Synthesis of Cr(IV)-GSH, its identification and its free hydroxyl radical generation: a model compound for Cr(VI) carcinogenicity.

Current models of Cr(VI) carcinogenesis suggest an important role for Cr(IV) as an intermediate, toxic, carcinogenic species, but direct chemical evidence has been lacking. This is because Cr(IV) is a highly reactive oxidation state of Cr and few Cr(IV)-based compounds are known that can be used as a model compound containing a biological ligand. This study reports the isolation of such a stable Cr(IV) complex. The Cr(IV)-GSH complex has been synthesized through the reaction of Cr(VI) with GSH. Its electron paramagnetic resonance (EPR) spectrum exhibits g = 1.9629 and a peak-to-peak line width

Sulfur dioxide and glutathione promote acetaldehyde accumulation under aerobic conditions in a model wine system: Impacts on hydroxyl radical formation.

The addition of antioxidants is essential in red wine aging, as it influences chemical oxidation processes. This study investigated the effects of free sulfur dioxide (SO2) and glutathione (GSH), both individually and combined, on oxidative changes in a model wine system. Dissolved oxygen (DO), acetaldehyde, hydroxyl radicals (HO·), ferrous ions (Fe2+), SO2, and GSH concentrations were monitored. Results showed that 30 mg/L SO2 treatments effectively inhibited the Fenton reaction and accumulation of HO· and acetaldehyde, while higher SO2 treatments (≥60 mg/L SO2) promoted the production of ace

Epitope mapping of SARS-CoV-2 RBDs by hydroxyl radical protein footprinting reveals the importance of including negative antibody controls.

Understanding protein-protein interactions is crucial for drug design and investigating biological processes. Various techniques, such as CryoEM, X-ray spectroscopy, linear epitope mapping, and mass spectrometry-based methods, can be employed to map binding regions on proteins. Commonly used mass spectrometry-based techniques are cross-linking and hydrogen‑deuterium exchange (HDX). Another approach, hydroxyl radical protein footprinting (HRPF), identifies binding residues on proteins but faces challenges due to high initial costs and complex setups. This study introduces a generally applicable

Ratiometric electrochemical determination of hydroxyl radical based on graphite paper modified with metal-organic frameworks and impregnated with salicylic acid.

Hydroxyl radical (•OH) detection is pivotal in medicine, biochemistry and environmental chemistry. Yet, electrochemical method-specific detection is challenging because of hydroxyl radicals' high reactivity and short half-life. In this study, we aimed to modify the electrode surface with a specific recognition probe for •OH. To achieve this, we conducted a one-step hydrothermal process to fabricate a CoZnMOF bimetallic organic framework directly onto conductive graphite paper (Gp). Subsequently, we introduced salicylic acid (SA) and methylene blue (MB), which easily penetrated the pores of CoZ

Electrocatalytic Deep Dehalogenation and Mineralization of Florfenicol: Synergy of Atomic Hydrogen Reduction and Hydroxyl Radical Oxidation over Bifunctional Cathode Catalyst.

It is difficult to achieve deep dehalogenation or mineralization for halogenated antibiotics using traditional reduction or oxidation processes, posing the risk of microbial activity inhibition and bacterial resistance. Herein, an efficient electrocatalytic process coupling atomic hydrogen (H*) reduction with hydroxyl radical (•OH) oxidation on a bifunctional cathode catalyst is developed for the deep dehalogenation and mineralization of florfenicol (FLO). Atomically dispersed NiFe bimetallic catalyst on nitrogen-doped carbon as a bifunctional cathode catalyst can simultaneously generate H* an

Redox Behavior of Secondary Solid Iron Species and the Corresponding Effects on Hydroxyl Radical Generation during the Pyrite Oxidation Process.

During the pyrite oxidation process, aqueous ferrous/ferric ions (Fe2+/Fe3+), as well as surface-adsorbed Fe2+/Fe3+, have been widely recognized to dominate hydroxyl radical (•OH) generation, while this study reveals that the secondary solid iron species also play non-negligible roles. Based on the different forms and the presence of sites, the secondary solid iron species were classified as Fecoat (iron-containing coating on the pyrite surface) and Fedep (ex situ-deposited iron (oxyhydr)oxide that is not in contact with pyrite). Instead of participating in building a stubborn passivation laye

The formation of reactive species having hydroxyl radical-like reactivity from UV photolysis of N-nitrosodimethylamine (NDMA): kinetics and mechanism.

This study focuses on the detailed mechanism by which N-nitrosodimethylamine (NDMA) is photolyzed to form oxidized products, i.e., NO(2)(-) and NO(3)(-), and reveals a key reactive species produced during the photolysis of NDMA. Under acidic conditions, NO(2)(-) formed from the photodecomposition of NDMA was more prevalent than NO(3)(-). In this result, key species for the formation of NO(2)(-) are presumably N(2)O(3) and N(2)O(4) as termination products as well as NO and O(2) as reactants. Conversely, under alkaline conditions, NO(3)(-) was more prevalent than NO(2)(-). For this result, a key

Hydroxyl radical mediates cisplatin-induced apoptosis in human hair follicle dermal papilla cells and keratinocytes through Bcl-2-dependent mechanism.

