Synergy of enediyne antibiotic lidamycin and temozolomide in suppressing glioma growth with potentiated apoptosis induction.

The present work evaluated the synergistic efficacy of an enediyne antibiotic lidamycin (LDM) plus temozolomide (TMZ) against glioma in vitro and in vivo. LDM plus TMZ inhibited the proliferations of rat glioma C6 cells and human glioma U87 cells more efficiently than the single usage of LDM or TMZ. In addition, LDM also potentiated the apoptosis inductions by TMZ in rat C6 cells and human U87 cells. Meanwhile, the results of TdT-mediated dUTP Nick End Labeling assay for subcutaneous U87 tumor sections indicated an enhanced apoptosis induction in vivo by LDM plus TMZ, which confirmed the high

Perfusion and diffusion MRI of glioblastoma progression in a four-year prospective temozolomide clinical trial.

PURPOSE: This study was performed to determine the impact of perfusion and diffusion magnetic resonance imaging (MRI) sequences on patients during treatment of newly diagnosed glioblastoma. Special emphasis has been given to these imaging technologies as tools to potentially anticipate disease progression, as progression-free survival is frequently used as a surrogate endpoint. METHODS AND MATERIALS: Forty-one patients from a phase II temolozomide clinical trial were included. During follow-up, images were integrated 21 to 28 days after radiochemotherapy and every 2 months thereafter. Assessm

O6-methylguanine-DNA methyltransferase in pretreatment tumour biopsies as a predictor of response to temozolomide in melanoma.

Resistance of tumour cells to methylating and monochloroethylating agents in vitro and in vivo has been linked to levels of the DNA repair protein O6-methylguanine-DNA methyltransferase (MGMT). In a clinical trial of temozolomide in advanced malignant melanoma, the relationship between pretreatment MGMT levels in biopsies of cutaneous tumours and involved lymph nodes and clinical response to the drug has been studied. Among 50 evaluable patients, there were three complete responses (CR), four partial responses (PR), six with stable disease (SD) and 37 with progressive disease (PD), with an ove

Targeting the Metabolic-Immune Regulator SLC35F6 Enhances Temozolomide and Anti-PD-L1 Therapy in Glioma.

Single-pathway targeted therapy is often insufficient to control malignant glioma progression, and the blood-brain barrier (BBB) remains a major obstacle to effective drug delivery. Therefore, discovering multifunctional target molecules and corresponding drugs that can penetrate the BBB can provide new strategies for targeted glioma therapy. Analysis of public databases and 179 clinical samples showed that solute carrier family 35 member F6 (SLC35F6) was upregulated in gliomas and associated with poorer clinical outcomes. Proteomic analyses and functional validation identified PIK3R2 as a pot

Primary Tumor Size and Tumor-Vessel Interface Following Capecitabine and Temozolomide for Pancreatic Neuroendocrine Tumor.

Capecitabine/temozolomide (CAPTEM) is an established regimen for patients with metastatic pancreatic neuroendocrine tumors (PanNET) that is being increasingly used for tumor volume reduction in patients with borderline anatomically resectable disease. We sought to understand the response of the primary tumor, defined as changes in the tumor-vascular interface (TVI). This is a retrospective, single-institution study of patients with locally advanced or metastatic PanNET treated with CAPTEM between 2010 and 2020. RECISTv1.1 measurements and TVI assessments of the primary tumor were performed on

Glioma Cell-Derived Apoptotic Extracellular Vesicles Promote Tumorigenesis and Temozolomide Resistance by Delivering LncRNA-XIST.

Gliomas are characterized by high incidence, high recurrence rate, high malignancy, high aggressiveness, high therapeutic difficulty, poor clinical prognosis, and high mortality. Therefore, overcoming temozolomide (TMZ) resistance in glioma has become a research hotspot. This study will investigate the effects of glioma cell-derived apoptotic extracellular vesicles (apoEVs) on tumorigenesis and TMZ resistance in glioma. apoEVs derived from TMZ-induced apoptotic glioma cells were isolated, extracted, and characterized. The effects of apoEVs and the extracellular vesicle inhibitor GW4869 on the

BET inhibition enhances temozolomide sensitivity in cervical cancer cells through ALDH suppression and epigenetic reprogramming.

BACKGROUND: Cervical cancer remains one of the most prevalent gynecological malignancies worldwide, and therapeutic resistance continues to limit the effectiveness of current treatment strategies. Aldehyde dehydrogenase 1 (ALDH1) has been associated with chemoresistance and cellular stress responses in cervical cancer, while bromodomain and extraterminal (BET) proteins have emerged as regulators of transcriptional and epigenetic programs involved in tumor progression. In this study, we investigated whether BET inhibition by JQ1 modulates cellular responses to temozolomide (TMZ) in cervical can

Rewiring cell death and evading repair: Evolution of temozolomide and emerging strategies against resistant glioblastoma.

Glioblastoma (GBM) is the most common and aggressive primary malignant brain tumor, characterized by a universally dismal prognosis. For over two decades, the alkylating agent temozolomide (TMZ) has remained the cornerstone of the standard-of-care "Stupp protocol" largely owing to its unique pH-dependent activation and favorable blood-brain barrier penetration. However, the clinical efficacy of TMZ is profoundly compromised by complex intrinsic and acquired resistance mechanisms. These are primarily driven by O6-methylguanine-DNA-methyltransferase (MGMT) overexpression, mismatch repair (MMR) d

Extracellular Vesicles From Glioblastoma Cells Reflect 2D vs. 3D Culture Adaptation and Resistance to Temozolomide.

