The acoustic startle response in DBA/2 and C57BL/6 mice: relationship to auditory neuronal response properties and hearing impairment.

The acoustic startle response (ASR) was elicited with tone pips in sensorineural hearing-impaired DBA/2 mice and in non-impaired C57BL/6 mice. The influence of stimulus frequency and intensity on ASR amplitude varied as a function of both strain and age. Previous neurophysiological findings indicate that ASR amplitude is correlated with the proportion of neurons in the cochlear nucleus and inferior colliculus that respond to the ASR stimulus frequencies and to the excitability of neurons in these structures. ASR amplitude is not highly correlated with threshold sensitivity. These observations

Acoustic stimulation alters deoxyglucose uptake in the mouse cochlea and inferior colliculus.

Deoxyglucose uptake and activities of hexokinase and glucose-6-phosphatase in auditory structures (organ of Corti, stria vascularis and spiral ligament, modiolar section of VIIIth nerve, inferior colliculus) and non-auditory tissues (heart, kidney, liver) of the mouse were analyzed. [3H]Deoxyglucose was given as a pulse into the tail vein and uptake was quantitated by microdissection of tissues and scintillation counting. Radioactivity in cochlear tissues was maximal after 45-60 min and declined with a half-life of 30-60 min. Deoxyglucose 6-phosphate represented ca. 60% of total radioactivity

Echolalic speech in childhood autism. Consideration of possible underlying loci of brain damage.

The speech of echolalic autistic children is (1) specifically lacking in appropriate use of expressive-intonational features, but (2) the echolalic child's clear articulation of words and phrases indicates that discrimination of phonemic features is intact. The impairment in aphasic disorders is just the reverse. Failure to attend to auditory stimuli and the characteristic language disorder are among the most consistent findings in autistic children; they could be related. Discrimination of differential stress emphasis is the way the normal young child extracts major morphemic word stems and s

Far-field acoustic response: origins in the cat.

Short-latency evoked potentials recorded from the vertex of adult cats in response to click stimulation (the far-field acoustic response) were analyzed in a series of lesion experiments to determine the origins of each component. The resultant data indicate that the primary generator of potential is the acoustic nerve; of potential 2, the cochlear nucleus; of potential 3, neurons of the superior olivary complex activated by projections crossing the midline; of potential 4, neurons of the ventral nucleus of the lateral lemniscus and preolivary region activated equally by crossed and uncrossed p

Reduced brain volume and white matter alterations in Shank3-deficient rats.

Mutations and deletions in the SHANK3 gene cause the major neurodevelopmental features of Phelan-McDermid syndrome (PMS), which is characterized by intellectual disability, autism spectrum disorder, and sensory hyporeactivity. SHANK3 encodes a key structural component of excitatory synapses important for synaptogenesis. Clinical assessments and limited brain imaging studies of patients with PMS have uncovered regional volume reductions and white matter thinning. While these impairments have been replicated ex vivo in pups of a rat model, brain structure has not been assessed in rats in vivo or

Brainstem neuropathology in a mouse model of Niemann-Pick disease type C.

Niemann-Pick disease type C (NPC) is a neurovisceral lipid storage disorder characterized by progressive and widespread neurodegeneration. Although some characteristic symptoms of NPC result from brainstem dysfunction, little information is available about which brainstem structures are affected. In this study, the brainstems of mutant BALB/c NPC1-/- mice with a retroposon insertion in the NPC1 gene were examined for neuropathological changes. In the midbrain, the integrated optic density (IOD) and cell count density of tyrosine-hydroxylase (TH) immunostained neurons were decreased in the subs

Infantile hypermethioninemia and hyperhomocysteinemia due to high methionine intake: a diagnostic trap.

Studies were carried out to identify the cause of combined severe hypermethioninemia and moderate hyperhomocysteinemia in a cluster of 10 infants ascertained between 1999 and early 2001. Although several were thought initially to have cystathionine beta-synthase (CBS) deficiency and treated accordingly, CBS deficiency and other known genetic causes of hypermethioninemia were ruled out by assay of CBS activity in fibroblasts of four patients and by assays of plasma cystathionine and S-adenosylmethionine. Retrospective data on dietary methionine intakes and plasma concentrations of methionine an

The human brain at stage 17, including the appearance of the future olfactory bulb and the first amygdaloid nuclei.

