Appropriate reduction of importin-α gene expression enhances yellow dwarf disease resistance in common wheat.

Barley yellow dwarf viruses (BYDVs) cause widespread damage to global cereal crops. Here we report a novel strategy for elevating resistance to BYDV infection. The 17K protein, a potent virulence factor conserved in BYDVs, interacted with barley IMP-α1 and -α2 proteins that are nuclear transport receptors. Consistently, a nuclear localization signal was predicted in 17K, which was found essential for 17K to be transported into the nucleus and to interact with IMP-α1 and -α2. Reducing HvIMP-α1 and -α2 expression by gene silencing attenuated BYDV-elicited dwarfism, accompanied by a lowered nucle

Introduction of barnase/barstar in soybean produces a rescuable male sterility system for hybrid breeding.

Hybrid breeding for increased vigour has been used for over a century to boost agricultural outputs without requiring higher inputs. While this approach has led to some of the most substantial gains in crop productivity, breeding barriers have fundamentally limited soybean (Glycine max) from reaping the benefits of hybrid vigour. Soybean flowers self-pollinate prior to opening and thus are not readily amenable to outcrossing. In this study, we demonstrate that the barnase/barstar male sterility/rescue system can be used in soybean to produce hybrid seeds. By expressing the cytotoxic ribonuclea

Comparing Multiple Reaction Monitoring and Sequential Window Acquisition of All Theoretical Mass Spectra for the Relative Quantification of Barley Gluten in Selectively Bred Barley Lines.

Celiac disease (CD) is a disease of the small intestine that occurs in genetically susceptible subjects triggered by the ingestion of cereal gluten proteins for which the only treatment is strict adherence to a life-long gluten-free diet. Barley contains four gluten protein families, and the existence of barley genotypes that do not accumulate the B-, C-, and D-hordeins paved the way for the development of an ultralow gluten phenotype. Using conventional breeding strategies, three null mutations behaving as recessive alleles were combined to create a hordein triple-null barley variety. Proteom

Development of diagnostic markers for the disease susceptibility gene Tsn1 in wheat reveals novel resistance alleles and a new locus required for ToxA sensitivity.

The wheat gene Tsn1 confers susceptibility to tan spot, septoria nodorum blotch, and spot blotch. The markers developed here may be immediately deployed in breeding programs to eliminate Tsn1. The wheat Tsn1 gene recognizes the necrotrophic effector ToxA, which is produced by three different necrotrophic fungal pathogens. A compatible Tsn1-ToxA interaction leads to host-induced responses that result in the development of disease. Therefore, marker-assisted elimination of functional Tsn1 alleles is an effective strategy for the development of disease resistant varieties. To develop such markers

Transcriptome and Coexpression Network Analyses Provide In-Sights into the Molecular Mechanisms of Hydrogen Cyanide Synthesis during Seed Development in Common Vetch (Vicia sativa L.).

The common vetch (Vicia sativa L.) seed is an ideal plant-based protein food for humans, but its edible value is mainly limited by the presence of cyanogenic glycosides that hydrolyze to produce toxic hydrogen cyanide (HCN), and the genes that regulate HCN synthesis in common vetch are unknown. In this study, seeds from common vetch at 5, 10, 15, 20, 25, 30, and 35 days after anthesis were sampled, and the seven stages were further divided into five developmental stages, S1, S2, S3, S4, and S5, based on morphological and transcriptome analyses. A total of 16,403 differentially expressed genes

Metabolite profiling characterises chemotypes of Musa diploids and triploids at juvenile and pre-flowering growth stages.

Bananas (Musa spp.) are consumed worldwide as dessert and cooking types. Edible banana varieties are for the most part seedless and sterile and therefore vegetatively propagated. This confers difficulties for breeding approaches against pressing biotic and abiotic threats and for the nutritional enhancement of banana pulp. A panel of banana accessions, representative of the diversity of wild and cultivated bananas, was analysed to assess the range of chemotypes available globally. The focus of this assessment was banana leaves at two growth stages (juvenile and pre-flowering), to see when duri

Engineering for disease resistance: persistent obstacles clouding tangible opportunities.