Glioblastoma (GBM) is an aggressive brain tumor marked by extensive heterogeneity, resistance to therapy, and dismal prognosis. Extracellular vesicles (EVs) have emerged as key players in GBM biology, mediating intercellular communication and therapy adaptation. However, the exact functions and molecular impact of EVs in GBM remain incompletely understood. In this study, we performed a comparative proteomic analysis of U87MG GBM cells grown in two-dimensional (2D) monolayers and three-dimensional (3D) spheroids following temozolomide (TMZ) treatment, alongside characterization of EVs derived f

DoE-Optimized Chitosan-Coated pH-Sensitive Liposomes for Targeted Nose-to-Brain Delivery of Temozolomide.

Glioblastoma is an aggressive brain tumor with limited treatment efficacy due to poor blood-brain barrier (BBB) penetration and intrinsic resistance. Temozolomide (TMZ), the frontline chemotherapeutic, undergoes rapid hydrolysis at physiological pH, with low systemic bioavailability and efflux-mediated clearance. To address this, we developed chitosan-coated pH-sensitive liposomes (SpH-TMZ-CS-LIPO) that were optimized via Box-Behnken design, achieving 223.48 ± 1.93 particle size, 35.72 ± 0.51 zeta potential, and 78.85 ± 2.62% entrapment. The formulation exhibited pH-sensitive release, with cum

Ferulic Acid Increases Temozolomide Sensitivity in Glioblastoma Cells, Causing DNA Damage and Inhibiting Cell Proliferation.

AIM: To investigate the antitumor effects of ferulic acid (FA) on glioblastoma multiforme (GBM) cells both alone and in combination with temozolomide (TMZ), and also to determine the potential of this synergy for treatment processes. MATERIAL AND METHODS: Human glioblastoma U87-MG cells were used in this study. To evaluate the potential constructive interaction between temozolomide (TMZ) and ferulic acid (FA), a sequential treatment protocol was applied in which cells were first treated with TMZ (20, 40, and 80 μM) for 48 hours, followed by FA (1000 μM and 1500 μM) for an additional 24 hours.

Decitabine Reprograms Temozolomide-Resistant Glioblastoma Through Epigenetic Reactivation and Mesenchymal Attenuation: A Multi-Omics Study.

Background/Objectives: Glioblastoma (GBM) is the most lethal primary brain malignancy in adults, with a median overall survival of approximately 15 months. Temozolomide (TMZ) resistance develops in virtually all patients, and no second-line regimen has improved outcomes over the past two decades. The DNA methyltransferase inhibitor decitabine (DAC) has attracted interest as a chemosensitizer, but whether it directly reverses the TMZ-resistance transcriptome or operates through distinct, complementary mechanisms has not been tested at multi-omics resolution. Methods: We performed an integrative

Feedforward miR-181d degradation modulates population variance of methyl-guanine methyl transferase and temozolomide resistance.

Intratumoral heterogeneity plays a pivotal role in cancer evolution, providing the substrate for adaptation to selective pressures, including chemotherapy treatment. Here, we demonstrate that miR-181d modulates variability in methyl-guanine methyl transferase (MGMT) expression, contributing to this heterogeneity in glioblastoma, the most common form of adult primary brain tumor. Treatment with standard-of-care temozolomide (TMZ) chemotherapy triggers a feedforward loop that accelerates polyribonucleotide nucleotidyltransferase 1 (PNPT1)-dependent miR-181d degradation. This degradation requires

DNA polymerase eta protects human cells against DNA damage induced by the tumor chemotherapeutic temozolomide.

Human DNA polymerases can bypass DNA lesions performing translesion synthesis (TLS), a mechanism of DNA damage tolerance. Tumor cells use this mechanism to survive lesions caused by specific chemotherapeutic agents, resulting in treatment relapse. Moreover, TLS polymerases are error-prone and, thus, can lead to mutagenesis, increasing the resistance potential of tumor cells. DNA polymerase eta (pol eta) - a key protein from this group - is responsible for protecting against sunlight-induced tumors. Xeroderma Pigmentosum Variant (XP-V) patients are deficient in pol eta activity, which leads to

Targeted and Synergic Glioblastoma Treatment: Multifunctional Nanoparticles Delivering Verteporfin as Adjuvant Therapy for Temozolomide Chemotherapy.

Despite advances in cancer therapies, glioblastoma multiforme treatment remains inefficient due to the brain-blood barrier (BBB) inhibitory activity and to the low temozolomide (TMZ) chemotherapeutic selectivity. To improve therapeutic outcomes, in this work we propose two strategies, (i) photodynamic therapy (PDT) as adjuvant treatment and (ii) engineering of multifunctional theranostic/targeted nanoparticles ( m-NPs) that integrate biotin as a targeting moiety with rhodamine-B as a theranostic agent in pluronic P85/F127 copolymers. These smart m-NPs can surmount the BBB and coencapsulate mul
