Apicomplexan parasites are responsible for a number of important human pathologies. Obviously, as Eukaryotes they share a number of cellular features and pathways with their respective host cells. One of them is autophagy, a process involved in the degradation of the cell's own components. These intracellular parasites nonetheless seem to present a number of original features compared to their very evolutionarily distant host cells. In mammals and other metazoans, autophagy has been identified as an important contributor to the defence against microbial pathogens. Thus, host autophagy also lik

Pathogens interact with their hosts at different spatial and temporal scales. Studying these interactions therefore requires a wide range of imaging tools and approaches that bridge physics and biology, as shown by this Minireview focusing on recent studies of the causative agents of malaria, toxoplasmosis, and sleeping sickness.

A breakdown of the human parasite fauna shows approximately 270 species, with almost equal numbers of nematodes, flukes and protozoa and smaller numbers of arthropods and tapeworms. More than 70% are 'adventitious' species for which man is an incidental host. Only 16% are 'core' species, dependent on man for their survival. For 26% man is the usual source of human infection, and for 51% the source is other mammals. A sequence is illustrated showing how parasites may arrive by the host-transfer or co-evolutionary pathways. Adventitious parasites have a narrower geographical range than core spec

Variation in infection dynamics across host species can profoundly influence parasite epidemiology, ecology, and evolution. However, because experiments involving multiple hosts and multiple parasite species that capture the full infection dynamics within hosts are rare, the underlying mechanisms of host heterogeneity effects across different parasite species remain largely unknown. We dissected the specific roles of host and pathogen effects in shaping within-host infection dynamics using a model system comprising three coexisting rodent species from Israel's northwestern Negev Desert and the

Schistosome parasites have co-evolved an intricate relationship with their human and snail hosts as well as a novel interplay between the adult male and female parasites. We review the role of the TGF-beta signaling pathway in parasite development, host-parasite interactions and male-female interactions. The data to date support multiple roles for the TGF-beta signaling pathway throughout schistosome development, in particular, in the tegument which is at the interface with the host and between the male and female schistosome, development of vitelline cells in female worms whose genes and deve…

Understanding the dynamics of host-parasite interactions is essential for uncovering the ecological and evolutionary processes shaping natural populations. In this study, we investigated the spatial and temporal patterns of prevalence of the cestode Schistocephalus solidus in 4 Alaskan populations of threespine stickleback (Gasterosteus aculeatus) over 11 years (1996 to 2006). Additionally, we assessed host tolerance through key indicators such as the Parasite Index and the presence of fibrotic scar tissue, a specialized form of resistance in this system. We observed fibrosis formation in both…

Toxoplasma gondii is an obligate intracellular parasite belonging to the phylum Apicomplexa that infects all warm-blooded animals, including humans. T. gondii can replicate in every nucleated host cell by orchestrating metabolic interactions to derive crucial nutrients. In this review, we summarize the current status of known metabolic interactions of T. gondii with its host cell and discuss open questions and promising experimental approaches that will allow further dissection of the host-parasite interface and discovery of ways to efficiently target both tachyzoite and bradyzoite forms of T.

Host specificity is a critical feature of the survival and proliferation of parasites. In the context of interactions with the host, numerous mechanisms have been identified, particularly in parasitic helminths, that enable manipulation of the host immune system to enhance their own survival. The evolutionary history of these interactions often results in hosts becoming disease-tolerant or asymptomatic, even when burdened with a high number of worms, until a disruption in the host's immune system can trigger a disease state. However, the molecular mechanisms underlying host specificity and the

BACKGROUND: The enteric myxozoan parasite Enteromyxum leei is an important problem in gilthead seabream aquaculture invading the intestinal epithelium and leading to chronic intestinal inflammation, poor food conversion rates, cachexia, and mortalities, with no treatments available, resulting in significant economic losses. It is known that myxozoan infections are affected by factors such as temperature, duration of exposure, stocking densities, and seasonality. Gut microbiota has key effects on host health, including disease resistance and immune system training and development, tightly inter

