Ultrastructural changes in the nephron and renal protein cleaving function in suckling rabbits with experimental cholera.

Immature nephrons and the presence of a proteolytic system mediating extracellular protein digestion in the epithelium of proximal tubules are characteristic features of renal cortical ultrastructure in intact 10-12-day-old suckling rabbits. Cholera infection is accompanied by intensive cleavage of exogenous protein, which starts in podocytes and is completed in the cytoplasm of the proximal tubule epitheliocytes. Overload to nephron associated (in addition to urine production) with intensive extracellular hydrolysis impairs nephrothelium reactivity and increases its vulnerability to cholera e…

A simple, nonradioactive method for evaluating single-nephron filtration rate using FITC-inulin.

The determination of inulin concentration in nanoliter fluid samples is fundamental to micropuncture investigations of renal function, and this is generally accomplished through the use of radioisotopes. We report here a simple and reliable alternative to the use of radioisotopes that employs FITC-labeled inulin. Samples containing FITC-inulin are stored between oil columns in constant-bore microcapillary tubes, which are then used as cuvettes to determine fluorescence on a microscope fluorometer. Standard curves were generated and found to be linear, with correlation coefficients (R) exceedin

Expression and function of calcineurin in the mammalian nephron: physiological roles, receptor signaling, and ion transport.

Protein phosphorylation is central to the regulation of sodium transport and other cellular processes in the nephron. Complex interactions between protein kinases and phosphatases catalyze the reversible phosphorylation of ion transporting proteins on the apical and basolateral surfaces of renal epithelia. Although the role of protein kinases in regulating sodium transport has been extensively studied, the function of phosphatases in the nephron is less well understood. Calcineurin is a serine-threonine phosphatase that was shown to be the target of cyclosporin A (CsA) and FK-506 in lymphocyte

Functional state of the nephron and diuretic dose-response--rationale for low-dose combination therapy.

The functions of the different nephron segments follow changes in the effective arterial blood volume and the extracellular fluid volume. In … ease in its fractional excretion as the glomerular filtration rate is reduced. The remaining intact nephrons exhibit a relatively increased response to diuretics. The efficacy of loop diuretics in renal failure can be optimized by combination with thiazides. The latter prevent early distal tubular hyperreabsorption following diuretic-induced blockade of so …

Distal nephron function in Bartter's syndrome: abnormal conductance to chloride in the cortical collecting tubule?

Five patients with the clinical patterns of Bartter's syndrome underwent a series of clearance studies in order to characterize the underlying tubule defect. Free water generation during maximal water diuresis (CH2O), expressed as percentage of the distal delivery (CH2O + CCl), was lower in the patients (72.5 +/- 3.2%) than in controls (84.4 +/- 5.5, p < 0.0001). During maximal water diuresis and furosemide administration (40 mg i.v. as bolus), NaCl reabsorption along the diluting nephron segments could be separated into 2 components, that occurring in the loop of Henle (DRNaHL) and that occur

Identification of a regulated Na/K/Cl cotransport system in a distal nephron cell line.

Lack of an adequate cell model has limited investigation of Na/K/Cl cotransporter regulation in the kidney. Using A6 cells, an amphibian distal renal cell line, we observed that 63% of rubidium uptake in confluent A6 monolayers was ouabain-insensitive. Ouabain-insensitive rubidium uptake was inhibited in a dose-dependent fashion by furosemide (IC50 6.6 microM) or bumetanide (IC50 1.7 microM). Kinetic studies confirmed that furosemide-sensitive rubidium uptake had features consistent with cotransporter activity in other cell lines. Furthermore, specific binding of [3H]bumetanide occurred with a

[The systemic disorder of connective tissue metabolism and hypoplasia of the nephron elements in hereditary nephritis in children].

Clinical biochemistry and morphological methods were employed to examine 25 children aged 3 to 15 years with hereditary nephritis. Measurements were made of morphological alterations in renal biopsy specimens, excretion with urine of connective tissue metabolites (hydroxyproline, hydroxylysine glycosides, glycosaminoglycans), the level of the same metabolites and characteristic features of the cellular growth of skin fibroblasts in culture. The early stages of nephritis development were marked by hypoplasia of nephron elements, followed by dystrophy and destruction of its ultrastructural eleme

Assessment of the nephron segments involved in post-obstructive diuresis in man, using lithium clearance.

