BACKGROUND: Spartin protein is involved in degradation of epidermal growth factor receptor and turnover of lipid droplets and a lack of expression of this protein is responsible for hereditary spastic paraplegia type 20 (SPG20). Spartin is a multifunctional protein that associates with many cellular organelles, including lipid droplets. Recent studies showed that spartin interacts with E3 ubiquitin ligases that belong to the neural precursor cell-expressed developmentally downregulated gene (Nedd4) family, including atrophin-1-interacting protein 4 (AIP4/ITCH). However, the biological importan

Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality worldwide, yet the underlying mechanisms driving its progression are not fully elucidated. Long non-coding RNAs (lncRNAs) have recently emerged as key regulatory molecules in tumor biology. In this study, we identified ABHD11-AS1 as a tumor-suppressive lncRNA that is significantly downregulated in CRC tissues, its low expression is correlated with poor patient prognosis. Functional assays demonstrated that ABHD11-AS1 inhibits CRC cell proliferation, migration, and invasion, and enhances sensitivity to oxalipl

The role of ubiquitin in post-translational modifications is important for tumor progression, but how these mechanisms regulate bladder cancer (BLCA) is not completely known. The study demonstrated that FBXL6, a member of the F-box protein family, could drive oncogenesis in BLCA, as shown by integrative bioinformatic analysis and clinical sample validation. Experiments demonstrated that FBXL6 speeds up the in vitro growth and migration of BLCA cells and contributes to tumor development and metastasis in vivo. Mechanistically, transcriptomic and metabolic studies indicate that FBXL6 regulates t

The tripartite motif (TRIM) family of E3 ubiquitin ligases is known to play a crucial role in the initiation, growth, and metastasis of various tumors. However, little is known about the biological features and relevant molecular mechanism of Tripartite motif-containing 63 (TRIM 63) in melanoma. The expression levels of TRIM63 and purinergic receptor P2Y1 (P2RY1) in melanoma were examined by online database. Cell counting kit-8 (CCK-8) and colony formation assay were carried out to explore the effects of TRIM63 on melanoma cells proliferation. Transwell assay was used to explore the influence

Ubiquitination is a pivotal regulatory mechanism in host-virus interactions, playing essential roles in both antiviral defense and viral immune evasion, with E3 ubiquitin ligases serving as critical regulatory nodes. In this study, we systematically evaluated 26 candidate host E3 ubiquitin ligases for their involvement in influenza A virus (IAV) infection. Among these, potassium channel modulatory factor 1 (KCMF1) was identified as a novel negative regulator of IAV replication in vitro and in vivo. Mechanistically, KCMF1 interacts with the viral polymerase subunit PB1 and mediates its ubiquiti

Photoaging is the major type of exogenous aging caused by chronic ultraviolet (UV) radiation, whose pathogenesis is complex and not fully understood. Recently, the involvement of ubiquitination in aging and aging-related disorders has aroused extensive attention. The present study aimed to investigate the relationship between ubiquitination and photoaging with bioinformatics. We utilized the GSE38308 dataset to screen the differentially expressed ubiquitination-related genes (UbR-DEGs), as well as immune infiltration and function, in the sun-exposed skin samples versus sun-protected controls.

Gastric cancer (GC) is a globally lethal malignancy, with invasion and metastasis driving treatment failure and poor prognosis. MX dynamin like GTPase 1 (MX1) shows tumor-specific functional heterogeneity, while its expression, biological functions and molecular mechanisms in GC remain unclear. Here, we explored MX1's clinical significance and its regulatory mechanism in GC cell migration. We integrated public databases and institutional paired clinical samples for bioinformatics analysis of MX1's correlation with clinical outcomes, and verified its pro-migratory effect via Transwell and wound

Cullin-RING E3 ligases (CRLs) ubiquitinate their substrates by recruiting them to complexes containing ubiquitin-conjugating enzymes (UCEs). Ubiquitination proceeds through the transfer of ubiquitin from UCEs to substrate lysines, but the specificity of lysine ubiquitination has not been widely investigated in cells. In this study, we use global ubiquitination profiling to identify sites in the kinase domains of receptor tyrosine kinases (RTKs), EGFR and Her2, that undergo CRL-mediated ubiquitination in cells treated with heterobifunctional degrader molecules. We find that VHL- and Cereblon-de

BACKGROUND: Globally, among gynecological malignancies, ovarian cancer (OC) stands as an important factor contributing to mortality. E3 ubiquitin ligase activity is a common characteristic found in the tripartite motif (TRIM) proteins, contributing significantly to post-translational modifications of various proteins. Among these TRIM proteins, TRIM15 has been linked to multiple cancer types. Nevertheless, the function of TRIM15 concerning OC has yet to be elucidated. We aimed to explore the role of TRIM15 in OC and shed light on the underlying mechanisms thereof.

