TBC1D21 Potentially Interacts with and Regulates Rap1 during Murine Spermatogenesis.

Few papers have focused on small guanosine triphosphate (GTP)-binding proteins and their regulation during spermatogenesis. TBC1D21 genes (also known as male germ cell RAB GTPase-activating protein MGCRABGAP) are related to sterility, as determined through cDNA microarray testing of human testicular tissues exhibiting spermatogenic defects. TBC1D21 is a protein specifically expressed in the testes that exhibits specific localizations of elongating and elongated spermatids during mammalian spermiogenesis. Furthermore, through co-immunoprecipitation (co-IP) and nano liquid chromatography⁻tandem

Mitochondria-targeted antioxidant SkQ1 improves spermatogenesis in Immp2l mutant mice.

Previous studies have confirmed that spermatogenesis in homozygous Immp2l mutant male mice was normal at the age of 6 months, but was significantly abnormal at the age of 13 months. Meanwhile, oxidative stress is reported to be involved in spermatogenic impairment in old mutant mice. However, it is unclear whether antioxidant treatment is a suitable intervention for improving spermatogenesis in old mutant mice. This study sought to investigate the effect of mitochondria-targeted antioxidant SkQ1 on spermatogenesis in homozygous Immp2l mutant mice. Immp2l mutant mice were treated with the mitoc

Cypermetherin toxic effects on spermatogenesis and male mouse reproductive organs.

Cypermetherin has been implicated in the development of a variety of reproductive disorders in humans and infertility in wild life, where it increases the death rate of the offspring and induces aggressiveness (Elbetieha et al., 2001). Studies in workers exposed to handling of agro pesticides indicate that they have defects in their reproduction capabilities characterized by infertility and/or a decrease in the fertilizing potential, fetal death. In this study, mice weighing 30-35 g were used, separated in 3 groups, (1) control (2) vehicle (oil) and (3) experimental (Cypermetherin and oil). Th

Gut-derived succinic acid potentiates high-altitude-related spermatogenesis dysfunction.

Exposure to high altitude (HA) is linked to male spermatogenesis impairment and microbial imbalance; however, the association and underlying mechanisms remain unexplored. Herein, we demonstrate that HA-induced gut microbiota changes in humans and mice lead to reduced sperm quality. Specifically, we observe an increase in intestinal Clostridium symbiosum colonization in HA human populations and mice exposed to HA-mimicking conditions. Furthermore, C. symbiosum causes a decline in sperm quality through succinic acid (su) production. Mechanistically, su targets G-protein-coupled receptor 91(GPR91

TNXB Regulates Spermatogenesis by Maintaining Cytoskeleton and Cell Junction Stability.

Tenascin-X (TNXB) is an extracellular matrix glycoprotein known for its structural and regulatory roles in connective tissues, yet its pathophysiological role in testicular development and spermatogenesis remains uncharacterized. Here, we identified TNXB as clinically implicated in human spermatogenic disorder, with a significant downregulation in the testes of patients with non-obstructive azoospermia (NOA) revealed by isobaric tags for relative and absolute quantification (iTRAQ)-based proteomics. Developmental profiling in mice revealed a progressive increase in TNXB expression with testicu

Systematic identification of Y-chromosome gene functions in mouse spermatogenesis.

The mammalian Y chromosome is essential for male fertility, but which Y genes regulate spermatogenesis is unresolved. We addressed this by generating 13 Y-deletant mouse models. In Eif2s3y, Uty, and Zfy2 deletants, spermatogenesis was impaired. We found that Uty regulates spermatogonial proliferation, revealed a role for Zfy2 in promoting meiotic sex chromosome pairing, and uncovered unexpected effects of Y genes on the somatic testis transcriptome. In the remaining single Y-gene deletants, spermatogenesis appeared unperturbed, but testis transcription was still altered. Multigene deletions, i

Historical Aspects of Testicular Function: Virility, Androgen Production, and Spermatogenesis.

From antiquity, man has been fascinated by at least 2 processes of testicular function: virility and reproduction; their biological basis was uncovered beginning in the mid-19th century. We have divided the search into 3 epochs: the speculative and observational, the experimental, and the biochemical/physiological. The first begins with Susruta, approximately 3000 years ago, and winds its way through the Greek, Roman, the Christian Bible, Arabic, Chinese, and Indian pathways before coalescing in Europe at the dawn of the Renaissance. The second began with. Thomas Willis, who postulated a viril

Dissecting the enhancer gene regulatory network in early Drosophila spermatogenesis.

Cellular decision-making and tissue homeostasis are governed by transcriptional networks shaped by chromatin accessibility. Using single-nucleus multi-omics, we jointly profile gene expression and chromatin accessibility in 10,335 cells from the Drosophila testis apical tip. This enables inference of 147 cell type-specific enhancer-gene regulons using SCENIC + . We functionally validate key transcription factors, including ovo and klumpfuss, known from other stem cell systems but not previously linked to spermatogenesis. CRISPR-mediated knockout reveals their essential roles in germline stem c

Multidimensional mechanistic analysis elucidates spermatogenesis disruption induced by bisphenol S.

