Prenatal ethanol exposure induces a sexually dimorphic effect on daily water consumption in prepubertal and adult rats.

Previous studies have shown that female rats consume significantly more water than males on a weight basis. Because exposure to alcohol during the last week of gestation is associated with incomplete behavioral defeminization in male rats, we examined daily water intake in fetal alcohol-exposed (FAE) males and females. Time-pregnant multiparous Sprague-Dawley dams were administered an ethanol liquid diet containing 35% ethanol-derived calories from day 14 through parturition. At 80 days of age, daily water consumption of FAE males and female litter representatives was measured for 7 days. FAE

Early events in the development of neuronal polarity in vitro are altered by ethanol.

Among the neuropathological effects of prenatal exposure to ethanol is the disruption of neuromorphogenesis. The effects of ethanol on early events in the development of axons and dendrites were studied using cultured embryonic rat hippocampal neurons, which develop in vitro in a stereotypical sequence of events that mimics their development in vivo. During the first 24 hr in culture, hippocampal neurons attach to the substrate and develop into one of three stages identified by phase-contrast microscopy: (i) neurons having lamellipodia and no processes (stage 1); (ii) neurons developing minor

Fetal alcohol exposure produces delay-dependent memory deficits in juvenile and adult rats.

The effects of prenatal alcohol exposure on both behavioral and neurobiological measures may be dependent, in part, on the age of the animal. Previous evidence from our laboratory has shown a delay-dependent memory deficit in young adult fetal alcohol exposure (FAE) rats. The present study examined the effects of FAE on an alternation task at three different ages of male rats: juvenile (days 38 to 44), young adult (days 82 to 89), and adult (days 173 to 180). In the present study, subjects were three age groups of male offspring of Sprague-Dawley rats fed 35% ethanol-derived calories, pair-fed

Effects of ethanol on biotransformation of all-trans-retinol and all-trans-retinal to all-trans-retinoic acid in rat conceptal cytosol.

Enzymatic catalysis of the oxidations of ethanol, all-trans-retinol (tretinol) and all-trans-retinal (t-retinal) were demonstrated in the cytosolic fractions of rat conceptal homogenates at day 12 of gestation. Products of the retinoid oxidation reactions were identified with HPLC by comparing elution times with those of authentic standard retinoids. NAD-dependent oxidations of each of the three substrates were demonstrable with assay conditions used; t-retinol and t-retinal each were converted to readily detectable quantities of all-trans-retinoic acid (t-RA). At 1.0 mM or higher concentratio

Cell cycle kinetics in fetal rat cerebral cortex: effects of prenatal treatment with ethanol assessed by a cumulative labeling technique with flow cytometry.

The effects of ethanol on the cell cycle kinetics of cortical precursor cells during the period of cortical neuronogenesis [between gestational day (G) 12 and G21] was systematically examined. Samples of dissociated cortical cells were harvested from the cerebral cortices of 13-, 15-, 17-, 19-, and 21-day-old fetuses. The fetuses were obtained from pregnant rats: (a) fed a liquid diet containing 6.7% (v/v) ethanol (Et) ad libitum, (b) pair-fed an isocaloric liquid control diet (Ct), or (c) fed chow and water (Ch) ad libitum. Before harvesting the cells, the fetuses were administered a series o

Loss of reproductive competence at an earlier age in female rats exposed prenatally to ethanol.

Previous studies have shown that prenatal ethanol exposure can partially masculinize or defeminize neurobehavioral development of female rats. An early age of onset of anovulation is one of the primary characteristics of partial defeminization. Consequently, we examined the occurrence of anovulation in fetal alcohol-exposed (FAE) female rats at 2, 6, and 12 months of age using both vaginal cytology as well as wheel-running behavior. We assessed the ability of estrogen and progesterone to elicit proprioceptive behaviors and lordosis at 2 and 17 months of age. Female subjects were derived from S

A revised estimate of the economic impact of fetal alcohol syndrome.

A revised estimate of the national economic impact of fetal alcohol syndrome (FAS)-related abnormalities was conducted. The present evaluation included "corrections" for background rates of low birth weight, and costs normally incurred for housing and food regardless of whether an individual requires institutionalization or not. Additional anomalies were included along with hospital costs provided for in diagnostic-related groups. The current estimate for annual costs related to FAS is $249.7 million, which is about $80 million less than our previous estimate. This estimate is lower due to a s

[Activity of Zn-, Cu-containing superoxide dismutase in the brain tissue of rats with fetal alcohol syndrome].

Zn(2+)-, Cu2+ superoxide dismutase activity was estimated in the brain of 14- and 30-day-old antenatally alcoholized rat offsprings. The enzyme activity in 14-day-old experimental offsprings. The enzyme activity in 14-day-old experimental offsprings was significantly decreased in the brain cortex, hippocampus and brain stem by 27.1; 31.1 and 33.4% respectively as compared with control animals. The results are discussed from the point of view of the activation of free radical processes in the brain of alcoholized offsprings.

Maternal genetic effects on ethanol teratogenesis and dominance of relative embryonic resistance to malformations.

Maternal genetic factors and/or fetal genetic factors contribute to variations in response to prenatal alcohol exposure. To assess the contribution of maternal genotype to ethanol teratogenesis, a reciprocal cross study was conducted in an animal model using C57BL/6J (B6) and long-sleep (LS) mice. B6 mice are more susceptible than LS mice to prenatal ethanol-induced malformations but both mouse stocks are susceptible to fetal weight deficits following in utero alcohol exposure. B6 and LS dams were reciprocally mated to B6 or LS males producing four embryonic genotype groups: the true-bred B6B6

Molecular changes associated with ethanol-induced growth suppression in the chick embryo.

