Male germ cell meiosis is essential for the generation of haploid spermatozoa in mammals. Oocyte-specific KO of Exosc10 results in female subfertility by impairing germinal vesicle breakdown and disrupting the transcriptome during oocyte maturation. However, the involvement of EXOSC10 in the regulation of male germ cell meiosis remains poorly understood. This study explores the pivotal role of EXOSC10 ribonuclease in male germ cell meiosis in mice. Conditional deletion of Exosc10 in premeiotic germ cells impedes spermatogonial differentiation and obstructs meiotic progression in spermatocytes,

We tracked prosaposin (PSAP), a trophic factor, using an antibody specific to its proteolytic portion and an antibody to sortilin that traffics PSAP only to the lysosome. Immunostaining revealed that PSAP was distributed mainly on the basal side of seminiferous tubules, where many Sertoli cells and pachytene spermatocytes contained PSAP and its distribution differed depending on the stage of the spermatogenic cycle. The PSAP-sortilin complex was sorted to large lysosomes in the basal cytoplasm of Sertoli cells, where it may be processed into saposins. In contrast, in the thinner apical cytopla

BACKGROUND: As a member of RNA-binding protein, CDKN2AIP has been shown to play a critical role in stem cell pluripotency and somatic differentiation. Recent studies indicate that Cdkn2aip is essential for spermatogonial self-renewal and proliferation through the activating Wnt-signaling pathway. However, the mechanisms of how Cdkn2aip regulate spermatogenesis is poorly characterized.
RESULTS: We discovered that the CDKN2AIP was expressed in spermatocyte as well as spermatids and participated in spermiogenesis. Cdkn2aip-/- mice exhibited multiple sperm head defects accompanied by age dependen

Amida was first isolated from a rat hippocampal cDNA library as an Arc-associated protein. Previous studies showed that Amida is a nuclear protein and overexpression of Amida induces cell apoptosis. In this study, we found that Amida mRNA was expressed predominantly in rat testis by Northern blot analysis. During the development of testis, Amida mRNA was barely detectable until postnatal days 24 to 29 during which it increased to levels found in adults. However, Amida protein was not detected until postnatal day 32. Amida mRNA was found to be enriched in spermatocytes and less in round spermat

We have presented a brief discussion of WHO International Histological Classification of Testicular Germ Cell Tumors. This classification divides the tumors into those of one histologic type and those of more than one type. Seven basic histological categories are recognized: seminoma, spermatocytic seminoma, embryonal carcinoma, yolk sac tumor, polyembryoma, choriocarcinoma and teratoma. Comparison of the WHO classification with the Pugh-Cameron classification reveals that except for seminoma and spermatocytic seminoma comparability between the 2 classifications is impossible. The relationship

Hybrids between closely related but genetically incompatible species are often inviable or sterile. Cattle-yak, an interspecific hybrid of yak and cattle, exhibits male-specific sterility, which limits the fixation of its desired traits and prevents genetic improvement in yak through crossbreeding. Transcriptome profiles of testicular tissues have been generated in cattle, yak, and cattle-yak; however, the genetic variations underlying differential gene expression associated with hybrid sterility have yet to be elucidated. We detected differences in the cellular composition and gene expression

Fertility restoration using autologous testicular tissue transplantation is relevant for infertile men surviving from childhood cancer and, possibly, in men with absent or incomplete spermatogenesis resulting in the lack of spermatozoa in the ejaculate (non-obstructive azoospermia, NOA). Currently, testicular tissue from pre-pubertal boys extracted before treatment with gonadotoxic cancer therapy can be cryopreserved with good survival of spermatogonial stem cells. However, strategies for fertility restoration, after successful cancer treatment, are still experimental and no clinical methods h

BACKGROUND: Cryopreservation of immature testicular tissue (ITT) is currently the only option to preserve fertility of prepubertal patients. Autologous transplantation of ITT may not be safe or appropriate for all patients. Therefore, methods to mature ITT ex vivo are needed.
OBJECTIVES: Aim to investigate the feasibility of inducing in vitro spermatogenesis from ITT cryopreserved for pediatric patients prior to initiation of gonadotoxic therapy.
MATERIALS AND METHODS: Cryopreserved-thawed ITT from prepubertal and peripubertal patients were cultured for 7, 16, and 32 days in medium with no h

The present study was designed to see the effects of parenterally administered drugs cimetidine and bromocriptine affecting serum prolactin upon the fertility of adult male albino rats. Ninety adult young male albino rats between the ages of 60 to 120 days were selected. The animals were divided into three groups. Cimetidine was administered in a dose of 200 mg/kg body weight to group B intramuscularly and in addition to cimetidine, bromocriptine in a dose of 2.5 mg/day intramuscularly was given to group C. Normal saline was administered intramuscularly to control group A. Plasma prolactin was

BRD8, a member of the bromodomain-containing protein family, is a chromatin-associated protein that recognizes acetylated histones and recruits regulatory factors to modulate gene expression. Despite extensive studies of BRD8 in tumor biology, its physiological roles in normal tissues remain largely unknown. Given its enriched expression in the testis, we investigated the role of Brd8 in male reproduction using a germ cell-specific Brd8 knockout mouse model. Brd8 deficiency resulted in a marked reduction in testicular size and sperm counts, accompanied by severe defects in germ cell differenti

