Sphingolipids and Male Reproductive Health: A Narrative Review of Their Roles in Spermatogenesis, Fertility, and Dysfunction.

Sphingolipids are bioactive lipids involved in cellular signaling and structural processes. Recent studies highlight their critical role in spermatogenesis, sperm functionality, and male infertility. Key sphingolipid metabolites, such as ceramide, ceramide-1-phosphate, and sphingosine-1-phosphate, mediate signaling pathways related to apoptosis, proliferation, stress responses, necrosis, inflammation, autophagy, aging, and differentiation. This review aims to summarize recent advancements in understanding the roles of sphingolipids in spermatogenesis, sperm motility, capacitation, the acrosome

[Mitochondrial damage and male reproductive dysfunction: Progress in research].

Mitochondria are the center of sperm metabolism and essential for all aspects of flagellar motility, capacitation, acrosome reaction and gametic fusion of sperm. Abnormalities in the mitochondrial ultrastructure, membrane potential, reactive oxygen species, Ca2+ homoeostasis, sperm apoptosis, mitochondrial DNA, mitochondrial enzymes and proteomic activity can decrease sperm quality and even cause male infertility. This paper elucidates the impact of mitochondrial dysfunction on sperm by reviewing and analyzing relevant literature on mitochondria and male reproductive function, aiming to provid

The impact of epididymal proteins on sperm function.

The epididymis is necessary for post-testicular sperm maturation as it provides the milieu required for spermatozoa to gain the ability for progressive movement and fertilization. In the epididymis the sperm protein, lipid and small RNA content are heavily modified due to interaction with luminal proteins secreted by the epididymal epithelium and extracellular vesicles, epididymosomes. This review focuses on epididymal proteins demonstrated to have an effect on sperm functions, such as motility, capacitation, acrosome reaction, sperm-zona pellucida binding and sperm-egg binding, as well as on

Distribution of glycoproteins on feline testicular sperm, epididymal sperm and ejaculated sperm.

Glycoproteins (GPs) are known to be involved in the phenomenon of sperm maturation and capacitation. In the present study, we investigated the attachment of GPs on sperm cell membrane during the process of feline sperm maturation from testicular sperm to ejaculated sperm by using 8 FITC-labeled lectins. The results showed that 3 types of GPs were presented on testicular sperm and 7 on caput epididymal sperm. Corpus and cauda epididymal sperm and ejaculated sperm had GPs detected by 8 FITC-labeled lectins used in the present study. This study demonstrates the part of the characteristic of GPs t

Cryopreservation of bull semen is associated with carbonylation of sperm proteins.

Artificial insemination with cryopreserved semen enables affordable, large-scale dissemination of gametes with superior genetics. However, cryopreservation can cause functional and structural damage to spermatozoa that is associated with reactive oxygen species (ROS) production, impairment of sperm motility and decreased fertilizing potential, but little attention has been paid to protein changes. The goal of this study was to investigate the oxidative modifications (measured as carbonylation level changes) of bull spermatozoa proteins triggered by the cryopreservation process. Flow cytometry

Changes in aquaporins mRNA expression and liquid storage at 17°C: A potential biomarker of boar sperm quality?

Artificial insemination (AI) for pigs relies on liquid storage of extended semen at 17°C, which preserves sperm quality and ensures its fertilizing capacity. Routine quality controls include the evaluation of sperm motility, viability and capacitation status. The physiological functions of all these features depend on transmembrane aquaporins (AQPs), proteins playing key roles in osmoadaptation. In this study, we made a relative quantification, using RT-qPCR, of the mRNA of several sperm AQPs in AI-liquid semen doses before and after a 48-hr incubation period, aiming to determine possible quan

Male infertility testing: reactive oxygen species and antioxidant capacity.

Reactive oxygen species (ROS) are an integral component of sperm developmental physiology, capacitation, and function. Elevated ROS levels, from processes such as infection or inflammation, can be associated with aberrations of sperm development, function, and fertilizing capacity. We review the impact of ROS on sperm physiology, its place in infertility evaluation, the implications for reproductive outcomes, and antioxidant therapy. Our systematic review of PubMed literature from the last 3 decades focuses on the physiology and etiology of ROS and oxidative stress (OS), evaluation of ROS, and

Molecular insights into the divergence and diversity of post-testicular maturation strategies.

Competition to achieve paternity has coerced the development of a multitude of male reproductive strategies. In one of the most well-studied examples, the spermatozoa of all mammalian species must undergo a series of physiological changes as they transit the male (epididymal maturation) and female (capacitation) reproductive tracts prior to realizing their potential to fertilize an ovum. However, the origin and adaptive advantage afforded by these intricate processes of post-testicular sperm maturation remain to be fully elucidated. Here, we review literature pertaining to the nature and the p

Deciphering the Nexus Between Oxidative Stress and Spermatogenesis: A Compendious Overview.

Oxidative stress (OS) and reactive oxygen species (ROS) are one of the main reasons for the multifactorial concern - male infertility. ROS are active components of cellular metabolism that are intrinsic to cellular functioning and are present at minimal and unreactive levels in normal cells. They are an integral component of the sperm developmental physiology, capacitation, and function. As said "anything in excess is poison," so is the case with ROS. These, when produced in excess to the antioxidants present in the seminal plasma, cause multiple malformations during the process of spermatogen

Hyaluronan-degrading activity of mouse sperm hyaluronidase is not required for fertilization?

Sperm hyaluronidase has long been believed to participate in sperm penetration through the cumulus matrix. However, our previous works using male mice lacking either one of two sperm hyaluronidases, SPAM1 and HYAL5, conclusively showed that neither of these hyaluronidases is essential for fertilization. In this study, we examined whether the hyaluronan-degrading activity of mouse epididymal sperm is indeed required for the fertilization process. When the oocyte-cumulus complex was incubated with sperm protein extracts or capacitated epididymal sperm in the presence of the hyaluronidase inhibit

Selected Metabolites Found in Equine Oviductal Fluid do not Modify the Parameters Associated to Capacitation of the Frozen-thawed Equine Spermatozoa In Vitro.

In the horse, a repeatable protocol for in vitro fertilization has not been developed, possibly due to incomplete sperm capacitation. We have previously identified the metabolites present in equine oviductal fluid (OF). We aimed to test the effects of different metabolites found in equine oviductal fluid on quality parameters of frozen-thawed spermatozoa. Different concentrations of myoinositol (5-25 mM), lactate (6-60 mM), glycine (0.1-5 mM), β-alanine (1-6 mM), and histamine (0.05-0.4 mM) were added independently to modified Whitten's medium (pH = 7.25). Thawed equine spermatozoa (three stal…

Redox regulation of sperm surface thiols modulates adhesion to the fallopian tube epithelium.

Sperm that adhere to the fallopian tube epithelium are of superior quality and adhesion extends their fertile life. It has been postulated that periovulatory signals, as yet undefined, promote sperm release. In the in vitro studies described here, we examined the effects of several antioxidants, reportedly present within oviductal fluid, on the modulation of sperm-oviduct adhesion in bovine species. Results showed that 1) the cell-permeant thiols (penicillamine, beta mercaptoethanol, cysteine, and dithiotreitol), as well as the nonpermeant thiol, reduced glutathione, cause adhering spermatozoa
