Dynamic regulation of FGF23 by Fam20C phosphorylation, GalNAc-T3 glycosylation, and furin proteolysis.

The family with sequence similarity 20, member C (Fam20C) has recently been identified as the Golgi casein kinase. Fam20C phosphorylates secreted proteins on Ser-x-Glu/pSer motifs and loss-of-function mutations in the kinase cause Raine syndrome, an often-fatal osteosclerotic bone dysplasia. Fam20C is potentially an upstream regulator of the phosphate-regulating hormone fibroblast growth factor 23 (FGF23), because humans with FAM20C mutations and Fam20C KO mice develop hypophosphatemia due to an increase in full-length, biologically active FGF23. However, the mechanism by which Fam20C regulate

A plant protein-based fat texturizer: Enzymatic bioproduction and application in non-fat yogurt.

The growing demand for low-fat or non-fat food products highlights the need for fat texturizers that replicate the mouthfeel and texture of fat. This study investigates the enzymatic modification of lentil protein isolates (LPI) to generate amphiphilic polypeptides for fat replacement in non-fat foods, exemplified by yogurt. LPI was hydrolyzed using Alcalase at varying enzyme-to-substrate ratios (1:100-1:2000) and hydrolysis times (2-240 min). Enzymatic hydrolysis significantly modified LPI, with partially hydrolyzed proteins (LPP5-10, <50 kDa) exhibiting balanced functional properties, includ

Enhanced nattokinase production by optimized fermentation conditions of douchi-derived Bacillus subtilis natto NK11 and evaluation of its thrombolytic activity.

Nattokinase is considered effective in preventing and treating thromboembolic diseases due to its significant fibrinolytic activity, thus demonstrating great application potential in the fields of pharmaceuticals and functional foods. In this study, nattokinase-producing strains were isolated and identified from traditional fermented soybean products. The fermentation conditions were optimized via orthogonal experiments, and the crude enzyme was then characterized by SDS-PAGE, zymogram analysis, and in vitro blood clot thrombolytic assay. The results indicated that a strain of Bacillus subtili

Combined enzyme-assisted aqueous extraction and supercritical CO₂ produces Rice bran oil with enhanced bioactive compounds and antioxidant activity.

Rice bran (RB), a by-product of rice milling, is rich in bioactive compounds like γ-oryzanol and vitamin E. This study aimed to optimize enzyme-assisted aqueous extraction (EAAE) of rice bran oil (RBO) by employing response surface methodology to produce RBO using a combination of EAAE and supercritical CO2 (EAAE+SC-CO2) and compared the results with hexane extraction (HE). Optimized EAAE conditions (4:1 alcalase to pentopan, pH 7, 40 °C) indicated enhanced antioxidant activity and oil quality. Although HE gave the highest yield (9.03 %), EAAE+SC-CO2 retained higher levels of γ-oryzanol (26.30

Alcalase-Assisted Mytilus edulis Hydrolysate: A Nutritional Approach for Recovery from Muscle Atrophy.

Muscle atrophy is a complex physiological condition caused by a variety of reasons, including muscle disuse, aging, malnutrition, chronic diseases, immobilization, and hormonal imbalance. Beyond its effect on physical appearance, this condition significantly reduces the quality of human life, thus warranting the development of preventive strategies. Although exercising is effective in managing this condition, it is applicable only for individuals who can engage in physical activities and are not bedridden. A combination of exercise and nutritional supplementation has emerged as a more advantag

PCSK9 and Inflammation: Their Role in Autoimmune Diseases, with a Focus on Rheumatoid Arthritis and Systemic Lupus Erythematosus.

Despite a clear epidemiological link between autoimmune disease and cardiovascular (CV) risk exists, pathophysiological explanations are extremely complex and far from being elucidated. Dysregulation of metabolic pathways and chronic low-grade inflammation represent common pathways, but CV risk still remains underestimated in patients with autoimmune diseases. Among different candidate mediators, pro-protein convertase subtilisin/kexin type 9 (PCSK9) is attracting growing attention, due to a combined effect on lipid metabolism and inflammatory response. Studies on PCSK9 inhibitors have establi

Antioxidant and Angiotensin I-Converting Enzyme (ACE) Inhibitory Peptides Obtained from Alcalase Protein Hydrolysate Fractions of Hemp (Cannabis sativa L.) Bran.

Proteins from hemp bran (HPB), a byproduct of the hemp seed food-processing chain, were chemically extracted, hydrolyzed by Alcalase, and separated by membrane ultrafiltration into four fractions (MW <1, 1-3, 3-5, and >5 kDa). The antioxidant and antihypertensive properties of the initial extract and the fractions were evaluated by in vitro assays for their ability to scavenge radical species, bind with metal ions, reduce ferric ions, and inhibit angiotensin-converting enzyme (ACE) activity. Bioactive peptides were identified by high-resolution mass spectrometry and sequence comparison with BI

Affinity purification of copper chelating peptides from chickpea protein hydrolysates.

Chickpea protein hydrolysates obtained with alcalase and flavourzyme were used for purification of copper chelating peptides by affinity chromatography using copper immobilized on solid supports. The chelating activity of purified peptides was indirectly measured by the inhibition of beta-carotene oxidation in the presence of copper. Two protein hydrolysates, obtained after 10 and 100 min of hydrolysis, were the most inhibitory of beta-carotene oxidation. Purified copper chelating peptides from these protein hydrolysates contained 19.7 and 35.1% histidine, respectively, in comparison to 2.7 an

A role for PACE4 in the proteolytic activation of anthrax toxin protective antigen.

