A method for light microscopy examination of cellular and structural interrelations in undemineralized tooth specimens.

This study examines ameloblasts and odontoblasts in relation to mineralizing enamel and dentin in thin undemineralized tooth sections from unerupted and partly erupted human third molars. The material comprised 12 freshly removed third molars. After fixation for 24 h the specimens were dehydrated and embedded in hydroxyethylmethacrylate. Undemineralized sections 10-15 microns thick were prepared with a cutting-grinding system, and radiomicrographs were taken. Postsecretory ameloblasts were seen in different morphologic stages ranging from ruffle- and smooth-ended ameloblasts to cuboidal and sq

Human root caries: histopathology of initial lesions in cementum and dentin.

The histopathology of human root caries was examined in extracted teeth by different optical methods. The present part of the study details the sequential stages of initial caries in both cementum and peripheral dentin. Significant differences are shown between the mechanisms operating on the various dental hard tissues during development of caries. Histologically distinguishable reaction patterns depended on both the degree of the cariogenic challenge and the respective structural features of cementum and peripheral dentin. Earliest lesions, histologically visible as small clefts traversing c

Acceleration of segmental bone regeneration in a rabbit model by strontium-doped calcium polyphosphate scaffold through stimulating VEGF and bFGF secretion from osteoblasts.

The development of suitable bioactive three-dimensional scaffold for the promotion of bone regeneration is critical in bone tissue engineeri … on a rabbit segmental bony defects model over a period of 16 weeks by histology combined with X-ray microradiography. And then, in order to avoid the influence from the other factors such as hypoxia which emerge in vivo study and affect the secretion of VEGF and bFGF from host cells, human osteoblast-like cells (MG63) were seeded to SCPP, CPP and HA scaffolds in v …

Mechanisms of the anabolic effects of teriparatide on bone: insight from the treatment of a patient with pycnodysostosis.

Pycnodysostosis is an extremely rare genetic osteosclerosis caused by cathepsin K deficiency. We hypothesized that teriparatide, a potent an … siliac bone biopsies were performed, and the degree of mineralization was evaluated by quantitative microradiography. In vitro assessment of bone resorption was performed after separation and differentiation of CD14(+) monocytes from peripheral blood. Bone structure assessed by HR-pQCT on the radius and tibia showed augmentation of cortical and trabecular density. …

Sinus floor elevation using autogenous bone or bone substitute combined with platelet-rich plasma.

OBJECTIVE: Sinus augmentation is a common approach for patients with severe alveolar ridge atrophy. However, autogenous bone sometimes results in donor site complications. Bone substitutes with platelet-rich plasma (PRP) promote early bone formation with autogenous bone. Use of PRP on autogenous bone and a bovine bone substitute were investigated in this split-mouth animal study. STUDY DESIGN: Premolars were extracted from minipigs. Each animal received sinus augmentation using a lateral approach with simultaneous insertion of 3 implants in each site. Groups were randomized using autogenous b

The relationship between general osteoporosis of the organism and periodontal diseases.

Osteoporosis and periodontitis are very prevalent diseases and are most common in middle-aged and elderly women. These diseases are related as both damage bone tissue and share common risk factors. Discussions about the association between these two bone-damaging diseases began in 1960. A hypothesis was raised that systemic imbalance in bone resorption and deposition might manifest itself in the alveolar bone earlier than in other bones. When analyzing systemic and local changes in bone density, a number of issues were investigated and attempted to answer the question of whether dental osteope

Remineralization of advanced root dentin lesions in vitro.

The possibility for remineralization of advanced root dentin lesions was investigated in vitro. Lesions were produced in acidic buffer solution with 0.1 and 0.5 ppm fluoride, for 3 and 5 weeks, respectively, using a single-section model. Next, the sections were immersed in a remineralization solution containing 0.1 ppm F (as NaF) for 20 weeks. Microradiographs were made after each 4-week-period. The lesions were around 500 micro m deep, with mineral loss values between 11,000 and 18000 vol%.microm. After 20 weeks of remineralization, the absolute amounts of deposited mineral ranged from 7500 t

Quantitative remineralization evolution kinetics of artificially demineralized human enamel using photothermal radiometry and modulated luminescence.

Human molars were subjected to demineralization in acid gel followed by incubation in remineralization solutions without or with fluoride (1 or 1000 ppm). Photothermal radiometry (PTR) and modulated luminescence (LUM) frequency scans were performed prior to and during de/remineralization treatments. Transverse Micro-Radiography (TMR) analysis followed at treatment conclusion to determine mineral loss and lesion depth. The remineralization process illustrated a complex interplay between surface and subsurface mineral deposition, confining the thermal-wave centroid toward the dominating layer. E

Physical properties of root cementum: Part 10. Comparison of the effects of invisible removable thermoplastic appliances with light and heavy orthodontic forces on premolar cementum. A microcomputed-tomography study.

INTRODUCTION: Orthodontic treatment with clear sequential removable thermoplastic appliances (TAs) is gaining popularity as an alternative to treatment with fixed appliances. The amount of orthodontically induced inflammatory root resorption generated by such appliances has not been investigated. In this prospective randomized clinical trial, we used x-ray microtomography to quantify resorption generated by treatment with ClearSmile appliances (ClearSmile, Woollongong, Australia) and compared the effects with those of heavy and light conventional orthodontic forces and no force. METHODS: The

Micro-CT of the human lung: imaging of alveoli and virtual endoscopy of an alveolar duct in a normal lung and in a lung with centrilobular emphysema--initial observations.

The appearance of human lung parenchyma at the structural level of alveoli was investigated by the use of micro-computed tomography (CT). Approval for use of autopsy lungs was given by the head of the pathology institute of the university, in accordance with the requirements of the State Ministry of Science and Arts and without the need for institutional review board approval. Two human lungs (one normal lung and one lung with centrilobular emphysema of a mild to moderate degree) were inflated and fixed with hot formalin vapor. Lung specimens excised from the superior segment of the left lower

Effect of surface zone deproteinisation on the access of mineral ions into subsurface carious lesions of human enamel.

It has been proposed that the so-called intact surface zone of carious lesions of enamel could restrict the ingress of mineral ions and hinder remineralisation. The present study was intended to determine the role of organic (proteinaceous) material in restricting the movement of mineral ions into carious lesions in vitro. Natural carious lesion surfaces were divided into two halves. The experimental half was de-proteinised using hypochlorite, the control half remained untreated. The whole tooth was exposed to 45Ca in solution, and 45Ca uptake into experimental and control tissue was measured

The role of parathyroids for the adaptation to a low calcium intake.

One-year-old selectively parathyroidectomized rats were found to have lost the ability to adapt themselves to a reduced calcium intake. Of importance in this respect was certainly the demonstrated inability to mobilize skeletal calcium to the blood. Thus, when the normal intake of calcium in the diet is reduced in the adult intact rat, skeletal calcium reserves must become mobilized with resulting osteoporosis. Histological and morphometric analyses of the adrenal glands showed no apparent effect of the parathyroidectomy upon the functional state of the adrenal cortex.