The phenomena of post-obstructive diuresis and natriuresis have been studied using the lithium clearance technique in 10 patients with high pressure chronic retention. Following relief of obstruction there was a significant increase in both sodium and water excretion. There was a coincident reduction in the fractions of sodium and water reabsorbed in both proximal and distal nephron segments. This study demonstrates for the first time that following relief of chronic obstructive uropathy in man, changes in sodium and water excretion are due to altered handling in both proximal and distal nephr

[Pathogenetic significance of lipid peroxidation in damage of the proximal segment of the nephron in acute Masugi's nephritis].

During the acute experimental nephritis a decrease of reabsorption by proximal tubules is combined with the activation of the lipid peroxidation into the renal cortex without changes in the activation of lysosomal enzyme of acid phosphatase and cathepsin D. Ionol, an inhibitor of oxygen products, exerts a protective action on the renal function and reabsorption by proximal tubules, decreasing concentration of the malondialdehyde into the renal cortex.

LRBA organizes distinct vesicular trafficking systems in distal nephron segments for water and sodium conservation.

Lipopolysaccharide-responsive and beige-like anchor protein (LRBA) deficiency is a rare genetic disorder characterized by immune dysregulation. The immune checkpoint molecule cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) fails to perform proper membrane trafficking in the absence of LRBA. In addition to immune cells, LRBA localizes to intracellular vesicles in various epithelial cells; however, its physiological roles have not been accurately deciphered. It was observed in this study that LRBA facilitates water and sodium transport by promoting vesicular trafficking of aquaporin-2 (AQP2

Nephron-Sparing Surgery for Unilateral Non-Syndromic Wilms Tumor in a 9-Day-Old Neonate.

Neonatal renal tumors are uncommon and generally associated with an excellent prognosis. We report the youngest patient to date with unilateral non-syndromic Wilms tumor successfully treated with nephron-sparing surgery rather than radical nephrectomy, which is the current standard of care. The patient subsequently received risk-adapted chemotherapy. At 18 months of follow-up, he remains asymptomatic and with symmetric differential renal volumes, consistent with preserved bilateral renal function. This case suggests that nephron-sparing surgery, when technically feasible, may represent a safe

Pathological Insights into Non-Neoplastic Renal Parenchyma in Wilms Tumor: Implications for Nephron-Sparing Surgery.

INTRODUCTION: This study aimed to evaluate the non-neoplastic renal parenchyma in Wilms tumor (WT) and investigate its impact on nephron-sparing surgery (NSS). MATERIALS AND METHODS: The non-neoplastic renal parenchyma of WT patients was prospectively collected for pathological examination. The histology of non-neoplastic renal parenchyma was assessed from two perspectives: nephrogenic rests (NRs) and nephrons. RESULTS: A total of 46 non-neoplastic renal parenchyma specimens were collected from 42 WT patients. The surgeons assessed the median proportion of non-neoplastic renal parenchyma as …

Nephron-sparing surgery for primary mucinous adenocarcinoma of renal pelvis in horseshoe kidney: A case report.

RATIONALE: Mucinous adenocarcinoma of the renal pelvis is extremely rare in malignancy of the renal pelvis, and <100 cases have been reported so far. However, horseshoe kidneys appear to be more prone to this mucinous gland metaplasia. Due to the lack of a specific diagnostic basis, it is difficult to confirm the diagnosis before surgery, which is easy to cause misdiagnosis. PATIENT CONCERNS: We report a 64-year-old male patient who found a cystic mass in the left kidney during physical examination. CT examination showed a horseshoe kidney with a cystic mass 9.5 × 8.0 cm in front of the left

Inflammatory myofibroblastic tumor of the kidney in patient with nephron-sparing surgery. Case report and review of the literature.

According to the WHO classification, mesenchymal tumors of the kidney are divided into mesenchymal tumors occurring mainly in adults and children. Accumulating evidence suggests that renal mesenchymal tumors represent a group of histologically heterogeneous diseases. We are reporting the case of a 58-year-old man with a history of nephron-sparing surgery due to renal cell carcinoma in 2014 in his left kidney. He was urgently admitted to the hospital due to left-sided nephrotoxic and anuria. Computed tomography (CT) imaging was performed and revealed irregular tumor mass. He underwent left-side

Effects of Environmental Conditions on Nephron Number: Modeling Maternal Disease and Epigenetic Regulation in Renal Development.

A growing body of evidence suggests that low nephron numbers at birth can increase the risk of chronic kidney disease or hypertension later in life. Environmental stressors, such as maternal malnutrition, medication and smoking, can influence renal size at birth. Using metanephric organ cultures to model single-variable environmental conditions, models of maternal disease were evaluated for patterns of developmental impairment. While hyperthermia had limited effects on renal development, fetal iron deficiency was associated with severe impairment of renal growth and nephrogenesis with an all-p
