Baculovirus expression vectors: the requirements for high level expression of proteins, including glycoproteins.

The requirements for high level expression of three foreign proteins using the polyhedrin gene promoter of Autographa californica nuclear polyhedrosis virus (AcNPV, Baculoviridae) have been investigated. In Spodoptera frugiperda cells infected with the appropriate recombinant baculoviruses, the synthesis of the two S RNA coded genes of lymphocytic choriomeningitis virus (LCMV; i.e. the nucleoprotein, N, and glycoprotein precursor, GPC), or the haemagglutinin gene of influenza A virus, appears to be related to the degree of integrity of the 5' upstream sequence of the polyhedrin gene. No effect

Receptors for the vasodilator maxadilan are expressed on selected neural crest and smooth muscle-derived cells.

Maxadilan is a potent vasodilator peptide isolated from salivary glands of the blood feeding sand fly Lutzomyia longipalpis. The peptide relaxes rabbit aortic rings in an endothelium independent manner while elevating levels of cAMP and has been found to bind to membrane homogenates from brain. These studies on tissues have now been expanded with an examination of binding and signaling of maxadilan to a number of established cell lines and primary cultures. The data reveal that maxadilan binds to and stimulates the accumulation of cAMP in the rat pheochromocytoma line PC12 and the human neurob

The somatic white-ivory eye spot test does not detect the same spectrum of genotoxic events as the wing somatic mutation and recombination test in Drosophila melanogaster.

A groups of six chemical compounds was tested in parallel in two different somatic genotoxicity assays in Drosophila melanogaster, the wing somatic mutation and recombination test (SMART) and the white-ivory eye spot test. The wing spot test makes use of the wing cell markers multiple wing hairs (mwh) and flare (flr) and detects both mitotic recombination and various types of mutational events. The white-ivory eye spot test makes use of the white-ivory (wi) quadruplication and detects the somatic reversion of the recessive eye color mutation wi to the wild-type (w+). SCAR/WAVE and Arp2/3 are crucial for cytoskeletal remodeling at the site of myoblast fusion.

Myoblast fusion is crucial for formation and repair of skeletal muscle. Here we show that active remodeling of the actin cytoskeleton is essential for fusion in Drosophila. Using live imaging, we have identified a dynamic F-actin accumulation (actin focus) at the site of fusion. Dissolution of the actin focus directly precedes a fusion event. Whereas several known fusion components regulate these actin foci, others target additional behaviors required for fusion. Mutations in kette/Nap1, an actin polymerization regulator, lead to enlarged foci that do not dissolve, consistent with the observed

Postembryonic patterns of expression of cut, a locus regulating sensory organ identity in Drosophila.

The cut locus is both necessary and sufficient to specify the identity of a class of sensory organs in Drosophila embryos. It is also expressed in and required for the development of a number of other embryonic tissues, such as the central nervous system, the Malpighian tubules and the tracheal system. We here describe the expression of cut in the precursors of adult sensory organs. We also show that cut is expressed in cells of the prospective wing margin and correlate the wing margin phenotype caused by two cut mutations with altered cut expression patterns. Finally, we observe cut-expressin

Microorganism-Based Larval Diets Affect Mosquito Development, Size and Nutritional Reserves in the Yellow Fever Mosquito Aedes aegypti (Diptera: Culicidae).

BACKGROUND: Mosquito larvae feed on organic detritus from the environment, particularly microorganisms comprising bacteria, protozoa, and algae as well as crustaceans, plant debris, and insect exuviae. Little attention has been paid to nutritional studies in Aedes aegypti larvae. OBJECTIVES: We investigated the effects of yeast, bacteria and microalgae diets on larval development, pupation time, adult size, emergence, survivorship, lifespan, and wing morphology. MATERIALS AND METHODS: Microorganisms (or Tetramin® as control) were offered as the only source of food to recently hatched first i

Serological reactivity of recombinant 1D autoantigen and its expression in human thyroid and eye muscle tissue: a possible autoantigenic link in Graves' patients.

