Putative M2 muscarinic receptors of rat heart have high affinity for organophosphorus anticholinesterases.

The M2 subtype of muscarinic receptor is predominant in heart, and such receptors were reported to be located in muscles as well as in presynaptic cholinergic and adrenergic nerve terminals. Muscarinic receptors of rat heart were identified by the high affinity binding of the agonist (+)-[3H]cis-methyldioxolane ([3H]CD), which has been used to label a high affinity population of M2 receptors. A single population of sites (KD 2.74 nM; Bmax of 82 fmol/mg protein) was detected and [3H]CD binding was sensitive to the M2 antagonist himbacine but much less so to pirenzepine, the M1 antagonist. These

Therapeutic comparison of MitoTEMPO and rosmarinic acid as mitochondria targeted and non targeted antioxidants evaluated in rat sciatic nerve crush injury model.

Mitochondria are key organelles involved in various pathophysiological processes. As the primary site of reactive oxygen species (ROS) production, they represent a major target for many therapeutic agents. In this study, the therapeutic effects of a mitochondria-targeted antioxidant (MitoTEMPO, MT) and a conventional yet potent antioxidant compound (rosmarinic acid, RA) were compared in a rat model of sciatic nerve crush injury, focusing on nerve conduction parameters and histological structures. A total of 28 adult Wistar Albino rats were used in the study, with seven animals assigned to each

Genotoxicity of selected pesticides in the mouse bone-marrow micronucleus test and in the sister-chromatid exchange test with human lymphocytes in vitro.

Selected pesticides (aldicarb, 1,3-dichloropropene, methidathion, parathion, triadimefon, vinclozolin) were tested for their clastogenic and aneugenic activities in the mouse bone-marrow micronucleus (MN) test in vivo and for their sister-chromatid exchange-inducing activities in human lymphocytes in vitro in the presence and absence of an exogenous metabolizing system from rat-liver S9. 1,3-Dichloropropene significantly increased the frequencies of micronucleated polychromatic erythrocytes (PCE) in bone-marrow cells of female mice from 3.3 MN/1000 PCE to 15.3 MN/1000 PCE (187 mg per kg body w

Cholinesterase inhibition by organophosphorus compounds and its clinical effects.

The clinical manifestations of acute poisoning by organophosphorus compounds in man are in accord with, initially, the stimulation and, later, the blocking of cholinergic transmission due to acetylcholinesterase inhibition. The manifestations involve mainly the para-sympathetic nerves, the neuromuscular junctions, and the central nerve synapses, and to a smaller degree the cholinergic sympathetic nerves. Miosis and muscle fasciculations are useful signs for diagnosis and for the control of therapy. Blood cholinesterase determination is the best diagnostic test. The cause of death is usually re

Copy number variants underlie major selective sweeps in insecticide resistance genes in Anopheles arabiensis.

To keep ahead of the evolution of resistance to insecticides in mosquitoes, national malaria control programmes must make use of a range of insecticides, both old and new, while monitoring resistance mechanisms. The outdoor-biting malaria vector Anopheles arabiensis is of increasing concern for malaria transmission because it is apparently less susceptible to many indoor control interventions, yet knowledge of its mechanisms of resistance remains limited. Furthermore, comparatively little is known in general about resistance to non-pyrethroid insecticides such as pirimiphos-methyl (PM), which

H2S donor GYY4137 mitigates sFlt-1-induced hypertension and vascular dysfunction in pregnant rats†.

Gestational hypertension, often associated with elevated soluble Fms-related receptor tyrosine kinase 1 (sFlt-1), poses significant risks to both maternal and fetal health. Hydrogen sulfide (H2S), a gasotransmitter, has demonstrated blood pressure-lowering effects in hypertensive animals and humans. However, its role in pregnancy-induced hypertension remains unclear. This study investigated the impact of GYY4137, a slow-release H2S donor, on sFlt-1-induced hypertension in pregnant rats . Pregnant rats were administered sFlt-1 (6 μg/kg/day, intravenously) or vehicle from gestation day (GD) 12-2

A slow-releasing donor of hydrogen sulfide inhibits neuronal cell death via anti-PANoptosis in rats with spinal cord ischemia‒reperfusion injury.