BACKGROUND: Cisplatin is a crucial chemotherapeutic agent used for treating various cancers; however, its excessive use can cause irreversible damage to the reproductive system, and the protein expression profile of cisplatin-induced testicular injury remains unclear.
METHODS: Male C57BL/6 mice were treated with cisplatin at various doses, and testes were collected for histological, immunofluorescence, and proteomic analyses. Germ cell loss and apoptosis were assessed using H&E staining, TUNEL assays, and immunofluorescence for LIN28A, SYCP3, MVH, and CDK1. Label-free quantitative proteomics

Klinefelter syndrome (KS) is a chromosome disorder characterized by small firm testes, gynecomastia, hypogonadism, and abnormally elevated concentrations of follicle-stimulating hormone (FSH). Most KS patients show a classic 47,XXY karyotype, while about 20% have other numeric sex chromosome abnormalities, including mosaicisms. 47,XXY/46,XX mosaicism is extremely rare, and has been reported in just 10 individuals with features suggestive of KS. None of these had any spermatozoa in their ejaculate or testicular samples. We describe the first case of sperm retrieval in a 19-year-old male patient

Long noncoding RNAs (lncRNAs) are important regulatory biomolecules responsible for many cellular processes. The aging of mammals is manifested by a slow and gradual decline of physiological functions after adulthood, progressively resulting in age-related diseases. Testis comprises different cell-types with defined functions for producing haploid gametes and androgens in males, contributing gene-pool to the next generation with genetic variations to species for evolutionary advantage. The LINC-RBE (long intergenic noncoding-rat brain expressed) RNA showed highest expression in the Leydig cell

Although exposure to environmental toxicants, including endocrine-disrupting chemicals, is thought to be a possible cause of male infertility, the pathogenesis of male reproductive disorders remains unclear. In the present study, we used Gulo-/- mutant mice, which are unable to synthesize ascorbic acid, to study the importance of dietary vitamin C (VC) on spermatogenesis. Regular chow containing approximately 110 mg/kg VC is unable to support the growth of these mutant mice, but a VC supplement in their drinking water (330 mg/L) is able to ameliorate the VC deficiency. Testes of Gulo-/- mutant

The present study examined the quantitative and qualitative effects of suppressed testicular testosterone concentration upon spermatogenesis. Adult male rats were given sc implants of 0.5- to 5-cm long testosterone filled silastic capsules (TC) for 10 weeks. Testicular testosterone concentrations were suppressed to below 5% of the control level in rats receiving 1.5- to 5-cm TC implants. Spermatogenesis was arrested at early spermatids in rats given 1.5-cm TC implants, but was maintained in those given 3- or 5-cm TC implants. The number of A1 spermatogonia was 10-15% higher in rats bearing 0.7

The goal of meiosis is typically to produce haploid gametes (eggs or sperm). Failure to do so is catastrophic for fertility. However, Lepidopteran (moths and butterflies) males produce two sperm morphs: nucleated (eupyrene) sperm and anucleated (apyrene) sperm, both of which are essential for fertilization. The meiotic differences in the two types of spermatogenesis are unclear, and our knowledge of the molecular differences between eupyrene and apyrene spermatogenesis is extremely limited in all systems. The only factor identified as being required for apyrene spermatogenesis is Sex-lethal (S

Haploid males of hymenopteran species produce gametes through an abortive meiosis I followed by meiosis II that can either be symmetric or asymmetric in different species. Thus, one spermatocyte could give rise to two spermatids with either equal or unequal amounts of cytoplasm. It is currently unknown what molecular features accompany these postmeiotic sperm cells especially in species with asymmetric meiosis II such as bees. Here we present testis single-cell RNA sequencing datasets from the honeybee (Apis mellifera) drones of 3 and 14 days after emergence (3d and 14d). We show that, while 3

A simple technique for the identification of the different spermatogenetic stages of Onchocerca volvulus is described. It is practicable for large-scale examinations in the field. The reproductivity of male worm populations can be assessed and directly correlated with the corresponding female worm populations. The activity of male filariae decreases with the increasing age of the worm population. Only few male worms transmit spermatozoa during mating, but spermatogenesis is continous also in very old male individuals. These old male filariae store high numbers of spermatozoa and cease their mi

Spermatogenesis is a highly organized process by which undifferentiated spermatogonia self-renew and differentiate into spermatocytes and spermatids. The entire developmental process from spermatogonia to sperm occurs within the seminiferous tubules. Spermatogenesis is supported by the close interaction of germ cells with Sertoli cells. In this study, testicular tissues were collected from Hu sheep at 8 timepoints after birth: 0, 30, 90, 180, 270, 360, 540, and 720 days. Immunofluorescence staining and histological analysis were used to explore the development of male germ cells and Sertoli ce

Thyroid hormone (T3 ) acts on the testis via thyroid hormone receptor alpha 1 (TRα1), though the cellular localization of TRα1 in testis remains controversial. Studies on the presence of TRα1 in the epididymis are also lacking. The present study, therefore, examined the cellular localization and expression pattern of TRα1 in testis and epididymis of Parkes mice during postnatal development. Immunohistochemical results showed localization of TRα1 in interstitial and tubular compartments of the testis all through the development. On postnatal day (PND) 14, only leptotene spermatocytes showed TRα