Thyroid-associated ophthalmopathy (TAO) is a potentially severe autoimmune disease, in and around the orbit, usually accompanied by Graves' disease. It was the goal of this study to develop a serological indicator for TAO and to characterize its expression in human thyroid and eye muscle tissue. Thus, we have recloned the full-length 1D-complementary DNA and assessed its expression levels in 90 healthy and diseased human thyroids. Only Graves' patients suffering from TAO (n = 29) displayed a significant, 2.1-fold increase of 1D expression levels (P = 0.029), compared with normal controls (n =

Metarhizium anisopliae pathogenesis of mosquito larvae: a verdict of accidental death.

Metarhizium anisopliae, a fungal pathogen of terrestrial arthropods, kills the aquatic larvae of Aedes aegypti, the vector of dengue and yellow fever. The fungus kills without adhering to the host cuticle. Ingested conidia also fail to germinate and are expelled in fecal pellets. This study investigates the mechanism by which this fungus adapted to terrestrial hosts kills aquatic mosquito larvae. Genes associated with the M. anisopliae early pathogenic response (proteinases Pr1 and Pr2, and adhesins, Mad1 and Mad2) are upregulated in the presence of larvae, but the established infection proces

Homeobrain, a novel paired-like homeobox gene is expressed in the Drosophila brain.

The homeobrain (hbn) gene is a new paired-like homeobox gene which is expressed in the embryonic brain and the ventral nerve cord. Expression of homeobrain initiates during the blastoderm stage in the anterior dorsal head primordia and the gene is persistently expressed in these cells which form parts of the brain during later embryonic stages. An additional weaker expression pattern is detected in cells of the ventral nerve cord from stage 11 on. The homeodomain in the Homeobrain protein is most similar to the Drosophila proteins DRx, Aristaless and Munster. In addition, the localized brain e

The hedgehog signalling pathway in tumorigenesis and development.

The hedgehog signalling pathway is responsible for the embryonic patterning of a range of tissues, and it is now known that dysregulation of this pathway can result in the formation of several tumour types. This cascade is regulated at the cell surface by the opposing actions of the patched and smoothened molecules which together form a receptor complex for hedgehog. The discovery that inactivation of the human patched gene is responsible for familial and sporadic forms of basal cell carcinoma firmly established a role for dysregulation of hedgehog signalling in tumorigenesis. Other key member

Structural Modification of Bifenox Derivatives: Discovery of Novel Diaryl Thioethers as Potent Insect Trehalase Inhibitors.

To discover novel noncarbohydrate trehalase inhibitors, 62 bifenox derivatives were designed and synthesized in this study. Among them, diaryl thioether 2b-1 exhibited optimal trehalase inhibitory activity with 72.5% inhibition rate at 100 μM and a Ki value of 36.3 μM, superior to bifenox (56.5 μM). Insecticidal activity assay showed that compound 2b-1 displayed stronger insecticidal potency against Ostrinia furnacalis (LC50 = 34.7 mg/L) than bifenox (LC50 = 61.2 mg/L). Molecular docking and density functional theory (DFT) calculations proved 2b-1 formed abundant hydrogen bonds and π-sulfur in

Targeting octopamine synthesis genes LsTDC and LsTβH impairs reproduction, feeding and virus transmission in the small brown planthopper Laodelphax striatellus.

BACKGROUND: Octopamine (OA) is an important biogenic amine in insects, and regulates multiple physiological and behavioral processes. Tyrosine decarboxylase (TDC) and tyramine β-hydroxylase (TβH) are critical enzymes in the biosynthesis of OA. The small brown planthopper Laodelphax striatellus is a major rice pest and causes serious damage by transmitting plant viruses, such as rice stripe virus (RSV). However, the functions of TDC and TβH remain largely unknown in rice planthoppers. RESULTS: We cloned LsTDC and LsTβH from L. striatellus. LsTDC was significantly enriched in the 4th instar nym

Research note: Antimicrobial properties of commercial black soldier fly (Hermetia illucens) meal against gram-positive and gram-negative bacteria.

