Acute effect of labetalol on blood pressure in relation to the sympathetic nervous system and plasma renin activity.

A single oral dose of 300 mg labetalol was given to 10 subjects with uncomplicated primary hypertension; its effects on blood pressure, heart rate (HR), plasma renin activity (PRA), and plasma concentration of catecholamines and of their major metabolites, octopamine ( OTP ), and dopamine-beta hydroxylase (DbH) were investigated. Although HR did not change after dosing, both systolic and diastolic blood pressure decreased in 2 hr and remained below control up to 12 hr. There was symptomatic orthostatic hypotension in two subjects in the first 2 hr after dosing. PRA decreased in the first 2 hr

Neurotoxicity studies with a tropomyosin-related kinase A inhibitor, ASP7962, on the sympathetic and sensory nervous systems in rats.

ASP7962 is a small molecule inhibitor for the nerve growth factor (NGF) receptor, tropomyosin-related kinase A (TrkA). NGF contributes to the survival of sensory and sympathetic neurons through TrkA receptor activation. Gross, microscopic, and quantitative effects to the nervous system were evaluated following oral ASP7962 administration to Sprague Dawley rats for 4 weeks and 13 weeks and after a recovery period. Histopathological findings included reversible neuronal atrophy but no neuronal death in the sympathetic ganglia (cervicothoracic ganglion, cranial mesenteric ganglion or superior [cr

Role of the sympathetic nervous system in vasovagal syncope and rationale for beta-blockers and norepinephrine transporter inhibitors.

Vasovagal or neurocardiogenic syncope is a common clinical situation and, as with other entities associated with orthostatic intolerance, the underlying condition is a dysfunction of the autonomic nervous system. This article reviews various aspects of vasovagal syncope, including its relationship with orthostatic intolerance and the role of the autonomic nervous system in it. A brief history of the problem is given, as well as a description of how the names and associated concepts have evolved. The response of the sympathetic system to orthostatic stress, the physiology of the baroreflex syst

The effects of drug which destroy the sympathetic nervous system on the retrograde transport of nerve growth factor.

It has been proposed that the drugs (6-hydroxydopamine, guanethidine, vinblastine) which are known to destroy sympathetic neurons in neonatal animals do so by preventing the accumulation of retrogradely transported nerve growth factor (NGF). It was found, consistent with the proposal, that administration of 6-hydroxydopamine (100 mg/kg s.c.) or vinblastine (0.4 mg/kg s.c.) 16 h prior to the administration of [125I]NGF complete prevented the accumulation of retrogradely transported [125I]NGF in superior cervical ganglia of neonatal rats. Administration of 6-hydroxydopamine or vinblastine to adu

Functional-Optical Coherence Tomography: A Non-invasive Approach to Assess the Sympathetic Nervous System and Intrinsic Vascular Regulation.

Sympathetic nervous system dysregulation and vascular impairment in neuronal tissue beds are hallmarks of prominent cardiorespiratory diseases. However, an accurate and convenient method of assessing SNA and local vascular regulation is lacking, hindering routine clinical and research assessments. To address this, we investigated whether spectral domain optical coherence tomography (OCT), that allows investigation of retina and choroid vascular responsiveness, reflects sympathetic activity in order to develop a quick, easy and non-invasive sympathetic index. Here, we compare choroid and retina…

Significance of cardiac sympathetic nervous system abnormality for predicting vascular events in patients with idiopathic paroxysmal atrial fibrillation.

PURPOSE: Neuronal system activity plays an important role for the prognosis of patients with atrial fibrillation (AF). Using (123)I metaiodobenzylguanidine ((123)I-MIBG) scintigraphy, we investigated whether a cardiac sympathetic nervous system (SNS) abnormality would be associated with an increased risk of vascular events in patients with paroxysmal AF. METHODS: (123)I-MIBG scintigraphy was performed in 69 consecutive patients (67 + or - 13 years, 62% men) with paroxysmal AF who did not have structural heart disease. SNS integrity was assessed from the heart to mediastinum (H/M) ratio on del…

Sympathetic nervous system overactivity in the Wistar rat with proliferative lesions of ventral prostate induced by chronic stress.

OBJECTIVES: The sympathetic nervous system and hypothalamic-pituitary-adrenal axis have been known to be reactive to psychological factors such as stress in which several humoral factors are released. Growing data suggest that stress conditions could be associated with the development or aggravation of prostatic disease. The present study aims to investigate the influence of chronic stress and blockade of the sympathetic nerve on rat prostate. METHODS: We used 2 weeks of restraint water-immersion stress to induce a chronic psychological stress model of Wistar rats. Denervation of the peripher…

Sympathetic nervous system in salt-sensitive and obese hypertension: amelioration of multiple abnormalities by a central sympatholytic agent.

Excess activity of the sympathetic nervous system (SNS) is linked to human obese hypertension and to salt-sensitive hypertension. Paradoxically, reduced SNS activity has been implicated as a contributor to obesity, particularly in animal models, and salt loading usually inhibits SNS activity. We have investigated the relationship between SNS activity, diet, and hypertension in the obese spontaneously hypertensive rat (SHROB), a model with a recessive obesity trait superimposed on a hypertensive background with multiple metabolic abnormalities resembling human syndrome X. We examined the role o…

[Electron microscopy of human nasal mucosa in disturbances of the autonomic nervous system. I. Sympathetic denervation (author's transl)].

