Copy number variation regions in sheep resistant, resilient and susceptible to gastrointestinal nematode infection.

Gastrointestinal nematode (GIN) infection in sheep is a major cause of production losses, and the inappropriate use of anthelmintics has promoted drug resistance and environmental contamination, highlighting the need for alternative control strategies. Copy number variations (CNVs) are important structural variations that can influence gene expression, adaptation, and disease susceptibility. Therefore, this study detected copy number variation regions (CNVRs) in resistant, resilient, and susceptible Santa Ines sheep, and identified genes associated with resistance to GIN. The estimated breedin

Neuronally sensed oxygen drives behavior and development in human-infective, skin-penetrating nematodes.

Parasitic nematodes infect over a billion people worldwide and cause some of the most prevalent neglected tropical diseases1-5. Many of these parasites are skin penetrating and have both extra-host life stages that inhabit host feces and surrounding soil, and intra-host life stages that inhabit host niches such as skin, vasculature, and intestine2,6-8. Across life stages, these parasites encounter oxygen (O2) levels that range from ~21% at the soilair interface to near-anaerobic levels in the host intestine9-12. However, whether parasitic nematodes detect and respond to changes in O2 levels wa

Immune response of root-knot nematode-infected tomato is improved by biogenic copper nanoparticles.

"Green" synthesized copper nanoparticles from hemp plant waste (Cu-G-NPs) identified as brochantite, together with chemically synthesized copper nanoparticles (Cu-C-NPs), and copper sulfate (CuSO4) were tested as foliar spray against Meloidogyne incognita in tomato (Solanum lycopersicum). The materials were evaluated in two separate greenhouse experiments designed to assess their effectiveness at the different temperatures: one under warmer growth conditions and another stimulating cold stress. In both experiments treatments were applied at 200 mg/L of Cu and effects were assessed over a 45-day

Effects of zinc oxide nanoparticle supplementation in young sheep naturally infected with gastrointestinal nematodes.

The experiment aimed to evaluate the effects of daily supplementation with 150 mg of ZnO-NP on performance, hematological and immunological responses of lambs naturally infected with GIN under grazing conditions. Given the field-based approach adopted, the study was designed as an exploratory trial to generate preliminary evidence and hypotheses for future controlled experiments, acknowledging that individual feed intake was not measured and, therefore, the effect of ZnO-NP cannot be fully isolated from variations in voluntary concentrate intake. Each animal of one group (n = 14) was daily sup

Maternal gastrointestinal nematode infection enhances spatial memory of uninfected juvenile mouse pups.

The developing brain is particularly vulnerable to factors including maternal infection during pregnancy. Establishment of neural networks critical for memory and cognition begins during the perinatal period, when Heligmosomoides bakeri, a gastrointestinal (GI) nematode restricted to the maternal mouse intestine, has been shown to upregulate expression of long-term potentiation genes in the young rodent pup brain. We explored the impact of maternal infection during pregnancy and early lactation on the spatial behavior of uninfected male and female juvenile mice. Pre-weaned pups of H. bakeri in

The Imaginal Disc Growth Factors 2 and 3 participate in the Drosophila response to nematode infection.

The Drosophila imaginal disc growth factors (IDGFs) induce the proliferation of imaginal disc cells and terminate cell proliferation at the end of larval development. However, the participation of Idgf-encoding genes in other physiological processes of Drosophila including the immune response to infection is not fully understood. Here, we show the contribution of Idgf2 and Idgf3 in the Drosophila response to infection with Steinernema carpocapsae nematodes carrying or lacking their mutualistic Xenorhabdus nematophila bacteria (symbiotic or axenic nematodes, respectively). We find that Idgf2 an

Gastrointestinal nematodes infections and anthelmintic resistance in grazing sheep in the Eastern Inner Mongolia in China.

Gastrointestinal nematodes (GIN) are a crucial restraint to grazing sheep production worldwide. This study was conducted to determine the infections and anthelmintic resistance (AR) of GIN in pasture-based sheep in the Eastern Inner Mongolia, China. GIN eggs were tested from 600 grazing sheep feces of 10 farms using saturated saline flotation method and McMaster's method. The egg hatch test (EHT) and the faecal egg count reduction test (FECRT) were used to evaluate resistance of GIN to anthelmintics. We found that the average infection rate was 79.2% (range: 45%-100%). The grand mean faecal eg

Resiniferatoxin modulates the Th1 immune response and protects the host during intestinal nematode infection.

In the early stage of the intestinal phase of Trichinella spiralis infection, the host triggers a Th1-type immune response with the aim of eliminating the parasite. However, this response damages the host which favours the survival of the parasite. In the search for novel pharmacological strategies that inhibit the Th1 immune response and assist the host against T. spiralis infection, a recent study showed that resiniferatoxin had anti-inflammatory activity contributed to the host in T. spiralis infection. In this study, we evaluated whether RTX modulates the host immune response through the i

Interactions Between Nutrition and Infections With Haemonchus contortus and Related Gastrointestinal Nematodes in Small Ruminants.

