Effects of subchronic parathion administration on sodium salicylate excretion kinetics in female rats.

Organophosphorus (OP) pesticides are considered to be environmental contaminants, and chronic exposure to low levels through the diet may affect drug action. To study this possible interaction, ethyl parathion was administered by intubation to female rats for 35 consecutive days at a dose of 0.05 or 0.2 mg/kg of body weight per day. At 7, 21 and 35 days after parathion was initiated, rats were administered a single dose of 20 mg/kg sodium salicylate intraperitoneally. Total salicylates, salicylic acid (SA), salicyluric acid (SU) and gentisic acid (GA) were determined in urine. At 7 days, parat

Direct measurement of salicylphenolic glucuronide in human urine.

Indirect measurement of salicylphenolic glucuronide (SPG) has suggested that the formation of this metabolite from therapeutic doses of salicyclic acid (SA) is capacity-limited in humans. A direct high performance liquid chromatographic (HPLC) assay for SPG in human urine is described. SPG was prepared by a published method and purified by HPLC. On treatment with beta-glucuronidase, SPG yielded the expected amount of SA. Spectroscopic data, melting point, and optical rotation of the glucuronide and/or its triacetyl dimethyl ester derivative were consistent with the proposed structure. SPG was

Studies on simultaneous determination of acetaminophen, salicylic acid and salicyluric acid in biological fluids by high performance liquid chromatography.

A rapid and sensitive high performance liquid chromatographic method was developed for separation and quantitation of acetaminophen, salicylic acid and salicyluric acid in human plasma and urine. The method involved ethyl acetate extraction of the three drugs from plasma or urine followed by evaporation of the organic solvent phase and dissolution of the residue in 100 microliters methanol. A 20 microliters aliquot was analysed on a reversed phase column using an isocratic system of 60 ml/l acetonitrile in 4 mmol/l phosphate buffer, pH 5.7 as a mobile phase and a variable wave length UV detect

Initial renal responses of nonhuman primate to immersion and intravascular volume expansion.

Experiments were performed in anesthetized Macaca fascicularis monkeys to determine if the initial renal responses of these animals to head-out vertical water immersion and isoncotic, isotonic volume expansion are similar, especially with regard to the onset of any changes in solute or water excretion. Significant increases in urine flow, sodium excretion, and osmolar clearance occurred after 10 min of immersion but not until 30 min after volume expansion. Potassium excretion increased during immersion but decreased after volume expansion. Mean arterial blood pressure increased after 30 min of

Oxidative Stress, Metabolic Impairment and Neuroinflammation are Associated With Target Organ Damage in SHRSP.

Stroke-prone spontaneously hypertensive rats (SHRSP) are widely used as a model to study cerebral small vessel disease (CSVD) and its association with chronic hypertension. This study investigated the relationship between metabolic, cardiovascular, and neuronal comorbidities in 32-week-old SHRSP rats versus Wistar-Kyoto (WKY) controls, with a focus on oxidative stress, inflammation, and metabolic alterations. Despite hypertension and cardiac and renal hypertrophy, no significant cerebral vascular changes or microbleeds and no cerebral edema were detected in SHRSP. NMR-based urinary metabolomic

Vancomycin-Induced Modulation of Gram-Positive Gut Bacteria and Metabolites Remediates Insulin Resistance in iNOS Knockout Mice.

The role of oxidative and nitrosative stress has been implied in both physiology and pathophysiology of metabolic disorders. Inducible nitric oxide synthase (iNOS) has emerged as a crucial regulator of host metabolism and gut microbiota activity. The present study examines the role of the gut microbiome in determining host metabolic functions in the absence of iNOS. Insulin-resistant and dyslipidemic iNOS-/- mice displayed reduced microbial diversity, with a higher relative abundance of Allobaculum and Bifidobacterium, gram-positive bacteria, and altered serum metabolites along with metabolic

First-Void Urine Microbiome in Women with Chlamydia trachomatis Infection.

BACKGROUND: Chlamydia trachomatis (CT) is the agent of the most common bacterial sexually transmitted infection worldwide. Until now, little information is available about the microbial composition of urine samples during CT urethritis. Therefore, in this study, we characterized the microbiome and metabolome profiles of first-void urines in a cohort of women with CT urethral infection attending an STI clinic. METHODS: Based on CT positivity by nucleic acid amplification techniques on urine samples, the enrolled women were divided into two groups, i.e., "CT-negative" (n = 21) and "CT-positive"

Beverages containing Lactobacillus paracasei LC-37 improved functional dyspepsia through regulation of the intestinal microbiota and their metabolites.

Functional dyspepsia (FD) is a common disease of the digestive system and probiotics have been reported to be effective in the treatment of functional gastrointestinal diseases. The aim of this study was to determine the effect of the beverage containing Lactobacillus paracasei LC-37 (LC-37) and its ability to relieve symptoms of FD by a randomized clinical trial. Due to the mechanistic complexity underlying FD, intestinal microbiota and stool metabolomes were also evaluated. The results showed that FD was relieved in participants after treatment with the beverage containing LC-37 for 14 and 2

Metabolic Response in Rabbit Urine to Occurrence and Relief of Unilateral Ureteral Obstruction.

Ureteral obstruction will lead clinically to hydronephrosis, which may further develop into partial or complete loss of kidney function and even cause permanent histological damage. However, there is little knowledge of metabolic responses during the obstructed process and its recoverability. In this study, a complete unilateral ureteral obstruction (CUUO) model was established in the rabbit, and 1H NMR-based metabolomic analysis of urine was used to reveal the metabolic perturbations in rabbits caused by CUUO and the metabolic recovery after the CUUO was relieved. Univariate and multivariate

Effects of diltiazem on renal blood flow and renal function during angiotensin II infusion in anesthetized dogs.

Effects of diltiazem on renal blood flow and renal function were studied under angiotensin II-loaded condition in anesthetized dogs. Intravenous infusion of angiotensin II at a rate of 0.25 microgram/kg/min produced an increase in blood pressure by 30 to 40 mmHg and a decrease in renal blood flow by 30 to 40%, resulting in a 100% increase in renal vascular resistance. Under this condition, diltiazem exhibited a dose related increase in renal blood flow not only by intrarenal administration (1-30 micrograms/kg) but also by intravenous administration (10-200 micrograms/kg). The effect was greatl