The fine structure of the human nasal mucosa is described shortly after unilateral stellate gang-lion blockage as well as in a case of sympathetic denervation of four weeks duration. For the clinical picture, corresponding to vasomotor rhinitis, there is a characteristic fine structural correlation consisting of dilatation and increased permeability of the vessels as well as stimulation of the nasal glands. In the glands, both increased activity and degenerative changes suggesting "secretory exhaustion" are observed. The findings differ from those seen in acute and chornic allergic rhinitis, b

Nos1 neurons in the paraventricular hypothalamic area modulate lipid metabolism via the sympathetic nervous system in male mice.

The selection of the appropriate energy substrate under different physiological conditions is a key aspect of the energy metabolism homeosta … neurons in the PVH area induces WAT lipolysis and fat oxidation in other peripheral tissues via the sympathetic nervous system, thereby increasing whole-body fat consumption. Inhibition of these neurons abolishes the increase in fat consumption during the light period, whereas long-term silencing lead to obesity independent of energy intake. These neurons are also necessary …

Long-term fasting-induced parasympathetic and sympathetic autonomic nervous system modulation in a subgroup of the GENESIS study.

Parasympathetic and sympathetic nervous systems are known to be modulated during long-term fasting, but temporal dynamics and persistence post-fasting are unclear. We analyzed daily heart rate variability (HRV), as a reflection of autonomic nervous system balance, as an exploratory analysis, in 16 participants from the GENESIS trial following the Buchinger fasting protocol (12 days of ~250 kcal/day). HRV was assessed via Polar H10 sensors daily 7 days before, during and 7 days after fasting. Overall, fasting led to an increase in parasympathetic activity, as indicated by higher root mean squar…

Autonomic nervous system reactivity to alcohol-related cues in cocaine use disorder: vagal-sympathetic balance and clinical implications.

RATIONALE: Chronic substance use disrupts autonomic nervous system (ANS) function, altering stress and reward processing. The clinical significance of autonomic responses to alcohol-related stimuli in cocaine use disorder (CUD) remains unclear. OBJECTIVE: This study investigated ANS reactivity to alcohol-related cues and emotional stimuli in individuals with CUD and healthy controls, aiming to characterize specific patterns of autonomic dysregulation and their relationship to craving. METHODS: Participants with CUD (n = 80) and healthy controls (n = 42) performed two tasks-alcohol cue exposu

Can a Sacrococcygeal Epidural of 0.25% Bupivacaine Prevent the Activation of the Sympathetic Nervous System during Feline Ovariectomy?

The ovariectomy (OVE) procedure can trigger somatosensory and visceral peritoneal nociception. Sacrococcygeal epidural (ScE) anesthesia may complement or replace systemic analgesia used for feline OVE, reducing opioid consumption and their related undesirable adverse effects and consequently reducing or completely blocking the sympathetic nervous system activation during this procedure. The present study aimed to evaluate the activation of the sympathetic nervous system resulting from adding an ScE injection of bupivacaine 0.25% (0.3 mL kg-1) in feline OVE and identify whether this translates …

Role of ventrolateral part of ventromedial hypothalamus in post-myocardial infarction cardiac dysfunction induced by sympathetic nervous system.

Psychological stress has been recognized as a contributing factor to worsened prognosis in patients with cardiac failure following myocardial infarction (MI). Although the ventrolateral part of the ventromedial hypothalamus (VMHVL) has been implicated in emotional distress, its involvement in post-MI cardiac dysfunction remains largely unexplored. This study was designed to investigate the effect of the VMHVL activation in the MI rat model and its underlying mechanisms. Our findings demonstrate that activation of VMHVL neurons enhances the activity of the cardiac sympathetic nervous system thr…

Schwann cell-specific Pten inactivation reveals essential role of the sympathetic nervous system activity in adipose tissue development.

There is increasing evidence that the sympathetic nervous system (SNS) plays an important role in adipose tissue development. However, the underlying molecular mechanism(s) associated with this remains unclear. SNS innervation of white adipose tissue (WAT) is believed to be necessary and sufficient to elicit WAT lipolysis. In this current study, mice with Schwann cell (SC)-specific inactivation of phosphatase and tensin homolog (Pten) displayed enlarged inguinal white adipose tissue (iWAT). This serendipitous observation implicates the role of SCs in mediating SNS activity associated with mous…

Pioglitazone treatment enhances the sympathetic nervous system response to oral carbohydrate load in obese individuals with metabolic syndrome.

CONTEXT: Insulin resistance is associated with blunted sympathetic nervous system (SNS) response to carbohydrate ingestion which may contribute to postprandial hypotension and impaired body weight homeostasis. OBJECTIVE: This study was conducted to examine the effects of pharmacological insulin sensitization on whole-body norepinephrine kinetics during a standard 75-g oral glucose tolerance test (OGTT) in obese, insulin resistant subjects with metabolic syndrome. METHODS: Un-medicated individuals (n=42, mean age 56±0.8 yrs, body mass index 34±0.6 kg/m(2)) were randomised to 12-weeks pioglita…
