Porcine zona pellucida proteins (ZPs) have been utilized as female immunocontraceptive antigens. The purpose of this study was to explore the potential use of silkworm recombinant bovine ZP4 as an alternative. When the protein was injected with monophosphoryl lipid A (MPL) - an immuno-stimulative agent - into two female goats, marked elevation of the anti-ZP4 titer was detected. Application of the purified specific IgG to a porcine in vitro fertilization system reduced the sperm penetration rate. In one goat, the cyclic profile of serum progesterone disappeared as the anti-ZP4 titer increased.

Enzymatically active stromal cells (EASC) in different ovarian tumors are concerned with hormon production. 198 cases of ovarian tumors were investigated by different histochemical methods. Distribution of lactate-and glucose-6-phosphate-dehydrogenase was investigated by plaimetric measurement.--EASC were found in benign ovarian tumors in 48%, in malignant in 30%. They are found exclusively in ovarian tumors and are completely absent in metastases. Incidence is dependent on histological type of tumor. With regard to untreated ovarian carcinoma containing EASC, these cells cover an aerea of 1.9

Fibrosis and tissue stiffening are hallmarks of ovarian ageing, linked to a decrease in fertility. However, the lack of three-dimensional (3D) characterization of ovary elasticity limits our understanding of localized elasticity patterns and their connection to tissue composition. Here, we developed an integrated approach to link ovarian elasticity, volume, and cell-matrix composition using quantitative micro-elastography (QME), a label-free, non-invasive method to study 3D microscale elasticity in conjunction with immunofluorescence microscopy. QME reveals distinct spatial elasticity patterns

Reliable prediction of reproductive toxicity remains a critical challenge in drug development and environmental safety. Here, a biomarker-integrated, fluorescent reporter-based reproductive organ-on-a-chip platform that recapitulates the multicellular composition, 3D architecture, endocrine signaling, and cyclic dynamics of the human menstrual cycle, is presented. The system is constructed using primary human theca, granulosa, endometrial stromal and stem cells, vascular endothelial cells, uterine macrophages, and myometrial smooth muscle cells, compartmentalized within collagen-hyaluronic aci

Single-cell atlas efforts have reshaped the way we understand cells across the human body. Despite their power, they have not been effectively used to study ovarian biology in the context of other tissues, nor have they comprehensively incorporated healthy tissues from pre-menopausal donors. A focused pre-menopausal single-cell atlas could both advance our understanding of this life stage and support identification of ovarian gene targets for indications prevalent among younger demographics, such as fertility management. Here, we present an integrated resource of single-cell datasets from pre-

This review discusses the management of ovarian function and participation in the process of a new class of cellular regulators--Gasotransmitters (NO, CO and H2S). According to modern concepts in regulatory processes, in addition to hypothalamic-pituitary mechanisms involved a large and diverse group of ovary cells (interstitial, internal theca and atretic bodies, follicular epithelium, teka-luteal and granulosa-luteal cells of the corpus luteum), which not only mediate the gonadotropic stimulation, but satisfy the for the production of a wide range of biologically active substances. STUDY QUESTION: Are key signalling pathways WNT, TGFβ/BMP, NOTCH, and HH involved in follicular atresia in the human adult ovary?
SUMMARY ANSWER: In this study, we used spatial transcriptomics to investigate the progression of follicular atresia, focusing on genes of interest associated with steroidogenesis and key signalling pathways WNT, TGFβ/BMP, NOTCH, and HH.
WHAT IS KNOWN ALREADY: While extensive research has focused on the mechanisms driving follicular growth, much less is known about the process of follicular atresia, despite its relevance for ovarian aging and reproductive longevity

1. The metabolism of 7,12-dimethylbenz(a)anthracene (DMBA) by primary cultures of human ovarian cells has been studied to identify the cell type(s) responsible for biotransformation of this carcinogen. The rate of DMBA metabolism was maximal in granulosa cells prestimulated in vivo with antiestrogen, hMG (human menopausal gonadotropin) and hCG (human chorionic gonadotropin), i.e., treatments required for maximal oocyte maturation and, thus, granulosa cell proliferation. In cells from unstimulated ovaries, the metabolism was maximal in granulosa-lutein cells isolated from corpus luteum. 2. Ster

Composite microassays for plasma progesterone (P), 17alpha-OH-progesterone (17P), estrone (E1), 17beta-estradiol (E2), and estriol (E3) were developed. Steroids were extracted from plasma samples with diethyl ether, and were separated from each other through two steps of sephadex LH-20 microcolumn chromatography (benzene:methanol 85:15, and n-hexane: benzene: methanol 80:10:10), prior to assays by radioimmunoassay (P, E1, E1, and E3) and competitive protein binding assay (17P). Steroids were recovered satisfactorily through these procedures (mean recovery; P:107.6%, 17P:102.3%, E1:88.2%, E2:84

