Effects of mycotoxin-producing fungi on the fitness and gut bacterial community of the soil springtail Folsomia candida.

UNLABELLED: Mycotoxin-producing fungi are widespread and their adverse effects on mammals have been investigated; however, their impacts on soil invertebrates are not fully understood. Folsomia candida is a model soil arthropod that represents an important part of the soil invertebrate community. This study investigated the consequences of F. candida grazing on mycotoxin-producing fungi Fusarium verticillioides, F. graminearum, Aspergillus ochraceus, and A. nidulans. Consuming mycotoxin-producing fungi affected the body size and reproductive ability of F. candida, and altered the gut bacterial

Deoxynivalenol mycotoxin dietary exposure on broiler performance and small intestine health: A comprehensive meta-analysis.

The effect of DON mycotoxins on broiler production performance and the small intestine is a critical factor in the health and well-being of broilers. Several studies have been conducted on this topic and have reported varying results and conclusions. Therefore, it is necessary to conduct systematic reviews and meta-analyses to thoroughly examine and draw unique conclusions. In this meta-analysis, we conducted a systematic review of multiple studies on the effects of DON mycotoxins in broilers. The analysis comprised 26 articles from reputable journals, and 14 parameters were identified based o

Extracellular ATP targets Arabidopsis RIBONUCLEASE 1 to suppress mycotoxin stress-induced cell death.

Extracellular ATP is a purinergic signal with important functions in regulating plant growth and stress-adaptive responses, including programmed cell death. While signalling events proximate to receptor activation at the plasma membrane have been characterised, downstream protein targets and the mechanism of cell death activation/regulation are unknown. We designed a proteomic screen to identify ATP-responsive proteins in Arabidopsis cell cultures exposed to mycotoxin stress via fumonisin B1 (FB1) application. Arabidopsis RIBONUCLEASE 1 (RNS1) was identified by the screen, and transgenic plant

1H-NMR metabolomics response to a realistic diet contamination with the mycotoxin deoxynivalenol: Effect of probiotics supplementation.

Low-level contamination of food and feed by deoxynivalenol (DON) is unavoidable. We investigated the effects of subclinical treatment with DON, and supplementation with probiotic yeast Saccharomyces cerevisiae boulardii I1079 as a preventive strategy in piglets. Thirty-six animals were randomly assigned to either a control diet, a diet contaminated with DON (3 mg/kg), a diet supplemented with yeast (4 × 109 CFU/kg), or a DON-contaminated diet supplemented with yeast, for four weeks. Plasma and tissue samples were collected for biochemical analysis,1H-NMR untargeted metabolomics, and histology.

Mycotoxin Deoxynivalenol Has Different Impacts on Intestinal Barrier and Stem Cells by Its Route of Exposure.

The different effects of deoxynivalenol (DON) on intestinal barrier and stem cells by its route of exposure remain less known. We explored the toxic effects of DON on intestinal barrier functions and stem cells after DON microinjection (luminal exposure) or addition to a culture medium (basolateral exposure) using three-dimensional mouse intestinal organoids (enteroids). The influx test using fluorescein-labeled dextran showed that basolateral DON exposure (1 micromolar (µM) disrupted intestinal barrier functions in enteroids compared with luminal DON exposure at the same concentration. In vitro and in vivo effects of a mycotoxin, deoxynivalenol, and a trace metal, cadmium, alone or in a mixture on the intestinal barrier.

Deoxynivalenol (DON), one of the most widespread mycotoxins in Europe, and cadmium (Cd), a widespread environmental pollutant, are common food contaminants. They exert adverse effects on different organs including kidney, liver, and intestine. The intestine is a common target of DON and Cd when they are ingested. Most studies have focused on their individual effects whereas their combined toxicity has rarely been studied. The aim of this study was thus to evaluate their individual and combined effects on the intestinal barrier function in vitro and in vivo. In vitro, Caco-2 cells were treated

A direct assessment of mycotoxin biomarkers in human urine samples by liquid chromatography tandem mass spectrometry.

Detection of mycotoxin biomarkers in urine of humans and animals provides a direct approach for assessing exposure to these mycotoxins as opposed to the indirect approach of food analysis, which in most cases is affected by the heterogeneity of the toxin in the food samples. Seven (7) mycotoxins and their metabolites (total 18 analytes) were selected and an LC-MS/MS method for their determination in human urine was developed and validated. The method consisted of direct analysis of two mycotoxin conjugates, deoxynivalenol-glucuronide and zearalenone-glucuronide without beta glucuronidase diges

Mycotoxin problem in Africa: current status, implications to food safety and health and possible management strategies.

Mycotoxins are toxic secondary metabolites of fungal origin and contaminate agricultural commodities before or under post-harvest conditions. They are mainly produced by fungi in the Aspergillus, Penicillium and Fusarium genera. When ingested, inhaled or absorbed through the skin, mycotoxins will cause lowered performance, sickness or death on humans and animals. Factors that contribute to mycotoxin contamination of food and feed in Africa include environmental, socio-economic and food production. Environmental conditions especially high humidity and temperatures favour fungal proliferation re

A TiO2@Au-Enhanced Portable Parallel Single-Electrode Electrochemiluminescence Array for High-Throughput Dual-Color Imaging Detection of Mycotoxins.

A single-electrode electrochemical system (SEES) shows outstanding advantages of high-throughput detection and portable imaging. Yet, its traditional series structure makes it difficult for some luminescent materials to achieve clear electrochemiluminescence (ECL) imaging, and it also suffers from uneven voltage distribution. Thus, altering the SEES structure and enhancing ECL performance are key challenges to achieve high-resolution ECL imaging. Here, this study innovatively designed a unique parallel ten-channel SEES device, introduced HOF-14 and Agm/ATT-Au NCs as dual-color ECL luminophors,

Metals, pesticides, and mycotoxins oral exposure via potato chips ingestion: A Monte Carlo-simulation risk assessment for Iranian consumers.

