The effect of histamine on the field stimulated rat vas deferens preparation was studied and compared to mouse vas deferens. Histamine produced a concentration dependent inhibition of rat vas deferens (ID50 14 microM) similar to that seen with mouse vas deferens (ID50 2.4 microM). Unlike mouse however, where only H2 receptors are observed, the histamine inhibition of rat vas deferens was attenuated by both H1, (diphenhydramine) and H2 (cimetidine) antagonists. It is concluded that rat vas deferens contains both H1 and H2 receptor types.

Intravenous histamine causes high amplitude repetitive phasic contractions of the in vivo cat pylorus but has little effect on the antrum and duodenum. The genesis of this phasic response was studied using a pinned perfused catheter with openings at the pylorus, antrum, and duodenum. 2-Pyridylethylamine, an H1 agonist, produced phasic contractions similar to histamine whereas dimaprit, an H2 agonist, did not. To elucidate further the role of histamine in pituitary regulation, TRH and L-DOPA stimulation tests were performed with and without diphenhydramine, cimetidine, or betazole pre-treatment. Betazole blunted the GH response to L-DOPA and slightly enhanced the T3 response to TRH without altering the TSH or PRL increments. Neither diphenhydramine nor cimetidine had any acute effect on the hormonal responses examined. Histamine apears to play only a limited role in these aspects of human pituitary regulation.

Because of certain side effects of cimetidine therapy which may be hormonally mediated (e.g. gynecomastia), there has been recent interest in the possible endocrine effects of this H2 histamine receptor-blocking agent used in the treatment of peptic ulcer disease. In mepyramine treated, ovalbumin sensitized anaesthetized guinea pigs protracted anaphylactic shock was produced by i.p. injection of antigen, and serum creatine kinase (CK) activities were determined 4, 6, or 17 hrs thereafter. Methylxanthines and catecholamines both inhibit antigenically induced histamine release from human leukocytes. They act synergistically to inhibit the reaction, but must be present when antigen is added; preincubation is not effective. Since both increase cellular levels of cyclic 3 ', 5'-adenosine monophosphate it is postulated that this compound plays a role in the regulation of allergic histamine release.
