Reversion and spread of vaccine-derived poliovirus (VDPV) to cause outbreaks of poliomyelitis is a rare outcome resulting from immunisation with the live-attenuated oral poliovirus vaccines (OPVs). Global withdrawal of all three OPV serotypes is therefore a key objective of the polio endgame strategic plan, starting with serotype 2 (OPV2) in April 2016. Development of inactivated poliovirus vaccine derived from Sabin strains.

In the course of Sabin-inactivated poliovirus vaccine (S-IPV) development, we have established high-yield virus production techniques based on Vero cell micro-carrier cultures. Development of specific ELISA tests to quantify the antigen content of S-IPV has been achieved. To adjust the immunogenicity of S-IPV so as to be comparable with the conventional-IPV, a new formulation was determined using a potency test using rats. The reformulated S-IPV was shown to be efficacious for the immunization of monkeys.

Indigenous wild poliovirus was last isolated in China in 1994. On October 13, 1999, a case of acute flaccid paralysis (AFP) in a 16-month-old boy was reported to public health authorities in Xunhua Autonomous County, Haidong Prefecture, Qinghai Province, China. Following onset of paralysis on October 12, the boy was no longer able to stand or walk. Two stool samples, taken within 14 days of onset of paralysis, were analyzed in the Qinghai provincial laboratory and yielded poliovirus. Circulation of poliovirus among risk groups in Ibadan, Nigeria.

Faecal samples collected from 114 fully vaccinated pre-school children and 32 unvaccinated infants in Ibadan, Nigeria, were assayed for poliovirus in Hep-2 and RD cell cultures. 8 strains of poliovirus type 1 were isolated from 146 samples--3 from 32 unvaccinated children aged less than 40 d and 5 from 114 fully vaccinated children aged between 9 and 60 months. Studies using Sabin and wild monoclonal antibodies and the polymerase chain reaction confirmed 7 of the 8 isolates to be of the wild type, a possible source of infection among vaccinated children.

Isolation of poliovirus from cases of acute flaccid paralysis.

OBJECTIVE: To evaluate the frequency of isolation of polio and other viruses from fecal samples in subjects with acute flaccid paralysis in northern parts of India. SETTING: Rural and Urban Immunization Centers used for Expanded Programme on Immunization. METHODS: 219 fecal samples were collected by anal tube in 1992 from Punjab, Haryana, Himachal Pradesh and Chandigarh and processed for virus isolation. Early immunization with inactivated poliovirus vaccine in premature infants.

The antibody titers of 41 premature infants receiving inactivated poliovirus vaccine at 2 months of age (control group) were compared with titers of 39 infants receiving an additional dose at 5 to 10 days of age (study group). At 1 month of age 97.4% of the study group but only 70.8% of the control group had protective antibodies against poliovirus 3 (p < 0.001).

Poliovirus-sensitive transgenic mice as a new animal model.

Transgenic mice susceptible to poliovirus infection were produced by introducing a human gene encoding cellular receptors for poliovirus into the mouse genome. Expression of receptor mRNAs in tissues of the transgenic mice was analysed using Northern blot hybridization. The results indicate that the human gene is expressed in many tissues of the transgenic mice just as in human tissues, and that the amount of the receptor mRNAs varies from tissue to tissue. Poliomyelitis remains an important public health problem in China. Most cases and outbreaks are associated with wild type 1 polioviruses. To obtain an overview of type 1 poliovirus transmission in China, partial genomic sequences were compared for 24 case isolates from 12 provinces. Because polioviruses evolve rapidly during infection of humans, the genetic relationships among isolates provide a measure of the extent of epidemiologic linkage among cases.
