Panic disorder (PD) is an anxiety disorder characterized by recurrent and unexpected panic attacks, subsequent worry and phobic avoidance. Although a number of association and linkage studies have been conducted, no gene has been identified as a susceptibility locus. We previously conducted a genome-wide association analysis of PD in 200 Japanese patients and the same number of controls, using a 500 K single nucleotide polymorphisms (SNPs) chip. In this study, we report a replication analysis of PD using the DigTag2 assay. The second stage sample consisted of 558 Japanese patients and 566 cont

To identify bipolar disorder (BD) genetic susceptibility factors, we conducted two genome-wide association (GWA) studies: one involving a sample of individuals of European ancestry (EA; n=1001 cases; n=1033 controls), and one involving a sample of individuals of African ancestry (AA; n=345 cases; n=670 controls). For the EA sample, single-nucleotide polymorphisms (SNPs) with the strongest statistical evidence for association included rs5907577 in an intergenic region at Xq27.1 (P=1.6 x 10(-6)) and rs10193871 in NAP5 at 2q21.2 (P=9.8 x 10(-6)). For the AA sample, SNPs with the strongest statist

Downbeat nystagmus (DBN) is a frequent form of acquired persisting central fixation nystagmus, often associated with other cerebellar ocular signs, such as saccadic smooth pursuit or gaze-holding deficits. Despite its distinct clinical features, the underlying etiology of DBN often remains unclear. Therefore, a genome-wide association study (GWAS) was conducted in 106 patients and 2609 healthy controls of European ancestry to identify genetic variants associated with DBN. A genome-wide significant association (p < 5 × 10-8) with DBN was found for a variation on chromosome 13 located within the…

The role of heritable genetic variation in hematologic traits in adults is well established, yet the genetic architecture of neonatal blood cell traits is unknown. Leveraging flow cytometry profiling in cord blood samples from 382 Hispanic newborns, we conducted genome-wide association studies (GWASs) of 24 blood cell phenotypes to assess the impact of genetic and birth-related characteristics. We identified six genome-wide significant loci, including a signal at chromosome 2p11.2 spanning CD8A and CD8B that was associated with CD4+CD8+ double-positive T cells (rs35505884-C: p = 6.83 × 10-26).

Langya chicken is a Chinese indigenous chicken breed with high genetic diversity. To systematically analyse the genetic basis of body size traits, eight traits (including BW, comb shape, and body size) of 2 952 Langya hens were measured at 130 days of age and at first egg of age. A total of 9 708 856 high-quality single-nucleotide polymorphisms (SNPs) were obtained through whole-genome resequencing and used for subsequent genetic parameter estimation and genome-wide association study (GWAS). The results of genetic parameter analysis revealed significant differences in the SNP heritability of d…

BACKGROUND: Colostrum yield (CY, L) and concentration of immunoglobulin G (IgG, g/L) are important phenotypes to monitor in dairy farms because of their association with the risk of failure of the passive transfer of immunity in the newborn calf. This can occur when the CY of the parturient cow is insufficient or when the IgG concentration is low. Given that both of these traits are heritable, the present study aimed to investigate their genetic determinism by identifying significant genomic regions. A genome-wide association study coupled with an exploratory functional enrichment analysis was…

UNLABELLED: <p>Introduction: Data from family and twin studies as well as prior genome-wide association meta-analyses suggest that hidradenitis suppurativa (HS) has a hereditary component. This GWAS identifies genetic variants associated with HS.
METHODS: Individuals with a diagnosis of HS (defined as at least one instance of ICD9 705.83 or ICD10 L73.2) were identified within the Million Veteran Program. Multi-population and population-specific (African, European, and Hispanic ancestries) case-control genome-wide association studies (GWAS) were performed. Lead single-nucleotide polymorphisms