Bisphenol S (BPS), a primary substitute for bisphenol A, has led to widespread environmental exposure globally. However, the effects of BPS on spermatogenesis disruption (SD) remain contentious, and the underlying mechanisms responsible for SD induced by BPS are not fully understood. In this study, we employed a multidimensional computational strategy, integrating network toxicology, bioinformatics, systematic machine learning, single-cell analysis, and biomolecular modeling, to comprehensively elucidate the molecular mechanisms by which BPS induces SD. Our findings suggest that BPS may disrup

Ex Vivo Spatiotemporal Characterization of Spermatogenesis in Mouse Testicular Organoids.

Testicular organoids (TOs) offer an opportunity to preserve fertility, but current TO protocols are limited by suboptimal maintenance of the organoid structure, meiotic defects, and incomplete in vitro spermatogenesis (IVS). Here, a strategy is developed for self-reconstitution of single-cell suspensions of neonatal mouse testes into TOs containing the major testicular cell types and generation of tubular-like structures and haploid spermatids. Morphological and lineage-specific marker analyses revealed that these TOs met the criteria for organoids, including germ cell differentiation and reca

ZDHHC5 deficiency impairs spermatogenesis and causes male infertility in mice.

IN BRIEF: S-palmitoylation is a key protein modification, but the role of ZDHHC5 in male fertility was unknown. This study shows ZDHHC5 loss impairs spermatogenesis, causing male infertility, and identifies ZDHHC5 as a key regulator, offering new insights into infertility mechanisms. ABSTRACT: S-palmitoylation is a reversible post-translational modification critical for protein localization and function. ZDHHC5, a member of the ZDHHC family of palmitoyltransferases, has well-established roles in cardiovascular, neural, and metabolic systems, but its function in male reproduction remains uncle

Deficiency of nucleosome-destabilizing factor GLYR1 dampens spermatogenesis in mice.

Aberrant sperm morphology hinders sperm motility and causes male subfertility. Spermatogenesis, a complex process in male germ cell development, necessitates precise regulation of numerous developmental genes. However, the regulatory pathways involved in this process remain partially understood. We have observed the widespread expression of Glyr1, the gene encoding a nucleosome-destabilizing factor, in mouse testicular cells. Our study demonstrates that mice experiencing Glyr1 depletion in spermatogenic cells exhibit subfertility characterized by a diminished count and motility of spermatozoa.

Loss of PMFBP1 Disturbs Mouse Spermatogenesis by Downregulating HDAC3 Expression.

PURPOSE: Polyamine modulating factor 1 binding protein (PMFBP1) acts as a scaffold protein for the maintenance of sperm structure. The aim of this study was further to identify the new role and molecular mechanism of PMFBP1 during mouse spermatogenesis. METHODS AND RESULTS: We identified a profile of proteins interacting with PMFBP1 by immunoprecipitation combined with mass spectrometry and demonstrated that class I histone deacetylases, particularly HDAC3 and chaperonin-containing TCP1 subunit 3 (CCT3), were potential interaction partners of PMFBP1 based on network analysis of protein-protei

The Decellularized Calf Testis: Introducing Suitable Scaffolds for Spermatogenesis Studies.

BACKGROUND: Men's infertility and lack of production of healthy and active sperm are concerns of recent years in most countries. Studies on the preparation of extracellular matrix (ECM) from decellularization of testis tissue and spermatogenesis could provide proper results to solve some of the men's infertility problems. This study aims to decellularize calf testis by different methods to reach a suitable scaffold and introduce it in spermatogenesis studies. MATERIALS AND METHODS: In this experimental study, calf testis were decellularized by a freeze-de freeze, 1% sodium deoxycholate (SD),

Elevated Id2 expression causes defective meiosis and spermatogenesis in mice.

BACKGROUND: Inhibitors of DNA binding (ID) proteins mainly inhibit gene expression and regulate cell fate decisions by interacting with E-proteins. All four ID proteins (ID1-4) are present in the testis, and ID4 has a particularly important role in spermatogonial stem cell fate determination. Several lines of evidence indicate that ID proteins are involved in meiosis; however, functional experiments have not been conducted to validate this observation. RESULTS: In this study, we report that ID2 is enriched in spermatocytes and that forced ID2 expression in germ cells causes defects in spermat

[Influence of platelets growth factors on spermatogenesis after electronic irradiation].

BACKGROUND: The number of cases of male infertility is steadily growing every year, and therefore it is necessary to develop new methods for the diagnosis and treatment of this disease. It is known that plasma enriched with platelets, the α-granules of which contain growth factors, possesses high regenerative activity; therefore, we can expect positive results from its use for the restoration of spermatogenic epithelium. OBJECTIVE: Morphological assessment of spermatogenesis after local β-irradiation with a dose of 8 Gy and the introduction of growth factors. MATERIAL AND METHODS: Wistar rat
