Quantitation of cytopathic honey bee viruses using the viral plaque assay method.

Viral pathogens are highly prevalent threats to our pollination workforce. Basic virological techniques for ascertaining infection intensity and quantifying infectious viral particles lag behind the research interest in pollinator-associated viruses. Here, we utilized the only available immortalized honey bee (Apis mellifera) cell line, AmE-711, to establish the first plaque assay protocol for a bee-infecting virus in honey bee cells. First, we demonstrate that AmE-711 is susceptible to acute bee paralysis virus (ABPV) despite being persistently infected with deformed wing virus. Then, we comp…

Agrochemical-induced stress in stingless bees: peculiarities, underlying basis, and challenges.

The toxicological stress induced by pesticides, particularly neonicotinoid insecticides, and its consequences in bees has been the focus of much recent attention, particularly for honey bees. However, the emphasis on honey bees and neonicotinoids has led to neglect of the relevance of stingless bees, the prevailing pollinators of natural and agricultural tropical ecosystems, and of other agrochemicals, including other pesticides and even leaf fertilizers. Consequently, studies focusing on agrochemical effects on stingless bees are sparse, usually limited to histopathological studies, and lack

Environmental antibiotic and fungicide exposure differentially affects survival and carbohydrate-fueled flight performance in Apis mellifera.

Environmental contamination by antibiotics and fungicides is an emerging concern due to their widespread occurrence in agricultural ecosyste … anisms and may consequently compromise bee survival and the sustained flight required for effective pollination; however, their effects at environmentally relevant concentrations remain poorly understood. In this study, we examined how exposure to tetracycline and boscalid affected honey bee survival, carbohydrate-fueled flight performance, and inferred carbo …

Pollen Diet Diversity does not Affect Gut Bacterial Communities or Melanization in a Social and Solitary Bee Species.

Pollinators face many stressors, including reduced floral diversity. A low-diversity diet can impair organisms' ability to cope with additional stressors, such as pathogens, by altering the gut microbiome and/or immune function, but these effects are understudied for most pollinators. We investigated the impact of pollen diet diversity on two ecologically and economically important generalist pollinators, the social bumble bee (Bombus impatiens) and the solitary alfalfa leafcutter bee (Megachile rotundata). We experimentally tested the effect of one-, two-, or three-species pollen diets on gut

Differences among six woody perennials native to Northern Europe in their level of genetic differentiation and adaptive potential at fine local scale.

The ability of perennial species to adapt their phenology to present and future temperature conditions is important for their ability to ret … while the relations are much less clear for the three insect-pollinated species. This supports that pollination system can influence the balance between genetic drift and natural selection and thereby influence the level of local adaptation in long-lived species. On the positive side, we find that the native populations of woody plant species have maintained …

Biomechanics of substrate boring by fig wasps.

Female insects of diverse orders bore into substrates to deposit their eggs. Such insects must overcome several biomechanical challenges to successfully oviposit, which include the selection of suitable substrates through which the ovipositor can penetrate without itself fracturing. In many cases, the insect may also need to steer and manipulate the ovipositor within the substrate to deliver eggs at desired locations before rapidly retracting her ovipositor to avoid predation. In the case of female parasitoid ichneumonid wasps, this process is repeated multiple times during her lifetime, thus

Effect of confounding cofactors on responses to pollens during natural season versus pollen challenge chamber exposure.

BACKGROUND: The severity of allergic rhinoconjunctivitis (AR) symptomatology elicited after exposure to pollen in the absence versus the presence of confounding cofactors, such as in a pollen challenge chamber (PCC) and the natural pollinating season, respectively, might differ. OBJECTIVE: We sought to determine the correlation of AR severity in the natural season versus out-of-season PCC exposures. METHODS: Twenty-four Virginia live oak (VLO)-positive, 14 VLO-negative, 16 mountain cedar (MC)-positive, 8 MC-negative, and 26 ragweed-positive participants recorded AR symptoms (total symptom sc

The Effect of Biogeographic Origin on the Adaptive Trajectories of Plants in Urban Environments.

Parallel adaptation is a hallmark of urban evolution, yet whether plant responses to urbanization are globally consistent or influenced by biogeographic origin remains unclear. We investigated pollinator communities, floral display and reproductive allocation in Trifolium repens along urbanization gradients in 11 European urban areas spanning boreal, continental, and Mediterranean biogeographic regions. We found strong differences, with boreal and continental gradients showing no consistent floral trait shifts, whereas Mediterranean gradients showed parallel reductions in flower and infloresce

The Effects of Diesel Exhaust Pollution on Floral Volatiles and the Consequences for Honey Bee Olfaction.

There is growing evidence of a substantial decline in pollinators within Europe and North America, most likely caused by multiple factors such as diseases, poor nutrition, habitat loss, insecticides, and environmental pollution. Diesel exhaust could be a contributing factor to this decline, since we found that diesel exhaust rapidly degrades floral volatiles, which honey bees require for flower recognition. In this study, we exposed eight of the most common floral volatiles to diesel exhaust in order to investigate whether it can affect volatile mediated plant-pollinator interaction. Exposure

A Global Analysis of the Distribution Patterns of Style-Length Polymorphisms Across Angiosperms.