The growth of the cerebral hemispheres rostrally and caudodorsally brings about a deepening of the interlongitudinal fissure, in which blood capillaries for the future choroid plexus develop. Accumulation of mesenchyme basally presages the formation of the nasal septum. The olfactory bulb and tubercle become outlined. The three main areas of the telencephalon, future archi-, paleo-, and neocortex, can be recognized. The amygdaloid body, which is related to the medial ventricular eminence, contains either one or two nuclei. Nerve fibres from the olfactory tubercle arrive and pass through the am

Molecular remodeling of the central auditory pathway in tinnitus revealed by RNA sequencing.

BACKGROUND: Tinnitus is a complex neurological condition affecting 10-15% of adults worldwide, characterized by phantom auditory perception without external sound sources. While traditional investigations have focused on discrete auditory structures, emerging evidence suggests tinnitus involves broader alterations across central auditory regions. OBJECTIVE: This study employed transcriptomic analysis to investigate molecular mechanisms underlying salicylate-induced tinnitus across multiple brain regions simultaneously. METHODS: Male C57BL/6 N mice received daily intraperitoneal injections of

Distinct forms of synaptic plasticity during ascending vs descending control of medial olivocochlear efferent neurons.

Activity in each brain region is shaped by the convergence of ascending and descending axonal pathways, and the balance and characteristics of these determine the neural output. The medial olivocochlear (MOC) efferent system is part of a reflex arc that critically controls auditory sensitivity. Multiple central pathways contact MOC neurons, raising the question of how a reflex arc could be engaged by diverse inputs. We examined functional properties of synapses onto brainstem MOC neurons from ascending (ventral cochlear nucleus, VCN) and descending (inferior colliculus, IC) sources in mice usi

Changes in microRNA Expression in the Cochlear Nucleus and Inferior Colliculus after Acute Noise-Induced Hearing Loss.

Noise-induced hearing loss (NIHL) can lead to secondary changes that induce neural plasticity in the central auditory pathway. These changes include decreases in the number of synapses, the degeneration of auditory nerve fibers, and reorganization of the cochlear nucleus (CN) and inferior colliculus (IC) in the brain. This study investigated the role of microRNAs (miRNAs) in the neural plasticity of the central auditory pathway after acute NIHL. Male Sprague-Dawley rats were exposed to white band noise at 115 dB for 2 h, and the auditory brainstem response (ABR) and morphology of the organ of

Transcriptomic profile analysis of brain inferior colliculus following acute hydrogen sulfide exposure.

Hydrogen sulfide (H2S) is a gaseous molecule found naturally in the environment, and as an industrial byproduct, and is known to cause acute death and induces long-term neurological disorders following acute high dose exposures. Currently, there is no drug approved for treatment of acute H2S-induced neurotoxicity and/or neurological sequelae. Lack of a deep understanding of pathogenesis of H2S-induced neurotoxicity has delayed the development of appropriate therapeutic drugs that target H2S-induced neuropathology. RNA sequencing analysis was performed to elucidate the cellular and molecular me

Scopolamine attenuates auditory cortex response.

CONCLUSION: Scopolamine, a tropane alkaloid drug that mainly acts as an antagonist of muscarinic acetylcholine receptors, was found to reduce the local field potentials (LFP) of auditory cortex (AC) evoked by tone and gap-offsets whose effects may compensate the cortical hyperexcitability related to tinnitus. OBJECTIVE: To study the effects of scopolamine on the AC and the inferior colliculus (IC) of awake rats in order to understand scopolamine's effect on tinnitus and gap detection. METHOD: Silent gaps (duration varied from 2-100 ms) embedded in otherwise continuous noise were used to elic

Thin-film micro-electrode stimulation of the cochlea in rats exposed to aminoglycoside induced hearing loss.