Induction of massive apoptosis of hair follicle cells by chemotherapy has been implicated in the pathogenesis of chemotherapy-induced alopecia (CIA), but the underlying mechanisms of regulation are not well understood. The present study investigated the apoptotic effect of cisplatin in human hair follicle dermal papilla cells and HaCaT keratinocytes, and determined the identity and role of specific reactive oxygen species (ROS) involved in the process. Treatment of the cells with cisplatin induced ROS generation and a parallel increase in caspase activation and apoptotic cell death. Inhibition

In vivo monitoring of hydroxyl radical generation caused by x-ray irradiation of rats using the spin trapping/EPR technique.

Measurement of hydroxyl radical (*OH) in living animals irradiated with ionizing radiation should be required to clarify the mechanisms of radiation injury and the in vivo assessment of radiation protectors, because generation of *OH is believed to be one of the major triggers of radiation injury. In this study, *OH generation was monitored by spin trapping the secondary methyl radical formed by the reaction of *OH with dimethyl sulfoxide (DMSO). Rats were injected intraperitoneally with a DMSO solution of alpha-phenyl-N-tert-butylnitrone (PBN). X-irradiation of the rats remarkedly increased t

Ultraviolet-B-induced oxidative DNA base damage in primary normal human epidermal keratinocytes and inhibition by a hydroxyl radical scavenger.

To evaluate the effects of ultraviolet-induced environmental trauma on human skin cells, primary normal human epidermal keratinocytes were exposed to ultraviolet-B radiation (290-320 nm). We found that relatively low doses of ultraviolet-B (62.5-500 mJ per cm2) caused dose-dependent increases in 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxo-dG), a biomarker of oxidative DNA damage. Unirradiated normal human epidermal keratinocytes contained 1.49 (+/- 0.11) 8-oxo-dG per 10(6) 2'-deoxyguanosine (dG) residues in cellular DNA, which increased linearly to as high as 6.24 (+/- 0.85) 8-oxo-dG per 10(6)

Gastroprotective effect of Neem (Azadirachta indica) bark extract: possible involvement of H(+)-K(+)-ATPase inhibition and scavenging of hydroxyl radical.

The antisecretory and antiulcer effects of aqueous extract of Neem (Azadirachta indica) bark have been studied along with its mechanism of action, standardisation and safety evaluation. The extract can dose dependently inhibit pylorus-ligation and drug (mercaptomethylimidazole)-induced acid secretion with ED(50) value of 2.7 and 2 mg Kg(-1) b.w. respectively. It is highly potent in dose-dependently blocking gastric ulcer induced by restraint-cold stress and indomethacin with ED(50) value of 1.5 and 1.25 mg Kg(-1) b.w. respectively. When compared, bark extract is equipotent to ranitidine but mo

alpha-Phenyl-tert-butyl-nitrone (PBN) attenuates hydroxyl radical production during ischemia-reperfusion injury of rat brain: an EPR study.

Alpha-phenyl-tert-butyl-nitrone (PBN) a spin adduct forming agent is believed to have a protective action in ischemia-reperfusion injury of brain by forming adducts of oxygen free radicals including .OH radical. Electron paramagnetic resonance (EPR) has been used to both detect and monitor the time course of oxygen free radical formation in the in vivo rat cerebral cortex. Cortical cups were placed over both cerebral hemispheres of methoxyflurane anesthetized rats prepared for four vessel occlusion-evoked cerebral ischemia. Prior to the onset of sample collection, both cups were perfused with

Intracranial microdialysis of salicylic acid to detect hydroxyl radical generation through dopamine autooxidation in the caudate nucleus: effects of MPP+.

Ringer's solution containing salicylic acid (5 nmol/microliters/min) was infused directly through an intracranial microdialysis probe to detect the generation of hydroxyl radicals (.OH) reflected by the formation of dihydroxybenzoic acids (DHBA) in the caudate nucleus of anesthetized rats. Brain dialysate was assayed for dopamine, 2,3-, and 2,5-DHBA by a high-pressure liquid chromatography-electrochemical (HPLC-EC) procedure. 1-Methyl-4-phenylpyridinium ions (MPP+, 0 to 150 nmol) increased dose-dependently the release of dopamine and the formation of DHBA. A positive linear correlation between

Detection of salicylate and its hydroxylated adducts 2,3- and 2,5-dihydroxybenzoic acids as possible indices for in vivo hydroxyl radical formation in combination with catechol- and indoleamines and their metabolites in cerebrospinal fluid and brain tissue.