Several bacterial protein toxins require activation by eukaryotic proteases. Previous studies have shown that anthrax toxin protective antigen (PA), Pseudomonas exotoxin A (PE), and diphtheria toxin (DT) are cleaved by furin C-terminal to the sequences RKKR, RQPR, and RVRR, respectively. Because furin-deficient cells retain some sensitivity to PA and DT, it is evident that other cellular proteases can activate these toxins. Whereas furin has been shown to require arginine residues at positions -1 and -4 for substrate recognition, another protease with an activity which could substitute for fur

Total Synthesis of Enlicitide Decanoate.

We report the total synthesis of enlicitide decanoate, an orally bioavailable inhibitor of proprotein convertase subtilisin/kexin type 9 that is being developed for the treatment of atherosclerotic cardiovascular disease. It is a highly complex macrocyclic peptide with a significant number of nonpeptide structural elements that presents a daunting synthetic chemistry challenge. We describe the development of a convergent, efficient, and robust manufacturing process that enables the large-scale production of enlicitide.

Lipid metabolism abnormalities induced by elevated PCSK9 in polycystic ovary syndrome mice lead to ovarian granulosa cell apoptosis via AMPK and GSK-3β.

In brief: The mechanism underlying granulosa cell apoptosis in polycystic ovary syndrome remains unclear. This study investigates the role of proprotein convertase subtilisin/kexin type-9, a lipid metabolism-regulating molecule, in inducing ovarian granulosa cell apoptosis through lipid metabolism abnormalities. Abstract: Polycystic ovary syndrome (PCOS) is a common reproductive endocrine disorder among women of childbearing age, significantly impacting their fertility needs. Granulosa cell apoptosis in the ovaries is one of the primary factors affecting follicular growth and development as we

Current State of Pediatric Hyperlipidemia: Epidemiology, Management, and Emerging Therapies.

Hyperlipidemia in children is an increasingly recognized public health concern. Both primary genetic disorders-notably familial hypercholesterolemia, which affects roughly 1 in 250 children-and secondary factors like obesity, diabetes, hypothyroidism, and nephrotic syndrome contribute to pediatric dyslipidemia. Unmanaged childhood hyperlipidemia can track into adulthood and drive premature atherosclerosis and cardiovascular disease. The American Academy of Pediatrics and National Heart, Lung, and Blood Institute recommend universal lipid screening at ages 9-11 years and again in late adolescen

Simvastatin Resistance of Leishmania amazonensis Induces Sterol Remodeling and Cross-Resistance to Sterol Pathway and Serine Protease Inhibitors.

The sterol biosynthesis pathway of Leishmania spp. is used as a pharmacological target; however, available information about the mechanisms of the regulation and remodeling of sterol-related genes is scarce. The present study investigated compensatory mechanisms of the sterol biosynthesis pathway using an inhibitor of HMG-CoA reductase (simvastatin) and by developing drug-resistant parasites to evaluate the impact on sterol remodeling, cross-resistance, and gene expression. Simvastatin-resistant L. amazonensis parasites (LaSimR) underwent reprogramming of sterol metabolism manifested as an inc

The hepatic WASH complex is required for efficient plasma LDL and HDL cholesterol clearance.

The evolutionary conserved Wiskott-Aldrich syndrome protein and SCAR homolog (WASH) complex is one of the crucial multiprotein complexes that facilitates endosomal recycling of transmembrane proteins. Defects in WASH components have been associated with inherited developmental and neurological disorders in humans. Here, we show that hepatic ablation of the WASH component Washc1 in chow-fed mice increases plasma concentrations of cholesterol in both LDLs and HDLs, without affecting hepatic cholesterol content, hepatic cholesterol synthesis, biliary cholesterol excretion, or hepatic bile acid me

Efficacy and safety of coadministration of rosuvastatin, ezetimibe, and colestimide in heterozygous familial hypercholesterolemia.

Aggressive low-density lipoprotein (LDL) cholesterol-lowering therapy is important for high-risk patients. However, sparse data exist on the impact of combined aggressive LDL cholesterol-lowering therapy in familial hypercholesterolemia (FH), particularly on side effects to changes in plasma coenzyme Q10 and proprotein convertase subtilisin/kexin type 9 levels. We enrolled 17 Japanese patients with heterozygous FH (12 men, 63.9 ± 7.4 years old) with single LDL receptor gene mutations in a prospective open randomized study. Permitted maximum doses of rosuvastatin (20 mg/day), ezetimibe (10 mg/d

Valorization of Camelina Seed Meal: Ethanolic Ammonium Hydroxide Extraction Coupled With Enzymatic Hydrolysis Enhances Protein Functionality.

Camelina is an emerging oilseed crop whose deoiled meal has considerable potential as a protein ingredient for food and feed applications. However, its utilization is limited by glucosinolates (GLs), which impart toxicity and undesirable flavors. Although conventional hexane extraction efficiently extracts oil, it retains GLs. Therefore, this study evaluated a novel ammonium hydroxide-ethanol-hexane (NH4OH-EtOH-hexane) extraction system to address the need for effective GL detoxification while simultaneously recovering oil and investigated its effects on the functional properties of protein hy