The accelerating pace of gene discovery, coupled with novel plant breeding technologies, provides tangible opportunities with which to engineer disease resistance into agricultural and horticultural crops. This is especially the case for potato, wheat, apple and banana, which are afflicted with fungal and bacterial diseases that impact significantly on each crop's economic viability. Yet public scepticism and burdensome regulatory systems remain the two primary obstacles preventing the translation of research discoveries into cultivars of agronomic value. In this perspective review, the potent…

New advances and future directions in plant polyspermy.

Plants have evolved a battery of mechanisms that potentially act as polyspermy barriers. Supernumerary sperm fusion to one egg cell has consequently long remained a hypothetical concept. The recent discovery that polyspermy in flowering plants is not lethal but generates viable triploid plants is a game changer affecting the field of developmental biology, evolution, and plant breeding. The establishment of protocols to artificially induce polyspermy together with the development of a high-throughput assay to identify and trace polyspermic events in planta now provide powerful tools to unravel…

Pathogen Effector-Mediated Reprogramming of Plant Alternative Splicing: From Immune Regulation to Crop Disease Resistance.

Plants have evolved sophisticated immune systems to defend against diverse pathogens, whereas pathogens deploy effectors to manipulate host cellular processes for successful infection. Recent studies have revealed that pathogen effectors can target host RNA processing, particularly pre-mRNA alternative splicing (AS), to reshape transcript isoform profiles and modulate plant immunity. However, the common principles and specific differences among pathogen effector-mediated regulation of host AS and their impacts on plant immunity have not been systematically summarized. In this review, we summar

Reactive Oxygen Species and Phytohormone Signalling: Redox Modifications Shaping Plant Growth and Stress Acclimation.

Reactive oxygen species (ROS) and phytohormones are essential components of a precisely coordinated signalling network that regulates plant growth, development and adaptation to environmental changes. This review synthesises current knowledge to elucidate the specific oxidative modification mechanisms by which ROS target core signalling components, biosynthetic enzymes, as well as transport proteins throughout all hormonal pathways and distils cross-pathway principles that define a coherent redox-guided regulatory architecture. We further delineate the reciprocal regulation where phytohormones

Iron homeostasis and plant immune responses: Recent insights and translational implications.

Iron metabolism and the plant immune system are both critical for plant vigor in natural ecosystems and for reliable agricultural productivity. Mechanistic studies of plant iron home-ostasis and plant immunity have traditionally been carried out in isolation from each other; however, our growing understanding of both processes has uncovered significant connections. For example, iron plays a critical role in the generation of reactive oxygen intermediates during immunity and has been recently implicated as a critical factor for immune-initiated cell death via ferroptosis. Moreover, plant iron s

Comparative transcriptome analysis reveals higher expression of stress and defense responsive genes in dwarf soybeans obtained from the crossing of G. max and G. soja.

BACKGROUND: Plant height is an important component of plant architecture and significantly affects crop breeding practices and yield. Dwarfism in plants prevents lodging and therefore it's a desired trait in crops. OBJECTIVE: To find differentially expressed genes to classify and understand the regulation of genes related to plant growth in mutant dwarf soybeans, which appeared in the F5 generation. METHODS: We obtained a few segregated dwarf soybeans in the populations derived from the crossing of Glycine max var. Peking and Glycine soja var. IT182936 in an F5 RIL population. These dwarf so

Optimizing multi-trait selection for profitability in Illinois wheat using a factor analytic mixed model and an economic selection index.

