The serine/threonine protein phosphatase Sit4p activates multidrug resistance in Saccharomyces cerevisiae.

Multidrug resistance in Saccharomyces cerevisiae is frequently associated with gain-of-function mutations in zinc finger-containing transcription factors Pdr1p and Pdr3p. These regulatory proteins activate the expression of several ATP-binding cassette transporter genes, leading to elevated drug resistance. Here, we report that loss of the type 2A-related serine/threonine protein phosphatase Sit4p renders yeast cells sensitive to cycloheximide, azoles, daunorubicin and rhodamine 6G. This effect is a consequence of the decreased transcriptional levels of mainly PDR3 and its target genes, PDR5, …

Contribution of Yap1 towards Saccharomyces cerevisiae adaptation to arsenic-mediated oxidative stress.

In the budding yeast Saccharomyces cerevisiae, arsenic detoxification involves the activation of Yap8, a member of the Yap (yeast AP-1-like) family of transcription factors, which in turn regulates ACR2 and ACR3, genes encoding an arsenate reductase and a plasma-membrane arsenite-efflux protein respectively. In addition, Yap1 is involved in the arsenic adaptation process through regulation of the expression of the vacuolar pump encoded by YCF1 (yeast cadmium factor 1 gene) and also contributing to the regulation of ACR genes. Here we show that Yap1 is also involved in the removal of ROS (react…

Vacuolar compartmentation of the cadmium-glutathione complex protects Saccharomyces cerevisiae from mutagenesis.

In the yeast Saccharomyces cerevisiae, gamma-glutamyl transferase (gamma-GT; EC 2.3.2.2) is a vacuolar-membrane bound enzyme. In this work we verified that S. cerevisiae cells deficient in gamma-GT absorbed almost 2.5-fold as much cadmium as the wild-type (wt) cells, suggesting that this enzyme might be responsible for the recycle of cadmium-glutathione complex stored in the vacuole. The mutant strain showed difficulty in keeping constant levels of glutathione (GSH) during the stress, although the GSH-reductase activity was practically the same in both wt and mutant strains, before and after m…

Engineering natural Saccharomyces cerevisiae isolates for enhanced one-step cellulosic ethanol production.

Engineering yeast strains for use as chassis organisms in second-generation (2G) bioethanol is a promising strategy to improve process economics. Natural isolates of Saccharomyces cerevisiae offer strain backgrounds with greater genetic diversity and enhanced robustness, with the potential for improved heterologous protein production capabilities. In this study, heterologous cellulase production using different expression strategies was evaluated in various process-relevant conditions. Enhanced cellulolytic activity was clearly demonstrated in a cell-tethered enzyme system, compared to a free …

CRISPR-Cas9 engineering for high-level β-galactosidase production in Saccharomyces boulardii.

Saccharomyces boulardii is a clinically validated probiotic yeast widely used to treat gastrointestinal disorders. Its natural probiotic effects, combined with the capacity of surviving the harsh gastrointestinal conditions, makes it an attractive chassis for therapeutic protein delivery. However, its engineering has relied on a limited set of genetic tools and lacks systematic validation, which has constrained its development as a next generation engineered probiotic. In this study, the compatibility of the EasyClone-MarkerFree vector system with CRISPR-Cas9 editing in S. boulardii was system…

Phosphatidylinositol 3-phosphate metabolism impacts cellular α-synuclein localization in Saccharomyces cerevisiae.

Alpha-synuclein (αSyn), a hallmark protein of synucleinopathies such as Parkinson's disease (PD), is likely to be involved in neuronal membrane trafficking and synaptic vesicle dynamics at axon terminals. Its specific binding to anionic phospholipids, such as phosphatidylinositol phosphates that are essential for intracellular signaling and membrane trafficking, suggests an involvement in vesicular transport processes. In Saccharomyces cerevisiae, a model organism for cell biological PD research, human αSyn localizes to the plasma membrane via the secretory machinery. Employing this yeast mode…

Transcriptomic responses of Saccharomyces cerevisiae and Hanseniaspora uvarum during separated co-fermentation.

