Silk Fibroin-Based Matrices for the Guidance of Cell Interaction, Tissue Regeneration, and Crosstalk.

The interactions between cells and the extracellular matrix are essential regulators of cell behaviors such as adhesion, proliferation, migration, differentiation, and function. From the perspective of tissue regeneration, some physicochemical characteristics of the material, including hydrophilicity, topology, and charge of the material surface, can significantly affect cell adhesion, proliferation, and differentiation. Many biomaterials are introduced for tissue engineering scaffolds, biomimicking natural tissues. Among the biomaterials, silk proteins (fibroin and sericin) have many excellen…

A medical adhesive used in a wet environment by blending tannic acid and silk fibroin.

A multifunctional and effective medical adhesive with a combination of high toughness and superior adhesion is highly desired in biomedical fields. However, clinical application of medical adhesives is still limited due to their weak adhesion to wet tissue. In this study, a novel medical adhesive called TASK composed of tannic acid (TA) and silk fibroin (SF) based on polyphenol-gel systems was developed. TASK powder was prepared by a simple physical mixture of pyrogallol-rich tannic acid and silk fibroin in aqueous solution and further freeze drying, which was stable and convenient for sterili…

The Nephila clavipes genome highlights the diversity of spider silk genes and their complex expression.

Spider silks are the toughest known biological materials, yet are lightweight and virtually invisible to the human immune system, and they thus have revolutionary potential for medicine and industry. Spider silks are largely composed of spidroins, a unique family of structural proteins. To investigate spidroin genes systematically, we constructed the first genome of an orb-weaving spider: the golden orb-weaver (Nephila clavipes), which builds large webs using an extensive repertoire of silks with diverse physical properties. We cataloged 28 Nephila spidroins, representing all known orb-weaver …

Anti-inflammation effects of corn silk in a rat model of carrageenin-induced pleurisy.

Pleurisy is an inflammation of the pleural layers that surround the lungs. Despite much research into inflammatory diseases, no drugs with favorable safety profiles are available yet for their treatment. Corn silk has been used in many parts of the world for the treatment of edema, cystitis, gout, kidney stones nephritis, and prostitutes. However, no scientific reports on the anti-inflammatory effects of corn silk were so far available. To test the anti-inflammatory efficacy of corn silk extract (CSEX) in a rat model of carrageenin (Cg)-induced pleurisy, exudate formation, and cellular infiltr…

A protocol for the production of recombinant spider silk-like proteins for artificial fiber spinning.

The extreme strength and elasticity of spider silks originate from the modular nature of their repetitive proteins. To exploit such materials and mimic spider silks, comprehensive strategies to produce and spin recombinant fibrous proteins are necessary. This protocol describes silk gene design and cloning, protein expression in bacteria, recombinant protein purification and fiber formation. With an improved gene construction and cloning scheme, this technique is adaptable for the production of any repetitive fibrous proteins, and ensures the exact reproduction of native repeat sequences, anal…

Studies on the tolerance of silk, nylon, dacron and collagen suture material in the cornea of the rabbit.

With rabbit corneas we did comparative biomicroscopical and histological investigations on material tolerance of the following threads at present used in microsurgery: Barraquer silk (virgin silk), nylon 30 mum (Tübinger nylon), dacron (polyester) 40 mum and plain collagen 7-0. The sutures were implanted into the cornea intra-lamellarly, completely under the microscope. The examinations showed only very little tissue reaction in all these materials.

Spider-silk-inspired dynamic hard-soft ionogels enabled by solvation-contrast-guided phase separation.

Stretchable ionogels are promising epidermal biointerfaces, yet integrating mechanical robustness with efficient viscoelastic dissipation remains challenging. Inspired by the hard-soft hierarchical architecture of spider silk, we develop a phase-separated ionogel by copolymerizing N,N-dimethylacrylamide (DMAA) and acryloxyethyl trimethylammonium chloride (DAC) in an ionic-liquid matrix. Differences in polymer/ionic-liquid affinity and local ionic association are proposed to promote nanoscale heterogeneity, as supported by AFM, USAXS, spectroscopy, and molecular simulations. The resulting dynam…

Eudragit E100/silk fibroin hybrid nanoparticles for the controlled oral delivery of amphotericin B.

AIMS: Amphotericin B (AmB) is the first-line antifungal compound for severe fungal infections, yet it possesses high renal toxicity, low water solubility, and acidic instability in the stomach. Herein, we developed novel nanoparticles from silk fibroin with/without Eudragit E100 (FNP and FNP/EE) for the controlled oral delivery of AmB. METHODS AND RESULTS: The particles were formulated by two different methods of co-condensation and adsorption, which yielded comparable particle sizes of ~1500 nm (FNP-AmB) and ~800 nm (FNP/EE-AmB) and AmB entrapment efficiencies of >70%. AmB was rapidly adsorb…

Interfacial Self-Polymerization of Tannic Acid for Functional Flame-Retardant Coatings on Silk Fibers.

Developing surface finishes that impart flame retardancy to silk while avoiding phosphorus-, metal-, and halogen-containing additives remains challenging, because biobased polyphenols often provide limited protection when directly deposited on fibers. Here, we report an aqueous oxidative self-polymerization strategy that converts tannic acid (TA) into surface-deposited poly(tannic acid) (pTA) particulates on silk. Under mild alkaline conditions, TA underwent self-polymerization and aggregation, and the resulting pTA particles formed a polyphenolic coating on silk fibers through dip coating. In…