CD47 is an innate immune checkpoint that inhibits phagocytosis by myeloid cells, contributing to immune evasion by cancer cells. CD47-blocking antibodies have limited efficacy in glioblastoma (GBM), and the cell-intrinsic role of CD47 is poorly understood. In this study, we show that CD47 is highly expressed at the invasive edge of GBM tumors, and its elevated expression correlates with poor patient survival. We demonstrate that CD47 loss impairs GBM cell proliferation, migration, and invasion, independent of immune activity, and leads to reduced tumor burden and prolonged survival in vivo. Ou

Inflammatory bowel disease (IBD) belongs to a group of idiopathic intestinal inflammatory disorders that affect the ileum, rectum, and colon. There is still no effective treatment methods, and with the increasing number of patients, IBD has brought a heavy burden to the medical system. Chronic inflammation mediated by abnormal activation of the intestinal mucosal immune system plays an important role in the occurrence and development of IBD. As a common intracellular sensor that activates the innate immune system, inflammasomes have a signaling cascade regulated by ubiquitination/deubiquitinat

BACKGROUND: Pathological cardiac hypertrophy frequently leads to heart failure (HF). UBA1, the key E1 ubiquitin-activating enzyme, initiates ubiquitin-proteasome signaling and contributes to various diseases, yet its mechanism in cardiac hypertrophy remains unclear.
METHODS: Cardiac hypertrophy model was induced by either Ang II stimulation or TAC in vitro and in vivo. Mice received rAAV9-UBA1-siRNA or rAAV9-UBA1 for UBA1 knockdown or overexpression, respectively.
RESULTS: We found UBA1 upregulated in murine and human hypertrophic hearts. Cardiomyocyte-specific UBA1 knockdown protected again

Tumor necrosis factor receptor-associated factor 4 (TRAF4) is a RING-type E3 ubiquitin ligase linked to vascular inflammation, but whether it drives endothelial ferroptosis in atherosclerosis (AS) remains unclear. TRAF4 expression was assessed in GEO dataset GSE166780, ApoE-/- mice fed a high-fat diet for 4 months, and oxidized low-density lipoprotein (ox-LDL)-treated human coronary artery endothelial cells (HCAECs; 50 μg/mL, 24 h). In vivo, mice received tail-vein delivery of shTRAF4 vector. Plaque burden, endothelial injury, inflammatory responses, and ferroptosis were evaluated by histology

BACKGROUND: Retinal ganglion cells (RGCs) degeneration is an early event in diabetic retinopathy (DR), tightly coupled to mitochondrial dysfunction. While the mitochondrial gatekeeper voltage-dependent anion channel 1(VDAC1) is a known mediator of apoptosis, its regulation and therapeutic targeting in diabetic RGCs remain unclear.
METHODS: We examined VDAC1 expression in human donor retinas, streptozotocin-induced diabetic mice, and high glucose-treated SH-SY5Y cells. VDAC1-knockdown SH-SY5Y cells were established to explore its functional role in the context. Using virtual screening and mole

Lung adenocarcinoma (LUAD) is the most common subtype of lung cancer; however, its prognosis remains poor, and the underlying molecular mechanisms have yet to be fully elucidated. In this study, a prognostic model was constructed based on manganese metabolism-related genes, which effectively stratifies patients with LUAD with different prognoses and drug sensitivity. Mendelian randomization analysis identified glutathione peroxidase 3 (GPX3) as a key prognostic gene, which was downregulated in LUAD and significantly correlated with favorable outcomes. Functionally, GPX3 was found to inhibit LU

In stressed cells, the recessive Parkinson disease (PD) associated gene products PINK1 and parkin mediate the autophagic removal of damaged mitochondria (mitophagy). Upon mitochondrial membrane potential disruption, PINK1 phosphorylation activates the ubiquitin ligase parkin which ubiquitinates various mitochondrial protein substrates. These feed-forward modifications on the mitochondria surface attract ubiquitin-binding autophagy receptors that target ubiquitinated mitochondria to autophagosomes and indirectly contribute to phagophore elongation. Investigating post-translational protein modif

Impaired cardiac microvascular function is a significant contributor to diabetic cardiomyopathy. Rnd3 expression is notably downregulated in cardiac microvascular endothelial cells under diabetic conditions. Rnd3 overexpression mitigates diabetic myocardial microvascular injury and improves cardiac function through the Rock1/myosin light chain signaling pathway. Rnd3 facilitates the recruitment and interaction with Trim40 to promote Rock1 ubiquitination, thereby preserving endothelial barrier integrity in the diabetic heart.

Neuraminidase (NA) protein of influenza A virus (IAV), a tetrameric envelope glycoprotein critical for viral release and spread, has well-characterized enzymatic functions but poorly resolved host-pathogen interactions. However, host factors targeted by NA or those regulating NA activity are largely uncharacterized. Here, we identified host heat shock protein A1L (HSPA1L) as a novel NA-binding partner that suppresses IAV replication. Subsequent studies revealed that HSPA1L promotes NA ubiquitination at K242, resulting in its NBR1-dependent autophagic degradation. Moreover, a recombinant H5N1 v

BACKGROUND: Alcohol-associated liver disease (ALD), one of the most frequent chronic liver diseases globally, is characterized by steatosis. HMG-CoA reductase-degrading protein 1 (HRD1) participates in the endoplasmic reticulum-associated protein degradation pathway through the recognition, translocation, and ubiquitination of substrate proteins. HRD1 is implicated in endoplasmic reticulum stress, oxidative stress and cell metabolism; however, the function of HRD1 in ALD remains unclear.
AIMS: We aimed to explore the contribution and underlying molecular mechanism of HRD1 in alcoholic liver d