Hanseniaspora uvarum is a predominant yeast species in vineyards and some strains demonstrate oenological potential by enhancing the aroma complexity and reducing the content of volatile acid and ethanol in wine. H. uvarum kept in our lab could improve the flavor and quality of wine when co-fermented with Saccharomyces cerevisiae, but its growth was inhibited by S. cerevisiae. To investigate the interaction mechanisms of S. cerevisiae and H. uvarum, double-compartment fermentor with dialysis tube of 1000 kDa MWCO was selected for separated co-fermentation (SCF) in synthetic grape juice at 25℃ …

Comparative adaptability of 307 Saccharomyces cerevisiae strains from winemaking and Mantou fermentation.

Domesticated Saccharomyces cerevisiae is one of the most significant microbial populations in human civilization due to its remarkable diversity and high adaptability to human environments. However, the adaptability mechanisms underlying this population ecological behavior remain unclear. This study explored the adaptive behaviors of S. cerevisiae strains from the Wine and Mantou genetic lineages under both artificial stress conditions and natural or near-natural environments. A total of 307 diploid S. cerevisiae strains were analyzed, including 169 strains derived from grape sources and 138 f…

Probiotic paradox: Saccharomyces cerevisiae fungemia after S. boulardii use in severe pancreatitis.

BACKGROUND: Saccharomyces boulardii is a widely used probiotic for managing antibiotic-associated diarrhea and other gastrointestinal disorders. While considered safe in immunocompetent individuals, its use in critically ill patients has been increasingly associated with invasive fungal infections, particularly Saccharomyces cerevisiae fungemia. CASE PRESENTATION: We report the case of a 46-year-old male admitted with severe necrotizing biliary pancreatitis, complicated by pancreatic necrosis, portal vein thrombosis, and a suspected duodenal fistula. Following emergency necrosectomy, the pati…

Evolutionary engineering of Saccharomyces cerevisiae: Crafting a synthetic methylotroph via self-reprogramming.

Methanol, as a non-edible feedstock, offers a promising sustainable alternative to sugar-based substrates in biochemical production. Despite progress in engineering methanol assimilation in nonmethylotrophs, the full transformation into methanol-dependent synthetic methylotrophs remains a formidable challenge. Here, moving beyond the conventional rational design principle, we engineered a synthetic methylotrophic Saccharomyces cerevisiae through genome rearrangement and adaptive laboratory evolution. This evolutionarily advanced strain unexpectedly shed the heterologous methanol assimilation p…

Sheep immune-stimulated with Saccharomyces boulardii show reduced prolificacy of Haemonchus contortus.

Haemonchus contortus is the most pathogenic parasite for sheep. The objective was to evaluate immunomodulation of the probiotic Saccharomyces boulardii in sheep experimentally infected with H. contortus. Twenty-four sheep were divided into three groups: one infected with 500 H. contortus larvae/day for 26 days and supplemented with S. boulardii (40 ml with 1 × 108  CFU/ml/day); a control group only infected with H. contortus but not supplemented; and a naïve group that never came into contact with either parasites or S. boulardii. To assess the humoral immune response, production of specific s…

Leptospiral lipopolysaccharide mediated Hog1 phosphorylation in Saccharomyces cerevisiae directs activation of autophagy.

Cells have developed a variety of mechanisms to counteract stress to give a specific and adaptive response. Yeast Hog1 is a homolog to mammalian p38, which is a mitogen-activated protein kinase. In this work, we analyze the Hog1 signaling during the induction of leptospiral LPS (100 ng/mL) and the hyperosmotic element NaCl (0.8 M). After the addition of stress elements, the stress-activated protein kinase was phosphorylated within 30 min of exposure and led to the expression of various genes responsible for cell survival. We found that leptospiral lipopolysaccharide mediated Hog1 phosphorylati

Treatment of Rhenium-Containing Effluents Using Environmentally Friendly Sorbent, Saccharomyces cerevisiae Biomass.