In humans and in animal models the most frequently observed alcohol-related birth defect (ARBD) is intrauterine growth retardation (IUGR). The central nervous system (CNS) is sensitive to the growth inhibitory effects of in utero ethanol exposure and neonatal CNS alterations with associated behavioral deficits are a likely result of maternal ethanol consumption. Presently, little information exists as to the biochemical mechanism by which ethanol inhibits fetal CNS growth. Further, it is unknown if there are genetic differences in maternal or fetal responses to ethanol. Ongoing research using

The effects of prenatal alcohol exposure on radial arm maze performance in adult rats.

The hippocampal formation is sensitive to the in utero exposure to ethanol. It is one brain area thought to play an important role in spatial memory. We examined radial arm maze performance in rats exposed to ethanol prenatally. Pregnant rats were placed into the following treatment groups: LC, 17% EDC (ethanol-derived calories), 35% EDC, PF 35% or PF 17%. The LC group was fed lab chow and water ad lib, the 17% EDC and 35% EDC groups were fed a liquid diet containing either 3.3% or 6.7% v/v ethanol, respectively. Pair-fed controls were fed the same volume of an isocaloric diet as was consumed

Maternal use of alcohol and other drugs during pregnancy and birth abnormalities: further results from a prospective study.

This paper presents an extended analysis of data produced during a prospective study of the possible association between maternal alcohol use during pregnancy and fetal harm. This new analysis examines the possible links between not only maternal alcohol consumption, but also the use of tobacco, prescribed and illicit drugs and birth abnormalities. There was no general association between birth abnormalities and maternal psychoactive drug use and misuse. Even so, women who neither smoked nor consumed alcohol during the first trimester produced offspring with significantly fewer birth abnormali

Maternal ethanol consumption: effect on skeletal muscle development in guinea pig offspring.

The effect of ethanol on developing skeletal muscle was analyzed by examining the gastrocnemius muscle from newborn guinea pigs, exposed to ethanol during the second half of gestation. Electron microscopy revealed vacuolated sarcoplasmic reticula, enlarged lipid droplets, decreased glycogen and mitochondrial abnormalities in the skeletal muscle samples from the ethanol-exposed newborn guinea pigs. None of these abnormalities were seen in the newborn controls who were born to dams which consumed the same amount of calories as the ethanol-treated dams. The ethanol-associated abnormalities, seen

Fetal alcohol syndrome associated with trisomy 21.

Five cases of Down syndrome in association with fetal alcohol syndrome (FAS) are described. Four of the babies were full term, one was premature. Phenotypically these babies (3M:2F) had manifestations of both Down syndrome and FAS; growth deficiency was more pronounced than is expected in Down syndrome. All measurements, birth weight, length and head circumference were below -2 SD. All had congenital heart disease. Chromosome analysis confirmed 21 trisomy in every case. Down syndrome and FAS may occur together, at random, in 1 in 525,0000 newborns in United States but 1 in 200,000 in our insti

Prenatal exposure to ethanol alters lateral plasma membranes and gap junctions of newborn rat hepatocytes as revealed by freeze-fracture.

The effects of prenatal exposure to ethanol on the lateral (contiguous) plasma membrane of newborn rat hepatocytes was investigated using qualitative and quantitative freeze-fracture. The number of free intramembranous particles decreased by 11% and 17% in P- and E-faces, respectively. Alcohol also induced the appearance of particle-free areas within the gap junctions, in addition to a three-fold increase in the mean area and an increment in the percent of plasma membrane occupied by these elements. However, no differences in either the size of gap junction particles or in the mean interpartic

Cerebral metabolic alterations in rats following prenatal alcohol exposure: a deoxyglucose study.

Numerous reports dealing with the gross morphological, microanatomical, and behavioral changes occurring with prenatal alcohol exposure in rats have appeared. In order to define the cerebral metabolic correlates of prenatal alcohol exposure, in vivo glucose utilization rates in brain were assayed using an autoradiographic [14C]deoxyglucose technique. Pregnant Long-Evans rats were fed liquid diets containing either 35% or 0% ethanol-derived calories during day 6 through day 20 of gestation. These liquid diets were isocaloric and a pair-feeding procedure was employed. The offspring were tested o

Reduced binding of basic somatomedin by mouse placental membranes following maternal alcohol administration.

The effects of 3 days of maternal ethanol administration on the placenta and on basic somatomedin (B-SM) were investigated in the mouse. Following administration of aqueous ethanol by gavage on days 13, 14 and 15 of gestation, there was no treatment-related difference in embyonic growth or placental weight as seen on day 15. There was a significant reduction in the specific binding of [125I]B-SM by day 15 placental membranes, but no difference in serum B-SM concentrations or in the frequency or severity of degenerative changes in the placenta. We have shown that changing levels of serum B-SM o

The study of brain function impairment in fetal alcohol syndrome: some fruitful directions for research.

Fetal alcohol syndrome (FAS) is the most prevalent known preventable health hazard to the human fetus by a noxious agent. It is associated with impairments of the central nervous system that are expressed in the forms of mental retardation of varying severity, learning disabilities, attentional deficits and an increased vulnerability to stress. Results of psychophysiological studies of the effects of ethanol on the central nervous system are reviewed, with the aim of exploring how conclusions derived from them can serve as testable hypotheses in FAS research. The experimental methods used in s
