Centrifugal projections to the main olfactory bulb revealed by transsynaptic retrograde tracing in mice.

A wide range of evidence indicates that olfactory perception is strongly involved in food intake. However, the polysynaptic circuitry linking the brain areas involved in feeding behavior to the olfactory regions is not well known. The aim of this article was to examine such circuits. Thus, we described, using hodological tools such as transsynaptic viruses (PRV152) transported in a retrograde manner, the long-distance indirect projections (two to three synapses) onto the main olfactory bulb (MOB). The ß-subunit of the cholera toxin which is a monosynaptic retrograde tracer was used as a contro

MRI findings in Kallmann syndrome.

Kallmann syndrome (KS) is a neuronal migration disorder characterised by hypogonadotrophic hypogonadism and anosmia or hyposmia. Five patients with clinical findings suggestive of KS were evaluated with MRI. All patients had abnormalities of olfactory system. Olfactory bulbs were absent in all patients. Olfactory sulci were absent in 3 patients and hypoplastic in 2 patients. Anterior pituitary was hypoplastic in two patients. The MRI findings in KS are characteristic and MRI is a useful adjunct to the diagnosis of KS.

Effect of anosmia on reproduction in male and female wolves (Canis lupus).

Anosmia was produced in two female and three male wolves by transection of the olfactory peduncle and was confirmed by their inability to detect meat, urine, feces, anal-gland secretions, and fish emulsion. All operated animals continued to investigate the environment with their noses, to interact normally with other pack members, and to feed at levels which maintained presurgical body weights. No effect was found on reproductive physiology (females: estradiol or progesterone concentrations, ovulation, pregnancy or parturition; males: testosterone, testicular recrudescence or sperm numbers, mo

Manipulations of the olfactory circuit highlight the role of sensory stimulation in regulating sleep amount.

STUDY OBJECTIVES: While wake duration is a major sleep driver, an important question is if wake quality also contributes to controlling sleep. In particular, we sought to determine whether changes in sensory stimulation affect sleep in Drosophila. As Drosophila rely heavily on their sense of smell, we focused on manipulating olfactory input and the olfactory sensory pathway. METHODS: Sensory deprivation was first performed by removing antennae or applying glue to antennae. We then measured sleep in response to neural activation, via expression of the thermally gated cation channel TRPA1, or i

Distribution of aminopeptidase P like immunoreactivity in the olfactory system and brain of frog, Microhyla ornate.

The enzyme aminopeptidase P (AP-P) is encountered in diverse vertebrate and invertebrate phyla and is known to act on proteins and peptides by releasing their N-terminal amino acid when the penultimate amino acid is proline. The present study is the first attempt at visualizing distribution of this polypeptide in the brain of a vertebrate species. The distribution of this enzyme was studied immunocytochemically in the forebrain of frog Microhyla ornata using antisera directed against cytosolic aminopeptidase P (DAP-P) of Drosophila melanogaster. Receptor cells in the olfactory epithelium exhib

An experimental study of the connections of the telencephalon in the rainbow trout (Oncorhynchus mykiss). I: Olfactory bulb and ventral area.

The fluorescent carbocyanine dye 1,1'-dioctadecyl 3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI) was used in fixed tissue to comprehensively analyze the connections of the olfactory bulbs and the different regions of the ventral (V) area of the telencephalic lobes (subpallium) of the rainbow trout. With this goal, DiI was applied to the different telencephalic nuclei and zones, as well as to the olfactory nerve, the olfactory bulb, the retina, and to several structures in the diencephalon and brainstem of juvenile trout. The olfactory bulbs maintain reciprocal connections with several

The cranial nerves of the Senegal bichir, Polypterus senegalus [osteichthyes: actinopterygii: cladistia].

The organization of the roots, ganglia and peripheral distribution of the rami of the cranial nerves of larval and juvenile Senegal bichirs was examined with a wide range of techniques, including gross dissection and histological preparations. The profundal nerve of bichirs is completely separate from the trigeminal nerve and innervates the skin overlying the orbit and snout, and there is no evidence that a superficial ophthalmic ramus of the trigeminal nerve exists. Bichirs appear to retain all six pairs of lateral line nerves that characterized the earliest gnathostomes, but the sensory gang

Uptake of 203Hg2+ in the olfactory system in pike.

Inorganic mercury (203Hg2+) was applied to the olfactory chambers or was given i.v. to pike (Esox lucius) and the uptake of the metal in the olfactory system and the brain was examined by autoradiography and gamma spectrometry. Application of 203Hg2+ in the olfactory chambers resulted in an accumulation of the metal in the olfactory nerves and the anterior parts of the olfactory bulbs of the brain. The levels of 203Hg2+ in other brain areas, such as the telencephalon, the optic tecti and the cerebellum, remained low. Application of 203Hg2+ in only one olfactory chamber resulted in an uptake of

Olfactory loss in chronic rhinosinusitis is associated with neuronal activation of c-Jun N-terminal kinase.

