Plant-driven microbiome reorganization sustains multifunctionality of iron-biochar constructed wetlands under chronic PFOA exposure.

Plant capacity to regulate treatment stability and ecological function of iron-biochar (IC) based constructed wetlands (CWs) operated under … vegetation-associated changes in enzyme activity, including higher dehydrogenase, phosphatase, and nitrate reductase activities during specific periods or in specific layers, greater EPS production, and improved biofilm stability with reduced bio-clogging risk. Microorganism-plant interaction also influenced greenhouse gases (GHG) emission, in which CO2 fluxes wer …

Mechanistic insights into iron cycling-driven nitrogen removal from biogas slurry via coupled iron-based denitrification and Feammox.

In this study, ferrous-based denitrification was combined with Feammox (Fe(III) reduction coupled with anaerobic ammonium oxidation) to trig … ities were also increased by 7.69%-16.08%. Metagenomic analysis showed that the NO3- group had more nitrate reductase genes but fewer downstream denitrification genes than the control group, indicating that NO2- accumulated as a key intermediate. NO3- could not directly oxidize Fe(II), and no nitrate-dependent Fe(II)-oxidizing microorganisms were detected. Moreover …

Efficacy and Possible Mechanism(s) of Action of Gallium Tetraphenylporphyrin Nanoparticles against HIV-TB Coinfection in an In Vitro Granuloma Structure Model.

Coinfection of Mycobacterium tuberculosis (Mtb) and human immunodeficiency virus-1 (HIV) is a significant public health concern. Treatment i … es the level of reactive oxygen species and inhibits catalase in Mtb. A significant increase in Mtb nitrate reductase activity was also observed when Mtb was incubated with GaTP and sodium nitrate. Overall, increased oxidative stress and nitrite levels induced by GaTP are consistent with the possibility that GaTP inhibits Mtb aerobic respiration, which leads to inc …

Reduction of nitrosative stress by methane: Neuroprotection through xanthine oxidoreductase inhibition in a rat model of mesenteric ischemia-reperfusion.

Our aim was to characterize the main components of the nitrosative response with quantitative changes of the nitrergic myenteric neurons in … duodenal nitrergic neuron ratio was re-established. The inhibitory effect of methane on XOR-linked nitrate reductase activity was also demonstrated in vitro. With segment-specific microcirculatory alterations, the risk for nitrosative stress is highest in transiently hypoxic tissues with high endogenous XOR activities. The XOR-inhibitory effect of methane can redu …

Enhanced NO production during Mg deficiency and its role in mediating red blood cell glutathione loss.

The effect of dietary Mg deficiency on nitric oxide (NO) production and its role in mediating oxidative depletion of red blood cell (RBC) glutathione in rats were investigated. Male Sprague-Dawley rats were placed on Mg-deficient or Mg-sufficient diets for up to 3 wk. Plasma nitrate plus nitrite levels, determined by the Escherichia coli reductase/Griess reagent procedures, increased 1.7-fold during the 1st wk and increased 2- to 2.4-fold during the 2nd and 3rd wk on the Mg-deficient diet. In association, substantial losses (approximately 50%) of RBC glutathione occurred during the 2nd and 3rd

Gasotransmitters (NO and HCN) alter phytoglobin-NO cycle during dormancy alleviation of apple (Malus domestica Borkh.) embryos.

Nitric oxide (NO) and hydrogen cyanide (HCN) are N-containing gasotransmitters of bimodal action depending on the concentration. Both gases exhibit a beneficial effect on seed dormancy removal. The aim of this work was to compare the mode of action of NO and HCN on the elements of phytoglobin (Pgb)-NO cycle during seed dormancy alleviation (under aerobic conditions). Short-term fumigation of apple (Malus domestica Borkh.) embryos with HCN or NO increased the formation of reactive nitrogen species (RNS) in embryonic axes. The induction of nitrite/nitrate (NO2-/NO3-) signalling pathway, together

Auxin Acts Downstream of Ethylene and Nitric Oxide to Regulate Magnesium Deficiency-Induced Root Hair Development in Arabidopsis thaliana.

This study examines the association of auxin with ethylene and nitric oxide (NO) in regulating the magnesium (Mg) deficiency-induced root hair development in Arabidopsis thaliana. With Mg deficiency, both ethylene and NO promoted the elevation of root auxin levels in roots by inducing the expression of AUXIN-RESISTANT1 (AUX1), PIN-FORMED 1 (PIN1) and PIN2 transporters. In turn, auxin stimulated ethylene and NO production by activating the activities of 1-aminocyclopropane-1-carboxylate (ACC) oxidase (ACO), ACC synthase (ACS), nitrate reductase (NR) and NO synthase-like (NOS-L). These processes…

Mycobacterium tusciae sp. nov.

