A public health worry as is Tuberculosis (TB) has been making more than 10 million people globally suffer from its terror and causing more than 2 million people worldwide to lose their lives every year. Mankind is putting all its efforts, since the discovery of the causative bacilli, to come up with some dramatic improvements in providing high-quality TB diagnostic services. Nevertheless, it poses a challenge and many people with TB remain use only smear microscopy for diagnosis. In an environment where transmission is becoming easier by the day the challenge becomes burdensome once disease ge

BACKGROUND: Reporter phage assay offers a low-cost and rapid approach for drug susceptibility testing (DST) of Mycobacterium tuberculosis (Mtb). However, their adoption in clinical laboratories has been limited by challenges such as low detection sensitivity and operational complexity. To address these challenges, we integrated a novel reporter phage into the clinical laboratory workflow for DST of Mtb.
METHODS: A novel high-sensitive reporter phage, ΦLSN, was constructed by integrating promoter Pleft, the ribosome-binding site (RBS) of promoter Psmyc, and nanoluciferase reporter gene (nluc)

Gentamicin is a promising antibiotic for the treatment of multidrug-resistant gonorrhea. The aim of this study was to analyze the suitability and reliably of disk diffusion to monitor the susceptibility to gentamicin. We studied 237 Neisseria gonorrhoeae isolates obtained in 2013 and 2015. Reference MICs were correlated with inhibition zone diameters (in millimeters) of gentamicin 10 µg disks manufactured by BBL and Oxoid. The Pearson correlation between disk diffusion and agar dilution was r = -.68 (P < 0.001) for BBL disk and r = -.71 (P < 0.001) for Oxoid disk. No very major or major discre

High-frequency electromagnetic waves (HFEMWs) have been shown to influence cellular functions, including bacterial behavior, potentially affecting growth and antibiotic sensitivity. This study evaluated the response of Escherichia coli and Staphylococcus aureus to HFEMWs across a frequency range of 900 MHz to 73 GHz. The Bacterial sensitivity to antibiotics, including ceftazidime, ceftaroline, gentamycin, doxycycline, and ciprofloxacin, was assessed. The growth rate was evaluated by measuring the optical density (OD) and the number of colony-forming units (CFUs). Our results revealed significa

This study analyzed the effect of high-pressure processing (HPP) on the frequency of conjugal gene transfer of antibiotic resistance genes among strains obtained from starter cultures. Gene transfer ability was analyzed in vitro and in situ in the food matrix. It was found that the transfer of aminoglycoside resistance genes did not occur after high-pressure treatment, either in vitro or in situ. After exposure to HPP, the transfer frequencies of tetracycline, ampicillin and chloramphenicol resistance genes increased significantly compared to the control sample, both in vitro and in situ. The

INTRODUCTION: Enterococcus faecium is a major cause of community and hospital-acquired infections. Due to limited options for infection with fluoroquinolones-resistant Enterococci, novel therapeutics are urgently needed. Efflux pumps are contributed to fluoroquinolones resistance phenotype in this bacterium and novel inhibitors that target these efflux pumps could be effective in patients. In this research, the possible synergistic effect of an efflux pump inhibitor (EPI), thioridazine, with ciprofloxacin was investigated against clinical isolates of E. faecium.

Information on causative diarrheal pathogens and their associated antimicrobial susceptibility remains limited for Cambodia. This study describes antimicrobial resistance patterns for Shigella and nontyphoidal Salmonella isolates collected in Cambodia over a 5-year period. Multidrug resistance was shown in 98% of Shigella isolates, with 70%, 11%, and 29% of isolates being resistant to fluoroquinolones, azithromycin, and cephalosporin, respectively. As many as 11% of Shigella isolates were resistant to nearly all oral and parenteral drugs typically used for shigellosis, demonstrating extreme dr

Contaminated sites are recognized as the "hotspot" for the development and spread of antibiotic resistance in environmental bacteria. It is very challenging to understand mechanism of development of antibiotic resistance in polluted environment in the presence of different anthropogenic pollutants. Uncertainties in the environmental processes adds complexity to the development of resistance. This study attempts to develop mathematical model by using stochastic partial differential equations for the transport of fluoroquinolone and its resistant bacteria in riverine environment. Neisseria gonorrhoeae is one of the leading antimicrobial resistance threats worldwide. This study determined the MICs of closthioamide to be 0.008 to 0.5 mg/liter for clinical N. gonorrhoeae strains and related species. Cross-resistance with existing antimicrobial resistance was not detected, indicating that closthioamide could be used to treat drug-resistant N. gonorrhoeae.

As part of the ongoing multinational SENTRY antimicrobial resistance surveillance program, a total of 1,047 respiratory tract isolates of Streptococcus pneumoniae, 845 from 27 United States medical centers and 202 from seven Canadian institutions, were collected between February and June 1997 and characterized in a central laboratory. In the United States, the overall percentages of penicillin-intermediate strains and strains with high-level resistance to penicillin were 27.8% and 16.0%, respectively. In Canada, these values were 21.8% and 8.4%, respectively. Among the 31 centers in the United

Staphylococcus aureus isolates which produce type A staphylococcal beta-lactamase have been associated with wound infections complicating the use of cefazolin prophylaxis in surgery. To further evaluate this finding, 215 wound isolates from 14 cities in the United States were characterized by antimicrobial susceptibility and beta-lactamase type and correlated with the preoperative prophylactic regimen. Borderline-susceptible S. aureus isolates of phage group 5 (BSSA-5), which produce large amounts of type A beta-lactamase and exhibit borderline susceptibility to oxacillin, comprised a greater
