An altered fecal microbial profiling in rosacea patients compared to matched controls.

BACKGROUND/PURPOSE: Rosacea has been linked to inflammatory bowel disease and small bowel bacterial overgrowth. We aimed to investigate the fecal microbial profiling and the potential gene functions between rosacea and non-rosacea subjects. METHODS: A case-control study. Fecal microbiome and predicted genetic function inferred from high-throughput 16S ribosomal RNA sequencing were analyzed between rosacea (n = 11) and age-, gender- and body mass index-matched non-rosacea subjects (n=110). The correlation between altered microbiome as well as lifestyle and diet were also investigated. RESULTS

Alterations in Skin Microbiota in Patients with Spinocerebellar Ataxia Type 3: A Pilot Study.

BACKGROUND: Spinocerebellar ataxia type 3 (SCA3) exhibits marked variability in age of onset and disease progression that cannot be fully explained by CAG repeat length, indicating the presence of additional modifiers. The skin, as a neuro-immune interface, has been linked to neurological diseases but remains understudied in SCA3. The study aimed to characterize the skin microbiota in SCA3 and assess its relationship with disease severity. METHODS: The study characterized the neck and armpit skin microbiota in 30 genetically confirmed SCA3 patients and 30 age and sex-matched healthy controls

Artificial extracellular electron transfer engineering in nonclassical electroactive microorganisms for microbial electrochemical technologies.

Microbial electrochemical technologies (METs) couple microbial metabolism with extracellular electronic interfaces, but their broader application is limited by the small repertoire of naturally electroactive microorganisms. Nonclassical-electroactive microorganisms (nonclassical-EAMs) offer diverse metabolic functions, mature genetic tools, and environmental robustness, yet they usually lack efficient extracellular electron transfer (EET) pathways. This review critically examines artificial EET engineering as a strategy to convert function oriented nonclassical-EAMs for bioresource and environ

Effects of composting on the fate of doxycycline, microbial community, and antibiotic resistance genes in swine manure and broiler manure.

Aerobic composting is an economical and effective technology that is widely used to treat animal manure. To study the fate of doxycycline (DOX), the microbial community, and antibiotic resistance genes (ARGs) during composting, aerobic composting of broiler manure and swine manure was carried out under natural environmental conditions. Aerobic composting effectively removed DOX (with a removal rate > 97%) and most ARGs from animal manure. The microbial diversity and the numbers of ARGs were higher in composted swine manure compared with composted broiler manure. The microbial community structu

Plasmid-linked resistance to inorganic salts in Staphylococcus aureus.

The penicillinase plasmids, a series of extrachromosomal resistance factors in Staphylococcus aureus, were found to carry determinants of resistance to a series of inorganic ions as well as resistance to penicillin and, in some cases, erythromycin. Most of the ions involved were inhibitory but not lethal to the bacteria; the resistance markers conferred an increase in resistance by comparison with susceptible organisms of between 3- and 100-fold, depending on the ion involved. Separate genetic loci for resistance to arsenate, arsenite, lead, cadmium, mercuric, and bismuth ions were demonstrate

Neonatal jaundice and the infant gut microbiome: an integrated shotgun metagenomics and bidirectional Mendelian randomization study in Xinjiang.

BACKGROUND: Neonatal jaundice is a common condition, yet inter-individual variation in its onset and severity cannot be fully explained by traditional clinical risk factors. Emerging evidence suggests that the infant gut microbiome may modulate bilirubin metabolism, but its compositional and functional signatures in jaundiced neonates remain incompletely defined. This study aimed to characterize the taxonomic and functional features of the gut microbiome in neonatal pathologic jaundice and to explore potential causal links using Mendelian randomization (MR). METHODS: We conducted a case-contr

Gnotobiotic and Conventional Mouse Systems to Support Microbiota Based Studies.

Animal models are essential to dissect host-microbiota interactions that impact health and the development of disease. In addition to providing pre-clinical models for the development of novel therapeutics and diagnostic biomarkers, mouse systems actively support microbiome studies by defining microbial contributions to normal development and homeostasis, and as well as their role in promoting diseases such as inflammatory auto-immune diseases, diabetes, metabolic syndromes, and susceptibilities to infectious agents. Mice provide a genetically tenable host that can be reared under gnotobiotic

Aggregatibacter actinomycetemcomitans: virulence of its leukotoxin and association with aggressive periodontitis.

Periodontitis is an infection-induced inflammatory disease that causes loss of the tooth supporting tissues. Much focus has been put on comparison of the microbial biofilm in the healthy periodontium with the diseased one. The information arising from such studies is limited due to difficulties to compare the microbial composition in these two completely different ecological niches. A few longitudinal studies have contributed with information that makes it possible to predict which individuals who might have an increased risk of developing aggressive forms of periodontitis, and the predictors

BBA52 facilitates Borrelia burgdorferi transmission from feeding ticks to murine hosts.

Borrelia burgdorferi, the pathogen of Lyme borreliosis, persists in nature through a tick-rodent transmission cycle. A selective assessment of the microbial transcriptome, limited to gene-encoding putative membrane proteins, reveals that bba52 transcription in vivo is strictly confined to the vector-specific portion of the microbial life cycle, with the highest levels of expression noted in feeding ticks and with swift down-regulation noted in mice. bba52 deletion did not affect murine disease as assessed by the genesis of arthritis and carditis or long-term persistence of pathogens in mice or

Gut Microbial Composition and Short-Chain Fatty Acid Metabolism in Cognitively Unimpaired Adults Stratified by Amyloid-β Status.

