons and the axonal extension of olfactory neurons, which both originate in the olfactory epithelium during embryogenesis, were impaired in a fetus affected by X-linked Kallmann Syndrome. By a positional cloning strategy, we have isolated the KAL gene, responsible for the X-linked form of the

Sex in mammals is genetically determined and is defined at the cellular level by sex chromosome complement (XY males and XX females). The Y chromosome-linked gene sex-determining region Y (SRY) is believed to be the master initiator of male sex determination in almost all eutherian and metatherian mammals, functioning to upregulate expression of its direct target gene Sry-related HMG box-containing gene 9 (SOX9). Data suggest that SRY evolved from SOX3, although there is no direct functional evidence to support this hypothesis. Indeed, loss-of-function mutations in SOX3 do not affect sex deter

In seed plants (angiosperms and gymnosperms), sex is typically determined in the diploid sporophyte phase of the life cycle by sex chromosomes. This contrasts with bryophytes (mosses, liverworts, and hornworts), in which sex chromosomes act in the haploid gametophyte, whereas vascular non-seed plants (ferns and their allies) are not currently known to possess sex chromosomes. In seed plants, sex is usually controlled by one or several dominant factors in a non-recombining region. The causes of recombination suppression remain debated, but its consequences are increasingly well understood. Thes

Plague, caused by Yersinia pestis, has affected human societies for millennia, yet the timing, scale and biological shifts underlying its earliest spread remain unclear. Here, we screened 1,400 ancient human remains from eastern Eurasia (8,000-400 cal. BP) and recovered three prehistoric Y. pestis genomes, including one from East Asia. Integrating these data with 136 published ancient and modern Y. pestis genomes, we resolved a trans‑Eurasian polytomy dated to ~5.3 ka BP that links Scandinavia, Central Europe, and northern China. Low SNP distances and tip-dating indicate splits over century‑sc

BACKGROUND: In order to provide a reliable non-invasive method for fetal sex determination in a routine setting, we evaluated the possibility of identifying the fetal Y chromosome-specific sequence in maternal plasma using conventional PCR analysis.
METHODS: Fetal gender was determined in 31 pregnant women including one with a dizygotic twin pregnancy between 7 and 32 weeks of gestation using DNA extracted from 200 microl of each maternal plasma. The 198 bp SRY gene-specific sequence on Y chromosome and the 261 bp ATL1 gene-specific sequence on X chromosome were co-amplified in a multiplex ne

BACKGROUND: During gamete development and spermatogenesis, certain genes on the Y chromosome (Yq) in the Male-Specific Region (MSR) are responsible for human gametes formation. The long arm Yq is composed of both euchromatin and the genetically inactive heterochromatin regions. This region contains the Azoospermia factors AZFa, AZFb and AZFc. In the case of male infertility, microdeletions on the Yq chromosome appear to be structural chromosomal anomalies linked to sperm abnormality.
METHODS: The present study aimed to look at the incidence, of Asthenospermia (AS), Teratospermia (TS), Oligosp

BACKGROUND: Deoxyribonucleic acid from invasive, non-invasive and 9th week embryo can be a resource for the determination of fetal sex using highly sensitive and specific multiplex PCR.
METHODS: A total of 402 DNA samples were used to test the newly developed novel multiplex PCR including male specific (3 genes: SRY, DAZ2 and TSPY1) Y-biomarkers and internal control, ACTB. The study isolated cffDNA (Cell-free fetal DNA; n = 73) from mother's plasma, serum and urine, fetal DNA from 9th week embryo and cord blood, and fetal DNA from CD71+ve nucleated red blood cells (fNRBC; n = 73). STUDY QUESTION: What is the influence of copy number variation (CNV) in functional RNA binding motif protein Y-linked family 1 (RBMY1) on spermatogenic phenotypes?
SUMMARY ANSWER: The RBMY1 functional copy dosage is positively correlated with sperm motility, and dosage insufficiency is an independent risk factor for asthenozoospermia.
WHAT IS KNOWN ALREADY: RBMY1, a multi-copy gene expressed exclusively in the adult testis, is one of the most important candidates for male infertility in the azoospermia factor (AZF) region of the Y-chromosome. RBMY1 encodes an RNA-binding protein that serves

