Activation of the Complement System and Changes in the Content of Modified Lipoproteins in Antiorthostatic Hypokinesia in Humans and Mice.

The correlation between the total activity of the complement system (TACS) and the content of modified LDL (mLDL) in blood serum when modeling microgravity in Earth conditions was evaluated. Healthy volunteers were placed in a state of antiorthostatic hypokinesia for 21 days. Microgravity in ICR mice was modeled by tail suspension for 14 days. TACS, activity of the alternative pathway (in humans), and the content of mLDL in serum from volunteers and mice were measured by turbidimetry. On day 7 of antiorthostatic hypokinesia, volunteers had a significant increase in TACS, which persisted until

The effect of microgravity and space flight on the chemical senses.

The effect of space flight and microgravity on the chemical senses is reviewed. Skylab-4 and Soyuz 30-31 studies revealed changes in taste thresholds while no effect was found in a Canadian investigation (41-G) and conflicting results were obtained on another Soyuz mission. Two simulated microgravity studies found no effect on taste or smell sensitivity; while 5 other studies found an effect. Microgravity induces physiological changes including an upward shift of body fluids toward the head, which may lead to an attenuation of the olfactory component in the flavor of foods. Chemosensory change

Spaceflight and the skeleton: lessons for the earthbound.

Loss of bone during extended space flight has long been a concern that could limit the ability of humans to explore the universe. Surprisingly the available data do not support the concept that weightlessness leads inexorably to a depleted skeleton unable to withstand the stress of a return to a 1g environment. Nevertheless, some bone loss does occur especially in those bones most stressed by gravity prior to flight, providing confirmation of the proposal formulated over a century ago by Julius Wolff that mechanical stress determines the form and function of bone. Although the phenomenon of bo

Re-evaluating pharmaceutical stability in space: A biomedical and regulatory science framework for long-duration missions.

Pharmaceutical stability is a critical determinant of crew health and mission success in long-duration spaceflight. Current regulatory frameworks governing drug stability are primarily based on terrestrial testing paradigms, including International Council for Harmonisation (ICH) guidelines, which assume controlled environmental conditions. However, spaceflight introduces unique stressors such as microgravity, ionizing radiation, exposure to the vacuum of space, and operational constraints that are not accounted for in existing models. This study systematically evaluates the limitations of curr

Spaceflight Associated Neuro-ocular Syndrome (SANS) and its countermeasures.

Astronauts can develop a distinct collection of neuro-ophthalmic findings during long duration spaceflight, collectively known as Spaceflight Associated Neuro-ocular Syndrome (SANS). These clinical characteristics include optic disc edema, hyperopic refractive shifts, globe flattening, and chorioretinal folds, which may pose a health risk for future space exploration. Obtaining knowledge of SANS and countermeasures for its prevention is crucial for upcoming crewed space missions and warrants a multidisciplinary approach. This review examines the potential causes and countermeasures of SANS, in

Defining spaceflight associated dry eye syndrome (SADES): Mechanisms, complications, and countermeasures.

There is a high prevalence of dry eye-related symptoms among astronauts during spaceflight. We propose naming this phenomenon: Spaceflight Associated Dry Eye Syndrome (SADES). In this study, we review the potential mechanisms of dry eye in the spaceflight environment, categorizing them into external (radiation, environmental hazards) and microgravity-related (meibum outflow) mechanisms. One key mechanism is that microgravity impedes the secretion of meibum, the primary contributor to the tear film lipid layer. SADES may impact the ocular surface due to the positive feedback loop of inflammatio

Omics Studies of Tumor Cells under Microgravity Conditions.

Cancer is defined as a group of diseases characterized by abnormal cell growth, expansion, and progression with metastasis. Various signaling pathways are involved in its development. Malignant tumors exhibit a high morbidity and mortality. Cancer research increased our knowledge about some of the underlying mechanisms, but to this day, our understanding of this disease is unclear. High throughput omics technology and bioinformatics were successful in detecting some of the unknown cancer mechanisms. However, novel groundbreaking research and ideas are necessary. A stay in orbit causes biochemi

Optimizing autonomous artificial intelligence diagnostics for neuro-ocular health in space missions.

Spaceflight-Associated Neuro-Ocular Syndrome (SANS) presents a critical risk in long-duration missions, with microgravity-induced changes that threaten astronaut vision and mission outcomes. Current SANS monitoring, limited to pre- and post-flight exams, lacks in-flight diagnostics, highlighting an urgent need for autonomous tools capable of real-time assessment. Grok, an AI platform by xAI, offers promising potential as an advanced diagnostic tool for space-based health monitoring. Originally developed for broader applications, Grok's high-resolution imaging capabilities could be adapted to d

Postural adaptation to microgravity underlies fine motor impairment in astronauts' speech.

Understanding the role of anti-gravity behaviour in fine motor control is crucial to achieving a unified theory of motor control. We compare speech from astronauts before and immediately after microgravity exposure to evaluate the role of anti-gravity posture during fine motor skills. Here we show a generalized lowering of vowel space after space travel, which suggests a generalized postural shift of the articulators. Biomechanical modelling of gravitational effects on the vocal tract supports this analysis-the jaw and tongue are pulled down in 1g, but movement trajectories of the tongue are o

Astronauts eye-head coordination dysfunction over the course of twenty space shuttle flights.

