Effect of a spatial repellent on malaria incidence in an area of western Kenya characterised by high malaria transmission, insecticide resistance, and universal coverage of insecticide treated nets (part of the AEGIS Consortium): a cluster-randomised, controlled trial.

BACKGROUND: Spatial repellent products are used for prevention of insect bites, and a body of evidence exists on spatial repellent entomological efficacy. A new option for vector control, spatial repellent products are designed to release active ingredient into the air for disruption of human-vector contact thereby reducing human exposure to mosquito-borne pathogens. Clinical trials have shown spatial repellent epidemiological efficacy against Aedes-borne viruses but inconclusive outcomes against malaria. We aimed to show and quantify the protective efficacy of spatial repellents in reducing m

The V1016G mutation of the voltage-gated sodium channel (VGSC) gene contributes to the insecticide resistance of Aedes aegypti from Makassar, Indonesia.

Aedes aegypti represents one of the main vectors of at least five relevant arthropod-borne viral infections in humans (i.e., Rift Valley fever, Dengue fever, Zika, chikungunya, and yellow fever) worldwide. Ae. aegypti control strategies are mostly based on using chemical insecticides (i.e., organophosphates, pyrethroids, carbamates, and organochlorines) and reducing larval sources. Furthermore, monitoring the growth activity and mapping the geographical distribution of insecticide resistance are mandatory, as recommended by the WHO. Accordingly, we conducted a study on the possible mechanism b…

Structural modification of fipronil-5-amino and insecticidal activity evaluation of its derivatives against resistant Chilo suppressalis.

BACKGROUND: The resistant rice stem borer (Chilo suppressalis) poses a serious threat to rice yield and food security. For two decades, fipronil was a key insecticide for its control, but its use has been restricted owing to the toxicity of its degradation products to aquatic life and bees. To develop superior alternatives, we structurally modified fipronil-5-amino through acylation, benzylation and carbamation, and evaluated the insecticidal activity of the resulting derivatives against resistant C. suppressalis, as well as their toxicity to zebrafish and honeybees. RESULTS: Twenty-seven fip

Do some families act as a reservoir of head lice in the community? Surveys for prevalence and insecticide resistance suggest this is the case.

In response to increasing complaints of head louse infestation in the area of Cambridge, UK, in the mid-1990s, a series of prevalence surveys in selected schools coupled with collection of lice were commissioned by the local health authority. Carers of infested children were provided with advice on treatment by school nurses. Lice collected during these surveys were tested in the laboratory for evidence of resistance to insecticides. These data were used by the health authority to make decisions about recommended insecticide use for control of head infestations. Surveys over 3 years 1995-1997

Identification of genes involved in pyrethroid-, propoxur-, and dichlorvos- insecticides resistance in the mosquitoes, Culex pipiens complex (Diptera: Culicidae).

Culex pipiens pallens and Cx. p. quinquefasciatus are important vectors of many diseases, such as West Nile fever and lymphatic filariasis. The widespread use of insecticides to control these disease vectors and other insect pests has led to insecticide resistance becoming common in these species. In this study, high throughout Illumina sequencing was used to identify hundreds of Cx. p. pallens and Cx. p. quinquefasciatus genes that were differentially expressed in response to insecticide exposure. The identification of these genes is a vital first step for more detailed investigation of the m…

Transcriptome Analysis of an Insecticide Resistant Housefly Strain: Insights about SNPs and Regulatory Elements in Cytochrome P450 Genes.

BACKGROUND: Insecticide resistance in the housefly, Musca domestica, has been investigated for more than 60 years. It will enter a new era after the recent publication of the housefly genome and the development of multiple next generation sequencing technologies. The genetic background of the xenobiotic response can now be investigated in greater detail. Here, we investigate the 454-pyrosequencing transcriptome of the spinosad-resistant 791spin strain in relation to the housefly genome with focus on P450 genes. RESULTS: The de novo assembly of clean reads gave 35,834 contigs consisting of 21,…

Agrobacterium tumefaciens-mediated transformation of rice with the spider insecticidal gene conferring resistance to leaffolder and striped stem borer.

Immature embryos of rice varieties "Xiushuill" and "Chunjiang 11" precultured for 4d were infected and transformed by Agrobacterium tumefaciens strain EHA101/pExT7 (containing the spider insecticidal gene). The resistant calli were transferred onto the differentiation medium and plants were regenerated. The transformation frequency reached 56% approximately 72% measured as numbers of Geneticin (G418)-resistant calli produced and 36% approximately 60% measured as numbers of transgenic plants regenerated, respectively. PCR and Southern blot analysis of transgenic plants confirmed that the T-DNA

No benefit of a mid-season foliar insecticide application to aphid-resistant soybeans when overall insect pressure is low.

Producers currently rely on insecticides to manage pest soybean aphids (Aphis glycines Matsumura), although there are growing concerns about insecticide-resistant aphid populations. Using resistant soybean varieties with naturally occurring, pyramided Rag genes can adequately control soybean aphids and preclude the need to use an insecticide. However, forgoing a mid-season insecticide application could lead to yield loss from other insect pests and thus discourage farmer adoption of aphid-resistant varieties. We conducted a multiyear, multilocation field trial using a pyramided rag1b/rag3b soy

Efficacy of Gel Bait Insecticides with Different Modes of Action Against Pyrethroid-Resistant Blattella germanica Populations in Türkiye.

The German cockroach, Blattella germanica (Linnaeus) (Blattodea: Ectobiidae), is a major urban pest that can cause allergic reactions and mechanically carry pathogenic microorganisms. Its control has become increasingly difficult because of widespread resistance to commonly used insecticides, particularly synthetic pyrethroids. This study compared the efficacy of six commercial gel bait formulations containing imidacloprid, dinotefuran, clothianidin, indoxacarb, spinosad, or fipronil against four field populations of B. germanica from Antalya, Türkiye, previously characterized and routinely mo

Multi-insecticide resistant malaria vectors in the field remain susceptible to malathion, despite the presence of Ace1 point mutations.

Insecticide resistance in Anopheles mosquitoes is seriously threatening the success of insecticide-based malaria vector control. Surveillance of insecticide resistance in mosquito populations and identifying the underlying mechanisms enables optimisation of vector control strategies. Here, we investigated the molecular mechanisms of insecticide resistance in three Anopheles coluzzii field populations from southern Côte d'Ivoire, including Agboville, Dabou and Tiassalé. All three populations were resistant to bendiocarb, deltamethrin and DDT, but not or only very weakly resistant to malathion. …

Indoor spraying with chlorfenapyr (a pyrrole insecticide) provides residual control of pyrethroid-resistant malaria vectors in southern Benin.

BACKGROUND: New classes of insecticides with novel modes of action, which can provide effective and prolonged control of insecticide-resistant malaria vector populations, are urgently needed for indoor residual spraying. Such insecticides can be included in a rotation plan to manage and prevent further development of resistance in mosquito vectors of malaria. Chlorfenapyr, a novel pyrrole insecticide with a unique mode of action, is being developed as a long-lasting IRS formulation. METHODS: The efficacy of several formulations of chlorfenapyr alone and as mixtures with alpha-cypermethrin wer