This study assesses the risk associated with oral exposure to metals, pesticides, and mycotoxins through the consumption of potato chips among Iranian consumers using Monte Carlo simulation. At the 50th, 80th, and 90th centiles, the calculated non-carcinogenic Hazard Index (HI) indicated no evident risk for adults but a moderate risk for children concerning oral exposure to these substances. Under carcinogenic scenario, for adults, the mean Incremental Life Time Cancer Risk (ILCR) for arsenic (As) and cadmium (Cd) at these centiles exceeded 10-4, indicating potential regulatory concerns under

Comparative analysis of metals, pesticides, mycotoxins, microbial contaminants and THC potency in illegal and regulated cannabis inflorescences in Canada.

BACKGROUND: Following the legalization of recreational cannabis in Canada under the Cannabis Act (2018), regulatory frameworks were implemented to ensure product safety, quality, and consistency within the legal market. Previous studies revealed significantly greater pesticide contamination in illegal cannabis than in legal products. In light of these findings, the present study expands contaminant surveillance to include the quantification of Δ⁹-tetrahydrocannabinol (THC) and a broader range of potential toxicants, including metals, pesticides, mycotoxins, and microbial agents. METHODS: Fifty

Preparation and Application of Magnetic Microporous Organic Networks for Rapid Adsorption Enrichment of Multiple Mycotoxins in Complex Food Matrices.

Mycotoxins commonly contaminate grains and traditional Chinese medicinal materials, posing serious health risks to humans and animals. To address this issue, a magnetic microporous organic network (MMON) was synthesized via an in situ growth method and Sonogashira-Hagihara coupling for the simultaneous adsorption of seven mycotoxins, followed by UPLC-MS/MS detection. The optimized MMON featured a high surface area, uniform micropores, and rapid magnetic separation within 5 s. Structural and compositional analyses confirmed its tailored architecture, while DFT calculations revealed a pore confi

Short-term exposure to the mycotoxins zearalenone or fumonisins affects rumen fermentation and microbiota, and health variables in cattle.

Zearalenone (ZEN) and fumonisins (FUM) jeopardize fertility and health in cattle; yet, their toxigenic effects on rumen health and microbiota, both being crucial for animal health, are not clarified. This study determined the effects of a short-term exposure to ZEN or FUM on the rumen ecosystem, and further evaluated acute implications on health parameters. Six cows were fed a basal diet with 40% grain (dry matter basis) and exposed to either 5 mg of ZEN or 20 mg of FUM daily for two consecutive days each, separated by a 7-days washout period. The exposure to ZEN or FUM led to a reduction of L

Fluorometric lateral flow immunoassay for simultaneous determination of three mycotoxins (aflatoxin B1, zearalenone and deoxynivalenol) using quantum dot microbeads.

A fluorometric lateral flow immunoassay (LFA) is described for the simultaneous determination of the mycotoxins aflatoxin B1 (AFB1), zearalenone (ZEN) and deoxynivalenol (DON). The method is based on the use of CdSe/SiO2 quantum dot microbeads (QBs) with a mean diameter of 106 nm. These have strong red luminescence (with excitation/emission peaks at 365/622 nm) which results in enhanced sensitivity. The QBs binding with monoclonal antibodies (mAbs) as the signal probes can react specifically with AFB1, ZEN and DON, respectively. There is an inverse correlation between the fluorescence signal i

Synergistic action of the nephrotoxic mycotoxins ochratoxin A and citrinin at nanomolar concentrations in human proximal tubule-derived cells.

Increased ochratoxin A (OTA) or citrinin (CIT) concentrations in food correlate with increased prevalence of tubule-interstitial nephropathy. We tested the hypothesis that co-exposure of human proximal tubule-derived epithelial cells (HK-2) to OTA and CIT promotes synergistic events indicative for inflammation, epithelial-to-mesenchymal-transition (EMT) or fibrosis. We measured markers of inflammation, EMT and fibrosis and investigated the role of MAP-kinases. Only concurrent but not individual exposure to OTA and CIT at nanomolar concentrations led to (i) an increase of TNF protein and mRNA,

Balkan endemic nephropathy and associated urinary tract tumours: a review on aetiological causes and the potential role of mycotoxins.

A series of publications in the 1950s described a kidney disease in Bulgaria, the former Yugoslavia and Romania that became known as Balkan endemic nephropathy (BEN). The disease was qualified by World Health Organisation (WHO) experts as 'progressive and very gradually developing renal failure with insidious onset.... The last stage shows marked fibrosis...'. BEN is characterized by tubular degeneration, interstitial fibrosis and hyalinization of glomeruli accompanied by enzymuria and impaired renal function without nephrotic syndrome. Later, an association between BEN and tumours of the kidn

Screening, risk assessment, and rapid detection of mycotoxins in edible herbs based on UPLC-MS/MS and FT-NIR.

Mycotoxin contamination in edible herbs poses substantial health risks and economic losses due to its accumulation. This study employed a modified QuEChERS-based UPLC-MS/MS method for qualitative screening of 19 mycotoxins in 1000 batches of 77 kinds of edible herbs. For three kinds of edible herbs with high positive rates, Sojae semen praeparatum (SSP), Lablab semen album (LSA), and Allii tuberosi semen (ATS), the QuEChERS parameters were optimized to quantify the ten most common mycotoxins, while Monte Carlo simulation was used to assess human health risks.