Black soldier fly larvae meal (BSFM) is increasingly recognised as a sustainable and functional protein source in the livestock feed industry, owing to its high crude protein and rich fatty acid content. Beyond its nutritional value, BSFM has demonstrated antibacterial properties, enhancing resistance in animals to pathogens such as Salmonella species. These findings are critical as antimicrobial resistance continues to rise in the livestock sector. The black soldier fly (BSF) genome encodes antimicrobial peptides (AMPs) that can be classified under families such as cecropins, defensins, and a

Projected late-century climate change alters reproductive gene expression pathways in the arbovirus vector Aedes aegypti.

Climate change is expected to alter the ecology, persistence, and geographic distribution of mosquito vectors, yet the molecular traits underlying vector responses to future environmental conditions remain insufficiently understood. Aedes aegypti, a major vector of arboviruses, depends on tightly regulated reproductive and lipid-transfer pathways that are linked to population replacement and persistence. Here, we experimentally evaluated whether projected late-century increases in temperature and atmospheric CO₂ affect the transcriptional regulation of lipophorin- and vitellogenin-associated g

A tyramine receptor gene LsTAR2 is involved in reproduction and feeding in the small brown planthopper Laodelphax striatellus.

Tyramine (TA) is an important biogenic amine present in the central nervous system of insects, and mediates a variety of physiological and behavioral functions via tyramine receptors (TARs). However, TARs have not yet been characterized in planthoppers, and their physiological functions remain poorly understood in rice pests. Here, we cloned a tyramine receptor gene (LsTAR2) from the small brown planthopper Laodelphax striatellus, one of the most destructive rice pests. LsTAR2 shares high sequence identity with its orthologous receptors, and is closely related to its corresponding receptor gro

DsiRNA-mediated silencing of Ceratitis capitata transformer or transformer-2 leads to masculinization of XX embryos and systemic gene silencing in ovaries.

Ceratitis capitata (medfly), a major agricultural pest, is predominantly controlled using chemical insecticides, which pose environmental risks. Ecosustainable alternatives, such as the Sterile Insect Technique (SIT), rely on the mass release of sterile male-only progeny. However, sexing of male offspring requires the elimination of females during development. To overcome the loss of 50% of the progeny, the sex reversal of females into XX fertile males at embryonic stages by dsRNA injections was effective but not scalable. This study demonstrates the efficacy of Dicer-substrate small interferi

Reduced Genetic Diversity of Key Fertility and Vector Competency Related Genes in Anopheles gambiae s.l. Across Sub-Saharan Africa.

BACKGROUND: Insecticide resistance challenges the vector control efforts towards malaria elimination and proving the development of complementary tools. Targeting the genes that are involved in mosquito fertility and susceptibility to Plasmodium with small molecule inhibitors has been a promising alternative to curb the vector population and drive the transmission down. However, such an approach would require a comprehensive knowledge of the genetic diversity of the targeted genes to ensure the broad efficacy of new tools across the natural vector populations. METHODS: Four fertility and para

Crystal structures of angiotensin-converting enzyme from Anopheles gambiae in its native form and with a bound inhibitor.

The mosquitoes of the Anopheles and Aedes genus are some of the most deadly insects to humans because of their effectiveness as vectors of malaria and a range of arboviruses, including yellow fever, dengue, chikungunya, West Nile and Zika. The use of insecticides from different chemical classes is a key component of the integrated strategy against An. gambiae and Ae. aegypti, but the problem of insecticide resistance means that new compounds with different modes of action are urgently needed to replace chemicals that fail to control resistant mosquito populations. We have previously shown that
