A novel sensor array combined with machine learning for the effective differentiation of multiple classes of antibiotics based on copper-based nanozymes with laccase-like activity.

A nanozyme sensor array was developed combining with machine learning algorithms for the effective identification and differentiation of multiple antibiotic classes and antibiotic individuals. The four channel sensor array was composed by four copper-based nanozymes with laccase-like activity. The significant structural differences between different classes of antibiotics (such as Aminoglycosides, Tetracyclines, β-lactams, Nitroimidazole, Sulfonamides, Quinolones) resulted in the different effect on the nanozymatic reactions through interact with copper、substrate and so on. In addition, the di…

A novel "laccase-degradation and GABA-compensation" strategy mediated by Lactococcus lactis subsp. lactis GLM-1 for the safe and targeted detoxification of ginkgo seeds.

Ginkgo biloba L. seeds are valued for their bioactive compounds but contain the neurotoxin 4'-O-methylpyridoxine (MPN), limiting their safe use. This study developed a novel "laccase-degradation and GABA-compensation" strategy using a food-grade bacterium, Lactococcus lactis subsp. lactis GLM-1, to achieve detoxification with bioactive preservation. The study initially confirmed that while free laccase effectively degrades MPN, it nonspecifically oxidizes total flavonoids and terpene lactones. Furthermore, using a model Caenorhabditis elegans, it was elucidated that MPN impedes GABA production…

Complementary integration of highly sensitive aptasensor and portable hydrogel for colorimetric detection of dhurrin in sorghum based on enhanced laccase-like activity of CuCo-MOFs.

As a cyanogenic glycoside commonly found in grass family plants such as sorghum, dhurrin poses a serious threat to the food safety due to its hydrolysis product, hydrogen cyanide. But the equipment is expensive and the procedures complex, making it difficult to meet the demand for rapid on-site screening. This study develops a colorimetric aptasensor and its hydrogel format for rapid dhurrin detection in sorghum, based on aptamer-enhanced laccase-like activity of copper‑cobalt metal-organic frameworks. Dhurrin binding triggers aptamer detachment, weakening the enhancing effect of the aptamer o…

4-Hydroxybenzoic Acid-Based Hydrazide-Hydrazones as Potent Growth Inhibition Agents of Laccase-Producing Phytopathogenic Fungi That Are Useful in the Protection of Oilseed Crops.

The research on new compounds against plant pathogens is still socially and economically important. It results from the increasing resistance of pests to plant protection products and the need to maintain high yields of crops, particularly oilseed crops used to manufacture edible and industrial oils and biofuels. We tested thirty-five semi-synthetic hydrazide-hydrazones with aromatic fragments of natural origin against phytopathogenic laccase-producing fungi such as Botrytis cinerea, Sclerotinia sclerotiorum, and Cerrena unicolor. Among the investigated molecules previously identified as poten…

Inducing laccase activity in white rot fungi using copper ions and improving the efficiency of azo dye treatment with electricity generation using microbial fuel cells.

This work presents a white rot fungus-microbial fuel cell (WRF-MFC) that uses WRF that is grown at its cathode. Adding Cu2+ to the fungi-containing solid medium stimulated WRF-secreting laccase, which catalyzed the redox reaction in the MFC and thereby promoting the generation of electricity. Adding 12.5 mg L-1 Cu2+ to a G. lucidum-containing medium provided the greatest laccase stimulation and increased the laccase activity by a factor of 1.6. Adding 12.5 mg L-1 Cu2+ to the WRF chamber of WRF-MFC increased its decolorization of Acid Orange 7 (AO-7) and increased its power density to 223 mW m-…

Carbon-oxygen bond formation by fungal laccases: cross-coupling of 2,5-dihydroxy-N-(2-hydroxyethyl)-benzamide with the solvents water, methanol, and other alcohols.

Laccase-catalyzed reactions lead to oxidation of the substrate via a cation radical, which has been described to undergo proton addition to form a quinonoid derivative or nucleophilic attack by itself producing homomolecular dimers. In this study, for the substrate 2,5-dihydroxy-N-(2-hydroxyethyl)-benzamide, we show that, besides the quinonoid form of substrate, all products formed are nonhomomolecular ones. Indeed, without addition of a reaction partner, heteromolecular products are formed from the quinonoid form of the laccase-substrate and the solvents water or methanol present in the incub…

A one-pot two-step synthesis of tertiary alcohols combining the biocatalytic laccase/TEMPO oxidation system with organolithium reagents in aerobic aqueous media at room temperature.

The one-pot/two-step combination of enzymes and polar organometallic chemistry in aqueous media is for the first time presented as a proof-of-concept study. The unprecedented combination of the catalytic oxidation of secondary alcohols by the system laccase/TEMPO with the ultrafast addition (3 s reaction time) of polar organometallic reagents (RLi/RMgX) to the in situ formed ketones, run under air at room temperature, allows the straightforward and chemoselective synthesis of tertiary alcohols with broad substrate scope and excellent conversions (up to 96%).

