PPARα-independent transcriptional targets of perfluoroalkyl acids revealed by transcript profiling.

Perfluoroalkyl acids (PFAAs) are ubiquitous and persistent environmental contaminants. Compounds such as perfluoroocanoic acid (PFOA), perfluorooctane sulfonate (PFOS), perfluorononanoic acid (PFNA), and perfluorohexane sulfonate (PFHxS) are readily found in the tissues of humans and wildlife. While PFOA and PFOS have been the subject of numerous studies since they were first described over a decade ago, less is known about the biological activity of PFHxS and PFNA. Most PFAAs are activators of peroxisome proliferator-activated receptor α (PPARα), although the biological effects of these compo

Prehospital suicide mortality in the Lake Van basin: rural-urban disparities from three-year EMS dispatch surveillance in eastern Türkiye.

BACKGROUND: Eastern Türkiye records high suicide mortality; however, prehospital emergency medical services (EMS) dispatch records from this region have not been systematically evaluated as a surveillance source. We aimed to describe EMS-attended suicide and self-harm cases in Van Province and to quantify the association between rural residence and prehospital suicide mortality. METHODS: We conducted a retrospective population-based surveillance study of all 'suicide'-coded records in the Van Province 112 EMS database (ASOS) from January 2023 to December 2025. After excluding duplicates and m

Arctic deep-sea hydrothermal microbiomes as a natural niche for novel antimicrobial peptides.

BACKGROUND: The escalating threat of antimicrobial resistance (AMR) has created an urgent need for new antimicrobial agents. Antimicrobial peptides (AMPs) are promising alternatives to conventional antibiotics due to their broad-spectrum activity and reduced risk of resistance development. While most AMP discovery efforts have focused on terrestrial microbes, extreme environments remain largely untapped. Deep-sea hydrothermal vent biofilms, such as those from the Arctic Mid-Ocean Ridges (AMOR), are unique ecosystems characterized by high pressure, temperature gradients, and chemical extremes.

Effect of cosmetic whitening ingredients' lipophilicity on melanocyte cytotoxicity: A systematic review.

Tyrosinase is an enzyme involved in the early stages of melanin synthesis. Melanin, which gives skin its pigment, is the most targeted molecule in the development of skin-whitening cosmetic products. Consequential risks of chemically induced skin diseases are associated with the usage of skin-whitening products that contain tyrosinase-inhibiting compounds. Currently, products containing tyrosinase inhibitors on the market lack collective analysis of their toxicity on melanocytes. Moreover, national regulatory agencies (e.g., FDA) place the burden of ensuring the safety of whitening products on

Neuroprotective Role of Flavonoids: Implications for Natural Epilepsy Therapy-A Systematic Review.

INTRODUCTION: Epilepsy is a chronic neurological disorder characterised by recurrent, unprovoked seizures due to abnormal neuronal hyperexcitability and dysfunction. Despite the availability of several Antiepileptic Drugs (AEDs), approximately one-third of epilepsy patients are drugresistant and often experience adverse effects. This has led to the search for plant- derived bioactive compounds such as flavonoids, which have been reported to have widespread neuroprotective effects. This study aims to evaluate the neuroprotective role of flavonoids and their therapeutic implications in the manag

Mechanisms of Qing-Shen-Du formula in treating diabetic kidney disease: Integrating network pharmacology, molecular docking, molecular dynamics, transcriptomics, and experimental validation.

Diabetic kidney disease (DKD) represents the major contributor to end-stage renal disease globally, but no curative therapies are currently available. Qing-Shen-Du formula (QSDF), a Traditional Chinese Medicine formula, confers notable protective effects on DKD. It is yet unknown, nevertheless, how QSDF influences DKD. In this study, an analysis of the chemical compounds in QSDF was performed using UPLC-Q-TOF-MS/MS. The potential mechanism of QSDF against DKD was predicted by network pharmacology and subsequently verified by molecular docking and molecular dynamics. RNA-seq and microarray tran

Research Article: In Silico Identification of Potential Inhibitors Targeting NUSAP1, A Hub Gene in Hepatocellular Carcinoma.

Nucleolar-spindle-associated protein 1 (NUSAP1) participates in the assembly of microtubules and the mitotic spindle. Mitotic spindles are microtubule-based structures that segregate chromosomes during mitosis. Its overexpression and knockdown caused alterations in gene expression programs linked to tumor progression. It was also identified as one of the potential hub genes in various cancers, including the mediation of hepatocellular carcinoma. The present research addresses the NUSAP1 protein structure refinement and its targeting by the lead molecules identified using various computational

Discovery of a novel microtubule destabilizing agent targeting the colchicine site based on molecular docking.

Although a number of microtubule-targeting agents have been used in tumour therapy, their resistance and drug toxicity pose clinical challenges. Microtubule destabilizing agents (MDAs) targeting the colchicine site were found to have the advantage of being able to overcome drug resistance. In our previous studies, we identified a novel MDA from the compound database based on virtual screening methods. Its chemical formula is C23H19N3O5S, abbreviated as C10. In this study, molecular docking methods confirmed that the binding pattern of C10 to tubulin is similar to that of colchicine. Immunofluo

Geospatial analysis of hypospadias and cryptorchidism prevalence rates based on postal code in a Canadian province with stable population.

INTRODUCTION: Hypospadias and cryptorchidism are hormone-mediated malformations that occur during male development. Prevalence rates of hypospadias and cryptorchidism are thought to be increasing worldwide. In-utero exposure to endocrine-disrupting chemicals (EDCs) may have a role in the occurrence of these malformations. Our group has reported significant clustering of hypospadias and cryptorchidism at the county level in areas of intense agricultural activity in the Canadian province of Nova Scotia (NS). Finer scale spatial analysis has shown clustering near urban centres. OBJECTIVES: The o
