Autoantibodies to human nuclear lamin B2 protein. Epitope specificity in different autoimmune diseases.

The nuclear lamina of mammalian cells consists of three major proteins, lamins A, C, and B, and a fourth minor protein, lamin B2. Lamins belong to the family of intermediate filaments and are highly similar both in structure and primary sequence. They are organized in three well-defined domains: 1) a central alpha-helical rod, which is a secondary structure shared by all types of intermediate filaments, formed by three alpha-helices (coils 1A, 1B, and 2) and surrounded by 2) an amino-terminal head and 3) a carboxyl-terminal tail. Autoantibodies toward major lamin have been described previously…

Effects of Curing Schedules on Carbon-Fiber-Reinforced Laminates with a Bio-Based Epoxy Matrix.

Carbon-fiber-reinforced polymer (CFRP) composites fabricated with bio-based epoxy resins represent a promising approach for sustainable lightweight structures produced by out-of-autoclave (OoA) technologies. The curing schedule influences the state of the epoxy matrix and, consequently, can affect the fiber-matrix interaction and laminate performance. This study investigates the effect of practical curing conditions on 2 × 2 twill woven carbon fiber laminates fabricated by vacuum infusion using a commercially available bio-based epoxy resin IB2. The manufacturer's recommended room-temperature

Mesenchymal transitions reduce lamin A expression and nuclear stiffness to enhance confined migration in glioblastoma.

The evolution of glioblastoma (GBM) is coupled with a proneural-to-mesenchymal transition (PMT) that exacerbates disease in part through heightened cell invasiveness and spread. PMT follows similar molecular and phenotypic features with epithelial to mesenchymal transitions (EMT) and promotes rapid GBM invasion despite tight physical barriers in the dense brain parenchyma. Recent studies suggest that efficient confined migration is key to navigating these physical constraints; however, the role of PMT in mediating rapid confined migration remain unclear. Using microchannel platforms, AFM, and

Genetically reprogrammed BMSCs with Lamin B1 depletion enhance bone regeneration for osteoporosis therapy.

BACKGROUND: Mesenchymal stem cell (MSC) dysfunction contributes to impaired bone regeneration in osteoporosis. Lamin B1, a nuclear lamina protein implicated in stem cell aging, has an unclear role in osteogenesis. METHODS: We examined Lamin B1 expression during osteogenic differentiation, assessed its pathological alterations in OVX-derived BMSCs, and generated Lamin B1-knockout MSCs to evaluate effects on osteogenesis, migration, and KAT2A regulation. We performed transcriptomic profiling and intra-bone marrow transplantation in OVX mice to determine functional relevance. RESULTS: Lamin B1

Loss of Postnatal Retinal Input Perturbs Cortical Lamination in the Developing Ferret Visual Cortex.

Neuroanatomical development of the visual system is influenced by activity-dependent processes. Removal of retinal input early in development in animal model experiments reproduces several structural characteristics of the primary visual cortex in humans who have been blinded early in life. Yet, many questions regarding how the loss of retinal activity affects the organization and cellular composition of the visual cortex remain to be answered. Here, we report alterations to primary visual cortex development in ferrets over the postnatal day 20 (P20) to P38 age range that arise following bilat

In Situ ZIF-8-Coated Copper Laminate System for Fluid-Phase Adsorptive Separation.

The use of structured adsorbents is emerging as a promising approach for adsorptive separation processes, and several ex situ structuring routes like extrusion, three-dimensional (3D) printing, and coating over substrates have been extensively investigated. However, in situ growth of adsorbents such as metal-organic frameworks (MOFs) on metal laminates remains underexplored. This study introduces a novel laminate system, where aluminum pieces, inspired by the "LEGO" concept, were designed through CNC milling and used to fabricate embossed/dented copper laminates. These laminates were then coat

Elastoplastic Modeling of Kevlar® Composite Laminates: A Cyclic Loading Approach for In-Plane Characterization.