The anticancer activity of laccase from white rot fungus Cerrena unicolor on the example of its action on Caov-3 and NIH:OVCAR-3 ovarian cancer cells.

INTRODUCTION AND OBJECTIVE: According to epidemiological data, ovarian cancer is diagnosed in 240,000 women per year, which is why it is the seventh most common cancer among women worldwide. In addition, among gynaecological cancers it is characterized by the highest mortality. However, there is still no effective treatment, which is why maintenance therapies have a decisive impact on the quality of life of patients. Cerrena unicolor is an example of a medical fungus that produces a laccase enzyme (LAC), as well as synthesizing low-molecular secondary metabolites with a wide spectrum of biolog…

The laccase-catalyzed domino reaction between catechols and heterocyclic 1,3-dicarbonyls and the unambiguous structure elucidation of the products by NMR spectroscopy and X-ray crystal structure analysis.

The laccase-catalyzed reaction between catechols and heterocyclic 1,3-dicarbonyls (pyridinones, quinolinones, thiocoumarins) using aerial oxygen as the oxidant delivers benzofuropyridinones, benzofuroquinolinones, and thiocoumestans in a simple fashion, highly regioselectively with yields ranging from 55 to 98%. With barbituric acid derivatives the exclusive formation of dispiropyrimidinone derivatives takes place. The unambiguous and complete structure elucidation of all reaction products has been achieved by means of NMR spectroscopic methods (HSQMBC and band-selective HMBC) as well as by X-…

Influence of a soil enzyme on iron-cyanide complex speciation and mineral adsorption.

Cyanide is commonly found as ferrocyanide [Fe(II)(CN)(6)](-4) and in the more mobile form, ferricyanide [Fe(III)(CN)(6)](-3) in contaminated soils and sediments. Although soil minerals may influence ferrocyanide speciation, and thus mobility, the possible influence of soil enzymes has not been examined. In a series of experiments conducted under a range of soil-like conditions, laccase, a phenoloxidase enzyme derived from the fungi Trametes versicolor, was found to exert a large influence on iron-cyanide speciation and mobility. In the presence of laccase, up to 93% of ferrocyanide (36-362ppm)…

Eco-friendly decolorization of synthetic dyes using radiation-induced whole cell biocatalyst with enhanced copper resistance.

Toxic and carcinogenic compounds, such as synthetic dyes and polyphenols, were widely employed and released as pollutants in a variety of industries, including textiles, food, and cosmetics. Biological oxidation process that used oxidizing enzymes to breakdown pollutant compounds were environmentally favorable. However, due to the cell toxicity of metal ions supplements used for the biosynthesis of oxidizing enzymes like laccase, their efficient application for biological degradation is limited. In this study, we aimed to boost laccase activity by introducing high copper resistance into whole-…

Investigation on the Detoxification of Indoxyl Sulfate (IS) and Indole-3-Acetic Acid (IAA) Protein-Bound Uremic Toxins (PBUTs) Using Trametes versicolor Biocompatible Laccase: In Situ Synchrotron Imaging, Experimental and Computational Studies.

The removal of protein-bound uremic toxins (PBUTs), such as indoxyl sulfate (IS) and indole-3-acetic acid (IAA), from hemodialysis (HD) patients remains a significant challenge due to their strong binding to serum proteins, such as albumin. This study aimed to evaluate the potential of using the enzyme laccase, derived from Trametes versicolor, for the decomposition and removal of IS and IAA during HD. Molecular docking was utilized to investigate the interactions between laccase and the toxins, identifying key functional groups involved. To assess the detoxification efficacy, liquid chromatog…

Exploring fungal-mediated solutions and its molecular mechanistic insights for textile dye decolorization.

Decolorization of textile dyes and study of their intermediate compounds is necessary to comprehend the mechanism of dye degradation. In the present study, different fungal mediated solutions were explored to provide an alternative to treat the reactive dyes. Growing biomass of Pleurotus sajor caju showed 83% decolorization (249.99 mg L-1 removal) of Reactive Blue 13 (RB 13) and 63% decolorization (188.83 mg L-1) of Reactive Black 5 (RB 5) at 300 mg L-1 initial concentration on 8 d. Higher laccase activity was positively correlated with increase in decolorization. However, increasing dye conce…

Advanced enzyme-assembled hydrogels for the remediation of contaminated water.

Enzyme-catalyzed biodegradation is an emerging green strategy for environmental remediation, although challenged by high cost and poor robustness. Herein, natural biopolymer (cellulose)-derived hydrogels concurrently doped with β-cyclodextrin and montmorillonite nanosheets that are synthesized in one-step demonstrate exceptional pollutant affinity and mechanical strength. Laccase is then stably and effectively assembled onto the hydrogels by a facile strategy based on charge-assisted H-bonding, which can be extended to other enzymes. The advanced laccase-assembled hydrogels display excellent s…
