Flavivirus infection activates the XBP1 pathway of the unfolded protein response to cope with endoplasmic reticulum stress.

The unfolded protein response (UPR) is a coordinated change in gene expression triggered by perturbations in functions of the endoplasmic reticulum (ER). XBP1, a key transcription factor of the UPR, is activated by an IRE1-mediated splicing event, which results in a frameshift and encodes a protein with transcriptional activity. Here, we report that XBP1 was activated during flaviviral infection, as evidenced by XBP1 mRNA splicing and protein expression, as well as induction of the downstream genes ERdj4, EDEM1, and p58(IPK) in Japanese encephalitis virus (JEV)- and dengue virus serotype 2 (DE…

Mitochondrial Unfolded Protein Response Regulates Copper-Induced Oxidative Stress via a Sirt3/FOXO3a Signaling Pathway in Chicken Hepatocytes.

Copper (Cu) is an essential trace element but also an agricultural pollutant with hepatotoxic risks. This study investigated the relationship among UPRmt, the Sirt3/FOXO3a pathway, and oxidative stress in chicken hepatocytes under Cu exposure. Here, broilers were fed diets containing 11, 110, 220, or 330 mg/kg Cu for 49 days, and primary hepatocytes were treated with Cu combined with OTC overexpression, Sirt3 agonist ADTL-SA1215, Sirt3 inhibitor 3-TYP, and FOXO3a overexpression or silencing. Cu exposure causes liver injury and activates UPRmt (upregulated HSP60 and LonP1) and was associated wi

Selenomethionine Attenuates Hydrogen Peroxide-Induced Oxidative Injury and Modulates Unfolded Protein Response-Related Markers in Murine C2C12 Myoblasts.

Oxidative stress disrupts skeletal muscle cell survival and proteostasis, thereby compromising animal skeletal muscle health. As an organic selenium source, selenomethionine (SeMet) participates in antioxidant defense and selenoprotein regulation. However, its protective role in hydrogen peroxide (H2O2)-induced injury in C2C12 myoblasts and its association with unfolded protein response (UPR)-related molecular changes remain incompletely understood. This study investigated whether SeMet alleviates H2O2-induced injury in C2C12 myoblasts and whether this effect is associated with UPR-related mar…

CRM-1 Aggravates Stroke Injury by Promoting BANF1-Mediated Unfolded Protein Response Through Inducing Nuclear Export of ALKBH5.

The activation of endoplasmic reticulum stress (ERS), specifically the PERK/eIF2α/CHOP signaling, is a recognized consequence of ischemic stroke. However, the roles and mechanisms of CRM-1 regulating ERS in stroke are poorly elucidated. A murine model of stroke was generated via transient middle cerebral artery occlusion (MCAO). The endpoints included TTC-derived infarct volume, H&E/TUNEL histopathology, p-PERK immunohistochemistry, and Western blot. Oxygen-glucose deprivation/reoxygenation (OGD/R) was employed in HT22 neurons. The CRM1-ALKBH5 interaction and subcellular distribution were asse

Flavivirus-Induced ER Stress and Unfolded Protein Response: A Central Hub Linking Lipid Droplet Remodeling and Viral Replication.

Endoplasmic reticulum (ER) stress and the unfolded protein response (UPR) represent fundamental cellular adaptive mechanisms that maintain protein homeostasis and metabolic balance. Many RNA viruses, particularly flaviviruses such as dengue virus (DENV), Zika virus (ZIKV), West Nile virus (WNV), yellow fever virus (YFV), and Japanese encephalitis virus (JEV), extensively remodel the ER to establish replication compartments and assemble progeny virions. This massive reorganization disrupts ER homeostasis, leading to UPR activation. Emerging evidence reveals that flaviviruses not only trigger bu…

The thiocarbonyl analog of chrysin induces apoptosis and oxidative stress-mediated unfolded protein response in breast cancer cells.

Natural flavones such as chrysin and baicalein were characterized by a wide range of biological activities including, among others, antioxid … n of superoxide. Thiochrysin-mediated oxidative stress in MCF-7 cells resulted in the activation of unfolded protein response (UPR) as judged by increased phosphorylated status of IRE1, elevated levels of ATF6 and ER-associated chaperone GRP78. Cytotoxic potential of thiochrysin was also documented using 3D breast cancer cell-based spheroid models. Thiochrysin was also act …

Unfolded protein response markers Grp78 and eIF2alpha are upregulated with increasing alpha-synuclein levels in Lewy body disease.

AIMS: Endoplasmic reticulum stress followed by the unfolded protein response is one of the cellular mechanisms contributing to the progression of α-synuclein pathology in Parkinson's disease and other Lewy body diseases. We aimed to investigate the activation of endoplasmic reticulum stress and its correlation with α-synuclein pathology in human post-mortem brain tissue. METHODS: We analysed brain tissue from 45 subjects-14 symptomatic patients with Lewy body disease, 19 subjects with incidental Lewy body disease, and 12 healthy controls. The analysed brain regions included the medulla, pons,…

Immunogenic landscape and risk score prediction based on unfolded protein response (UPR)-related molecular subtypes in hepatocellular carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is the most common type of cancer and causes a significant number of cancer-related deaths worldwide. The molecular mechanisms underlying the development of HCC are complex, and the heterogeneity of HCC has led to a lack of effective prognostic indicators and drug targets for clinical treatment of HCC. Previous studies have indicated that the unfolded protein response (UPR), a fundamental pathway for maintaining endoplasmic reticulum homeostasis, is involved in the formation of malignant characteristics such as tumor cell invasiveness and treatment re…

Mixed copper(ii)-phenanthroline complexes induce cell death of ovarian cancer cells by evoking the unfolded protein response.

There is an ongoing need for the development of new cancer therapeutics that combine high cytotoxic efficiency with low side effects, and al … hese complexes induce endoplasmic reticulum (ER) stress and subsequently cell death mediated by the unfolded protein response. Alleviation of the ER-stress by tauroursodeoxycholic acid (TUDCA) attenuated the cytotoxic effects. In summary, we have identified a novel, ER-dependent, molecular mechanism mediating cytotoxic effects of copper(ii)-phenanthroline complexes.

Altered unfolded protein response is implicated in the age-related exacerbation of proteinuria-induced proximal tubular cell damage.

Aging is a dominant risk factor for end-stage renal disease. We analyzed the mechanism involved in age-related exacerbation of proteinuria-induced proximal tubular cell (PTC) damage by focusing on endoplasmic reticulum-related unfolded protein response (UPR). After equal-degree induction of proteinuria in 24-month-old (aged) and 3-month-old (young) mice by intraperitoneal free fatty acid-bound albumin overload, tubulointerstitial lesions were more severe in aged than in young mice. In aged PTCs, proteinuria-induced cell-adaptive UPR resulting from induction of the molecular chaperone BiP was s…

Viral vaccines promote endoplasmic reticulum stress-induced unfolding protein response in teleost erythrocytes.

Most available evidence points to a proviral role for endoplasmic reticulum (ER) stress, as many viruses exploit it to promote viral replication. In contrast, few studies have linked ER stress to the antiviral immune response, and even fewer to the vaccine-induced immune response. In this work, we demonstrated that ER stress is a key molecular link in the immune response of teleost erythrocytes or red blood cells (RBCs) under vaccine stimulation. Moreover, the unfolded protein response (UPRER) triggered by ER stress may work together with autophagy and related cellular mechanisms as part of a …