Short-chain fatty acids (SCFAs) produced by gut microbial fermentation influence host metabolism and neuroinflammatory processes implicated in Alzheimer's disease (AD). However, the relationship between fecal SCFAs, microbial taxa, and cerebral amyloid-β (Aβ) burden in cognitively unimpaired individuals remains unclear. Fecal SCFAs were quantified using GC-MS, and microbial species were profiled by shotgun metagenomics in 87 participants. Associations between SCFAs, demographics, APOE ε4 status, and Aβ burden were tested using nonparametric statistics and multivariable regression. Microbial-SC

Ketogenic diet synergistic reprogramming of both host and microbiome promotes tissue regeneration.

Nutrition influences host physiological processes, yet how diets reshape host physiology, microbial functions, or host-microbe interactions to promote regeneration remains poorly explored. Here, we show that a ketogenic diet (KD), enriched in fats and low in carbohydrates, reprograms both skin microbial and immune functions to promote tissue repair. KD enhances IL-17A activity in γδ T cells and mucosal-associated invariant T (MAIT) cells, accelerating tissue repair, while KD-induced skin lipidomic alterations enhance both the abundance and metabolic output of Staphylococcus epidermidis. Temporal gut microbiota dysbiosis links metabolic impairment, LDL desialylation, and accelerated atherosclerosis in LDLR-/- mice.

INTRODUCTION: This study used low-density lipoprotein receptor-deficient (LDLR-/-) mice fed a high-cholesterol diet (HCD) to establish an accelerated atherosclerosis model and to characterize the temporal dynamics of gut microbiota remodeling during disease progression. METHODS: Male C57BL/6J wild-type and LDLR-/- mice were fed either a normal diet (ND) or HCD for 12 weeks, with gut microbiota profiled at weeks 0, 8, and 12. HCD markedly accelerated atherogenesis in genetically susceptible mice, as evidenced by increased body weight, visceral adiposity, aggravated atherosclerotic lesions, and

Biosurfactants direct the evolutionary dynamics of surface-associated microbial systems.

Surface-associated microbial systems experience spatial constraints that can intensify stochastic lineage loss through genetic drift and alter how natural selection acts on competing lineages. A key feature of these systems is that steep resource gradients typically confine growth to a narrow band of cells located at the biomass periphery. This restricts the effective population size and constrains lineage interactions, thereby accelerating stochastic lineage exclusion. Here, we demonstrate that biosurfactants, which are secreted by many bacteria, can modulate the evolutionary dynamics of thes

From Circuits to Symphonies: A Systems-Engineering Blueprint for Multimicrobial Synthetic Biology.

Synthetic biology is expanding from single-species engineering toward the design of synthetic microbial consortia capable of distributed metabolism, coordinated behavior, and enhanced functional robustness. However, engineering frameworks derived from intracellular genetic circuits remain insufficient to capture the ecological, temporal, and spatial dynamics that govern multispecies systems. Here, a symphony-inspired systems-engineering framework is presented for multimicrobial synthetic biology, organizing consortium design across four application-dependent coordination layers: communication,

Single nucleotide variants drive evolutionary phage-host arms race in anaerobic carbon dioxide-converting microbiome.

Microbial bioconversions are shaped by environmental perturbations and the adaptation of resident microbiomes. Prokaryotes coexist with bacteriophages, yet their coevolutionary trajectories remain underexplored. Here, we investigate the effects of a cultivation vessel leak on an anaerobic consortium performing carbon dioxide reduction. Using time-series shotgun metagenomic sequencing, we reconstruct microbial and viral genomes to track community shifts. We further apply single-nucleotide variant profiling and CRISPR array analysis to monitor viral microdiversity and host defense mechanisms. Af

Microbial degradation of antibiotic: future possibility of mitigating antibiotic pollution.

Antibiotics are the major pharmaceutical wastes that are being exposed to the environment from the pharmaceutical industries and for the anthropogenic activities. The use of antibiotics for disease prevention and treatment in humans has been surpassed by the amount used in agriculture, particularly on livestock. It is stipulated that the overuse of antibiotics is the single largest reason behind the rise of bacterial anti-microbial resistance (AMR). The development of alternative therapy, like gene therapy, immunotherapy, use of natural products, and various nanoparticles, to control bacterial

Gut Microbiota and Metabolome Dynamics Along Gastric Cancer Progression: An Exploratory Multi-Omics Analysis.

BACKGROUND: Gastric cancer (GC) remains a major global health burden, particularly in East Asia, with complex etiologies involving Helicobacter pylori infection, diet, host genetics, and environmental exposures. GC development follows the Correa sequence (CS), a multistep cascade from gastritis to atrophy, erosion, and carcinoma. Although gut microbiota (GM) dysbiosis and metabolic reprogramming have each been implicated in GC, their integrated dynamics across CS remain incompletely defined. METHODS: We recruited participants across five groups: normal controls (G1), gastritis (G2), atrophy (

Recent Perspectives on Gene-Microbe Interactions Determining Predisposition to Otitis Media.

A comprehensive understanding about the pathogenesis of otitis media (OM), one of the most common pediatric diseases, has the potential to alleviate a substantial disease burden across the globe. Advancements in genetic and bioinformatic detection methods, as well as a growing interest in the microbiome, has enhanced the capability of researchers to investigate the interplay between host genes, host microbiome, invading bacteria, and resulting OM susceptibility. Early studies deciphering the role of genetics in OM susceptibility assessed the heritability of the phenotype in twin and triplet st
