Broadening SARS-CoV-2 Immunity by Combining ORFV and Protein-Based Vaccines.

BACKGROUND: Emerging immune-evasive viral variants threaten the efficacy of current vaccines, underscoring the need for strategies that elicit broad and durable protection. Heterologous prime-boost regimens combining distinct vaccine platforms can enhance humoral and cellular immunity through complementary mechanisms. METHODS: Using an intramuscular immunization scheme aligned with clinical vaccination practice, CD-1 mice received homologous or heterologous prime-boost regimens combining a replication-deficient Orf virus (Parapoxvirus orf, ORFV)-based spike vaccine (ORFV-S) with the licensed

SphB1 export system for antigen presentation to the respiratory tract by a live pertussis vaccine.

BPZE1 is a live attenuated vaccine currently in advanced clinical development for nasal immunization against pertussis. The ability of this vaccine to induce mucosal and systemic, humoral and cellular immunity make it an attractive mucosal delivery system for heterologous antigens to induce immunity especially in the respiratory tract. Here, we have developed a heterologous antigen expression system based on the pertactin-deficient BPZE1 derivative IB-V002 by using the SphB1 secretion system to present the SARS-CoV-2 nucleocapsid protein (N) as a model antigen to the respiratory mucosa. SphB1

Influenza vaccination post-COVID-19 expands vaccine-specific effector CD4 T-cells and Tregs under positive influence of host trained innate immunity.

SARS-CoV-2 immunity and innate immune training may influence influenza vaccine immunogenicity. We investigated this in India. Adult volunteers with hybrid SARS-CoV-2 immunity were administered FluarixTM Tetra (GlaxoSmithKlein) 2022/2023 NH Vaccine in 2022. Significant induction of hemagglutinin inhibition-specific antibodies and polyfunctional central memory CD4+ T-cells (TCM) were observed 1-week post-vaccination with variable induction of CD8+T-cell and innate effectors. Vaccination also expanded Flu-specific regulatory T-cells (Treg), which negatively correlated with CD4 responses, highligh

BCG vaccination induces innate immune memory in γδ T cells in humans.

Bacillus Calmette-Guérin vaccine is well known for inducing trained immunity in myeloid and natural killer cells, which can explain its cross-protective effect against heterologous infections. Although displaying functional characteristics of both adaptive and innate immunity, γδ T-cell memory has been only addressed in a pathogen-specific context. In this study, we aimed to determine whether human γδ T cells can mount trained immunity and therefore contribute to the cross-protective effect of the Bacillus Calmette-Guérin vaccine. We investigated in vivo induction of innate memory in γδ T cell

Both innate and adaptive immunity mediate protective immunity against hepatitis C virus infection in chimpanzees.

UNLABELLED: Understanding the immunological correlates associated with protective immunity following hepatitis C virus (HCV) reexposure is a prerequisite for the design of effective HCV vaccines and immunotherapeutics. In this study we performed a comprehensive analysis of innate and adaptive immunity following HCV reexposure of two chimpanzees that had previously recovered from HCV-JFH1 infection. One of the chimpanzees, CH10274, became protected from active viremia by repeated challenges with homologous HCV-JFH1 and developed neutralizing antibodies, but was later infected with high-level vi

Optimizing COVID-19 vaccination strategies for high-risk populations: potential and challenges of combining heterologous boosting with respiratory mucosal delivery.

Despite the decline of the global peak of COVID-19, SARS-CoV-2 continues to circulate, evolve, and undergo immune escape, posing persistent threats, particularly to older adults, individuals with comorbidities, and immunocompromised populations. Although existing vaccination strategies have substantially reduced the risks of severe disease, hospitalization, and death, their ability to block infection and transmission remains limited, and protection wanes over time. Current heterologous booster strategies still rely largely on combinations of different parenteral vaccine platforms. Although the

Learning from the omicron BA.4/5 dominance: Strategies for future vaccination preparedness.