BACKGROUND: Spinal cord ischemia‒reperfusion injury (SCIRI) can lead to paraplegia, which leads to permanent motor function loss. It is a disastrous complication of surgery and causes tremendous socioeconomic burden. However, effective treatments for SCIRI are still lacking. PANoptosis consists of three kinds of programmed cell death, pyroptosis, apoptosis, and necroptosis, and may contribute to ischemia‒reperfusion-induced neuron death. Previous studies have demonstrated that hydrogen sulfide (H2S) exerts a neuroprotective effect in many neurodegenerative diseases. However, whether H2S is ant

H2S counteracts proinflammatory effects of LPS through modulation of multiple pathways in human cells.

BACKGROUND: Hydrogen sulfide donors reduce inflammatory signaling in vitro and in vivo. The biological effect mediated by H2S donors depends on the kinetics of the gas release from the donor molecule. However, the molecular mechanisms of H2S-induced immunomodulation were poorly addressed. Here, we studied the effect of two different hydrogen sulfide (H2S)-producing agents on the generation of the LPS-induced inflammatory mediators. Importantly, we investigated the transcriptomic changes that take place in human cells after the LPS challenge, combined with the pretreatment with a slow-releasing

Hydrogen sulphide facilitates exocytosis by regulating the handling of intracellular calcium by chromaffin cells.

Gasotransmitter hydrogen sulphide (H2S) has emerged as a regulator of multiple physiological and pathophysiological processes throughout. Here, we have investigated the effects of NaHS (fast donor of H2S) and GYY4137 (GYY, slow donor of H2S) on the exocytotic release of catecholamines from fast-perifused bovine adrenal chromaffin cells (BCCs) challenged with sequential intermittent pulses of a K+-depolarizing solution. Both donors caused a concentration-dependent facilitation of secretion. This was not due to an augmentation of Ca2+ entry through voltage-activated Ca2+ channels (VACCs) because

Degradation of methamidophos by Hyphomicrobium species MAP-1 and the biochemical degradation pathway.

Methamidophos is one of the most widely used organophosphorus insecticides usually detectable in the environment. A facultative methylotroph, Hyphomicrobium sp. MAP-1, capable of high efficiently degrading methamidophos, was isolated from methamidophos-contaminated soil in China. It was found that the addition of methanol significantly promoted the growth of strain MAP-1 and enhanced its degradation of methamidophos. Further, this strain could utilize methamidophos as its sole carbon, nitrogen and phosphorus source for growth and could completely degrade 3,000 mg l(-1) methamidophos in 84 h un

Combined suicide by oxydemeton-methyl (Metasystox) ingestion and hanging.

A case of a combined suicide by oxydemeton-methyl (Metasystox) ingestion and hanging is reported. Dimethyl phosphate (DMP) proved to be a stable product of oxydemeton-methyl; for its determination a gas chromatographic/mass spectrometric procedure was performed. DMP levels were quantified in several liquids and tissue samples: measured concentrations were 103.31 microg/ml DMP in gastric contents, 0.10 microg/ml in blood, 1.40 microg/g in liver, 2.87 microg/ml in bile and 0.80 microg/g in kidney. No DMP was found in either urine or cerebrum. On the basis of the distribution of the poison in the

Effect of methamidophos and urea application on microbial communities in soils as determined by microbial biomass and community level physiological profiles.

In this study, we evaluated the effect of the application by two agrochemicals, methamidophos (O,S-dimethyl phosphoroamidothioate) and urea, on microbial diversity in soil, using the combined approaches of soil microbial biomass analysis and community level physiological profiles (CLPPs). The results showed that both a low and a high level of methamidophos application (CS2 and CS3) and urea application (CS4) significantly decreased microbial biomass C (Cmic) by 41-83% compared with the control (CS1). The soil organic C (Corg) values of CS3 and CS4 were significantly higher and lower by 24% and

Clinical, biochemical and neurobehavioural studies of workers engaged in the manufacture of quinalphos.

59 workers exposed to different chemicals during the manufacture of quinalphos, an organophosphate pesticide (OP) and 17 control subjects were studied. Despite similar blood acetylcholinestarase (AChE) levels in both the exposed and control subjects, a significant number of exposed subjects had altered plantar and ankle reflexes. Higher nervous functions such as memory, learning and vigilance were also found to be affected in these subjects. These findings were attributed to chronic low dose combined exposure to different chemicals used/formed in the manufacture of quinalphos. The study raises

Effect of WR-2721 on fetal development in the rat.