BACKGROUND: Coordination of motor activity is adapted to Earth's gravity (1 g). However, during space flight the gravity level changes from Earth gravity to hypergravity during launch, and to microgravity (0 g) in orbit. This transition between gravity levels may alter the coordination between eye and head movements in gaze performance. OBJECTIVE: We explored how weightlessness during space flight altered the astronauts' eye-head coordination (EHC) with respect to flight day and target eccentricity. METHODS: Thirty-four astronauts of 20 Space Shuttle missions had to acquire visual targets wi

Effects of long-term simulated microgravity on liver metabolism in rhesus macaques.

The liver is an essential multifunctional organ and constantly communicates with nearly all the tissues in the body. Spaceflight or simulated microgravity has a significant impact on the livers of rodent models, including lipid accumulation and inflammatory cell infiltration. Whether similar liver lipotoxicity could occur in humans is not known, even though altered circulating cholesterol profile has been reported in astronauts. Using a 42-day head-down bed rest (HDBR) model in rhesus macaques, the present study investigated whether simulated microgravity alters the liver of non-human primates

Cardiac function, structural, and electrical remodeling by microgravity exposure.

Space medicine is key to the human exploration of outer space and pushes the boundaries of science, technology, and medicine. Because of harsh environmental conditions related to microgravity and other factors and hazards in outer space, astronauts and spaceflight participants face unique health and medical challenges, including those related to the heart. In this review, we summarize the literature regarding the effects of spaceflight on cardiac structure and function. We also provide an in-depth review of the literature regarding the effects of microgravity on cardiac calcium handling. Our r

Effects of aerospace environments on the cardiovascular system.

Certain physical and physiological changes occur in the atmospheric levels where flight and space activities take place. Air pressure decreases with increasing altitude and the partial pres¬sure of O2 decreases in parallel with the atmospheric pressure drop and creates hypoxia in the flight crew and in the passen¬gers. In case of acute hypobaric hypoxia, blood is redistributed to the brain and the heart, whereas blood supply to internal organs, such as kidney and skin is reduced. Peripheral cyanosis can be observed on the fingertips and the lips during hypoxia-induced blood redistribution. Tac

Origins of Cerebral Edema: Implications for Spaceflight-Associated Neuro-Ocular Syndrome.

BACKGROUND: Spaceflight-associated neuro-ocular syndrome (SANS) was first described in 2011 and is associated with structural ocular changes found to occur in astronauts after long-duration missions. Despite multiple insufficient potential terrestrial models, an understanding of the etiology has yet to be described. EVIDENCE ACQUISITION: A systematic review was conducted on literature published about the pathophysiology of cerebral edema. Databases searched include PubMed, Scopus, and the Texas Medical Center Online Library. This information was then applied to create theories on mechanisms o

Drugs in space: Pharmacokinetics and pharmacodynamics in astronauts.

Space agencies are working intensely to push the current boundaries of human spaceflight by sending astronauts deeper into space than ever before, including missions to Mars and asteroids. Spaceflight alters human physiology due to fluid shifts, muscle and bone loss, immune system dysregulation, and changes in the gastrointestinal tract and metabolic enzymes. These alterations may change the pharmacokinetics and/or pharmacodynamics of medications used by astronauts and subsequently might impact drug efficacy and safety. Most commonly, medications are administered during space missions to treat

Feasibility of dried blood spot collection for caffeine pharmacokinetic studies in microgravity: Insights from parabolic flight campaigns.

AIMS: Therapeutic drug monitoring for astronauts faces limitations in conventional blood sampling and sample management onboard the international space station. Here, we explore the feasibility of dried blood spot (DBS) collection during parabolic flights (PF) to overcome these constraints. METHODS: We assessed the feasibility of blood deposition on blotting paper for preanalytical aspects in a PF using synthetic blood. Subsequently, DBS sampling validation was carried out in another PF campaign. Twenty volunteers participated in a pharmacokinetic study on caffeine and its metabolite, paraxan

Combined Exposure to Simulated Microgravity and Acute or Chronic Radiation Reduces Neuronal Network Integrity and Survival.

During orbital or interplanetary space flights, astronauts are exposed to cosmic radiations and microgravity. However, most earth-based studies on the potential health risks of space conditions have investigated the effects of these two conditions separately. This study aimed at assessing the combined effect of radiation exposure and microgravity on neuronal morphology and survival in vitro. In particular, we investigated the effects of simulated microgravity after acute (X-rays) or during chronic (Californium-252) exposure to ionizing radiation using mouse mature neuron cultures. Acute exposu

Neurovestibular considerations for sub-orbital space flight: A framework for future investigation.

Commercial sub-orbital operators will soon offer the excitement of traveling to space to thousands of people. Based on previous experience in space flight and parabolic flight, sensorimotor disruptions in eye movements, postural stability, and motor coordination are likely in these travelers. Here we propose a framework for developing strategies to overcome these sensorimotor disruptions. We delineate how approaches should differ from those applied to orbital flight and between sub-orbital passengers and pilots based on differing frequency of flights and mission objectives. Sensorimotor adapta

Preliminary results of medical investigations during manned flights of the Salyut 4 orbital station.

In 1975 the Soviet Union performed two manned flights on board the orbital station Salyut 4. The first mission of 30 days was flown by A. A. Gubarev (Commander) and G. M. Grechko (Flight Engineer) and the second mission of 63 days by P.I. Klimuk (Commander) and V.I. Sevastyanov (Flight Engineer). The present paper gives preliminary data on the general health and cardiovascular investigations of the crew members during and after flight. The clinical and physiological examinations of the cosmonauts showed that the work-rest cycles and counter-measures used helped to maintain their good health an