OBJECTIVES: To investigate the protective effectiveness of various COVID-19 booster strategies against the Omicron BA.4/5 variant and examine the impact of vaccination-infection sequence on subsequent immunity, focusing on individuals with waned immunity (>6 months since last vaccination/infection). METHODS: We conducted a territory-wide observational study of 1,737,475 adults in Hong Kong during the BA.4/5-dominant period (November 2022-January 2023). The Andersen-Gill model was employed to assess infection risks across different vaccination and infection history cohorts, comparing homologou

Induction of specific CD8+ memory T cells and long lasting protection following immunization with Salmonella typhimurium expressing a lymphocytic choriomeningitis MHC class I-restricted epitope.

Numerous studies have shown the potential of Salmonella typhimurium as a vector for delivery of heterologous proteins for vaccination against other pathogens. Earlier studies showed that the inefficient elicitation of MHC class I-restricted responses could limit the use of S. typhimurium as a heterologous antigen delivery vector for vaccination. We recently developed an approach to overcome this limitation by using a bacterial-encoded specialized protein secretion system, termed type III, to deliver proteins into the class I antigen presenting pathways. Thus, peptides of interest fused to prot

Gamma-irradiated scrub typhus immunogens: development and duration of immunity.

The development and duration of immunity to lethal scrub typhus infection was studied in BALB/c mice vaccinated with gamma-irradiated Rickettsia tsutsugamushi, strain Karp. One intraperitoneal injection containing approximately 10(8) 50% mouse lethal doses (MLD(50)) of irradiated organisms elicited an immune response protective against challenge with 10(5) MLD(50) of viable Karp. The same mass of immunogen given in three injections at 5-day intervals increased homologous (Karp strain) protection 25-fold and heterologous (Kato strain) protection 60-fold. Further temporal expansion of the immuni

New insights into an old vaccine: potency and breadth of neutralizing antibodies elicited by the yellow fever vaccine 17D are boosted by heterologous Orthoflavivirus infection.

The yellow fever vaccine 17D has controlled yellow fever for almost 90 years and is considered highly successful. However, with recent outbreaks in South America and Africa, yellow fever continues to represent a significant threat to public and global health. Despite its longstanding success, few studies have characterized the capacity of 17D-immune sera to neutralize wild-type (WT) yellow fever viruses (YFVs). Using a panel of WT YFVs that represent the known YFV genotypes and a unique cohort of non-endemic vaccinees with diverse Orthoflavivirus infection histories, we extensively characteriz

Immunomodulation by nematodes: a review.

There is current evidence that infections with Trichinella spiralis, Ascaris suum, Nippostrongylus brasiliensis, Nematospiroides dubius (syn. Heligmosomoides polygyrus) and diverse filariae affect the immune responsiveness of their hosts. T. spiralis, or its extracts, can depress or enhance the heterologous humoral or cell-mediated immunities, and affect macrophage activity or the response to other invaders. These effects are induced by products of the migratory and early muscle larvae and appear to obey more than one single-mechanism. A suum acute infections or extracts depress responses invo

Immunization against geographical isolates of Trichinella spiralis in mice.

Partially purified antigen preparations from six isolates of Trichinella spiralis were used to immunize mice. Immunogenicity of the antigens was assessed in terms of antibody and lymphocyte responses and ability to stimulate protective immunity against challenge. Isolate antigens showed considerable cross-reactivity, and all elicited protective responses. Two major patterns of immunizing ability could be distinguished: (a) isolates that immunized well against heterologous challenge and elicited good immunity in London isolate-immunized mice (C-76 and Laso), and (b) isolates that immunized poor

Cross-infection enhancement among African flaviviruses by immune mouse ascitic fluids.

Cross-infection enhancement of seven African flaviviruses by subneutralising concentrations of antibody in immune ascitic fluids was investigated in P388D1 cell culture. Infection by all the seven flaviviruses tested was enhanced by homologous and at least one of six heterologous immune mouse ascitic fluids (IMAF) tested. Enhancement ratios and enhancing antibody titres were higher in homologous than in heterologous enhancement. Zika, Wesselsbron, Uganda S and West Nile viruses were enhanced in culture by all the IMAF tested. Enhancement of Dakar bat and Yellow fever viruses was produced by fi

"Trained immunity" from Mycobacterium spp. exposure or BCG vaccination and COVID-19 outcomes.