A new, slow-growing, scotochromogenic mycobacterium was isolated from a lymph node of an immunocompromised child and subsequently from tap water and from a respiratory specimen of a patient with chronic fibrosis. Alcohol-acid-fastness, lipid patterns and the G + C content clearly support the placement of this organism in the genus Mycobacterium. The isolates grew very slowly at temperatures ranging from 25 to 32 degrees C and showed activities of nitrate reductase, catalase, urease, arylsulfatase and Tween 80 hydrolysis. The organism was susceptible to all antimycobacterial drugs tested. The 1…

[Corynebacterium group D2. Clinical study, biochemical identification and antibiotic sensitivity].

The comparative study of 44 isolates of Corynebacterium group D2, from urine, most frequently, shows the pathogenic role of these bacteria in urinary tract infection, with or without urinary stones. These microorganisms have an opportunistic behaviour in other non-urinary sites, and become pathogen in immunosuppressed conditions. The rapid tests as urease, glucose acidification, nitrate reductase, associated with multiple resistance to antibiotics (beta-lactams and aminosides) identify easily Corynebacterium group D2, from 48 h cultures under CO2 conditions. The results of MIC determination of…

Nucleic acid and protein synthesis in discs cut from mature leaves of Nicotiana tabacum L. and cultured on nutrient agar with and without kinetin.

The effect of kinetin on aspects of the metabolism of discs cut from mature leaves of Nicotiana tabacum and cultured in the light on agar containing mineral salts and sucrose was studied. In the first few days of culture there was a rapid decline in chlorophyll content. Discs treated with kinetin in the light began to resynthesise chlorophyll after 3-4 days and this was correlated with chloroplast replication. Kinetin promoted chloroplast replication but was not always essential. An increase in fresh weight also occurred, due mainly to cell expansion. Nitrate reductase activity increased rapid…

Collaborative metabolic curation of an emerging model marine bacterium, Alteromonas macleodii ATCC 27126.

Inferring the metabolic capabilities of an organism from its genome is a challenging process, relying on computationally-derived or manually … ATCC 27126. We postulate that demonstrated growth on nitrate as sole nitrogen source proceeds via a nitrate reductase pathway that is a hybrid of known pathways. Our evidence highlights the value of joint and interactive curation efforts, but also shows major knowledge gaps regarding Alteromonas metabolism. The manually-curated metabolic reconstruction is available …

Occurrence and diversity of the oxidative hydroxyhydroquinone pathway for the anaerobic degradation of aromatic compounds in nitrate-reducing bacteria.

The nitrate-reducing betaproteobacteria Azoarcus anaerobius and Thauera aromatica AR-1 use an oxidative mechanism to anaerobically degrade resorcinol and 3,5-dihydroxybenzoate (3,5-DHB), respectively, rendering hydroxyhydroquinone as intermediate. The first pathway step is performed by a dimethylsulphoxide-reductase family hydroxylase. The gene cluster coding for the pathway is homologous in these strains. Only these two Rhodocyclales are known to follow this anaerobic pathway, and nothing is known about its distribution in prokaryotes. To determine the relevance and diversity of this strategy

Dissimilatory nitrate reduction to ammonium-induced false stability masks dysfunctional Candidatus Brocadia failure during ANAMMOX start-up from municipal activated sludge.

Enriching anaerobic ammonium oxidation (ANAMMOX) bacteria from conventional municipal activated sludge remains difficult despite reported feasibility, particularly without highly enriched anammox cultures (HeAnAC). This study examined ANAMMOX start-up in an anaerobic-membrane-bioreactor fed ammonium (NH4+) and nitrite (NO2-) under organic carbon-free conditions. Initial nitrogen removal (NH4+: 78%; NO2-: 98%) declined sharply after day 70 (<11% and <28%, respectively), coinciding with stable hydrazine synthase gene (hzs) abundance, elevated DNRA nitrite reductase gene (nrfA) abundance, and hig

A potential novel Brucella species isolated from mandibular lymph nodes of red foxes in Austria.

The wild red fox (Vulpes vulpes) is a known indicator species for natural foci of brucellosis. Here, we describe phenotypic and molecular characteristics of two atypical Brucella strains isolated from two foxes hunted 2008 in Eastern Austria. Both strains agglutinated with monospecific anti-Brucella A serum and were positive in ELISA with monoclonal antibodies directed against various Brucella lipopolysaccharide epitopes. However, negative nitrate reductase- and negative oxidase-reaction were atypical traits. Affiliation to the genus Brucella was confirmed by 16S rRNA gene sequencing and by de…

In vivo nephrotoxicity induced in mice by chromium(VI). Involvement of glutathione and chromium(V).

The role of glutathione (GSH) and chromium (V) in chromium (VI)-induced nephrotoxicity in mice was investigated at 24 h after K2Cr(VI)2O7 ip injection. Nephrotoxicity was assessed by measurements of relative kidney weight and serum urea nitrogen. Cr(VI) nephrotoxicity was accompanied by decreased renal GSH and glutathione reductase (GSSG-R) levels. Pretreatment with buthionine sulfoximine, an inhibitor of GSH biosynthesis, enhanced Cr(VI)-induced nephrotoxicity, and remarkably diminished kidney GSH and GSSG-R levels. In contrast, pretreatment with glutathione methyl ester, a GSH-supplying agen