Host manipulation is a common parasite strategy to alter host behavior in a manner to enhance parasite fitness usually by increasing the parasite's transmission to the next host. In nature, hosts often harbor multiple parasites with agreeing or conflicting interests over host manipulation. Natural selection might drive such parasites to cooperation, compromise, or sabotage. Sabotage would occur if one parasite suppresses the manipulation of another. Experimental studies on the effect of multi-parasite interactions on host manipulation are scarce, clear experimental evidence for sabotage is elu

Examples of parasite-related effects on intermediate crustacean hosts are numerous but their ecological consequences on their vertebrate hosts are scarce. Here, we address the role of macroparasite infections on the trophic niche structure of definitive hosts and its potential physiological consequences using wild fish populations infected with an acantochephalan parasite Pomphorhynchus sp., a trophically transmitted intestinal worm. Infected and uninfected fish were sampled from six populations on the Marne River, France and the prevalence of intestinal parasites in the host populations range

The ability to distinguish these host-parasite interactions was then utilized to dissect the events that lead to the production of interleukin-12 p40 (IL-12p40), which is required for resistance to T. gondii. In vivo, the use of invasion-competent or invasion-inhibited (phagocytosed) parasites with IL-12p40 …

Giardia intestinalis is a unicellular protozoan parasite that infects the small intestines of humans and animals. Giardiasis, the disease caused by the parasite, occurs globally across socioeconomic boundaries but is mainly endemic in developing countries and particularly within young children, where pronounced effects manifests in a failure to thrive condition. The molecular pathogenesis of Giardia has been studied using in vitro models of human and rat intestinal epithelial cells (IECs) and parasites from the two major human genotypes or assemblages (A and B). High-quality, genome sequencing

Noninvasive approaches are increasingly reshaping parasite and disease surveillance by reducing stress and harm to hosts while expanding opportunities for ecological and epidemiological research. In this opinion article, we discuss these emerging approaches, which rely on molecular, citizen-science, and computational methods, for monitoring parasites, vectors, and hosts. These tools can improve spatial and temporal coverage, support the surveillance of rare or threatened hosts and parasites, and contribute to the understanding of transmission pathways and disease dynamics. However, their relia

Protozoan pathogens such as Plasmodium spp., Leishmania spp., Toxoplasma gondii, and Trypanosoma spp. are often associated with high-mortality, acute and chronic diseases of global health concern. For transmission and immune evasion, protozoans have evolved diverse strategies to interact with a range of host tissue environments. These interactions are linked to disease pathology, yet our understanding of the association between parasite colonization and host homeostatic disruption is limited. Recently developed techniques for cellular barcoding have the potential to uncover the biology regulat

Combining the biogeography and phylogenetic patterns of parasite-host associations allows a better understanding of the history of parasite–host interactions, which can be achieved via biogeographic regionalization incorporating phylogenetic information. Recently, the concepts of evoregions (regions where a majority of species evolved from one or several ancestors inhabiting these regions) and evolutionary transition zones (regions of high phylogenetic turnover) have been proposed, coupled with a classification approach for these concepts. We applied this approach to 206 flea species and 265 h

Within a single organism, numerous parasites often compete for space and resources. This competition, together with a parasite's ability to locate and successfully establish in a host, can contribute to the distribution and prevalence of parasites. Coinfection with trematodes in snail intermediate hosts is rarely observed in nature, partly due to varying competitive abilities among parasite taxa. Using a freshwater snail host (Biomphalaria glabrata), we studied the ability of a competitively dominant trematode, Echinostoma caproni, to establish and reproduce in a host previously infected with

Predation on parasites is a common interaction with multiple, concurrent outcomes. Free-living stages of parasites can comprise a large portion of some predators' diets and may be important resources for population growth. Predation can also reduce the density of infectious agents in an ecosystem, with resultant decreases in infection rates. While predator-parasite interactions likely vary with parasite transmission strategy, few studies have examined how variation in transmission mode influences contact rates with predators and the associated changes in consumption risk. To understand how tra