The multi-channel cochlear implant (CI) provides sound and speech perception to thousands of individuals who would otherwise be deaf. Broad activation of auditory nerve fibres when using a CI results in poor frequency discrimination. The CI also provides users with poor amplitude perception due to elicitation of a narrow dynamic range. Provision of more discrete frequency perception and a greater control over amplitude may allow users to better distinguish speech in noise and to segregate sound sources. In this research, thin-film (TF) high density micro-electrode arrays and conventional plati

Social regulation of serotonin in the auditory midbrain.

The neuromodulator serotonin regulates auditory processing and can increase within minutes in response to stimuli like broadband noise as well as nonauditory stressors. Little is known about the serotonergic response in the auditory system to more natural stimuli such as social interactions. Using carbon-fiber voltammetry, we measured extracellular serotonin in the auditory midbrain of resident male mice during encounters with a male intruder. Serotonin increased in the inferior colliculus (IC) over the course of a 15 minute interaction, but not when mice were separated with a perforated barri

Immunocytochemical and neurochemical evidence for age-related loss of GABA in the inferior colliculus: implications for neural presbycusis.

The present study describes substantial, selective, age-related loss of the putative inhibitory neurotransmitter GABA in the central nucleus of the inferior colliculus (CIC) of rat based on immunocytochemical and neurochemical data. For immunocytochemistry, neurons in the CIC were immunolabeled using an antibody against a GABA conjugate in young adult (2- to 7-month-old) and aged (18- to 29-month-old) Fischer-344 rats. Computer-assisted morphometry was then used to generate maps of GABA-immunoreactive neurons in the CIC. The number of GABA-positive neurons was reduced 36% in the ventrolateral

[Neurophysiological characteristics of early auditory waves of the brain stem and their clinical utilization].

The auditory brainstem electric response yields information on both the neurological and the audiological status of infants and adults. These early waves have particular properties as: non-habituation, absence of latency modifications between sleep, awake state, or under high doses of barbiturates. We have developed a procedure for extracting each type of information by measuring the latencies of wave I (auditory nerve) and wave V (inferior colliculus), then the speed of conduction between these two waves.

Diagnostic utility of tectal plate measures in clinical variants of progressive supranuclear palsy.

BACKGROUND: The length of the tectal plate, which sits above the midbrain, is reduced in both the Richardson's syndrome (RS) and parkinsonism (P) variants of progressive supranuclear palsy (PSP). However, it is unclear whether tectal measurements could be useful biomarkers in other PSP variants or predict PSP pathology. METHODS: The Neurodegenerative Research Group, Mayo Clinic, enrolled 115 PSP (including seven variants), 19 corticobasal syndrome (CBS), 21 Parkinson's disease (PD), and 50 controls. Forty-seven PSP patients had PSP at autopsy, and 15 had a non-PSP pathology. Tectal plate leng

Reduction in Constitutively Activated Auditory Brainstem Microglia in Aging and Alzheimer's Disease.

BACKGROUND: Alzheimer's disease (AD) pathology is considered to begin in the brainstem, and cerebral microglia are known to play a critical role in AD pathogenesis, yet little is known about brainstem microglia in AD. Translocator protein (TSPO) PET, sensitive to activated microglia, shows high signal in dorsal brainstem in humans, but the precise location and clinical correlates of this signal are unknown. OBJECTIVE: To define age and AD associations of brainstem TSPO PET signal in humans. METHODS: We applied new probabilistic maps of brainstem nuclei to quantify PET-measured TSPO expressio

[The central neural mechanism of the gastric motility in the dog's medulla oblongata on the vagal inhibitory and excitatory reflexes (author's transl)].

The influences of the vagal inhibitory and excitatory reflexes on the gastric motor centers in the dog's medulla oblongata were investigated. Dogs were anaesthetized with Nembutal, and supplemented Gallamine at need. The brain stem was transected on the level of inferior colliculi of midbrain, the spinal cord transected on the level of between C1 and C2. Bilateral splanchnic nerves were also severed. Electrical activities from the inhibitory and excitatory areas in medulla oblongata were recorded by using a concentric circle electrode which was inserted into medulla oblongata from the dorsal s