It has been suggested that salicylate (SA) hydroxylation can be used to detect hydroxyl radical formation in vivo. Here we describe a rapid and sensitive HPLC method using ultraviolet absorbance (UV) and electrochemical detection (EC) to detect SA (UV), its hydroxylated adducts 2,3- and 2,5-dihydroxybenzoic acids (DHBA) and catechol in combination with catechol- and indoleamines and related metabolites (EC) in one isocratic run. These compounds were measured in acidified cerebrospinal fluid (CSF) and perchlorate extracts of striatal tissues of untreated and SA-loaded rats (300 mg/kg SA, i.p.).

Atmospheric Oxidation Mechanism of Benzimidazole initiated by Hydroxyl Radicals (•OH): Nitrosamine Formation from •OH-Addition Pathways.

Atmospheric oxidation of nitrogen-containing volatile chemical products (N-VCPs) significantly affects air quality, with the formation of carcinogenic nitrosamines being particularly concerning. Benzimidazoles (BZIs) are high-production chemicals and a type of N-VCP. However, their atmospheric oxidation mechanisms and subsequent impacts remain unclear. Here, using the simplest benzimidazole (BZI) as a representative compound, we employed theoretical calculations and toxicological modeling to investigate the •OH-initiated reaction mechanism of BZI and assess the toxicity of their oxidation prod

Hydroxyl radicals produced from oxidation of ferrous sulfides promote mobilization of mercuric sulfide in soil-water system.

Mercuric sulfide nanoparticles (HgS-NPs) are recognized as a significant source of bioavailable mercury in paddy fields. The factors influencing the mobilization and bioavailability of HgS-NPs formed in flooded or drained paddy field-like systems are complicated and remain unexplored to date. Here, we show that ferrous sulfide (FeS) as an important mineral substance plays a crucial role in the dissolution and transformation of HgS-NPs in overlying water or during the drainage stage, as well as their bioavailability toward rice. Specifically, we found that oxidation of FeS significantly enhance

Contaminant transformation during sediment oxygenation: Temporal variation of oxidation mechanisms mediated by hydroxyl radicals and aerobic microbes.

Sediment oxidation by oxygen is ubiquitous, whereas the mechanisms of concurrent contaminant oxidation, particularly the temporal variation of chemical and biological oxidation, remain inadequately understood. This study investigated the oxidation of two contaminants (phenol and trichloroethylene) with different responses during the oxygenation of four natural sediments with different redox properties. Results showed that contaminant oxidation was initially dominated by hydroxyl radicals (•OH) (first stage), stabilized for different time for different sediments (second stage), and was re-start

Role of the Hydroxyl Radical-Generating System in the Estimation of the Antioxidant Activity of Plant Extracts by Electron Paramagnetic Resonance (EPR).

The scavenging activity of hydroxyl radicals, produced by the Fenton reaction, is commonly used to quantify the antioxidant capacity of plant extracts. In this study, three Fenton systems (Fe/phosphate buffer, Fe/quinolinic acid and Fe/phosphate buffer/quinolinic acid) and the thermal degradation of peroxydisulfate were used to produce hydroxyl radicals; the hydroxyl radical scavenging activity of plant extracts (ginger, blueberry juices and green tea infusion) and chemical compounds (EGCG and GA) was estimated by spin trapping with DMPO (5,5-dimethyl-1-pyrroline N-oxide) and EPR (Electron Par

Oxidative degradation of commingled trichloroethylene and 1,4-dioxane by hydroxyl radicals produced upon oxygenation of a reduced clay mineral.

Improper disposal of chlorinated solvents such as trichloroethylene (TCE) and its stabilizer 1,4-dioxane has resulted in extensive contamination in soils and groundwater. Oxidative degradation of these contaminants by strong oxidants has been proposed recently as a remediation strategy, but specific mechanisms and degradation efficiencies are still poorly understood, especially in commingled systems. In this study, a reduced iron-bearing clay (RIC), nontronite (rNAu-2), was oxygenated to produce hydroxyl radicals (•OH) for degradation of TCE and 1,4-dioxane under circumneutral and dark conditi

Gas-phase reaction of methyl isothiocyanate and methyl isocyanate with hydroxyl radicals under static relative rate conditions.

Gaseous methyl isothiocyanate (MITC), the principal breakdown product of the soil fumigant metam sodium (sodium N-methyldithiocarbamate), is an inhalation exposure concern to persons living near treated areas. Inhalation exposure also involves gaseous methyl isocyanate (MIC), a highly reactive and toxic transformation product of MITC. In this work, gas-phase hydroxyl (OH) radical reaction rate constants of MITC and MIC have been determined using a static relative rate technique under controlled laboratory conditions. The rate constants obtained are 15.36 × 10(-12) cm(3) molecule(-1) s(-1) for
