Diagnosing xeroderma pigmentosum group C by immunohistochemistry.

Xeroderma pigmentosum (XP) is a group of rare inherited human neurocutaneous diseases, and the group C (XPC) is the major group of patients with XP in Europe, North America, and South America. Current molecular diagnostic methods for XP require specialized, expensive, and time-consuming UV sensitivity and DNA repair assays followed by gene sequencing. To determine whether immunohistochemistry (IHC) would be a robust alternative method to diagnose patients with XPC, we stained sections of paraffin-embedded skin biopsies for XPC by IHC, using 69 archived blocks from confirmed or clinically suspe…

Coexistent Bullous Darier's Disease with Xeroderma Pigmentosum.

A case of bullous Darier's disease had typical brown follicular papules, greasy crusted papules discrete keratotic papules and vesiculo- bullous lesions on the dorsum of the hands and feet. In addition, characteristic lesions of xeroderma pigmentosum as extensivee, freekles intermingled with atrophic white, spots were present on the, face, trunk and limbs. This combination of bullous Darier's diseaese with xeroderma pigmentosum has not been reported earlier.

Xeroderma pigmentosum, defective DNA repair--and schistosomiasis?

Xeroderma pigmentosum (XP) is more common in Japan and Egypt than in Europe and America. Some unidentified selective advantage for the carrier of an XP gene is the probable explanation for these regional increases. The question then arises as to what unusual environmental factor(s) Japan and Egypt, otherwise so disparate, might share that is not shared by most of the Western World. The dependence on flooded lands for crop production is one such feature, and human schistosomiasis has been a consequence in both places. The second question becomes, therefore, how might this parasitism be responsi…

Xeroderma Pigmentosum-Associated Childhood Interstitial Lung Disease.

Chromosomal breakage syndromes are a group of genetic disorders that are ascribable to the autosomal recessive mode of inheritance. Xeroderma pigmentosum is one of the chromosomal breakage syndromes which is due to the involvement of deformity in the deoxyribonucleic acid's nucleotide excision repair. Xeroderma pigmentosum is a genetic disorder, which is autosomal recessive, heterogeneous, and more common in cases of consanguinity, caused by mutations in at least 10 genes and 9 complementation groups. The disorder is very rare. Patients experience photophobia and extreme photosensitivity and h…

Xeroderma Pigmentosum: Gene Variants and Splice Variants.

The nucleotide excision repair (NER) is essential for the repair of ultraviolet (UV)-induced DNA damage, such as cyclobutane pyrimidine dimers (CPDs) and 6,4-pyrimidine-pyrimidone dimers (6,4-PPs). Alterations in genes of the NER can lead to DNA damage repair disorders such as Xeroderma pigmentosum (XP). XP is a rare autosomal recessive genetic disorder associated with UV-sensitivity and early onset of skin cancer. Recently, extensive research has been conducted on the functional relevance of splice variants and their relation to cancer. Here, we focus on the functional relevance of alternativ…

Scalp squamous cell carcinoma in xeroderma pigmentosum.

CONTEXT: Xeroderma pigmentosum is a rare autosomal-recessive disorder that appears in early childhood. Squamous cell carcinoma is not uncommon in patients with xeroderma pigmentosum and mostly involving the face, head, neck, and scalp. However, squamous cell carcinoma of the scalp may exhibit an aggressive course. CASE REPORT: Here, we present a huge squamous cell carcinoma of the scalp in a three-years-old child with xeroderma pigmentosum. In addition, we illustrate the challenges of a child with xeroderma pigmentosum who grows up in a sunny environment where the possibility of early onset o…

Xeroderma pigmentosum variant associated with multiple cancers.

A 62-year-old Japanese man with xeroderma pigmentosum (XP) variant is reported. The patient had developed at least 6 basal cell carcinomas, a squamous cell carcinoma, and a malignant melanoma on sun-exposed areas, and an atypical carcinoid on the right lung. In vivo phototesting showed a normal response. The minimal erythema dose of ultraviolet B (UVB) was not lowered and no delayed peaking of the erythema reaction was observed. His skin fibroblasts exhibited higher sensitivity to UV irradiation, but a normal level of unscheduled DNA and RNA synthesis. Cell fusions with XP group A, C, D, E, F,…

Xeroderma pigmentosum: recent clinical and photobiological aspects.

In Japan, more than 400 patients with xeroderma pigmentosum (XP) have been registered. The major groups are XP-A and variant, while clinically mild types of XP with intermediate levels of unscheduled DNA synthesis (UDS) have recently been increasing. The classical type of XP-A and some of the XP-D patients exhibit neurologic abnormalities. XP individuals display a marked increase in the frequency of skin malignancy. Development of skin malignancies appears to be related to the level of DNA repair capacity; the lower the capacity, the earlier and more frequently the skin tumors develop. Further…