This study investigates the elastoplastic behavior of phenol formaldehyde/polyvinyl butyral matrix (70% PF/30% PVB) reinforced with Kevlar® fibers through comprehensive in-plane tensile testing. Cyclic loading-unloading tests were conducted at a 100%/min strain rate using a universal testing system at room temperature on 04, 904, and ±45s laminates. The experimental results revealed significant nonlinear hardening behavior beyond yield stress, accompanied by yarn stiffening effects during loading cycles. A novel elastoplastic constitutive model was developed, incorporating Hill's yield criteri

Ultrahigh-Water-Content yet Robust Hydrogels Enabled by Bioinspired Laminated Membranous Network.

Hydrogels, water-rich polymer networks, are important materials for application as structural biomaterials. More water-rich networks are typically less mechanically robust, which is manifested as softness and low stress to fracture. Unusually, jellyfish mesoglea exhibits paradoxical combination of high stiffness and strength with ultrahigh water content. Here, it is discovered that jellyfish mesoglea possesses a long-range-ordered laminated membranous network, which features crystal orientation along membrane plane and collagen chain spanning at the junction of membranes. Such a laminated memb

Decreasing Lamin A Triggers Cell Fate Transitions through Heterochromatin-Nuclear Periphery Detethering.

The interplay between nuclear architecture and extracellular matrix stiffness orchestrates cell fate decisions, yet the molecular mechanisms remain poorly defined. Here, we investigate the role of Lamin A (LMNA), a nuclear structural protein whose expression correlates with tissue stiffness, in regulating cellular differentiation and fate decision. Using myoblasts and fibroblasts as models, it was observed that cells with low LMNA expression showed that higher cell deformation elevated expression of neurological genes and exhibited potential for differentiation into a neural-like fate. Evaluating peritoneal elastic laminal invasion to improve stratification of patients with pT3 gastric cancer.

BACKGROUND: Elastic laminal invasion (ELI), defined as tumor invasion beyond the peritoneal elastic lamina, may affect gastric cancer (GC) prognosis, though limited data exist on this relationship. METHODS: We retrospectively reviewed representative pathological slides from 396 patients with pT3 or pT4a GC who underwent curative resection to assess the association between ELI and relapse-free survival (RFS) and overall survival (OS). RESULTS: The 5-year RFS of pT3 GC with negative ELI was 85.9%, which was better than the 55.9% of that of ELI-positive pT3 (P < 0.001) or pT4a (51.3%) GC (P < 0

Cytomegalovirus disrupts Lamin A/C to control microtubule-mediated nuclear movement and cell migration.

Human cytomegalovirus (HCMV) disrupts the inner nuclear Lamin A/C meshwork to facilitate virion egress. Here, we show that targeting Lamin A/C also plays a previously unrecognized role in HCMV-induced rewiring of cytoskeletal connections that control nuclear movement and cell migration at later stages of infection. Specifically, HCMV was found to employ distinct strategies to downregulate Lamin A/C and SUN2, which together regulate linker of nucleoskeleton and cytoskeleton (LINC) complexes that most often connect the nuclear membrane with cytoplasmic actin filaments. Unexpectedly, inhibition o

Lamin A K97E leads to NF-κB-mediated dysfunction of inflammatory responses in dilated cardiomyopathy.

BACKGROUND INFORMATION: Lamins are type V intermediate filament proteins underlying the inner nuclear membrane which provide structural rigidity to the nucleus, tether the chromosomes, maintain nuclear homeostasis, and remain dynamically associated with developmentally regulated regions of the genome. A large number of mutations particularly in the LMNA gene encoding lamin A/C results in a wide array of human diseases, collectively termed as laminopathies. Dilated Cardiomyopathy (DCM) is one such laminopathic cardiovascular disease which is associated with systolic dysfunction of left or both …

SIRT1 Ameliorates Lamin A/C Deficiency-Induced Cardiac Dysfunction by Promoting Mitochondrial Bioenergetics.