The radioprotector WR-2721 has been shown to radioprotect all tissues studied except the central nervous system. However, it has not yet been used to radioprotect the fetus. In this study we determined that [14C]WR-2721 injected into pregnant rats quickly passed the placenta and was concentrated by the fetus. In addition, we evaluated the toxicity of WR-2721 (2.5-600 mg/kg) to pregnant rats and their fetuses during the period of major organogenesis at Days 9, 11, and 14 postconception. Pregnant animals were only slightly more (10%) sensitive (LD50, 580 mg/kg) to WR-2721 than nonpregnant cohort

Inhibition of hepatic drug-metabolizing enzymes by thiophaophate insecticides and its toxicological evaluation.

Thiophosphate insecticides, fenitrothion, diazinon and methylparathion, inhibit hepatic drug-metabolizing enzyme activity which was assayed using aminopyrine and aniline as substrates. About 50% inhibition was observed 4 hrs after the injection of fenitrothion into mice in a dose of 25 mg/kg. The addition in vitro of thiophosphates into a reaction mixture of drug metabolizing enzyme also produced an effective inhibition and IC50 was around 10(-5)M for fenitrothion. DDVP and oxygenated metabolite of fenitrothion did not show any inhibiting effect either in vitro or in vivo. The inhibition of th

In Vitro Evaluation of the Combined Toxicity of Pirimiphos-methyl and Piperonyl Butoxide.

Pirimiphos-methyl (PMM) and piperonyl butoxide (PBO) are commonly used in crops to combat pests. This study examined their cytotoxicity and genotoxicity in various cell lines (RAW 264-7, H9C2, HepG2, HEK 293) across different exposure times. HepG2 cells showed heightened sensitivity to the combination (MIX) at 100 µg/mL after 48 and 72 hours, with no genotoxic effects observed in both HepG2 and mouse whole blood cells. HEK 293 cells experienced cytotoxicity at higher concentrations of PMM, PBO, and MIX (50 and 100 µg/mL) after 48 and 72 hours. H9C2 cells exhibited the most pronounced cytotoxic

GYY4137, a hydrogen sulfide donor, protects against endothelial dysfunction in porcine coronary arteries exposed to myeloperoxidase and hypochlorous acid.

BACKGROUND AND AIMS: Myeloperoxidase (MPO) and its principal reaction product hypochlorous acid (HOCl) are part of the innate immune response but are also associated with endothelial dysfunction, thought to involve a reduction in nitric oxide (NO) bioavailability. We aimed to investigate the effect of MPO and HOCl on vasorelaxation of coronary arteries and to assess directly the involvement of NO. In addition, we hypothesised that the slow release hydrogen sulfide (H2S) donor GYY4137 would salvage coronary artery endothelial function in the presence of MPO and HOCl. METHODS AND RESULTS: Contr

Characterization and Biological Activity of a Hydrogen Sulfide-Releasing Red Light-Activated Ruthenium(II) Complex.

Hydrogen sulfide (H2S) is a biological gasotransmitter that has been employed for the treatment of ischemia-reperfusion injury. Despite its therapeutic value, the implementation of this gaseous molecule for this purpose has required H2S-releasing prodrugs for effective intracellular delivery. The majority of these prodrugs, however, spontaneously release H2S via uncontrolled hydrolysis. Here, we describe a Ru(II)-based H2S-releasing agent that can be activated selectively by red light irradiation. This compound operates in living cells, increasing intracellular H2S concentration only upon irra

Protective effects of exogenous and endogenous hydrogen sulfide in mast cell-mediated pruritus and cutaneous acute inflammation in mice.

The recently described 'gasomediator' hydrogen sulfide (H2S) has been involved in pain mechanisms, but its effect on pruritus, a sensory modality that similarly to pain acts as a protective mechanism, is poorly known and controversial. The effects of the slow-releasing (GYY4137) and spontaneous H2S donors (Na2S and Lawesson's reagent, LR) were evaluated in histamine and compound 48/80 (C48/80)-dependent dorsal skin pruritus and inflammation in male BALB/c mice. Animals were intradermally (i.d.) injected with C48/80 (3μg/site) or histamine (1μmol/site) alone or co-injected with Na2S, LR or GYY4