Economic selection indices can improve the efficiency of selection for multiple traits, but they are underutilized in plant breeding due to limited research on the economic values of traits and a lack of well-documented case examples employing economic indices. This study evaluated a genomic-based Smith-Hazel index incorporating empirically derived economic weights for grain yield, test weight, and maturity in soft red winter wheat (Triticum aestivum L.) in Illinois. Economic weights used in the index were estimated using a function describing expected profit considering the predominant croppi…

How Does Cloning Change Phenolics, Minerals and Antioxidant Activity in Helichrysum arenarium (L.) Moench (Clone A, Clone B)?

Tissue culture and cloning are valuable techniques in plant biology research as they enable the production of genetically identical plants, which may be utilized for many purposes, such as crop enhancement, plant breeding, and genetic manipulation. Helichrysum arenarium (L.) Moench, also known as immortelle or strawflower (Asteraceae), is a species of flowering plant in the sunflower family. In this study, the aerial part of two Clones (Clone A, Clone B) of H. arenarium were used to consider the effect of Clones on the phenolics, minerals, and antioxidant activities. The methanol extracts were…

Photorespiration and plant immunity: Interactions and implications under a changing climate.

Climate change has profound impacts on plant growth, productivity, and immunity. Photosynthesis, a key biological process that sequesters atmospheric CO2, plays a crucial role in mitigating the effects of climate change. However, photorespiration, a process that consumes O2 instead of CO2 and is metabolically linked with photosynthesis, is often viewed as a wasteful process that reduces photosynthetic efficiency by nearly 48 % in C3 plants. This reduction has a significant impact on crop yield, given the direct contribution of photosynthesis to biomass accumulation. Over the years, numerous ef

Starch and tuber traits of diploid potato lines B26 and B100 and their hybrids.

Diploid potato breeding enables faster genetic improvement via selection against deleterious alleles in inbred lines, unlike breeding by intercrossing tetraploid varieties. Starch is the major source of calories in potato tubers, but the starch properties of diploid lines have rarely been reported. In this study, we provide a comprehensive characterisation of tuber and starch properties in two diploid lines that are early isolates from the Solynta breeding program, B26 and B100, and their F1 hybrids. B100 produced fewer, but larger tubers compared to B26, and both diploid lines produced tubers

Breeding jassid-resistant okra (Abelmoschus esculentus): from morpho-biochemical traits to precision breeding.

Okra, an economically important vegetable in tropical and subtropical regions, faces severe yield constraints due to sap-sucking pests, particularly jassids (Amrasca spp.). These insects cause hopper burn, leaf curling, and chlorosis, leading to estimated yield losses of 30-100% under severe infestation. Farmers' heavy reliance on chemical control to manage these insects poses environmental, economic, and health risks, underscoring the need for host-plant resistance as a sustainable component of integrated pest management. Successful breeding for jassid resistance requires in-depth knowledge o

Enhancing agronomic traits and bioactive compound accumulation in Thai Andrographis paniculata via EMS-induced variation.

BACKGROUND: Andrographis paniculata is a self-pollinating medicinal plant with limited natural genetic diversity, which restricts the development of improved cultivars with desirable agronomic traits and stable accumulation of key bioactive compounds, particularly andrographolide (AP1). To overcome this limitation, ethyl methanesulfonate (EMS) mutagenesis was applied to native Thai germplasm to induce phenotypic and phytochemical variation and to generate novel genetic resources for breeding improvement and gene function analysis. RESULTS: Optimal EMS treatment conditions (1.0% and 1.5% EMS fo

Improving the efficiency of isolated microspore culture in six-row spring barley: II-exploring novel growth regulators to maximize embryogenesis and reduce albinism.

Two alternative cytokinins, thidiazuron and meta-topoline, were tested in isolated microspore culture on recalcitrant barley genotypes (six-row, spring), and green plant regeneration was improved substantially. Doubled-haploid (DH) plants are coveted in plant breeding and in genetic studies, since they are rapidly obtained and perfectly homozygous. In barley, DHs are produced mainly via androgenesis, and isolated microspore culture (IMC) constitutes the method offering the greatest potential efficiency. However, IMC can often be challenging in some genotypes because of low yield of microspores…