Yeast Saccharomyces cerevisiae biomass was applied for rhenium and accompanying elements (copper and molybdenum) removal from single- and multi-component systems (Re, Re-Mo, Re-Cu, and Re-Mo-Cu). Yeast biomass was characterized using X-ray diffraction, scanning electron microscopy, and Fourier transform infrared spectroscopy. The effects of biosorption experimental parameters such as solution pH (2.0-6.0), rhenium concentration (10-100 mg/L), time of interaction (5-120 min), and temperature (20-50 °C) have been discussed in detail. Maximum removal of rhenium (75-84%) and molybdenum (85%) was a…

Mitochondrial Genome Variation Affects Multiple Respiration and Nonrespiration Phenotypes in Saccharomyces cerevisiae.

Mitochondrial genome variation and its effects on phenotypes have been widely analyzed in higher eukaryotes but less so in the model eukaryote Saccharomyces cerevisiae Here, we describe mitochondrial genome variation in 96 diverse S. cerevisiae strains and assess associations between mitochondrial genotype and phenotypes as well as nuclear-mitochondrial epistasis. We associate sensitivity to the ATP synthase inhibitor oligomycin with SNPs in the mitochondrially encoded ATP6 gene. We describe the use of iso-nuclear F1 pairs, the mitochondrial genome equivalent of reciprocal hemizygosity analysi…

Engineering Saccharomyces cerevisiae fatty acid composition for increased tolerance to octanoic acid.

Biorenewable chemicals such as short and medium chain fatty acids enable functional or direct substitution of petroleum-derived building blocks, allowing reduction of anthropogenic greenhouse gases while meeting market needs of high-demand products like aliphatic alcohols and alpha olefins. However, producing these fatty acids in microorganisms can be challenging due to toxicity issues. Octanoic acid (C8) can disrupt the integrity of the cell membrane in yeast, and exogenous supplementation of oleic acid has been shown to help alleviate this. We recently engineered the Saccharomyces cerevisiae…

Probiotic yeast Saccharomyces boulardii (nom. nud.) modulates adhesive properties of Candida glabrata.

Following the widespread use of immunosuppressive therapy together with broad-spectrum antimycotic therapy, the frequency of mucosal and systemic infections caused by the pathogenic yeast Candida glabrata has increased in the past decades. Due to the resistance of C. glabrata to existing azole drugs, it is very important to look for new strategies helping the treatment of such fungal diseases. In this study, we investigated the effect of the probiotic yeast Saccharomyces boulardii (nom. nud.) on C. glabrata adhesion at different temperatures, pH values, and in the presence of fluconazole, itra…

Dithiocarbamates with potent inhibitory activity against the Saccharomyces cerevisiae β-carbonic anhydrase.

Dithiocarbamates (DTCs) prepared from primary or secondary amines, which incorporated amino/hydroxyl-alkyl, mono-/bicyclic aliphatic/heterocyclic rings based on the quinuclidine, piperidine, hydroxy-/carboxy-/amino-substituted piperidine, morpholine and piperazine scaffolds, were investigated for the inhibition of α- and β-carbonic anhydrases (CAs, EC 4.2.1.1) of pharmacologic relevance, such as the human (h) isoform hCA I and II, as well as the Saccharomyces cerevisiae β-CA, scCA. The yeast and its β-CA were shown earlier to be useful models of pathogenic fungal infections. The DTCs investiga…

Ocular albinism 1 protein: trafficking and function when expressed in Saccharomyces cerevisiae.

The ocular albinism 1 (Oa1) protein is believed to be involved in the biogenesis of melanosomes, but its cellular localization is controversial and its function is unknown. Based upon sequence homology, it has been predicted that Oa1 belongs to the G protein coupled receptor (GPCR) superfamily. We used the yeast Saccharomyces cerevisiae as a genetically amenable system to study the localization and function of Oa1. Sucrose gradient and immunofluorescence studies revealed that when expressed in yeast, Oa1 localizes to the prevacuolar compartment, the functional equivalent of the mammalian late …