Male-to-female sex reversal in horses is a developmental disorder in which phenotypic females have a male genetic constitution. Male-to-female sex reversal is the second most common genetic sex abnormality, after X chromosome monosomy. All male-to-female sex reversal cases studied to date have been found to be infertile. Therefore, a screening test is particularly useful in laboratories doing DNA genotyping in horses. Our laboratory has tested > 209,000 horses for parentage using a panel of microsatellite markers and the sex marker gene amelogenin (AMEL). Suspect XY sex reversal cases are repo

Mutations in the X-linked gene encoding the transcriptional modulator methyl-CpG-binding protein 2 (MeCP2) impair postnatal development of the brain. Here we use neuronal-type specific gene deletion in mice to show that conditional Mecp2 deletion in GABAergic parvalbumin-expressing (PV) cells (PV-Mecp2(-/y)) does not cause most Rett-syndrome-like behaviours, but completely abolishes experience-dependent critical period plasticity of primary visual cortex (V1) that develops normal visual functions. However, selective loss of Mecp2 in GABAergic somatostatin-expressing cells or glutamatergic pyra

The molecular characteristics of the nude phenotype (alopecia and thymic aplasia) in humans and rodents are unknown. The nude locus encodes Whn, a transcription factor of the forkhead/winged-helix class. Expression of Whn in HeLa cells induced expression of human hair keratin genes Ha3-II and Hb5. Correspondingly, in nude mice, which are homozygous for a loss-of-function mutation of Whn, expression of mouse mHa3 and mHb5 hair keratin genes is severely reduced. Characterization of a previously identified nude allele, nu(Y), revealed a mis-sense mutation (R320C) in the DNA binding domain of Whn.

Potato (Solanum tuberosum) is a staple food crop that supports global food security, ranking as the world's third most important food crop after rice and wheat in terms of human consumption, and it is threatened by Potato virus Y (PVY), which causes severe yield losses. This study integrates bioinformatics analysis and experimental approaches to elucidate molecular defense mechanisms against PVY infection. Using transcriptomic data from PVY-infected potato plants, we constructed protein-protein interaction (PPI) networks and identified hub genes central to defense responses. The qPCR validatio

Microdeletions of the Y chromosome (YCMs), Klinefelter syndrome (47,XXY), and CFTR mutations are known genetic causes of severe male infertility, but the majority of cases remain idiopathic. Here, we describe a novel method using single molecule Molecular Inversion Probes (smMIPs), to screen infertile men for mutations and copy number variations affecting known disease genes. We designed a set of 4,525 smMIPs targeting the coding regions of causal (n = 6) and candidate (n = 101) male infertility genes. After extensive validation, we screened 1,112 idiopathic infertile men with non-obstructive

Among the relatively few flowering plants with strongly heteromorphic XY chromosomes are dioecious species of Hippophae (Elaeagnaceae). To understand the evolution of these sex chromosomes, we generated haplotype-resolved genomes for H. rhamnoides and H. tibetana, carried out ancestral karyotype reconstruction using Elaeagnus mollis (with bisexual flowers) as the outgroup, inferred the location of the sex-linked regions (SLRs), and used transcriptome data to test for dosage compensation. In H. rhamnoides, the X chromosome is 30% larger than the Y and in H. tibetana 15%, a difference primarily

Reference-quality genomes of both sexes are essential for studying sex determination and sex-chromosome evolution, as their gene contents and expression profiles differ. Here, we present independent chromosome-level genome assemblies for the female (XX) and male (XY) genomes of desert poplar (Populus euphratica), resolving a 22.7-Mb X and 24.8-Mb Y chromosome. We also identified a relatively complete 761-kb sex-linked region (SLR) in the peritelomeric region on chromosome 14 (Y). Within the SLR, recombination around the partial repeats for the feminizing factor ARR17 (ARABIDOPSIS RESPONSE REGU

In daily life, myopia is a frequent cause of reduced visual acuity (VA) due to missing or incomplete optical correction. While the genetic cause of high myopia itself is not well understood, a significant number of cases are secondary to hereditary malfunctions or degenerations of the retina. The mechanism by which this occurs remains yet unclear. Two female siblings, 4 y and 2 y, respectively, from a consanguineous Pakistani family were referred to our department for reduced VA and strabismus. Both girls were highly myopic and hence were further examined using standard clinical tests and elec