Protective variables for Coronavirus Disease 2019 (COVID-19) are unknown. "Trained immunity" of the populace as a result of Bacille Calmette-Guérin (BCG) vaccination policy implementation and coverage had been suggested to be one of the factors responsible for the differential impact of COVID-19 on different countries. Several trials are underway to evaluate the potential protective role of BCG vaccination in COVID-19. However, the lack of clarity on the use of appropriate controls concerning the measures of "trained immunity" or the heterologous cell-mediated immunity conferred by BCG vaccina

The impact of interferon-γ pathway on trained immunity induction by vaccination with Bacille Calmette-Guérin.

Bacillus Calmette-Guérin (BCG) has multiple heterologous off-target effects which extend beyond tuberculosis (TB) prophylaxis, which include protection against other non-tuberculous infections, autoimmune diseases, and tumor development. These heterologous effects are at least partially mediated by induction of trained immunity. In this study, we aimed to investigate the impact of IFNγ production capacity on induction of trained immunity in human volunteers vaccinated with BCG. We evaluated inflammation and immune activation-specific cytokine responses (IFNγ, TNF, IL-1, and IL-6) in PBMCs isol

"Bystander" recruitment of systemic memory T cells delays the immune response to respiratory virus infection.

The concept of heterologous T cell immunity postulates that nonspecific memory T cells recruited and reactivated in the context of an unrelated virus infection may contribute to protective antiviral immunity. Pulmonary infection with respiratory syncytial virus (RSV) of mice immune to lymphocytic choriomeningitis virus (LCMV) leads to substantial recruitment of systemic LCMV-specific memory CD8 T cells to the lung. Using a sensitive TCR-transgenic model, we show that such "bystander" recruitment to the RSV-infected lung could induce a single round of cell division among LCMV-specific T cells.

Long-Term Durability and Dynamics of Anti-S-RBD IgG Response in Healthcare Workers: A Comparative Analysis of Homologous and Heterologous SARS-CoV-2 Vaccination Schedules in a 1-Year Serial Cross-Sectional Study.

BACKGROUND The waning of post-vaccination immunity necessitates effective booster strategies, especially for high-risk healthcare workers (HCWs). This study aimed to compare the 1-year antibody response dynamics of homologous and heterologous SARS-CoV-2 vaccination schedules in Turkish HCWs. MATERIAL AND METHODS This prospective, serial cross-sectional study included 307 HCWs grouped by vaccination history reflecting the national vaccination program: homologous inactivated (SSS, 3 doses of CoronaVac), homologous mRNA (BBB, 3 doses of mRNA-BNT162b2), and heterologous mRNA-boosted (SSB, SSBB, SS

Hepatitis C cross-genotype immunity and implications for vaccine development.

While about a quarter of individuals clear their primary hepatitis C (HCV) infections spontaneously, clearance (spontaneous or treatment-induced) does not confer sterilizing immunity against a future infection. Since successful treatment does not prevent future infections either, an effective vaccine is highly desirable in preventing HCV (re)infection. However, development of an effective vaccine has been complicated by the diversity of HCV genotypes, and complexities in HCV immunological responses. Smaller studies on humans and chimpanzees reported seemingly opposing results regarding cross-n

Seasonal variation in BCG-induced trained immunity.

The Bacille Calmette-Guerin (BCG) vaccine is a well-established inducer of innate immune memory (also termed trained immunity), causing increased cytokine production upon heterologous secondary stimulation. Innate immune responses are known to be influenced by season, but whether seasons impact induction of trained immunity is not known. To explore the influence of season on innate immune memory induced by the BCG vaccine, we vaccinated healthy volunteers with BCG either during winter or spring. Three months later, we measured the ex vivo cytokine responses against heterologous stimuli, analyz
