Mapping dominant-negative mutations of anthrax protective antigen by scanning mutagenesis.

The protective antigen (PA) moiety of anthrax toxin transports edema factor and lethal factor to the cytosol of mammalian cells by a mechanism that depends on its ability to oligomerize and form pores in the endosomal membrane. Previously, some mutated forms of PA, designated dominant negative (DN), were found to coassemble with wild-type PA and generate defective heptameric pore-precursors (prepores). Prepores containing DN-PA are impaired in pore formation and in translocating edema factor and lethal factor across the endosomal membrane. To create a more comprehensive map of sites within PA

Anthrax: safe treatment for children.

Information regarding the treatment of anthrax infection is scarce in adults and is even more limited in children. Children, however, may be at a greater risk for developing an infection and systemic disease if exposed to anthrax than adults. The Centers for Disease Control and Prevention (CDC) recommends the use of doxycycline or ciprofloxacin for prophylaxis and treatment in children. Doxycycline currently is not indicated for use in children < 8 years old, due to staining of teeth and inhibition of bone growth associated with tetracyclines. Doxycycline, however, may have less adverse effect

The microbiology laboratory's role in response to bioterrorism.

CONTEXT: Bioterrorism has existed since before the 14th century; however, the specter of such an attack is much greater today than ever befo … observed. CONCLUSIONS: The most likely microorganisms to be utilized as biological weapons include Bacillus anthracis (anthrax), Brucella species (brucellosis), Clostridium botulinum (botulism), Francisella tularensis (tularemia), Yersinia pestis (plague), and variola major (smallpox). While knowledge of the potential of these microorganisms is critical, clinical m …

Search for correlates of protective immunity conferred by anthrax vaccine.

Vaccination by anthrax protective antigen (PA)-based vaccines requires multiple immunization, underlying the need to develop more efficacious vaccines or alternative vaccination regimens. In spite of the vast use of PA-based vaccines, the definition of a marker for protective immunity is still lacking. Here we describe studies designed to help define such markers. To this end we have immunized guinea pigs by different methods and monitored the immune response and the corresponding extent of protection against a lethal challenge with anthrax spores. Active immunization was performed by a single

Surveillance for adverse events associated with anthrax vaccination--U.S. Department of Defense, 1998-2000.

Concerns about the potential use of anthrax as a biologic weapon prompted the U.S. Department of Defense (DoD) to announce on December 15, 1997, anthrax vaccination of all U.S. military personnel. This effort is coordinated by the Anthrax Vaccine Immunization Program (AVIP). AVIP plans a phased vaccination process to achieve total force protection against anthrax by 2004. The current phase of implementation includes vaccination of all service members and mission-essential DoD civilian employees assigned or deployed to high-threat areas. On the basis of program monitoring, as of April 12, 2000,

[Effect of anthrax toxin on the chemiluminescence of human leukocytes].

The effect of substrate containing crude anthrax toxin on the phagocytosing leukocyte chemiluminescence has been studied. Preliminary toxin incubation with leukocytes for 60 min blocks cell chemiluminescence in the linear ratio effect concentration in the protein component of the toxin; the minimum concentration of the toxin protein component inhibiting the phagocytosing leukocyte luminescence is 3-5 micrograms per 5 x 10(5) cells. The substrate pure mixture of the oedema factor and protective antigen inhibits the chemiluminescence more intensively than crude toxin does. Inhibiting chemilumine

An epizootiologic study of anthrax in Falls County, Texas.

In June and July, 1974, an anthrax epizootic in Falls County, Texas, resulted in the death of 236 animals (228 cattle, 5 horses, 2 mules, and 1 pig) on 48 premises. Death rates were highest for horses (18.2%) and bulls (16.8%). The epizootic was apparently precipitated by drought, and infection appeared to be the result of ingesting intrinsically contaminated soil and grass. Human illness was not associated with the epizootic.

Quantification of immunoglobulin G as an alternative method to the Miles Evans blue assay to measure vascular hyperpermeability induced by infection and sterile inflammation.

Vascular hyperpermeability is conventionally evaluated by the Miles Evans blue assay. The assay is based on the intravenous injection of Eva … the increase in vascular permeability associated with inflammation due to cutaneous infection with Bacillus anthracis and to topical application of the pro-inflammatory agent, phorbol 12-myristate 13-acetate. We found the immunoglobulin G assay easier to perform and requires fewer animals when additional tissue samples are required.

Development and evaluation of recombinant protective antigen (rPA)-based indirect-ELISA for sero-monitoring of anthrax-specific antibodies in sheep.

Protective antigen (PA), a key exotoxin of Bacillus anthracis, is involved in the pathogenesis of anthrax, leading to 'sudden death' of affected herbivores. PA is also known to induce protective neutralizing antibodies following vaccination. Although an effective anthrax live spore vaccine is used to immunize susceptible animals, pre- and post-vaccination antibody titres are not regularly monitored, which is of immunological significance in endemic areas, and it is important to evaluate the vaccine efficacy at the field level. Hence, this study describes a standardized indirect-ELISA, a simple…

Awareness of anthrax disease and the knowledge of its transmission and symtoms identification: A cross sectional study among butchers in ile-ife.

Anthrax is a zoonotic disease of public health significance as it has led to the morbidity and mortality of human and livestock. Anthrax is caused by Bacillus anthracis which causes contamination of soil and water. Due to the grazing nature of cattle, they are mainly affected by anthrax among other herbivores. Butchers, as frontline actors in the meat value chain, are at high occupational risk. However, there is a paucity of data on their awareness and knowledge of this disease in urban settings in Nigeria, a gap this study aims to address. The study was a cross sectional study among 380 respo…

Sex-Specific Elk Resource Selection during the Anthrax Risk Period.

Anthrax, caused by the spore-forming bacterium Bacillus anthracis, is a zoonosis affecting animals and humans globally. In the United States, anthrax outbreaks occur in wildlife and livestock, with frequent outbreaks in native and exotic wildlife species in Texas, livestock outbreaks in the Dakotas, and sporadic mixed outbreaks in Montana. Understanding where pathogen and host habitat selection overlap is essential for anthrax management. Resource selection and habitat use of ungulates may be sex-specific and lead to differential anthrax exposure risks across the landscape for males and female…

Constructing and transient expression of a gene cassette containing edible vaccine elements and shigellosis, anthrax and cholera recombinant antigens in tomato.

Shigella dysenteriae causing shigellosis is one of the diseases that threaten the health of human society in the developing countries. In Shigella, IpaD gene is one of the key pathogenic genes causing strong mucosal immune system reactions. Anthrax disease is caused by Bacillus anthracis. PA protective antigen is one of the subunits in anthrax toxin complex responsible for the transfer of other subunits into the cytosol of host cells. The 20 kDa subunit of PA (PA20) has the property of immunogenicity. CTxB or B subunit of Vibrio cholerae toxin (CT) is a non-toxic protein and has the function t…

Anthrax lethal factor as an immune target in humans and transgenic mice and the impact of HLA polymorphism on CD4+ T cell immunity.

Bacillus anthracis produces a binary toxin composed of protective antigen (PA) and one of two subunits, lethal factor (LF) or edema factor (EF). Most studies have concentrated on induction of toxin-specific antibodies as the correlate of protective immunity, in contrast to which understanding of cellular immunity to these toxins and its impact on infection is limited. We characterized CD4+ T cell immunity to LF in a panel of humanized HLA-DR and DQ transgenic mice and in naturally exposed patients. As the variation in antigen presentation governed by HLA polymorphism has a major impact on prot…
