BACKGROUND: Natural bioactive compounds derived from plants have gained considerable attention due to their potential therapeutic and medicinal applications. In the context of antimicrobial resistance, these compounds are being explored as potential efflux pump inhibitors (EPIs) and antibiotic adjuvants to enhance antimicrobial efficacy. Their low toxicity and broad availability make them promising candidates for the development of alternative therapeutic strategies against multidrug-resistant (MDR) pathogens such as Pseudomonas aeruginosa.
METHODS: This study evaluated the efflux pump inhibi

BACKGROUND: The bacterium Staphylococcus aureus, particularly its methicillin-resistant form (MRSA), presents significant public health challenges due to its resistance to β-lactam antibiotics and frequent multidrug resistance. Livestock-associated MRSA (LA-MRSA), especially clonal complex 398, has become a concern in intensive pig farming, with emerging evidence suggesting flies may act as vectors. This study investigates the occurrence and molecular characterisation of MRSA in house flies (Musca domestica) and stable flies (Stomoxys calcitrans) on Austrian piglet-producing farms to understan

OBJECTIVE: Antibiotic resistance genes (ARGs) in bacteria are highly prevalent in the poultry industry. These genes can transfer not only within the same species but also across different species, posing significant threats to both the poultry industry and human health. However, limited research has been conducted on the prevalence and transmission links between extended-spectrum beta-lactamase (ESBL)-producing diarrheagenic Escherichia coli (E. coli) in chickens and their living environments in Shanghai's poultry industry.
METHODS: In this study, 600 samples were collected from the cloacal s

Hospital-acquired infections caused by multidrug-resistant (MDR) pathogens such as Methicillin-resistant Staphylococcus aureus (MRSA) and Carbapenem-resistant Klebsiella pneumoniae (CRKP) are a major health concern. In this study, ceftizoxime (CEF) and curcumin (CUR) were co-encapsulated into niosome nanoparticles (using thin-film hydration), and then embedded into a 3D-printed gelatin-alginate scaffold (Nio-CUR/CEF@SC). The Nio-CUR/CEF@SC were characterized by dynamic light scattering (DLS), transmission electron microscopy (TEM), scanning electron microscopy (SEM), and Fourier-transform infr

Staphylococcus aureus is a leading cause of ocular infections, resulting in vision loss in severe cases. Understanding the antibiotic resistance profiles of ocular S. aureus can help customize treatments. However, there is a lack of global data on the resistance patterns of ocular isolates and comparative regional analyses. Hence, WGS data from 195 ocular S. aureus isolates across six continents were analysed to identify antibiotic resistance genes (ARGs) and predict antibiotic resistance phenotypes in this study. A total of 40 ARGs were detected, involving resistance mechanisms against aminog

Tigecycline is considered a last-line therapeutic option for treating infections caused by multidrug-resistant (MDR) Gram-negative bacteria. The emergence of tmexCD1-toprJ1, a plasmid-mediated resistance-nodulation-division (RND) efflux pump gene cluster, poses a growing threat to tigecycline efficacy. While this gene cluster has primarily been identified in Klebsiella pneumoniae, its presence in other species remains poorly characterized. In this study, we investigated the occurrence, genetic features, resistance profile, and virulence potential of tmexCD-toprJ-positive Raoultella ornithinoly

Pet food have been considered as possible vehicles of bacterial pathogens. The sudden boom of the pet food industry due to the worldwide increase in companion animal ownership calls for pet food investigations. Herein, this study aimed to determine the frequency, antimicrobial susceptibility profile, and molecular characteristics of coagulase-negative staphylococci (CoNS) in different pet food brands in Brazil. Eighty-six pet food packages were screened for CoNS. All isolates were identified at species level by MALDI-TOF MS and species-specific PCR. Antimicrobial susceptibility testing was per

Globally, avian colibacillosis is a leading cause of morbidity and mortality in poultry, associated with economic losses and welfare problems. Here, clinical avian pathogenic E. coli isolates (CEC; n = 50) and faecal E. coli isolates from healthy (FEC; n = 187) Australian meat chickens collected between 2006 and 2014 were subjected to antimicrobial susceptibility testing, phylogenetic grouping, plasmid replicon (PR) typing, multilocus sequence typing, and virulence gene (VG) profiling. Extended-spectrum cephalosporin (ESC)- and fluoroquinolone (FQ)-resistant E. coli isolates underwent further