BACKGROUND: Olfactory inflammation in chronic rhinosinusitis (CRS) is associated with cytokines that may result in the death of olfactory sensory neurons. The principal signaling molecules involved in the apoptotic pathway are c-Jun N-terminal kinases (JNK). Although the JNK pathway has emerged as a key player in programmed cell death in neuroinflammation, its specific role in CRS-associated olfactory loss has not been thoroughly investigated. METHODS: JNK activation was studied in human tissue samples from 9 control and 11 CRS patients by immunohistochemical staining for phosphorylated c-Jun

Control of innate olfactory valence by segregated cortical amygdala circuits.

Animals exhibit innate behaviors that are stereotyped responses to specific evolutionarily relevant stimuli in the absence of prior learning or experience. The posterolateral cortical amygdala (plCoA) mediates innate attraction and aversion to odor. Here, we sought to define the circuit features of plCoA that give rise to innate attraction and aversion to odor in mice. First, we examined odor-evoked responses in these areas and found sparse encoding of odor identity, but not valence. We next considered a topographic organization and found that optogenetic stimulation of the anterior and poster

Undoing of firing rate adaptation enables invariant population codes.

Neural adaptation supports coding efficiency by tuning responses to prevailing stimulus statistics. However, when information is represented by populations of neurons, adaptation of individual units could degrade behaviorally relevant signals. Here, we investigate how the fly olfactory system implements adaptation to background odors in olfactory receptor neurons (ORNs) and the consequences for combinatorial coding in downstream circuits. We show that adaptation of ORN firing rate is compensated at the axon terminal, where inhibitory presynaptic feedback supports background-invariant calcium r

Comparison between olfactory bulb and olfactory tract implantations through an endoscopic supraorbital keyhole approach: a cadaveric study.

PURPOSE: Since the COVID-19 pandemic, olfactory loss has been recognized as a highly prevalent condition that greatly impacts the quality of life. Similar to other sensory implants, the idea of an olfactory implant has emerged. Evidence indicates that electrical stimulation of specific olfactory structures can evoke smell sensations. However, debates on the most appropriate anatomical target and surgical technique for implantation are still ongoing. By extrapolating data from other surgical indications, transcranial approaches appear to carry a lower risk of cerebrospinal fluid leakage and inf

Origin of wiring specificity in an olfactory map revealed by neuron type-specific, time-lapse imaging of dendrite targeting.

How does wiring specificity of neural maps emerge during development? Formation of the adult Drosophila olfactory glomerular map begins with the patterning of projection neuron (PN) dendrites at the early pupal stage. To better understand the origin of wiring specificity of this map, we created genetic tools to systematically characterize dendrite patterning across development at PN type-specific resolution. We find that PNs use lineage and birth order combinatorially to build the initial dendritic map. Specifically, birth order directs dendrite targeting in rotating and binary manners for PNs

[Evaluating function and disorders of smell].

Firstly the review deals with the olfactometry after discussing the olfactory and trigeminal sensitivity of the sense of smell. The term olfactometry will be newly fixed concerning the present problems of odor analysis. Under clinical aspects the methods of subjective olfactometry are discussed and valued. Olfactory tests basing on registering several psychosomatic reflexes (e.g. cardial and/or respiratory frequencies) cannot be described as "objective". Rather methods are objective which record poststimulatory electrophysiological events at different steps of olfactory pathways. The electric …

Nervus terminalis in dogfish (Squalus acanthias, Elasmobranchii) carries tonic efferent impulses.

Recordings from the intact nervus terminalis with a hook electrode or from a stump of the divided nerve with a suction electrode show a tonic, irregular discharge of broad, low frequency spikes in ca. 4-6 units. These nerve impulses are efferent from the brain. The mean frequency of discharge is not influenced by various chemical, thermal, tactile, acoustic, photic, vibratory and electric field stimuli but is decreased by certain forms of mechanical stimuli, presumably acting on the lateral line organs of the lateral aspect of the head. We have not succeeded in recording from afferents. The ne

Olfactory information converges in the amygdaloid cortex via the piriform and entorhinal cortices: observations in the guinea pig isolated whole-brain preparation.

The amygdaloid cortex (AC) has reciprocal connections with the entorhinal cortex (EC) and also receives projections from the olfactory bulb and the piriform cortex (PC). To assess the possibility that the AC and EC represent functionally coupled structures in the olfactory stream of information, we investigated the propagation pattern of neural activity in olfactory cortices--PC, AC and EC--using optical recordings with voltage-sensitive dyes in the guinea pig in vitro isolated whole-brain preparation. We observed two distinct pathways that convey neural activation evoked by olfactory nerve st

[Cross-regulation between olfaction and energy homeostasis in obesity and type 2 diabetes: from mouse models to humans].

This review highlights the crucial role of olfaction in regulating energy homeostasis. Neurons in the olfactory system stimulate the limbic and hedonic pathways involved in food-related behaviors. These neurons are modulated by the body's energy metabolism status, which they, in turn, help to regulate. Olfactory sensitivity is reduced in obesity, while bariatric surgery restores olfactory function. In type 2 diabetes, the onset of olfactory dysfunction is predictive of cognitive decline. Finally, in a mouse model of obesity, local activation of glucagon-like peptide 1 (GLP-1) and its receptors