Unraveling the evolutionary and biogeographical factors that drive the widespread occurrence of outcrossing mechanisms in plants is key to understanding their prevalence. Style-length polymorphisms (SLP) are labile outcrossing traits that have evolved repeatedly in several angiosperm lineages sharing moderately specialized pollination features, reflecting a convergent strategy to optimize outcrossing efficiency. Notably, over 50% of known genera exhibiting SLP belong to the Rubiaceae family, which, beyond harboring floral architectures that promote pollination precision, is characterized by a …

Functional characterization of terpene synthases SmTPS1 involved in floral scent formation in Salvia miltiorrhiza.

Plants attract beneficial insects and promote pollination by releasing floral scents. Salvia miltiorrhiza, as an insect-pollinated flowering plant, which has been less studied for its floral aroma substances. This study revealed that S. miltiorrhiza flowers produce various volatile terpenoids, including five monoterpenes and ten sesquiterpenes, with the sesquiterpene compound (E)-β-caryophyllene being the most abundant, accounting for 28.1% of the total volatile terpenoids. Y-tube olfactometer experiments were conducted on the primary pollinator of S. miltiorrhiza, the Apis ceranas. The result…

Risk assessment for side-effects of neonicotinoids against bumblebees with and without impairing foraging behavior.

Bombus terrestris bumblebees are important pollinators of wild flowers, and in modern agriculture they are used to guarantee pollination of vegetables and fruits. In the field it is likely that worker bees are exposed to pesticides during foraging. To date, several tests exist to assess lethal and sublethal side-effects of pesticides on bee survival, growth/development and reproduction. Within the context of ecotoxicology and insect physiology, we report the development of a new bioassay to assess the impact of sublethal concentrations on the bumblebee foraging behavior under laboratory condit…

Sexually concordant selection on floral traits despite greater opportunity for selection through male fitness.

Pollinators are important drivers of floral trait evolution, yet plant populations are not always perfectly adapted to their pollinators. Such apparent maladaptation may result from conflicting selection through male and female sexual functions in hermaphrodites. We studied sex-specific mating patterns and phenotypic selection on floral traits in Aconitum gymnandrum. After genotyping 1786 offspring, we partitioned individual fitness into sex-specific selfed and outcrossed components and estimated phenotypic selection acting through each. Relative fitness increased with increasing mate number,

The Extraordinary Alkali Bee, Nomia melanderi (Halictidae), the World's Only Intensively Managed Ground-Nesting Bee.

Among the ground-nesting bees are several proven crop pollinators, but only the alkali bee (Nomia melanderi) has been successfully managed. In <80 years, it has become the world's most intensely studied ground-nesting solitary bee. In many ways, the bee seems paradoxical. It nests during the torrid, parched midsummer amid arid valleys and basins of the western United States, yet it wants damp nesting soil. In these basins, extensive monocultures of an irrigated Eurasian crop plant, alfalfa (lucerne), subsidize millions of alkali bees. Elsewhere, its polylectic habits and long foraging range al

Are bee diseases linked to pesticides? - A brief review.

The negative impacts of pesticides, in particular insecticides, on bees and other pollinators have never been disputed. Insecticides can directly kill these vital insects, whereas herbicides reduce the diversity of their food resources, thus indirectly affecting their survival and reproduction. At sub-lethal level (<LD50), neurotoxic insecticide molecules are known to influence the cognitive abilities of bees, impairing their performance and ultimately impacting on the viability of the colonies. In addition, widespread systemic insecticides appear to have introduced indirect side effects on bo

Mapping the eco-holobiont: tripartite interactions of small carpenter bee, floral, and soil microbiomes across an urbanization gradient.

Interactions between plants and pollinating insects are essential to the health of many ecosystems, and are also of concern to human agriculture. Work in recent years has increasingly approached pollinator health from a holobiont perspective, considering communities of microbes alongside their hosts. Yet, little research has directly compared microbiomes between these insects and the environmental substrates from which they are largely acquired. We investigated the microbiome composition and structure of small carpenter bees (Ceratina calcarata) alongside directly associated samples of host fl

Sublethal effects of cyfluthrin, atrazine, and prothioconazole on the development of European bumblebee (Bombus terrestris) colonies.

BACKGROUND: Bumblebees (Bombus spp.) are essential pollinators of agricultural crops, relying primarily on nectar and pollen for nutrition. However, these floral resources often contain pesticide residues that may exert sublethal effects on colony performance. While acute impacts are difficult to observe, chronic exposure can undermine colony development and increase the risk of collapse. The field-level effects of commonly used pesticides such as cyfluthrin, atrazine, and prothioconazole on the European bumblebee (Bombus terrestris) remain poorly understood. To address this gap, we evaluated