BACKGROUND: Individuals with autosomal dominant Alzheimer's disease (ADAD) arising from mutations in PSEN1, PSEN2, or APP exhibit variability in clinical presentation. Genetic studies of ADAD have shaped our understanding of the disease, and the discovery of genetic modifiers can inform therapeutic interventions and improve patient outcomes. We aimed to discover new genetic modifiers in individuals with mutations in the three ADAD genes.
METHODS: In this genome-wide association study, we analysed data from participants in three study cohorts (the Knight Alzheimer Disease Research Center [Knig…

BACKGROUND: Dry eye disease (DED) affects up to 25% of the adult population, with chronic symptoms of pain and dryness often negatively impacting quality of life. Twin studies indicate a substantial heritability. However, the genetic architecture of DED is largely unknown.
METHODS: We devised a case-control algorithm using a combination of ICD-9/10 codes and prescription records and validated its performance through manual chart review, identifying 132,637 DED cases and 352,201 controls across four ancestry groups in the Million Veteran Program biobank. We then performed multi-ancestry genome

Background/Objectives: The anti-Müllerian hormone (AMH) is a key biomarker of the ovarian reserve, correlating with ovarian follicle count, fertility outcomes, and menopause timing. Understanding its genetic determinants has broad implications for female reproductive health. However, prior genome-wide association studies (GWASs) have focused exclusively on women of European ancestry, limiting insights into diverse populations. Methods: We conducted a GWAS to identify genetic loci associated with circulating AMH levels in a sample of 1185 Samoan women from two independently recruited samples. U

OBJECTIVES: Periodontitis is linked with many health conditions, but its genetic basis is not yet understood. This genome-wide association study (GWAS) aimed to investigate the genetic variants associated with periodontitis.
MATERIALS AND METHODS: This study utilised UK Biobank participants of European descent. Individuals were categorised as "having periodontitis" if they self-reported having 'painful gums', 'bleeding gums' or 'loose teeth' (n = 68,482), or as "controls" for those without these symptoms (n = 307,342). We conducted GWAS of this binary periodontitis phenotype using logistic re…

Lewy body dementia (LBD) is the second most prevalent dementia, however most genetic risk remains uncharacterized. Here, we performed the largest LBD genome-wide association study (GWAS) meta-analysis including 4252 LBD cases and 189,290 controls. We confirmed four previously known risk loci APOE, GBA, BIN1, and SNCA-AS1, and highlighted a novel locus SYT16. We further integrated LBD GWAS with multi-omics datasets, and identified 85 LBD risk genes that were enriched in eight functional clusters including 51 statistically significant pathways (e.g., SYT16 was enriched in the phospholipid bindin…

The pineal gland is the primary source of melatonin synthesis and secretion. Reduced pineal gland volume results in lower melatonin levels and has been linked to sleep impairment, as well as many (age-related) metabolic and neurodegenerative disorders. However, its genetic architecture remains unknown. Using brain imaging and genotype data from the UK Biobank (N=38,254) and Rhineland Study (N=5,286), we conducted the first genome-wide association study (GWAS) of pineal gland volume, demonstrating 19% heritability and identifying 34 genome-wide significant loci, with highly concordant results b…

Artificial insemination plays a crucial role in pig production, particularly in enhancing the genetic potential of elite boars. To accelerate genetic progress for semen traits in pigs, it is vital to understand and identify the underlying genetic markers associated with desirable traits. Herein, we genotyped 1238 Landrace boars with GeneSeek Porcine SNP50 K Bead chip and conducted genome-wide association studies to identify genetic regions and candidate genes associated with 12 semen traits. Our study identified 38 SNPs associated with the analyzed 12 semen traits. Furthermore, we identified s

BACKGROUND: Productive life (PL) of a cow is the time the cow remains in the milking herd from first calving to exit from the herd due to culling or death and is an important economic trait in U.S. Holstein cattle. The large samples of Holstein genomic evaluation data that have become available recently provided unprecedented statistical power to identify genetic factors affecting PL in Holstein cows using the approach of genome-wide association study (GWAS).
METHODS: The GWAS analysis used 1,103,641 Holstein cows with phenotypic observations on PL and genotypes of 75,282 single nucleotide po…