Uhrf1 is a multi-domain and multifunctional epigenetic regulator playing key roles in DNA methylation, cell metabolism, and cell proliferation. To investigate the role of Uhrf1 in the reproductive physiology of female yaks, we collected three reproductive organs (ovaries, oviducts, and uteri) from healthy yaks during three reproductive phases (follicular, luteal, and gestational phases), with a total of nine groups. Real-time fluorescence quantitative PCR (RT-qPCR), Western blotting, and immunohistochemistry (IHC) were employed to determine the expression levels of Uhrf1 and the subcellular lo

The coypu (Myocastor coypus bonariensis) is a semi-aquatic hystricomorph rodent native to South America, widely distributed across wetlands from southern Brazil to Tierra del Fuego. The general features of the ovary of the coypu older than 6 months are similar to those described for other hystricomorphs. The objective of this research was to expand the qualitative and quantitative characterization of the follicular typology of coypu at sexual maturity. We worked with 36 ovaries from females in different stages of the oestrous cycle. The follicles were analysed based on qualitative and quantita

The ovary is a dynamic organ where mechanical forces profoundly regulate follicular development, oocyte maturation, and overall reproductive function. These forces, originating from the extracellular matrix (ECM), granulosa and theca cells, and ovarian stroma, influence cellular behavior through mechanotransduction, translating mechanical stimuli into biochemical responses. This review explores the intricate interplay between mechanical cues and ovarian biology, focusing on key mechanosensitive pathways such as Hippo signaling, the PI3K/AKT pathway, and cytoskeletal remodeling, which govern fo

Excessive androgens may lead to abnormal folliculogenesis and anovulation. To explore the link between hyperandrogenism (HA) and anovulation - related infertility, we performed a two - sample Mendelian randomization analysis, followed by multiple validations. We also delineated potential pathogenic pathways and assessed druggability. The inverse variance weighted (IVW) combined with the meta analysis results showed sex hormone-binding globulin (SHBG, IVW: OR1 = 0.561; 95% CI: 0.363 to 0.866; P1 = 0.009; meta: OR = 0.817, P = 0.023) and bioavailable testosterone (BIOT, meta: OR = 1.298, P = 0.0

All girls presenting with virilization (which signifies severe hyperandrogenism) warrant thorough investigation. Ovarian hyperthecosis (OHT) is a rare cause of virilization in premenopausal women. Here, we report the case of a previously healthy 12-year-old Chinese girl with signs of virilization at puberty. Her serum total testosterone was elevated at 5.1 nmol/L (146.97 ng/dL) (normal: <1.4 nmol/L, <40.35 ng/dL). Workup for Cushing syndrome, sex development disorders, congenital adrenal hyperplasia, and adrenal and ovarian androgen-secreting tumors was unrevealing. Ovarian and adrenal venous

A significant fraction of couples around the world suffer from polycystic ovarian syndrome (PCOS), a disease defined by the characteristics of enhanced androgen synthesis in ovarian theca cells, hyperandrogenemia, and ovarian dysfunction in women. Most of the clinically observable symptoms and altered blood biomarker levels in the patients indicate metabolic dysregulation and adaptive changes as the key underlying mechanisms. Since the liver is the metabolic hub of the body and is involved in steroid-hormonal detoxification, pathological changes in the liver may contribute to female endocrine

The pathophysiology of polycystic ovary syndrome (PCOS) is confusing until today as it is a multifactorial endocrine disorder. It is presented with altered gonadotropin levels, bulky multi-follicular ovaries, infertility, and obesity. This complex pathophysiology is linked with insulin resistance and hyperandrogenism. Hyperandrogenemia significantly contributes towards cosmetic anomalies including hirsutism, acne, and alopecia in the PCOS women. The preexisting insulin resistance in women with PCOS is likely to aggravate the increased levels of androgen. The review findings have shown that in

This study tested the hypothesis that oxidative stress could be an underlying mechanism for APs-induced ovarian cytotoxicity and reproductive dysfunction. Rat ovarian theca interstitial cells (TICs) were isolated and treated with four APs [chlorpromazine (CPZ), haloperidol (HAL), risperidone (RIS) and clozapine (CLZ)]. MTT assay was used to test the effects of these antipsychotics on TICs viability and to estimate their 50% inhibitory concentrations (IC50s). The effects of APs (IC50s and 1μM concentrations) on the activities of caspases-3, -8 and -9, reactive oxygen species (ROS) production, t

This study evaluated the follicular atresia, cell proliferation, and anti-Mullerian hormone action in Aotus nancymae and Sapajus macrocephalus during three sexual phases (follicular, luteal, and gestational). Follicular quantification and immunolocalization of Caspase-3 protein, B-cell lymphoma 2 (BCL-2), proliferating cell nuclear antigen (PCNA), and anti-Mullerian hormone (AMH) were performed. A significant difference in the quantification between preantral and antral follicles, with a progressive decrease in the antrals, was identified. Protein and hormonal markers varied significantly betw

Research has been focused on determining the follicular microenviroment produced by the theca and granulosa cells since the molecular characterisation of this body fluid could lead to the understanding of several fertility problems. Oxidative stress may be one of the factors involved in female infertility since it plays a key role in the modulation of oocyte maturation and finally pregnancy. An increase in oxidative stress is correlated with inflammation and intense research was developed to understand the interaction between inflammation and adiponectin, based on the fact that many adipokines