Avian pathogenic Escherichia coli (APEC) is responsible for colibacillosis in poultry. APEC remains a constant problem for the poultry industry, despite the use of antimicrobials and disinfectants in farms. The endemicity of APEC in poultry farms is associated with its biofilm-forming ability, which is further aggravated by various virulence factors and resistance to multiple drugs that help bacteria to thrive under different environmental conditions. To characterize APEC from affected broiler chickens and their environments, samples (n=114) from dead birds (heart, liver, lungs, and cloacal sw

Hydrogen sulfide (H2S) has recently been recognized as a novel gaseous transmitter with several anti-inflammatory properties. The role of host- derived H2S in infections by Pseudomonas aeruginosa was investigated in clinical and mouse models. H2S concentrations and survival was assessed in septic patients with lung infection. Animal experiments using a model of severe systemic multidrug-resistant P. aeruginosa infection were performed using mice with a constitutive knock-out of cystathionine-γ lyase (Cse) gene (Cse-/-) and wild-type mice with a physiological expression (Cse+/+). Experiments we

OBJECTIVE: The emergence of extended-spectrum β-lactamase (ESBL)-producing bacteria is now a critical concern. The ESBL-producing Klebsiella pneumoniae constitutes one of the most common multidrug-resistant (MDR) groups of gram-negative bacteria involved in nosocomial infections worldwide. In this study we report on the molecular characterization through whole genome sequencing of an ESBL-producing K. pneumoniae strain, LAU-KP1, isolated from a stool sample from a patient admitted for a gastrointestinal procedure/surgery at the Lebanese Amrican University Medical Center-Rizk Hospital (LAUMCRH)

Heterogeneity is known to be present to varying degrees in cancer cell groups. There have been no reports, however, of studies in which a single cell clone was prepared from a cancer cell group to examine heterogeneity with respect to anticancer drug sensitivity. Thus, the authors herein report an investigation into the heterogeneity of cancer cells within the same tumor with respect to anticancer drug sensitivity. Anticancer drug sensitivity was investigated in primary tumors, metastatic lymph node tumors, recurrent tumors and established cell lines obtained from four cases of tongue cancer u

BACKGROUND: Human migration significantly contributes to the global spread of infectious diseases. In recent years, Ethiopia has experienced an increased influx of returnees from the Kingdom of Saudi Arabia. These migrants were often held in densely populated and inadequately ventilated detention centers and faced a heightened risk of tuberculosis transmission. This study investigates the genotypic drug resistance and transmission clusters of M. tuberculosis among returnees from various detention centers in the Kingdom of Saudi Arabia. Whole genome sequencing (WGS) is employed to characterize

Nosocomial infections caused by multidrug-resistant (MDR) Enterobacter cloacae complex (ECC) pathogens are on the rise. However, the virulence strategies employed by these pathogens remain elusive. Here, we study the interaction of ECC clinical isolates with human serum to define how this pathogen evades the antimicrobial action of complement, one of the first lines of host-mediated immune defense. We identified a small number of serum-sensitive strains, including Enterobacter hormaechei strain NR3055, which we exploited for the in vitro selection of serum-resistant clones. Comparative genomic

Antibiotic resistance (AMR) is an increasing public health crisis worldwide. This threatens our ability to adequately care for patients with infections due to multi-drug-resistant (MDR) pathogens. As such, there is an urgent need to develop new classes of antimicrobials that are not based on currently utilized antibiotic scaffolds. One promising avenue of antimicrobial research that deserves renewed examination involves the use of peptides. Although antimicrobial peptides (AMPs) have been studied for a number of years, innovations in peptide design and their applications are increasingly makin

This review focuses on drug-drug interactions with three major groups of antimicrobial agents: macrolides (including azalides and ketolides), quinolones, which are widely used for the treatment of bacterial infections, and azoles, which are used for antifungal therapy. Macrolides and the ketolide telithromycin are potent inhibitors of CYP3A4 and thus interfere with the pharmacokinetics of many other drugs that are metabolised by this enzyme. In contrast, although closely related, azithromycin is not a cytochrome inhibitor. All quinolones form complexes with di- and trivalent cations and, there

Livestock-associated methicillin-resistant Staphylococcus aureus (LA-MRSA) sequence type 9 (ST9) has emerged and disseminated in Asia. It is associated with colonization or infection in both humans and animal hosts; however, the genetic factors underpinning its adaptation to animal and human population remain to be determined. Here, we conducted a genomic analysis of 191 ST9 S. aureus genomes collected from 12 different countries, including 174 genomes retrieved from public databases and 17 sequenced in this study.