Dilated cardiomyopathy (DCM) is associated with high mortality despite advanced therapies. The LMNA gene encodes lamin A/C and is the second most frequently mutated gene associated with DCM, for which therapeutic options are limited. Here we generated Lmna -/- mice and found they exhibited cardiac dysfunction at the age of 1 month but not at 2 weeks. Proteomics showed down-regulation of mitochondrial function-related pathways in Lmna -/- hearts. Moreover, early injured mitochondria with decreased cristae density and sirtuin 1 (SIRT1) down-regulation were observed in 2-week-old Lmna -/- hearts.

A lamin A/C variant causing striated muscle disease provides insights into filament organization.

The LMNA gene encodes the A-type lamins, which polymerize into ∼3.5-nm-thick filaments and, together with B-type lamins and associated proteins, form the nuclear lamina. Mutations in LMNA cause a wide variety of pathologies. In this study, we analyzed the nuclear lamina of embryonic fibroblasts from LmnaH222P/H222P mice, which develop cardiomyopathy and muscular dystrophy. Although the organization of the lamina appeared unaltered, there were changes in chromatin and B-type lamin expression. An increase in nuclear size and consequently a relative reduction in heterochromatin near the lamina al…

Characterization of Oligofructose-Induced Acute Rumen Lactic Acidosis and the Appearance of Laminitis in Zebu Cattle.

The objective of this study was to characterize oligofructose-induced acute rumen lactic acidosis and its consequences in zebu cattle. We used 29 Nellore heifers which were submitted to experimental induction of laminitis by oligofructose excess. During the induction period, the animals underwent clinical examination, including laminitis diagnosis (hoof pressure testing and locomotion score) and blood and ruminal fluid sampling every six hours (over the initial 24 h) and every 12 h (up to 72 h), after the highest dose. Almost half of the animals (48.1%) required treatment with bicarbonate and

PCAF Involvement in Lamin A/C-HDAC2 Interplay during the Early Phase of Muscle Differentiation.

Lamin A/C has been implicated in the epigenetic regulation of muscle gene expression through dynamic interaction with chromatin domains and epigenetic enzymes. We previously showed that lamin A/C interacts with histone deacetylase 2 (HDAC2). In this study, we deepened the relevance and regulation of lamin A/C-HDAC2 interaction in human muscle cells. We present evidence that HDAC2 binding to lamina A/C is related to HDAC2 acetylation on lysine 75 and expression of p300-CBP associated factor (PCAF), an acetyltransferase known to acetylate HDAC2. Our findings show that lamin A and farnesylated pr

Lamin-Related Congenital Muscular Dystrophy Alters Mechanical Signaling and Skeletal Muscle Growth.

Laminopathies are a clinically heterogeneous group of disorders caused by mutations in the LMNA gene, which encodes the nuclear envelope proteins lamins A and C. The most frequent diseases associated with LMNA mutations are characterized by skeletal and cardiac involvement, and include autosomal dominant Emery-Dreifuss muscular dystrophy (EDMD), limb-girdle muscular dystrophy type 1B, and LMNA-related congenital muscular dystrophy (LMNA-CMD). Although the exact pathophysiological mechanisms responsible for LMNA-CMD are not yet understood, severe contracture and muscle atrophy suggest that muta…

Neuron type-specific increase in lamin B1 contributes to nuclear dysfunction in Huntington's disease.

Lamins are crucial proteins for nuclear functionality. Here, we provide new evidence showing that increased lamin B1 levels contribute to the pathophysiology of Huntington's disease (HD), a CAG repeat-associated neurodegenerative disorder. Through fluorescence-activated nuclear suspension imaging, we show that nucleus from striatal medium-sized spiny and CA1 hippocampal neurons display increased lamin B1 levels, in correlation with altered nuclear morphology and nucleocytoplasmic transport disruption. Moreover, ChIP-sequencing analysis shows an alteration of lamin-associated chromatin domains …