Interactions between host nutrition and feeding behaviour are central to understanding the pathophysiological consequences of infections of the digestive tract with parasitic nematodes. The manipulation of host nutrition provides useful options to control gastrointestinal nematodes as a component of an integrated strategy. Focussed mainly on the Haemonchus contortus infection model in small ruminants, this chapter (1) illustrates the relationship between quantitative (macro- and micro-nutrients) and qualitative (plant secondary metabolites) aspects of host nutrition and nematode infection, and

Gnathostomiasis: An Emerging Infection of Raw Fish Consumers in Gnathostoma Nematode-Endemic and Nonendemic Countries.

BACKGROUND: Gnathostomiasis, a helminthic infection commonly reported in Southeast Asia and Latin America, may follow consumption of raw seafood infected with muscle-encysted larvae of Gnathostoma species nematodes. As a result of increasingly exotic tastes for local ethnic dishes, including raw seafood, some regions outside of gnathostome-endemic areas import live species for raw consumption. This may facilitate imported human gnathostomiasis or potentially the establishment of this zoonosis in formerly nonendemic regions. Traveling to a gnathostome-endemic area is no longer a criterion for d

Gastrointestinal nematode infection and performance of weaned stocker calves in response to anthelmintic control strategies.

Gastrointestinal nematode (GIN) parasite control recommendations are in a state of flux because of the increase in anthelmintic resistant cattle parasites, such as Cooperia spp. In addition, Cooperia spp. infection is typically high in warm-season grass pastures and can affect growth performance of grazing stocker calves in the Gulf Coast Region. This study evaluated the effects of moxidectin pour-on, oxfendazole oral suspension, or a combination of the two given at separate times on infection and performance of weaned beef calves grazing summer forages. Steers (n=42) and heifers (n=31) were s

Strongyloides stercoralis age-1: a potential regulator of infective larval development in a parasitic nematode.

Infective third-stage larvae (L3i) of the human parasite Strongyloides stercoralis share many morphological, developmental, and behavioral attributes with Caenorhabditis elegans dauer larvae. The 'dauer hypothesis' predicts that the same molecular genetic mechanisms control both dauer larval development in C. elegans and L3i morphogenesis in S. stercoralis. In C. elegans, the phosphatidylinositol-3 (PI3) kinase catalytic subunit AGE-1 functions in the insulin/IGF-1 signaling (IIS) pathway to regulate formation of dauer larvae. Here we identify and characterize Ss-age-1, the S. stercoralis homo

Wastewater quality and the risk of intestinal nematode infection in sewage farming families in hyderabad, India.

Use of sewage or wastewater in agriculture is becoming increasingly common as a result of a global water scarcity. Intestinal nematode infections have been identified as the main health risk associated with this practice. To protect consumer and farmer health, the World Health Organization (WHO) has established an intestinal nematode water quality standard. However, because of a lack of well-designed studies, the validity of this guideline is questioned. This report presents the findings of a study on the risk of intestinal nematode infections in farming families occupationally exposed to untr…

Immune-mediated alteration in gut physiology and its role in host defence in nematode infection.

Activation of the mucosal immune system of the gastrointestinal tract in nematode infection results in altered intestinal physiology, which includes changes in intestinal motility and mucus production. These changes are considered to be under direct immunological control rather than a non-specific consequence of the inflammatory reaction to the infective agent. However, little is known about the immunological basis for the changes in intestinal physiology accompanying nematode infection, or the precise role of these changes in host defence, which remains an important area to explore. In this r

Neutral endopeptidase (EC 3.4.24.11) downregulates the onset of intestinal inflammation in the nematode infected mouse.

BACKGROUND AND AIMS: Substance P (SP) release from sensory nerves induces neurogenic inflammation. Neutral endopeptidase (NEP) degrades SP, thereby limiting its proinflammatory effects. Intestinal inflammation following Trichinella spiralis infection markedly downregulates NEP, resulting in diminished SP degradation, with unknown functional consequences. We hypothesised that diminished expression of NEP would exacerbate T spiralis induced enteritis. METHODS: NEP knockout (NEP-/-) and wild-type (NEP+/+) mice were infected with T spiralis and studied at 6, 12, 24, and 48 hours post infection (P

Temperature is a cue for gene expression in the post-infective L3 of the parasitic nematode Brugia pahangi.

The temporal expression pattern of two genes, Bp-cdd and Bp-S3, was studied at defined points throughout the life cycle of Brugia pahangi. Both mRNAs were up-regulated to coincide with the transition of the L3 from the vector to the mammalian host. Bp-cdd was expressed almost exclusively in the post-infective (p.i.) L3 and L4 stages of the life cycle while Bp-S3 was also expressed in adult worms, but at a much lower level than in the larval stages. Immunogold labelling with an antiserum raised to the recombinant Bp-CDD localised the native antigen to the hypodermis in the p.i. L3 and L4. Speci

Protective mechanisms against the intestinal nematode Strongyloides venezuelensis in Schistosoma japonicum-infected mice.

Mice infected with Schistosoma japonicum were resistant to the intestinal nematode, Strongyloides venezuelensis. The numbers of adult S. venezuelensis recovered from mice were significantly decreased when infections were given from 6 weeks after S. japonicum infection. Larval recovery from the lungs showed that significant numbers of subcutaneously inoculated S. venezuelensis larvae were eliminated by 3 days in S. japonicum-infected mice (P < 0.0001), while histology revealed that this was associated with massive eosinophilic infiltration in the lungs. In addition, adult S. venezuelensis worms
